Resistance Function of Rice Lipid Transfer Protein LTP110

  • Abstract Plant lipid transfer proteins (LTPs) are a class of proteins whose functions are still unknown. Some are proposed to have antimicrobial activities. To understand whether LTP110, a rice LTP that we previously identified from rice leaves, plays a role in the protection function against some serious rice pathogens, we investigated the antifungal and antibacterial properties of LTP110. A cDNA sequence, encoding the mature peptide of LTP110, was cloned into the Impact-CN prokaryotic expression system. The purified protein was used for an in vitro inhibition test against rice pathogens, Pyricularia oryzae and Xanthomonas oryzae. The results showed that LTP110 inhibited the germination of Pyricularia oryzae spores, and its inhibitory activity decreased in the presence of a divalent cation. This suggests that the antifungal activity is affected by ions in the media; LTP110 only slightly inhibited the growth of Xanthomonas oryzae. However, the addition of LTP110 to cultured Chinese hamster ovarian cells did not retard growth, suggesting that the toxicity of LTP110 is only restricted to some cell types. Its antimicrobial activity is potentially due to interactions between LTP and microbe-specific structures.

  • Sporulation potential and conidia release phase of Pyricularia oryzae on lesions under the natural conditions were measured in 1985 and 1986 leaf blast seasons. The amount of conidia produced in lesions on detached leaves and conidia released under the field condition were reached peak at $5{\sim}7$ days after lesion appearance. The maximum numbers of conidia produced and released were 16,200 and 15,900, respectively. Conidia release under the natural conditions lasted for 30 days.

Monitoring of Physiological races of Pyricularia oryzae in Gyeongnam Province (경남지역 벼 도열병균 Race의 분포추이)

  • 경남지방에서 벼도열병균 race의 분포상황을 알기 위하여 1990∼1994년 5개년간 경남지역의 재배품종 잎동열병 병반에서단포자 분리하여 얻은 균주 509 균주를 사용하여 연도별 race 분포상황을 조사한 결과는 다음과 같다. 도내에 분포한 race의 종류는 총 33종류였다. 이 중 KI 100군 race가 16종류로 연차간 분포변동이 심하였고, KI 400군 race가 6종류, KJ 100군 race가 4종류, KI 300군 race가 3종류, KJ 200군 race 2종류, race KJ-301 및 race KJ-401이 각각 1종류씩이었다. KI 및 KJ race의 분포 비율은 각각 11.5∼29.9%와 70.1∼88.5%로 KJ race의 분포밀도가 높았고 분포밀도가 높은 race는 KJ-401 및 KJ-301로 전 race의 26.7∼57.3%를 차지하였다. 지대별 race 구성 특징은 산간 지대에서는 KI race의 분포밀도가 27.3%로 해안지대 6.8%에 비해 훨씬 높았고 KJ 100군 race도 27.7%로 해안지대 11.7%보다 분포비율이 높았다. 반대로 KJ 200군 race는 해안지대가 22.0%로 산간지대 9.7%보다 아주 높았다. 조사기간중 재배된 벼품종과 race의 분포변동을 미루어 보아 race의 분포는 재배된 벼의 도열병균 race에 대한 반응형과 관련이 있을 것으로 사료된다.

Antifungal Activity of Extract of Common Purslane (Portulaca oleracea L.) (쇠비름 즙액(汁液)의 항균작용(抗菌作用))

  • Extracts of common purslane(Portulaca oleracea L.) showed to possess some antifungal substances which inhibited the mycelial growth of the phytopathogenic fungi tested;Valsa mali, Alternaria kikuchiana and Pyricularia oryzae. These antifungal substances were found to be soluble in methanol and were regarded as kinds of lipid. In order to isolate the antifungal substances, the extracts of common purslane were concentrated by evaporation under reduced pressure and extracted with methanol The methanol solution was subjected to silica gel-florisil column and divided into lipid and non-lipid fractions. Lipid fractions only showed antifungal activity against the fungi tested. The effective substances contained in the extracts of common purslane inhibited not only the mycelial growth but also the spore germination of the fungi.

  • Four types of spore trap (Kim's original, improved Kim's original, Yoshino's original and mixed type of Kim's and Yoshino's original) were evaluated for their efficacy to "estimate the amount of spores released from leaf blast lesions under the natural conditions. It was found that all four types had one or two defects in allowance for adequate sporulation/release, spore catch or spore counting. Thus, an improved type of spore trap was devised considering that it could cover the defects mentioned above. As a result, newly developed spore trap was quite satisfactory in above mentioned aspects and it could be used for pursuit of spore release phase under the natural conditions.

