제주지역 해수, 수족관물, 유통수산물에서 분리된 장염비브리오균의 분자생물학적 특성 (Molecular Characterization of Vibrio parahaemolyticus Isolates from Seawater, Fish Tanks, and Distributed Fishery Products in Jeju)

  • 장염비브리오균은 급성 설사와 같이 수인성·식품매개 질병을 일으키는 것으로 알려져 있다. 본 연구에서는 제주지역 해수, 수족관수, 유통 수산물에서 분리한 장염비브리오균을 real-time PCR을 이용하여 잠재적인 독소 유전자 또는 종특이성 유전자(tdh, trh, tlh, toxR)를 조사하고, 이 균주의 유전적인 특성을 PFGE로 분석하였다. 총 245개 시료 중 해수에서 33주, 수족관수에서 7주, 수산물에서 50주로 90주(36.7%)의 장염비브리오균을 분리하였다. 모든 장염비브리오균에서 tdh 유전자는 검출되지 않았지만, tlh 유전자 또는 toxR 유전자는 검출되었다. 또한, 해수에서 3주와 수산물에서 1주 분리된 균주에서 trh 유전자가 검출되었다. 매달 해수를 정량검사한 결과 장염비브리오균은 수온과 양의 상관관계를 가지고 있었다. 90주의 장염비브리오균은 64.0-97.3% 범위에서 유사한 유전자 상동성을 보였다. 이중에서 13개 유형에서 유전자 상동성이 100%였다. 이러한 결과는 일부 독소 유전자를 가진 장염비브리오균이 분리되었고, 해수, 수족관물, 유통 수산물에서 분리된 장염비브리오균 사이에 유전적 유사성이 있기 때문에 식중독 역학조사에 도움이 되도록 지속적인 모니터링이 필요함을 나타낸다.

탈구치 저장 매체로서 치주인대 세포에 미치는 Polydeoxyribonucleotide의 효과에 대한 연구 (Effect of Polydeoxyribonucleotide on Human Periodontal Ligament Cells as a Storage Medium for Avulsed Tooth)

  • 본 연구는 탈구된 치아의 저장 매체로서 polydeoxyribonucleotide (PDRN)의 적합성을 평가하고자 하였다. HBSS와 10, 25, 50, 100 ㎍/mL 농도의 PDRN 용액과 수돗물에 저장된 인간 치주인대 세포의 생존율을 측정하기 위해 Cell Counting Kit-8 assay와 Live/Dead assay를 시행하였다. 또한, PDRN의 항염증 효과를 평가하기 위한 NO 검출 및 qRT-PCR 실험을 진행하였다. 100 ㎍/mL 농도의 PDRN 용액에 저장된 치주인대 세포의 생존율이 다른 용액보다 유의하게 높았다(p < 0.01). 또한, 100 ㎍/mL 농도의 PDRN 용액은 유의하게 NO의 생산을 줄였다(p < 0.0001). 그리고, HBSS 용액에 비하여 50 및 100 ㎍/mL 농도의 PDRN 용액에서 유의하게 tumor necrosis factor α, interleukin (IL)-4, IL-6, 그리고 IL-10의 발현이 낮았다(p < 0.01). 이 연구를 통해 PDRN은 치주인대 세포에 세포 보존 및 항염증 효과를 가진 것으로 밝혀졌다. 이 연구는 효과적인 탈구치 저장매체의 개발을 위한 향후 추가적인 실험의 기반이 될 수 있을 것이라 생각한다.

Transcriptome profiling identifies immune response genes against porcine reproductive and respiratory syndrome virus and Haemophilus parasuis co-infection in the lungs of piglets

  • Background: Co-infections of the porcine reproductive and respiratory syndrome virus (PRRSV) and the Haemophilus parasuis (HPS) are severe in Chinese pigs, but the immune response genes against co-infected with 2 pathogens in the lungs have not been reported. Objectives: To understand the effect of PRRSV and/or HPS infection on the genes expression associated with lung immune function. Methods: The expression of the immune-related genes was analyzed using RNA-sequencing and bioinformatics. Differentially expressed genes (DEGs) were detected and identified by quantitative real-time polymerase chain reaction (qRT-PCR), immunohistochemistry (IHC) and western blotting assays. Results: All experimental pigs showed clinical symptoms and lung lesions. RNA-seq analysis showed that 922 DEGs in co-challenged pigs were more than in the HPS group (709 DEGs) and the PRRSV group (676 DEGs). Eleven DEGs validated by qRT-PCR were consistent with the RNA sequencing results. Eleven common Kyoto Encyclopedia of Genes and Genomes pathways related to infection and immune were found in single-infected and co-challenged pigs, including autophagy, cytokine-cytokine receptor interaction, and antigen processing and presentation, involving different DEGs. A model of immune response to infection with PRRSV and HPS was predicted among the DEGs in the co-challenged pigs. Dual oxidase 1 (DUOX1) and interleukin-21 (IL21) were detected by IHC and western blot and showed significant differences between the co-challenged pigs and the controls. Conclusions: These findings elucidated the transcriptome changes in the lungs after PRRSV and/or HPS infections, providing ideas for further study to inhibit ROS production and promote pulmonary fibrosis caused by co-challenging with PRRSV and HPS.

