Construction of hsf1 Knockout-mutant of a Thermotolerant Yeast Strain Saccharomyces cerevisiae KNU5377 (고온내성 연료용 알코올 효모균주 Saccharomyces cerevisiae KNU5377에서 HSF1 유전자의 변이주 구축)

  • HSF1 is the heat shock transcription factor in Saccharomyces cerevisiae. S. cerevisiae KNU5377 can ferment at high temperature such as $40^{\b{o}}C$. We have been the subjects of intense study because Hsf1p mediates gene expression not only to heat shock, but to a variety of cellular and environmental stress challenges. Basing these facts, we firstly tried to construct the hsf1 gene-deleted mutant. PCR-method for fast production of gene disruption cassette was introduced in a thermotolerant yeast S. cerevisiae KNU5377, which allowed the addition of short flanking homology region as short as 45 bp suffice to mediate homologous recombination to kanMX module. Such a cassette is composed of linking genomic DNA of target gene to the selectable marker kanMX4 that confers geneticin (G418) resistance in yeast. That module is extensively used for PCR-based gene replacement of target gene in the laboratory strains. We describe here the generation of hsf1 gene disruption construction using PCR product of selectable marker with primers that provide homology to the hsf1 gene following separation of haploid strain in wild type yeast S. cerevisiae KNU5377. Yeast deletion overview containing replace cassette module, deletion mutant construction and strain confirmation in this study used Saccharomyces Genome Deletion Project (http:://www-sequence.standard.edu/group/yeast_deletion_project). This mutant by genetic manipulation of wild type yeast KNU5377 strain will provide a good system for analyzing the research of the molecular biology underlying their physiology and metabolic process under fermentation and improvement of their fermentative properties.

  • Overexpression of P-glycoprotein (Pgp) is associated with multidrug resistance (MDR) of tumor cells to a number of chemotherapeutic drugs. Pgp inhibitors have been shown to effectively reverse Pgp-mediated MDR. We prepared a series of phenoxy-N-phenylacetamide derivatives and tested for their ability to inhibit Pgp as potential MDR reversing agents, using a Pgp over-expressing MCF-7/ADR cell line. Some of the synthesized compounds exhibited moderate to potent reversal activity. Of note, compound 4o showed a 3.0-fold increased inhibition compared with verapamil, a well-known Pgp inhibitor. In addition, co-treatment of the representative compound 4o and a substrate anticancer agent doxorubicin resulted in a remarkable increase in doxorubicin's antitumor effect and inhibition of DNA synthesis in the MCF-7/ADR cell line. Taken together, these findings suggest that compound 4o could be a useful lead for development of a novel Pgp inhibitor for treatment of MDR.

  • The present notes summarize the results obtained from the effect of soil-solution ratio on pH values of soils having widely different physico-chemical characteristics. The pH was determined in deionized water N-KCl and 0.01 M $CaCl_2$ solution The results obtained are: 1. With deionized water, the pH values increase with the increase in soil-solution ratio. The increase is more in upland soils than in paddy soils. 2. With N-KCl solution, there is also an increase in pH values with the increase in soil-solution ratio but the increase is less than the corresponding increase with deionized water. 3. With 0.01 M $CaCl_2$ solution, there is practically no change in pH values with the increase in soil-solution ratio except for saline soils. 4. In case of saline soils, the pH increase even in case of 0.01 M $CaCl_2$ solution with increase in soil-solution ratio, the reason for increase may be due to decrease of electric potential by high concentration of salts.

COMPARATIVE RESPONSES OF RICE (ORYZA SATIVA) STRAW TO UREA SUPPLEMENTATION AND UREA TREATMENT

  • Twenty five 75% Holstein Friesian cross bred bullocks fed rice straw (Oryza sativa) of long form, were fed with the following five treatments. 1. Rice straw, untreated (RS) 2. RS + water (1:1), stored for 24 hours (WRS) 3. RS (100 kg) + urea solution (4 kg urea/100 litre water) and dried (USRS) 4. RS (100 kg) + urea solution (as in 3) stored in wet condition for 24 hours (UWRS) 5. RS (100 kg) + urea solution (as in 3) stored in pit for 21 days (UTRS). Potential digestibility of treatments of RS was evaluated by monitoring (in vitro) Simulating Rumen like Fermentation (SRLF). The results indicated that Dry Matter Intake (DMI), digestibility of nutrients, N utilization were of the order UTRS > UWRS > USRS > WRS and RS (p < 0.05 to p < 0.01). SRLF index was high (255.84) for UTRS and least (145.58) for USRS. It was intermediary (199.66) for UWRS. The acetyl content (AC) of UTRS with higher hemicellulose (HCE) digestibility (80.8%) was low compared to UWRS, USRS, RS and WRS. The acetate content was of the order UTRS < UWRS < USRS < WRS and RS thereby indicating that reduction in acetyl content was an index of positive response of urea-treatment of RS. In addition, the ratio of HCE/AC in faeces of UTRS was 0.87 as against the ratios (2.26-2.48) observed in other treatments recording reduction in AC due to urea-treatment. Among the treatments, USRS only supplemented N while UTRS in addition to utilization N, increased the digestibility of structural carbohydrates. Reduction in treatment time from 21 days to 1 day (UWRS) resulted in improvements similar to those of UTRS.

