Effect of Polyamines on Formation of Adventitious Roots, Trichomes and Calli by NAA in Leaf Segment Cultures of Arabidopsis thaliana (애기장대 잎 절편 배양시 NAA 농도에 따른 부정근, 모용 및 캘러스 형성에 미치는 Polyamine의 영향)

  • In order to study the role of polyamines on the formation of adventitious roots, trichomes and calli, the effects of putrescine, spermidine, spermine, cyclohexylamine (CHA) and methylglyoxal-bis(guanylhydrazone) (MGBG) were investigated in the leaf segment cultures from ecotype Columbia of Arabidopsis thaliana. When the leaf segments were cultured on the media for forming adventitious roots (0.1 mg/L NAA), trichomes (2.0 mg/L NAA) and calli (10.0 mg/L NAA), and then each cultures was treated with 1-100 mg/L of putrescine, spermidine and spermine, respectively. On the adventitious root-forming medium treated with polyamines the trichomes were induced with adventitious roots. And on the trichome-forming medium with polyamines calli were induced with trichomes. In orther hand each cultures was treated with 1-100 mg/L of CHA and MGBG, respectively. CHA promoted adventitious roots on the medium for adventitious roots, was not effected on media for trichomes and calli. MGBG inhibited adventitious roots, trichomes and calli in all cultures, and induced adventitious roots on medium for trichomes in high concentration.

Effect of Strontium on Polyamine Synthesis and Diamine Oxidase during the Germination of Mung Bean (Vigna radiata L.) (숙주의 발아과정 동안 폴리아민 생합성과 Diamine 산화효소에 대한 스트론티움 효과)

  • Objective of this experiment was to investigate the role of strontium in intracellular processes in mung bean (Vigna radiata L.). Diamine oxidase (DAO) induction by $Sr^{2+}$ appeared to a decrease in putrescine levels correspondently. DAO activities in the hypocotyls were in a range of 0.5 to $1.8unit{\cdot}mg^{-1}\;protein{\cdot}min^{-1}$. The decrease in Put levels in the cotyledons might be partly resulted from Put degradation by DAO. It was observed that the accumulation of spermidine and spermine by $Sr^{2+}$ was in the range of 1 mM to 10 mM. Spermidine levels were 2 to 3 fold higher than in the absence of strontium. The increase in polyamine levels was observed not only on a basis of g fresh weight but also a RNA basis. These results demonstrated that the inhibitory action of $Sr^{2+}$ may be closely related with polyamine metabolism as well as diamine oxidation and polyamine accumulation.

  • The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

The Effect of Polyamines on the DNase Activity in Cultured Carrot Cells (당근(Daucus carota L.)배양세포의 DNase활성에 미치는 Polyamines의 영향)

  • The present study was attempted to investigate the effects of polyamines such as putrescine, spermidine and spermine on protein content and DNase activity in vivo and in vitro in carrot embryos. It was also investigated whether polyamines could replace role of cations required for DNase activity in vitro. The results obtained are as follows. Putrescine, spermidine and spermine increased protein content, although response to spermine reached plateau at the concentration of 0.1 mM. DNase activity was inhibited by polyamines, the inhibition being concentration-dependent and the highest att he concentration of 10 mM. The inhibition of DNase activity was the most prominent with spermine. Similar inhibitory effect to polyamines which was concentration-dependent was found in DNase activity but no change was shown on time-course in vitro. Putrescine and spermidine enhanced the DNase activity at low Mg2+ and Mn2+ concentrations, suggesting that the role of Mg2+ and Mn2+ for DNase activity could be, in part, replaced by these polyamines. These results, therefore, suggest that plyamines can modulate DNase activity through binding to DNA rather than direct effect on DNase activity.

  • A rapid and sensitive high performance liquid chromatographic method with UV detection for the analysis of polyamines(putrescine, spermidine, spermine) has been developed. The benzoyl chloride derivatives of putrescine, spermidine and spermine are separated on a mc-Bondapak C18 reverse-phase column with 50% methanol as the mobile phase. This method was applied to the analysis of polyamines in the serum of normal human and uterine cancer patients. The results show that the mean level of polyamines in cancer patients serum is much higher than that in normal human serum.

  • Substrate specificity of a flow-injection-analysis (FIA)-type biogenic amine sensor with enzyme reactor was determined. The enzyme reactor was prepared with a diamine oxidase immobilized on preactivated chitosan porous beads (Chitopearl) by intramolecular cross-linking via glutaraldehyde. The sensor showed a rapid response to putrescine and a quasi-linear calibration curve was obtained up to 15.0 mM. The optimal pH and temperature of the enzyme reactor system were 7.5 and $35^{\circ}C$. Interferences due to ATP-related compounds and trimethylamine, and the effects of NaCl and amino acids were measured. Inhibitory effects owing to these components could be mitigated by sample extraction with perchloric acid. Polyamines except putrescine were determined by a putrescine calibration range within 26.7%. This system was confirmed as rapid and convenient for biogenic amine determination.