miR-380-3p promotes β-casein expression by targeting αS1-casein in goat mammary epithelial cells

  • Objective: αS1-Casein is more closely associated with milk allergic reaction than other milk protein components. microRNA (miRNA) is a class of small non-coding RNAs that modulate multiple biological progresses by the target gene. However, the post-transcriptional regulation of αS1-casein expression by miRNA in ruminants remains unclear. This study aims to explore the regulatory roles of miR-380-3p on αS1-casein synthesis in goat mammary epithelial cells (GMEC). Methods: αS1-Casein gene and miR-380-3p expression was measured in dairy goat mammary gland by quantitative real-time polymerase chain reaction (qRT-PCR). miR-380-3p overexpression and knockdown were performed by miR-380-3p mimic or inhibitor in GMEC. The effect of miR-380-3p on αS1-casein synthesis was detected by qRT-PCR, western blot, luciferase and chromatin immunoprecipitation assays in GMEC. Results: Compared with middle-lactation period, αS1-casein gene expression is increased, while miR-380-3p expression is decreased during peak-lactation of dairy goats. miR-380-3p reduces αS1-casein abundance by targeting the 3'-untranslated region (3'UTR) of αS1-casein mRNA in GMEC. miR-380-3p enhances β-casein expression and signal transducer and activator of transcription 5a (STAT5a) activity. Moreover, miR-380-3p promotes β-casein abundance through target gene αS1-casein, and activates β-casein transcription by enhancing the binding of STAT5 to β-casein gene promoter region. Conclusion: miR-380-3p decreases αS1-casein expression and increases β-casein expression by targeting αS1-casein in GMEC, which supplies a novel strategy for reducing milk allergic potential and building up milk quality in ruminants.

In-silico annotation of the chemical composition of Tibetan tea and its mechanism on antioxidant and lipid-lowering in mice

  • BACKGROUND/OBJECTIVES: Tibetan tea is a kind of dark tea, due to the inherent complexity of natural products, the chemical composition and beneficial effects of Tibetan tea are not fully understood. The objective of this study was to unravel the composition of Tibetan tea using knowledge-guided multilayer network (KGMN) techniques and explore its potential antioxidant and hypolipidemic mechanisms in mice. MATERIALS/METHODS: The C57BL/6J mice were continuously gavaged with Tibetan tea extract (T group), green tea extract (G group) and ddH2O (H group) for 15 days. The activity of total antioxidant capacity (T-AOC) and superoxide dismutase (SOD) in mice was detected. Transcriptome sequencing technology was used to investigate the molecular mechanisms underlying the antioxidant and lipid-lowering effects of Tibetan tea in mice. Furthermore, the expression levels of liver antioxidant and lipid metabolism related genes in various groups were detected by the real-time quantitative polymerase chain reaction (qPCR) method. RESULTS: The results showed that a total of 42 flavonoids are provisionally annotated in Tibetan tea using KGMN strategies. Tibetan tea significantly reduced body weight gain and increased T-AOC and SOD activities in mice compared with the H group. Based on the results of transcriptome and qPCR, it was confirmed that Tibetan tea could play a key role in antioxidant and lipid lowering by regulating oxidative stress and lipid metabolism related pathways such as insulin resistance, P53 signaling pathway, insulin signaling pathway, fatty acid elongation and fatty acid metabolism. CONCLUSIONS: This study was the first to use computational tools to deeply explore the composition of Tibetan tea and revealed its potential antioxidant and hypolipidemic mechanisms, and it provides new insights into the composition and bioactivity of Tibetan tea.

