Protective Effects of a Ginseng Component, M altol(2- M ethyl-3- Hydroxy-4- Pyrone) against Tissue Damages Induced By Oxygen Radicals

  • Maltol(2-methyl-3-hydroxy-r-pyrone), a component known to be present in Korean Ginseng root showed an antioxidant action but its potency as an antioxidant was low; about 1150th that of other antioxidants such as p-phenylenediamine , BHA and BHT. However, maltol was able to protect the oxidation adamants in biological systems such as adriamycin-induced membrane damage in isolated cardiomyocytes, parquet-induced toxicities in isolated hepatocytes and repercussion injury in isolated hearts. The antioxidant action of maltol was also shown to be effective in vivo. The antioxidant action of this compound was probably due to the removal of hydroxyl radicals. In view of the roles of oxygen radical in various pathological processes, Korean Ginseng root, which contains several antioxidants including maltol, is expected to have beneficial efforts on the oxygen radical-involved processes.   • Intramolecular Diels-Alder cycloadditions of 2-pyrone-3-carboxylates with trans-vinyl silaketal groups tethered via a chiral, non-racemic 1,3-butanediol auxiliary proceeded in unexpected stepwise cycloadditions through ionic intermediates to provide cis-disubstituted bicylolactones. The ratio of two isomers, exo and endo, was 5 to 1, and each isomer was found to be diastereomerically pure (>99% de). Their relative and absolute stereochemistries were determined by $^1H$ NMR spectroscopy and confirmed by X-ray crystallography of minor, endo-adduct 9. The major exo-adduct was successfully transformed to (-)-2-butyl substituted A-ring phophine oxide 16, a key element for the synthesis of 2-butyl vitamin D3.

  • Extensive screening for cellulose-producing bacteria was done using differential media. Fifty seven strains were isolated totally from the fruits and the vinegar, respectively; the isolate A9 strain from apples was selected and examined to determine its taxonomical characteristics. The bacterium was identified as the genus Acetobacter sp_ based on morphological, cultural and biochemical properties. A9 strain produced acetic acid from ethanol and decomposed acetic acid to $CO_2$ and $H_2O$. They produced dihydroxyacetone from glycerol but did not produce y-pyrone from glucose and fructose. When A9 strain was cultivated statically in Hestrin and Schramm liquid medium(HS medium). thick cellulose pellicle was formed_ Higher cellulose production was obtained in the shaken culture using HS medium at 100 rpm.

  • An antioxidant was isolated from pine needles. This compound was identified as 4-hydroxy-5-methyl-3[2H]-furanone on the basis of spectroscopic evidences and by the comparison with the synthetic one from the Maillard reaction of xylose and glycine. It scavenged 1,1-diphenyl-2-picrylhydrazyl free radicals more efficiently than 4-hydroxy-2,5-dimethyl-3[2H]-furanone and $3-hydroxy-2-methyl-{\gamma}-pyrone$ did. It exhibited inhibitory effects on the autoxidation of 3,4-dihydroxyphenylalanine and linolenic acid.

  • The crystal structure of the potential active 4,6-dimethyl-9-phenyl-8,12-dioxa-4,6-diazatetracyclo [8.8.0.02,7.013,18] octadeca-2(7),13,15,17-tetraene-3,5,11-trione 2-ethoxyphenyl (2E)-but-2-enoate ($C_{22}H_{18}N_2O_5$) has been determined from single crystal X-ray diffraction data. In the title compound crystallizes in the monoclinic space group $P_12_1/c_1$ with unit cell dimension a=15.2039(8), b=12.3888(6) and c= 9.8162(5) [alpha & $gamma=90^{\circ}$ beta=98.113(2)]. In the structure fused pyrone and pyran rings each adopt a sofa/envelop conformation. The crystal structure is stabilized by intramolecular C-H... O hydrogen bond interaction.

  • The photoreaction of 5,7-dimethoxycoumarin with adenosine has been carried out in a dry film state. The mixture of DMC and adenosine was irradiated with 350 nm UV light and two major products were isolated. The structure was determined by various spectroscopic measurements involving $^{13}C$ nuclear magnetic resonance and fast atom bombardment mass spectrometry. These addition products were produced by covalent bond formation between the pyrone ring at carbon 3 or 4 and the sugar ring moiety of adenosine at carbon 5'.

  • The michael adducts 2a, b were obtained from the reaction of the phenylacelyl derivative 1 with benzadehyde and p-anisaldehyde and p-anisaldehyde respectively. 2a and 2b were subjected to react with cyanoethanoic acid hydrazide, malononitrile, cyanothioacetamide, cyanoacetamide and 1, 1, 3-tricyano-2-amino propene to yield 4a-h and 5a, b respecitively. Hydrogen peroxide oxidation of 2a, b gave the aurone derivative 6a, b. The pyone derivatives 8a, b were obtained from 2a, b by addition of chioroacetyl chioride followed by dehydrochlorination.
