Screening of Antifungal Activities of Medicinal Plants for the Control of Turfgrass Fungal Disease (잔디 병해 방제를 위한 약용식물의 항균작용 탐색)

  • Seven medicinal plant extracts were tested for antifungal activities against six species of the major turfgrass pathogenic fungi (Colletotrichum graminicola, Pythium spp., Rhizoctonia cerealis, Rhizoctonia solani AG1-1, Rhizoctonia solani AG2-2, and Sclerotinia homoeocarpa) using paper disk diffusion method. Three medicinal plant extracts, including Pinus densiflora showed antifungal activities. In suppression of mycelium growth test, on medium adding P. densiflora extract showed that inhibition rate of mycelium growth were above 80% in 10 mg/10 ml concentration of the extract. The inhibition rate of Pythium spp. was 100% and C. graminicola was 84.3% in 10 mg/10 ml concentrations of P. densiflora extract, respectively. In particularly, the inhibition rate of Pythium spp. was 89.5% in 2 mg/10 ml concentrations of P. densiflora extract. As a result, P. densiflora extract showed high antifungal activity to Pythium spp. and C. graminicola of the turfgrass pathogen in in vitro test.

  • In order to select ${\gamma}-Linolenic$ acid (GLA)-producing fungi, a total of forty-four strains of 4 genera such as Phytophthora, Pythium, Mucor and Rhizopus were obtained from Koran Agricultural Culture Collection (KACC) and then analysed by using GC-FID and GC-MS. GLA was detected on 39 fungal strains, and the highest rate of GLA was found as 24.8% of total fatty acids on Mucor hiemalis f. sp. hiemalis KACC 40264. Total GLA content of Zygomycota was comparatively high - Mucor (14.2%) and Rhizopus (14.3%), whereas that of Oomycetes was low - Phytophthora (3.3%) and Pythium (3.0%). Moreover, total fatty acids of the Zygomycota fungi such as Mucor (15.4 mg/100 ml) and Rhizopus (7.1 mg/100 ml) were higher compared with the Oomycetes such as Phytophthora (2.6 mg/100 ml) and Pythium (4.5 mg/100 ml). Thus, two genera such as Mucor and Rhizopus have higher potential as an useful microbial resource. The total fatty acid content varies even within the strains of the same genus e.g. Mucor. M. blumbeus KACC 40935 showed the highest values on productivity (18.2%) of GLA and total fatty acid contents (50.8 mg/100 ml liquid medium).

Determination of Carbon Source Utilization of Bacillus and Pythium Species by Biolog$^{(R)}$ Microplate Assay

  • The carbon utilizations of Bacillus species and Pythium species were investigated by using a Biolog$^{(R)}$ microplate assay to determine if there are differences in the carbon utilizations of selected strains of these species. It may be possible to afford a competitive advantage to bacterial biological control agents by providing them with a substrate that they can readily use as a carbon source, for example, in a seed coating formulation. Microplates, identified as SFP, SFN and YT were used to identify spore-forming bacteria, nonspore-forming bacteria, and yeast, respectively. Bacterial and mycelial suspensions were adjusted to turbidities of 0.10 to 0.11 at 600 nm. One hundred microliters of each of the bacterial and mycelial suspension were inoculated into each well of each of the three types of microplates. L-arabinose, D-galactose, D-melezitose and D-melibiose of the 147 carbohydrates tested were found to be utilized only by bacteria, and not by Pythium species, by Biolog$^{(R)}$ microplate assay, and this was confirmed by traditional shake flask culture. Thus, it indicated that the Biolog$^{(R)}$ microplate assay could be readily used to search for specific carbon sources that could be utilized to increase the abilities of bacterial biological control agents to adapt to contrived environments.

Aphelenchus avenae and Antagonistic Fungi as Biological Control Agents of Pythium spp.

  • To examine the control effect of damping-off on radish caused by Pythium spp., researchers used the isolates of a fungivorous nematode, Aphelenchus avenae, and antagonistic fungi, Trichoderma spp. These were used as biocontrol agents, either alone, or in combination. Growth rates of the A. avenae isolates and fungal damages by the nematodes varied depending on Trichoderma spp., which contained lower T. koningii and T. virens cultures than other Trichoderma cultures. Phythium spp. were damaged by all five Aphelenchus isolates, but the multiplication rate of nematode isolate Aa-3 was very poor. Antibiotic activity of T. virens and T. harzianum to Pythium spp. was stronger than that of T. koningii. Control efficacy against damping-off of radish was most enhanced under the treatment using the nematode-T. harzianum combination. On the contrary, the combinations of the nematodes and T. virens or T. koningii mostly did not increase or decreased their control effect vis-$\`{a}$-vis that of the nematodes or antagonistic fungi being used alone. The results suggest that the fungivorous nematodes may play a leading role in the disease control, and that the activity of the fungivorous nematodes may be activated by T. harzianum, but inhibited by T. koningii and T.virens.