  • 2-Imino-1,3-thiazolines 1 show selective antifungal activity against pyricularia oryzae by new mode of action. A synthesis of novel 2-cyano-l,3-thiazolines 2 in which cyano moiety is substituted in imino group at C-2 of 1 is described. The lone pair electrons of sulfur and nitrogen as well as strong electron withdrawing cyano substituent in 1,3-thiazoline scaffold would effect the biological activity of 2-imino-1,3-thiazoline series. Changes in Endophyte Communities across the Different Plant Compartments in Response to the Rice Blast Infection

  • The rice blast disease, caused by the fungal pathogen, Magnaporthe oryzae (syn. Pyricularia oryzae), poses a significant threat to the global rice production. Understanding how this disease impacts the plant's microbial communities is crucial for gaining insights into host-pathogen interactions. In this study, we investigated the changes in communities of bacterial and fungal endophytes inhabiting different compartments in healthy and diseased plants. We found that both alpha and beta diversities of endophytic communities do not change significantly by the pathogen infection. Rather, the type of plant compartment appeared to be the main driver of endophytic community structures. Although the overall structure seemed to be consistent between healthy and diseased plants, our analysis of differentially abundant taxa revealed the specific bacterial and fungal operational taxonomic units that exhibited enrichment in the root and leaf compartments of infected plants. These findings suggest that endophyte communities are robust to the changes at the early stage of pathogen infection, and that some of endophytes enriched in infected plants might have roles in the defense against the pathogen.

Comparision of Peroxidase, Superoxide dismutase and Diterpene cyclase Activity under Environmental Stress Conditions in Rice (환경 스트레스에 의한 벼의 Peroxidase, Superoxide dismutase 및 Diterpene cyclase 활성비교)

  • This study carried out to investigate activities of the superoxide dismutase(SOD), peroxidase(POD) and diterpene cyclase of rice plant treated by environmental stress conditions. The rice leaves of Ilpum was shown to have the most highest specific activity of POD. POD activity in the susceptible cv. Ilpum and Hwasung during the disease progress show a increase, while for a decreasing activity in the resistant cv. Dasan. SOD activity of rice plants treated by chitosan were increased as the treatment concentration increased. At the chitosan application, the SOD activity in the susceptible cv. Hwansung at the rice leaves 14days after treatment was higher in treatment than chitosan-untreated control. The activity of POD in the susceptible cv. Ilpum decreased as long as UV-B irradiation, while that in the resistant cv. Dasan increased. Futhermore, activity of that in Dasan were similar to as time on UV irradiation, Hwasung showed two times higher than untreated control at 40 minutes after UV-B exposure. The activity of diterpene cyclase in rice plants inoculated with Pyricularia oryzae has much higher tendency than UV-B irradiation. The activity of diterpene cyclase in the rice leaves gradually increased until 5 days after inoculation with Pyricularia oryzae.

Effects of 2-Mercaptoethanol on the Protoplast Formation and Osmotic Stabilizers on the Protoplast Reversion of Pyricularia oryzae Cavara (벼 도열병균(稻熱病菌)의 원형질체(原形質體) 생성(生成)에 미치는 2-Mercaptoethanol과 복귀(復歸)에 미치는 삼투압 안정제(安定劑)의 영향(影響))

  • The mycelia of Pyricularia oryzae were treated with enzyme solution mixture consisting of Driselase, ${\beta}-Glucuronidase$, Cellulase, and Macerozyme R-10 for the production of protoplasts. More protoplasts were formed from mycelia of race KJ 101 of P. oryzae than that of race KI 315a in the enzyme mixtures. The number of protoplasts was decreased in the untreated control three hrs after the enzyme treatment, whereas the number was increased in the treatments with 10, 50 and 100 mM 2-mercaptoethanol, respectively. The number of protoplasts increased to reach maximum at five hrs after treatment of 10 mM 2-mercaptoethanol, but the least was found in 200 mM. The protoplasts of P. oryzae in a liquid medium containing 2.5% yeast extract, and 2% dextrose reverted to the mycelia after five hrs shaking incubation at $27^{\circ}C$. Some protoplasts produced yeast like buds and the buds were developed to irregularly shaped chains of cell protruded a germ tube like hypha from the distal cell. Once in a while a germ tube like hypha protruded directly from the protoplasts. Except in the first type of reversion, other protoplasts reverted to the normal mycelia. The reversion frequency was highest on PDA with stabilizer of 0.6 M KCl. No reversion of protoplasts occurred on water agar regaardless oftreaatments.

Studies on antibiotic against wheat black rust (II) (밀의 항흑수병 항생물질의 연구 II)

  • The substance produced by Bacillus subtilis $480HS_{20}$ showed specific anti Puccinia graminis activity and narrow antimicrobial activity. The anti Puccinia graminis substance was found to be peptide antibiotic which had molecular weight of about 1,500 by sephadex LH-60 gel-filtration. The amino acid composition of the substance waas composed of Ser (1), Glu(3), Ile(1), Gyr(2) and His (1). The compound was yellowish powder and it's melting point was $214{\sim}216^{\circ}C$. UV spectrum in methanol showed single peak at 278(E 1% 1cm 15.2) and important peak at 3,350-3,320, 2,940, 1,635, 1,520-155, 1,240-1, $230cm^{-1}$ were observed in IR Spectrum. The substance was soluble in methanol but insoluble in water, ethyl acetate and chloroform. The antibiotic $480HS_{20}$ showed a little antimicrobial activity against Pyricularia aryzae but no antimicrobial activity was observed against Gram + and Gram - bacteria and molds except pyricularia oryzae.