Association study and expression analysis of olfactomedin like 3 gene related to meat quality, carcass characteristics, retail meat cut, and fatty acid composition in sheep

  • Objective: The objective of this study was to identify polymorphism in olfactomedin like 3 (OLFML3) gene, and association analysis with meat quality, carcass characteristics, retail meat cut, and fatty acid composition in sheep, and expression quantification of OLFML3 gene in phenotypically divergent sheep. Methods: A total of 328 rams at the age of 10 to 12 months with an average body weight of 26.13 kg were used. A novel polymorphism was identified using high-throughput sequencing in sheep and genotyping of OLFML3 polymorphism was performed using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Among 328 rams, 100 rams representing various sheep genotypes were used for association study and proc general linear model was used to analyse association between genotypes and phenotypic traits. Quantitative real-time polymerase chain reaction (qRT-PCR) was used for the expression analysis of OLFML3 mRNA in phenotypically divergent sheep population. Results: The findings revealed a novel polymorphism in the OLFML3 gene (g.90317673 C>T). The OLFML3 gene revealed three genotypes: CC, CT, and TT. The single nucleotide polymorphism (SNP) was found to be significantly (p<0.05) associated with meat quality traits such as tenderness and cooking loss; carcass characteristics such as carcass length; retail meat cut such as pelvic fat in leg, intramuscular fat in loin and tenderloin, muscle in flank and shank; fatty acids composition such as tridecanoic acid (C13:0), palmitoleic acid (C16:1), heptadecanoic acid (C17:0), ginkgolic acid (C17:1), linolenic acid (C18:3n3), arachidic acid (C20:0), eicosenoic acid (C20:1), arachidonic acid (C20:4n6), heneicosylic acid (C21:0), and nervonic acid (C24:1). The TT genotype was associated with higher level of meat quality, carcass characteristics, retail meat cut, and some fatty acids composition. However, the mRNA expression analysis was not different among genotypes. Conclusion: The OLFML3 gene could be a potential putative candidate for selecting higher quality sheep meat, carcass characteristics, retail meat cuts, and fatty acid composition in sheep.

RAW264.7 대식세포에서 미선나무 잎 추출물의 항산화, 항염증 효능 및 기전연구 (Antioxidant and anti-inflammatory effects and mechanism of Abeliophyllum distichum leaf extract in RAW264.7 macrophages)

  • 본 연구에서는 잠재적인 기능성 소재로의 개발 가능성을 검증하기 위하여 미선나무 잎 추출물의 항산화능과 항염증 효능을 확인하였다. 시료의 총 폴리페놀함량, DPPH 및 ABTS 라디칼 소거능, FRAP value를 측정하여 미선나무 잎 추출물의 항산화 활성을 확인하였으며 또한 RAW264.7 세포에 미선나무 잎 추출물 처리 후 HO-1의 발현이 증가하는 것을 통해 미선나무 잎 추출물의 항산화 기전을 확인하였다. 한편, LPS로 유도된 염증 상태의 RAW264.7 세포에서 미선나무 잎 추출물은 NF-κB 활성 억제를 통한 NO 생성 억제, iNOS, COX-2 발현 억제 및 염증성 사이토카인의 발현과 생성을 억제하는 것을 확인하였다. 이러한 본 연구 결과는 향후 미선나무 잎 추출물의 기능성 소재로의 개발을 위한 기초자료 마련에 그 의의가 있다.

목서 꽃 추출물의 항염, 항노화 및 피지 억제 효능 (Anti-inflammatory, Anti-aging, and Sebum Inhibitory Effects of Osmanthus fragrans Flower Extract)

  • 본 연구에서는 목서(Osmanthus fragrans, O. fragrans) 꽃 추출물을 화장품 소재로 활용하기 위해 피부에 대한 다양한 효능을 확인하고자 하였다. 이를 위해 제주산 목서 꽃 추출물(O. fragrans flower extract, OFFE)을 제조하여 실험에 이용하였다. 실험은 실시간 중합효소 연쇄반응인 qRT-PCR과 lipid droplet 염색법으로 평가하였다. 먼저 OFFE는 표피 각질세포에서 poly I:C로 유도된 3 종의 대표적인 전염증성 사이토카인 (IL-8, IL-6, IL-1α)과 염증 관련 효소인 PTGS2의 유전자 발현을 감소시켰다. 또한 OFFE는 진피 섬유아세포에서 콜라겐(COL1A1)과 엘라스틴(ELN) 유전자 발현을 증가시켰다. 더 나아가 OFFE는 피지세포에서 linoleic acid에 의해 유도된 피지 생성을 억제하는 효능 보여주었다. 따라서 본 연구를 통해 OFFE은 항염, 항노화, 그리고 피지억제 효능을 위한 천연 화장품 소재로써 적용이 가능할 것으로 기대된다.