Identification and Pathogenicity of Microorganisms Associated with Seed-Rhizome Rot of Gingers in Underground Storage Caves (토굴저장 생강의 부패에 관여하는 미생물의 동정 및 병원성)

  • Microorganisms associated with seed-rhizome rot of gingers preserved in three underground storage caves were identified with respect to rot types. Rot patterns were grouped into 4 different types : yellow soft rot, brown rot, localized ring rot, and water-soaked rot. Water-soaked rot was highest in frequency with 40% and ring rot the least with 14%. Causal pathogens differed with rot type, yellow soft rot by Erwinia carotovora and Pseudomonas aeruginosa, brown rot by Fusarium solani and Pseudomonas aeruginosa, localized ring rot by F. solani, and water-soaked rot by Pythium spinosum and P. ultimum. Pythium myriotylum, the causal pathogen of ginger rhizome rot which occurs severely in fields was rarely detected from storage seed-rhizomes suggesting its minor involvement with storage rot. Pathogenic Pythium isolates were frequently obtained from both rhizome surface and inner tissues of rotten rhizomes. Detection frequency of Pythium isolates in inner tissues decreased as increasing distance from rhizome surface. In wound-inoculation tests, above pathogens caused a varying degree of rot on healthy rhizomes at 15$^{\circ}C$, 2$0^{\circ}C$ and 3$0^{\circ}C$ with increasing severity at higher temperatures.

The Structural Chracteristics That Influence on the fungicidal activity of N-Phenylbenzenesulfonamide Derivatives against Dapping-off (Pythium ultimum) (모잘록병균(Pythium ultimum)에 대한 N-Phenylbenzenesulfonamide 유도체의 살균활성에 미치는 구조적 특성)

  • The structural characteristics that influence on the fungicidal activities of N-phenylbenzenesulfonamide (1~24) and N-phenyltheinylsulfonamide (25~30) derivatives against Dampping-off (Pythium ultimum) were discussed quantitatively using the models of molecular holographic quantitative structure-activity relationships (HQSAR). From the based on these findings, the statistical results of the optimized HQSAR F-3 model showed better predictablity ($r^2_{cv.}$ or $q^2=0.581$) and correlation coefficient ($r^2_{ncv.}=0.963$). And, from the analytical results of the atomic contribution maps on the fungicidal activities, the most active compound is $R_1=2-fluoro-4-chloro$ substituent (4) and the most inactive one is $R_1=4-methoxy$ substituent (20). It was found that the 2-fluoro-4-chlorophenyl group as a $R_1$-group was an important structure and a characteristic factor not only for herbicidal activity but also for fungicidal activity.

Biological Control of Seed and Seedling Rot Caused by Pythium arrhenomanes in Water-Seeded Rice (벼 담수직파재배에서 Pythium arrhenomanes에 의한 벼모썩음병의 생물학적 방제)

  • 벼 담수직파재배에서 Pythium arrhenomanes 1398에 의한 벼목썩음병의 발병 억제효과가 높은 biological control agent를 배지와 growth chamber 검정을 통하여 선발하고 생물학적 방제 효과를 온실에서 검정한 결과 공시한 66 세균 중 91-110 균주에 의해서만 벼모썩음병의 발병이 억제되었다. CMA 배지 검정에서는 P. arrhenomanes 1398 균주만을 접종한 볍씨는 100% 발병율을 보인 반면 91-110 균주와 P. arrhenomanes 1398 균주를 함께 처리한 경우에는 25.0%가 발병되었으며, 이 효과는 metalaxyl을 처리한 볍씨에서의 21.7% 발병율과 유사하였다. Growth chamber 검정에서는 P. arrhenomanes 1398 균주만을 접종한 볍씨는 27.7% 모 출아율을 보인 반면 91-110 균주와 함께 처리하면 59.1%가 출아되었다. 그러나 91-110 균주에 의한 발병 억제효과는 50% household bleach로 표면 소독한 볍씨에서만 인정되었으며, 이 소독한 볍씨를 91-110 균주의 세균 부유액에 침지하여 3시간 진탕한 후 그 부유액을 따라내고 24시간 배양한 것이 진탕하지 않고 배양한 것에 비하여 모출아율이 54.3%에서 65.7%로 증가되었다. 91-110 균주와 P. arrhenomanes 1398 균주를 함께 처리한 볍씨의 출아율은 62.3%로서 P. arrhenomanes 1398 균주만을 접종한것의 30.8%에 비하여 높았다. 이상과 같이 담수직파한 벼모썩음병의 발병 억제효과가 높은 91-110 균주는 Bacillus brevis로 동정되었다.

