A report of six unrecorded bacterial species isolated from soil samples in Korea

  • During an investigation of unrecorded prokaryotic species in Korea, six unrecorded bacterial strains were isolated from soil samples collected from Uljin-gun. Based on a similarity search using the 16S rRNA gene sequence of the isolated strains and the construction of the neighbor-joining phylogenetic tree, five strains were identified to the genus Pseudomonas of the family Pseudomonadaceae, while one strain was identified as a species belonging to the genus Paenibacillus of the family Paenibacillaceae. The details of these unreported species, including gram staining reaction, colony and cell morphology, basic biochemical characteristics, strain ID, and isolation source, are described in the description of the strains.

  • A marine bacterium (strain BKl) that produces extracellular enzymes capable of decomposing complex polysac-charides, such as agar, chitin, carboxymethylcellulose, xylan and mannan, was isolated from the marine red alga Porphyra dentata. Strain BKl was gram-negative, aerobic, catalase- and oxidase-positive, polarly flagellated bacilli that produce gelatinase and urease, but not decarboxylases. The G+C content of the DNA was 51.6 mol%. The major isoprenoid quinone component was identified as an ubiquinone-8, and the major cellular fatty acids were C16:0, C16:1 w6c and C18:1 w7c. Comparative 16S rRNA sequence analysis placed strain BK1 with members of the genus Pseudomonas. On the basis of phenotypic and genotypic data, the strain BK1 was shown to be a member of the subgroup of Pseudomonas, and named as Pseudomonas sp. BK1.

Deproteinized Mulberry Leaf Juice - A New Media for Growth of Microorganisms

  • Mulberry being a foliage crop is grown extensively for feeding of silkworms and are also used for cattle feeding. These loaves are highly nutritious, which contain various mineral elements and bio-molecules such as carbohydrates, proteins, lipids and other essential amino acids, etc. In the present study, deproteinized mulberry leaf juice was used for preparation of the medium for cultivation of various types of microbes. Results revealed that deproteinized mulberry leaf juice medium is best for isolation of fungi, bacteria and actinomycetes and this medium can be substituted with synthetic media, which are haying the costly ingredients for isolation and identification of bacteria, fungi and actinomycetes. Further, this deproteinized mulberry juice medium can also be used in mass multiplication of useful/beneficial microbes to enhance soil microflora to improve soil fertility and to avoid root diseases. Perspective enterprises can take up the mass multiplication/large-scale production of useful microbes such as Trichoderma, Rhizobium, Pseudomonas and Bacillus to use in mulberry and in other agricultural crops using deproteinized mulberry leaf juice.

  • Objectives; Bioassay-directed fractionation of the dried roots of Salvia miltiorrhiza led to the isolation of abietane tanshinones, crptotanshinone and dihydrotanshinone. Methods: Their structures were elucidated using 1H-and 13C-NMR, UV, IR and mass spectral analysis. Result and Conclusions : These compounds exhibited a moderate antimicrobial activities against Staphylococcus epidemidis, Staphylococcus aureus, Staphylococcus pyogene. Actinobacillus actinomycetemcomitans, Escherichia coil, Enterbacter cloacae, Pseudomonas aeruginosa, Citrobacter freundii, and Samonella typhimurium.

  • Isolation and identification of mesophilic and psychrotrophic bacteria distributed in Korean refrigerated beef were attempted. Total isolated colonies were 192, and identified as 5 genera and 10 species. Among them, mesophilic bacteria were Enterobacter aerogenes, E. agglomerans, Serratia liquefaciens, Proteus mirabilis, and "psychrotrophic" bacteria were Pseudomons fluorescens, P. putida, P. pickettii, P. mendocina, P. stutzeri, Alcaligenes faecalis. Dominant species was Serratia liquefaciens as mesophiles, and Pseudomonas putida as psychrotroph.chrotroph.

  • Two flavanones, (2S) -2'-methoxykurarinones (1) and kurarinones (2) , were isolated from the ethyl acetate extract of the roots of Sophora flavescense Ait. Their structrures were elucidated using NMR, UV and IR spectral analysis. These compounds exhibited a moderate antimicrobial activities against Staphylococcus aureus, Staphylococcus epidermidis Pseudomonas Putida and a weak anti-fungal activity against Candida albicans.

  • A phenolic compound isolated from the ethylacetate extract of Duchesnea indica (Andr.) Focke (Rosaceae) showed antimicrobial activities against Staphylococcus aureus, Shigella dysentriae and Pseudomonas aeruginosa. This compound was identified as ellagic acid by spectral analysis.

  • Detergent-degrading bacteria isolated from the Han River downstream running through the center of Seoul area were identified. Samples were monthly taken from the surface waters at four designated sites from October, 1980 to August, 1981. Isolated strains were characteristically Gram-negative bacteria in all, and they were classified within 18 genera, 43 spceies. The dominant strains of detergent -degrading bacteria were within geneus Pseudomonas. On the regional distribution of detergent-degrading bacteria, Euterobacteriaceae and genus Aeromonas were suddenly increased at the Third Bridge of site 2 and the First Bridge of site 3, and others were almost the same regardless of sites.

  • Isolated the heterotrophic aerobic bacteria in sandfiltered water on NA and TSBA solid medium, selected 8 dominant species and identified by Sherlock System. Each samples are irradiated 0, 5, 16, 40 and $60\;mJ/cm^2$ using on CBD (Collimated Beam Device) Medium Pressure UV lamp after these identified bacterium did liquid culture how to make $10^6{\sim}10^7\;cells/mL$ suspended in dilution water. Then cultured bacteria are estimated inactivation rate on plate media. Identified Gram positive group are Bacillus Subtilus, Bacillus megaterium, Rhodococcus erythropolis and Microbacterium laevaniformans; Gram negative group are Pseudomonas vesicularis, Pseudomonas pseudoflava, Alcaligenes paradoxus and Zooglea ramigera. These isolation of bacterium are more stronger reference strain and high resistance of MP UV irradiation, Besides Gram negative bacterium are more sensitive Gram positive bacterium on MP UV dose. Now we are estimating to $60{\sim}100\;mJ/cm^2$ MP UV dose for efficient disinfection in water treatment plant.

Identification and Bioassay of Nucleation Active Bacteria from Branch Rot of Mulberry and Their Population (뽕나무가지 썩음증상 분리한 빙핵활성세균의 동정, 생물검정 및 그 분석)

  • Isolation, indentification, bioassay and distribution of ice nucleation active(INA) bacteria were done on branch rot of mulberry which was severely developed after harvest of mulberry branches in autumn. Twelve isolates and two isolates out of thirty-six isolates had ice nucleation activity from -5$^{\circ}C$ to -1$0^{\circ}C$, and over -5$^{\circ}C$, respercively. Isolates which formed ice nucleation from -5$^{\circ}C$ to -1$0^{\circ}C$ were not inclined to injure tomato and corn seedlings. However, Two isolates, SE9316 and SE9338 which formed ice nucleation over -5$^{\circ}C$ injured mulberry, tomato and corn seedlings. SE9316 and SE9338 were identified as Pseudomonas syringae based on the morphological, cultural and biochemical characteristics. Populations of ice nucleation active bacteria, fluorecent pseudomonads, were higer in February and April, but they decreased in May. Populations decreased as they become distant from the center of the symptom. The bacterial populations of all sampling times and sites were higer than 105 cfu/g which was enough to induce frost injury.