  • Superconducting Gravimeter(SG) was installed and has been successfully operated at MunGyung, Kyungsang province in Korea in March 2005. It was registered as the 21st observatory of the Global Geodynamics Project. Since SG can precisely measure the gravity variations below the 1mHz frequency band, it has the outstanding capability to sense and resolve many different periodic gravity components from each other. From the raw data collected between 18 March 2005 and 21 February 2006 diurnal and semi-diurnal tidal band's residual gravity components were analyzed. During this process, the instrumental noises, air pressure, and ground water corrections were carried out. Values of $-3.18nm/s^2/hPa\;and\;17nm/s^2/m$ were used respectively in the air pressure and groundwater corrections. Hartmann-Wenzel and Whar-Dehant Earth tide models were adopted to compute the residual gravity for Q1, O1, P1, K1, M2, N2, S2, K2 tidal bands. For the ocean loading correction, SCW80, FES952, and FES02 models were used and compared. As a result, FES02 ocean loading model has shown the best match for the data processing at MunGyung SG MunGyung SG gravity was compared with GRACE satellite gravity. The correlation coefficient between the two gravity after groundwater correction was 0.628, which is higher than before ground water correction. To evaluate sensitivity at MunGyung SG gravity statition, the gravity data measured during 2005 Indodesian earthquake was compared with STS-2 broad band seismometer data. The result clearly revealed that the SG could recorded the same period of earthquake with seismometer event and a few after-shock events those were detected by seismometer.

Effect of Toothbrushing Facilities on PHP Index and Oral Health-Related Behaviors in Middle Schools (양치시설 설치여부에 따른 중학생의 구강환경관리능력 및 구강건강관리 행태 변화)

  • This study aims to analyze the effects of tooth-brushing facilities (TBF) at middle schools that installed TBF in 2012 as part of the oral health project in Seongdong-gu, Seoul. The subjects were the first graders at A middle school that installed TBF previously and B middle school that installed TBF in 2012. The results are as follows: DMFT, DMFS and CPI didn't show significant difference by schools (p>0.05), but code (0) was higher and code (2) was lower in female students than male students (p<0.05). As to PHP index, the students at A school with TBF indicated better oral hygiene (p<0.01). In the research on before and after installing TBF, the oral hygiene was improved significantly more after the installation than before (p <0.01). In the comparison on the oral health-related behavior, the percentage of not brushing after lunch was higher B school than A school and the frequency of toothbrushing after lunch was higher A school than B school (p<0.01). About the reasons why not to brush after lunch, there was difference in 'lack of the place' as A school showed 2.6% while B school without TBF indicated 14.2% (p<0.01). About the changes of the oral health-related behavior before and after TBF installation, the percentage of not brushing after lunch increased from 78.7% to 83.8% after the installation. The average frequency of tooth-brushing after lunch for one week also decreased from 0.50 to 0.34 (p>0.05). Among the reasons why not to brush after lunch, 'lack of the place' significantly reduced, but 'because other friends don't do it' increased greatly after the installation (p <0.05). Only with the improvement of oral health knowledge, we cannot expect either clinical or behavioral significance. Not only schools but policies of governmental agencies and financial support, cooperation of community-related groups for program development and evaluation will contribute altogether to helping teenagers to habitualize proper oral health management.