Biological Control of Turfgrass Diseases by Pseudomonas cepacia AF-2001 (Pseudomonas cepacia AF-2001을 이용한 잔디병의 생물학적 방제)

  • Effect of an antagonistic bacterium AF-2001, Pseudomonas cepacia, on control of turfgrass diseases as brown patch, Pythium blight, dollar spot, and large patch were evaluated in vitro and in vivo. Results obtained in this study are summarized as follows. 1. Pseudomonas cepacia AF-2001 showed antagonism against to the pathogens causing brown patch, Pythium blighe, dollar spot and large patch. Especially, the biological agent showed strong antagonistic effect on the causal pathogens of brown patch, dollar spot, and anthracnose, but weak on Pythium blight 2. Populotion density of P. cepacia AR-2001 decreased ra;idly in turfgrass soils. Initial population of the agent was 2.4$\times$107 cfu/g soil, however, decreased to 1.4$\times$103, 6$\times$102 and 0 cfu/g soil on 10, 20, and 30 days after application, respectively. 3. Under the controlled controlled conditions of $27^{\circ}C$ and 95% RH, P. cepacia AR-2001 showed 100% control efficacy on brown patch either by pre-treatment or post-treatment of infection. However, Pythium blight was controlled about 94% by pre-treatment and only 29% by post-treatment 4. In field trials, P. cepacia AF-2001 did not suppress large patch and the control efficacy on other turfgrass diseases was lower than agro-chemicals such as tebuconazole and metalaxyl. Control efficacy of brown patch, Pythium and dollar spot by the biological agent was 57.4%, 40.4%, 61.5~87%, respectively. 5. Growth, color and texture of creeping bentgrass were not differ significantly between AF-2001 treatment and untreated control.

Comparative Study of Cool-Season Turfgrass Varieties Introduced (한지형 잔디의 품종비교)

  • These experiments were conducted to compare the growth characteristics, cold and drought tolerances, disease tolerance and color of the 23 varieties of four different species of cool-season turfgrass introduced from the United States of America and Europe at Yonam College in 1995. Newport and Glade of 8 Kentucky bluegrass varieties were recorded better marks than others. Newport variety was recorded the good marks in the disease tolerance of summer seasons, uniformity, coverage and texture, but the bad marks in color, drought tolerance and rust tolerance and stay green of automn seasons. Glade variety was recorded the good marks in the disease tolerance of summer seasons, uniformity and coverage, but the bad marks in drought tolerance, texture and Pythium blight tolerance. Revel and Revel Jr of 5 tall fescue varieties were recorded better marks than others in color, shoot density, uniformity, coverage, and tolerance of Pythium and Fusarium blight disease. All of red fescue varieties died of disease for rainy season however texture was the finest of all the species in spring season. Dandy of 4 ryegrass varieties was recorded better marks than others in color, shoot density, coverage, drought tolerance and tolerance of Pythium and Fusarium blight disease but texture.

Potato Basal Stem Rot Caused by Pythium myriotylum in Hydroponic Cultural System (양액 재배시 발생하는 Pythium myriotylum에 의한 감자 줄기기부썩음병)

  • A basal stem rot disease of potato (Solanum tubersum) grown on hydroponic system in greenhouse was found in Jeju-city, Jeju Province of Korea in January 2001. The symptoms occurred on basel stem and root, and than plants eventually were died. The casual fungus isolate from basal stem rot symptom and identified to be Pythium myriotylum on the basis of mycological characteristics; Main hyphae wide were 8.4 ${\mu}{\textrm}{m}$ in size. Oogonia were spherical, smooth, mostly terminal in shape and 26.4∼31.2 ${\mu}{\textrm}{m}$ in size. Antheridia were 9.6∼14,4${\times}$4.8-9.6 ${\mu}{\textrm}{m}$ in size. Oospores were spherical, smooth, aplerotic in shape and 21.6∼26.4 ${\mu}{\textrm}{m}$ in size. This is the first report on the basal stem rot of potato caused by P. myriotylum in Korea.