  • We have purified and characterized a deoxyribonuclease from zygotes of the eukaryotic green alga, Chlamydomonas reinhardtii and investigated effects of the polyamines, putrescine, spermidine and spermine on the purifed endonuclease-catalyzed cleavage of plasmid DNA. The enzyme has a molecular weight of about 37 kDa as measured by gel filtration and SDS-polyacrylamide gel electrophoresis. There is no requirement for a divalent cation. The activity is sensitive to ionic strength, as NaCl and KCl result in inhibition. The cleavage of plasmid DNA by the purified endonuclease was effectively inhibited by polyamines. The enzyme activity was inhibited more effectively by spermine than by spermidine. The inhibition by putrescine was lower than the other two polyamines.

Foliar Application of Growth Bioregulators Influences Floral Traits, Cormassociated Traits and Chemical Constituents in Gladiolus grandiflorus L.

  • Gladiolus is one of the most popular cut flowers in the world floral market and is highly attractive to consumers. The production of poor-quality gladiolus spikes is one of the most important hurdles to obtaining high prices for this crop. The main objectives of this study were to improve the quantitative characteristics of gladiolus spikes, to increase propagule production and to enhance chemical constituent levels in leaves to help improve plant growth. We performed foliar application of bioregulators, including polyamines (putrescine and spermine) and vitamins (ascorbic acid and thiamine), at different concentrations (0, 0.1, 0.4, 0.7 and 1 mM) to gladiolus plants at the three leaf, five leaf and slipping stages. Application of 1 mM putrescine was the most effective treatment, with prominent effects on economically important spike traits including spike length (55.53 cm), spike diameter (7.53 mm), florets per spike (13.99), first floret diameter (9.66 cm) and first floret weight (4.90 g), followed by spermine treatment. The 1 mM putrescine treatment also had positive effects on corm-associated traits including corm diameter and corm weight, with values of 4.57 cm and 26.16 g compared to 3.47 cm and 17.16 g in control plants, respectively. The contents of chemical constituents in leaves, including total chlorophylls ($8.06mg{\cdot}g^{-1}$), total carotenoids ($1.66mg{\cdot}g^{-1}$), total soluble sugars ($4.75 mg{\cdot}g^{-1}$) and phenolics ($0.89mg{\cdot}g^{-1}$) increased in response to foliar application of 1 mM putrescine compared to the control. Ascorbic acid and thiamine also had positive effects on various plant morphological traits and chemical constituents in leaves compared to control plants, but the growth-promoting effects of polyamines were more pronounced. Hence, applying polyamines to gladiolus leaves will help improve spike production and increasing propagule (cormel) yields.

Influence of Salinity Treatment on Seed Germination and Polyamine Synthesis in Barnyard Grass(Echinochloa hispidula) (강피종자의 발아와 폴리아민 생합성에 대한 염류의 영향)

  • To illuminate the physiological response to salinity, barnyard grass (Echinochloa hispidula) was germinated with high concentration of NaCl and KCL. Duration and promptness of seed germination were observed. Under salt stress, lipid peroxidation and polyamine biosynthesis were also analyzed. It appeared that high salt treatments per se did not provoke an inhibition of germination although the process of germination was significantly delayed. In context of lipid peroxidation and polyamine biosynthesis, we would imply that barnyard grass is tolerant to salinity. The increase in lipid peroxidation and putrescine content was prolonged only for 1 day after saline treatment. It could be concluded that these early acciimulation of putrescine and production of lipid peroxide seems to be associated with salt tolerance in the short-term. The physiological interest of these responses was discussed.

  • A Corynebacterium sp. capable of utilizing diaminododecane (DAD) were isolated from the soil by enrichment culture. Among 9 different kinds of substituted alkanes containing CN, $NH_2$, Cl, and SH groups (monoterminally or diterminally substituted) tested as carbon source, the isolate, designated as DAD 2-3, utilized DAD, putrescine dihydrochloride, dodecane and laurylamine. Dodecanethiol, thioanisole, decanedithiol, dicyanooctane, laurylcyanide,and dichlorodecane were not utilized. When emulgen 950 was added to the medium, the growth of DAD 2-3 was slightly accelerated. Isolate DAD 2-3 grown in the medium with DAD as carbon source formed .alpha.-ketoglutaric acid. Metabolic product of DAD 2-3 grown in a medium without nitrogen source was different from that of grown in a medium with $NH_4NO_3$. When glucose, putrescine, n-dodecane and other alkane derivatives were tested in place of DAD, isolate DAD 2-3 yielded products different from those they formed with DAD suggesting specificity of DAD as a carbon source.