A Comparison of the Soil Physicochemical Properties of the Forest Stands in the Young-il Erosion Control District (영일사방사업지의 임분별 토양특성 비교분석)

  • In this study, the physicochemical properties of the soils in the forest stands in the ecological restoration project in the Young-il Erosion Control District were investigated according to the type of plant growing therein. The soil texture was mostly sandy loam (SL), and the sand content was 59.7% on average while the average soil pH was 5.0, which was lower than the average pH of Korea's forest soil (5.5). Moreover, the average carbon, total nitrogen, and available phosphorus ($P_2O_5$) contents were 1.6%, 0.11%, and 3.7 ppm respectively. The C/N ratio was 15.1, and the average cation exchange capacity (CEC) was 13.2 cmol/kg. The physicochemical properties of the soils in the different forest stands were compared. Among all the stands, the Larix leptolepis stand had the highest pH 6.3 and the most stable C/N ratio (11.7). The Alnus firma-Styrax stand had the lowest pH 4.6 while the Pinus rigida stand had the lowest total nitrogen content. The Alnus firma-Styrax stand had the highest CEC (17.4 cmol/kg). The results showed that the Young-il Erosion Control Districtwas devastated and deprived of soil nutrients over a long period. Therefore, sustainable forest management, suchas tending and regeneration, are recommended for the Young-il Erosion Control District.

The effect of the self-measurement frequency levels on SmartCare obesity management (스마트케어 비만관리에서 자가 측정 빈도 수준의 효과)

  • This study's purpose was to analyze the effect of the SmartCare obesity management pilot project, which was conducted between 09 March 2011 and 03 April 2013 in Korea (South). Of 212 subjects who were enrolled in the study, the final analysis was conducted with 147 subjects who completed their weight self-measurements and SmartCare services until the end of a 6-month service period. The self-measurement group for evaluation was classified into 3 groups (Low, Middle, and High) by evenly dividing the monthly average frequency of measurement for 6 months and the evaluation indices were weight, and BMI (Body Mass Index) transmitted from the SmartCare System. The monthly average weight and BMI were decreased more in the High Group than the Middle Group and Low Group (M5: High>Middle>Low), and they are showing statistically significant differences between groups (p< 0.05). In conclusion, analysis of the weight and BMI improvement effect showed a greater improvement effect when the self-measurement frequency level was higher.

  • This study was conducted to inform basic data for management of abandoned mine that was based on soil heavy metal concentration of the non-rearing and the rearing sites in Guundong Mine which had been completed of improve contamination soils, and examined into standard plant which had tolerances about pollution that analyzed into its accumulated heavy metals contents in the wild plant of naked soil. Consider form the result of contents by depth of soils heavy metals in the non-rearing sites, S1 point judged by contaminated to heavy metals of tailing leach after weathering caused by rainfall and permeate in S1 point soils when restored rest of tailing in 1996 restoration project. Lead of S2 point was not contaminated seriously by restoration and S3 point considered that heavy metals which leach by rainfall in the contaminated soils in mine vicinity accumulated in the abandoned paddy through river When heavy metals of rearing sites soils were examined from the view point of upside of mine point (SP1, SP2, and SP3) of same above the sea level, zinc (537.5 mg/kg) content of SP1 point was highest, and copper (535 mg/kg) and lead (141.5 mg/kg) in the SP3 point were high. To considered as orderly plenty of heavy metals in the plant were respectively in order, Perilla frutescens var. cauta KUDO (SP1 and SP4) >Artemisia princeps PAMPAN (SP2) > Miscanthus sinensis ANDERSSON(SP5) > Vicia angustifolia var. segetilis (THUILL.) K. KOCH. (SP2). Especially, there were plenty of heavy metals in P. frutescens var. acuta, so it judged as a standard plant which have tolerances about heavy metals pollution sites. Therefore, we need more study about the heavy metals accumulate ability of P frutescens var. acuta in future.

Nurses' Perception, Knowledge, and it's Affecting Factors on Performance of Nursing Practice in Hospital with No Guardian (보호자 없는 병실 간호사의 간호업무에 대한 인식, 지식과 수행도에 영향을 미치는 요인)

  • The purpose of this study was to investigate on paid hospital-nurses' work perception, knowledge, performance and the performance-related factors of a nurse who works in a hospital with no guardian which is phased in its beginning as well as interactions between the factors. Data were collected from nurses working in wards without guardian at 9 hospitals among a total of 10 Korean hospitals carrying out a national pilot project which is designed for institutionalization of national caregiving services. Self reported questionnaires were used to collect data from 167 nurses, and 135 returned forms were analyzed(December, 2010). The score of perception was 4.24/5.00, knowledge 4.25/5.00, and performance 3.49/5.00. The performance was significantly different depending on the hospital types(p=.002). Significant correlations were found among perception(p<.001), knowledge(p=.004) and performance. The degree of perception(p<.001) and types of hospital(p<.001) attributed to 16.5% of variance in the practice. To enhance level of performance by taking account of perception and types of hospital, can be effective for hospital-nurses when there is no guardian.