Identification of relevant differential genes to the divergent development of pectoral muscle in ducks by transcriptomic analysis

  • Objective: The objective of this study was to identify candidate genes that play important roles in skeletal muscle development in ducks. Methods: In this study, we investigated the transcriptional sequencing of embryonic pectoral muscles from two specialized lines: Liancheng white ducks (female) and Cherry valley ducks (male) hybrid Line A (LCA) and Line C (LCC) ducks. In addition, prediction of target genes for the differentially expressed mRNAs was conducted and the enriched gene ontology (GO) terms and Kyoto encyclopedia of genes and genomes signaling pathways were further analyzed. Finally, a protein-to-protein interaction network was analyzed by using the target genes to gain insights into their potential functional association. Results: A total of 1,428 differentially expressed genes (DEGs) with 762 being up-regulated genes and 666 being down-regulated genes in pectoral muscle of LCA and LCC ducks identified by RNA-seq (p<0.05). Meanwhile, 23 GO terms in the down-regulated genes and 75 GO terms in up-regulated genes were significantly enriched (p<0.05). Furthermore, the top 5 most enriched pathways were ECM-receptor interaction, fatty acid degradation, pyruvate degradation, PPAR signaling pathway, and glycolysis/gluconeogenesis. Finally, the candidate genes including integrin b3 (Itgb3), pyruvate kinase M1/2 (Pkm), insulin-like growth factor 1 (Igf1), glucose-6-phosphate isomerase (Gpi), GABA type A receptor-associated protein-like 1 (Gabarapl1), and thyroid hormone receptor beta (Thrb) showed the most expression difference, and then were selected to verification by quantitative real-time polymerase chain reaction (qRT-PCR). The result of qRT-PCR was consistent with that of transcriptome sequencing. Conclusion: This study provided information of molecular mechanisms underlying the developmental differences in skeletal muscles between specialized duck lines.

토종 종계를 이용한 이면 교배조합 계통 간 스트레스 반응정도 비교 분석 (Comparison of Stress Response in Diallel Crossed Korean Domestic Chicken Breeds)

  • 본 연구에서는 국산종계 개발을 위하여 5계통의 토종닭 순계를 이용하여 $5{\times}5$ 이면교배조합 시험을 수행하고, 공시된 25계통에 대한 스트레스 반응 정도를 비교 분석하였다. 스트레스 반응정도 표지로 텔로미어 함량 및 감축율, 열 스트레스 단백질 유전자(HSPs)로 HSP-70, $HSP-90{\alpha}$, $HSP-90{\beta}$의 발현율 및 DNA 손상율을 이용하여 분석하였다. 텔로미어 함량은 양적 형광접합보인법을 이용하고, HSPs 발현율은 quantitative real-time PCR 기법으로, DNA 손상율은 Comet assay법을 적용하였다. 분석 결과, 교배조합 25계통 간 텔로미어 함량과 감축율, HSPs 유전자 발현율 및 DNA 손상율 모두에서 유의한 차이를 보였고, 더불어 계통 간 체중 및 생존율에도 유의한 차이가 나타났다. 텔로미어 감축율, HSPs 유전자 발현율 및 DNA 손상율 모든 분석 값에서 W 및 Y 계통과의 교잡구가 다른 교잡구에 비해 유의하게 낮은 값을 보였고, 반면 G계통의 순계구가 가장 높은 값을 나타내었다. 생존율과 스트레스 표지 값들 간의 상관계수를 추정한 결과, 텔로미어 함량과는 저도의 정(+)의 상관, HSP-70, $HSP-90{\alpha}$, $HSP-90{\beta}$ 및 DNA 손상율과는 중도 및 저도의 부(-)의 상관을 나타내었다. 또한 폐사 개체와 생존 개체 간 HSP-70, $HSP-90{\alpha}$$HSP-90{\beta}$의 발현율을 비교 분석한 결과, 폐사 개체의 HSPs 발현율이 생존 개체에 비해 유의하게 높은 발현율을 보였다. 이상 스트레스 표지 값의 분석 결과를 종합할 때, 교잡계들이 순계에 비해 스트레스 저항성이 높은 것으로 판단되고, 더불어 W, Y와 같은 경량종 계통의 교잡계들이 외적 스트레스에 강한 반면, G, H, F와 같은 고체중 계통의 교잡계들은 스트레스에 민감한 것으로 사료된다.
