Exposures to environmental chemicals that mimic endogenous hormones are proposed for a number of adverse health effects, including infertility, abnormal prenatal and childhood development and above all cancers. In addition, recently miRNA (micro RNA) has been recognized to play an important role in various diseases and in cellular and molecular responses to toxicants. In this study, endocrine disrupting environmental toxicant, nonylphenol (NP) was treated to MCF-7 (Human breast cancer cell) and HepG2 (Human hepatocellular liver carcinoma) cell line at 3 hrs and 48 hrs time point and miRNA analysis using $mirVana^{TM}$ miRNA bioarray was performed and compared with total mRNA microarray data for the same cell line and treatment. Robust data quality was achieved through the use of dye-swap. Analysis of microarray data identifies a total of 20 and 11 miRNA expressions at 3 hrs and 48 hrs exposure to NP in MCF-7 cell line and a total of 14 and 47 miRNA expression at 3 hrs and 48 hrs exposure respectively to NP in HepG2 cell line. Expression profiling of the selected miRNA (let-7c, miR-16, miR-195, miR-200b, miR200c, miR-205, and miR-589) reveals changes in the expression of target genes related to metabolism, immune response, apoptosis, and cell differentiation. The present study can be informative and helpful to understand the role of miRNA in molecular mechanism of chemical toxicity and their influence on hormone dependent disease. Also this study may prove to be a valuable tool for screening potential estrogen mimicking pollutants in the environment.

Scour means the erosion of bed material by flow change when a bridge is constructed in a stream. Scour is one of the critical factors of a bridge failure. There are several methods for the monitoring of scour near bridge foundations; Sounding rods, Magnetic sliding collar System, Sonar system, underwater camera system and so on. In general, Sonar system is preferred due to its convenience and good accuracy. In this study, the new scour monitoring system was developed using profiling sonar sensor. The new system can measure a line profile of a seabed and has small size due to the effectively designed data logger. The performance of the new scour monitoring system was evaluated at a bridge pier in tidal environment. The measured local scour depths were discussed with the result of the empirical formulas; CSU, Froehlich, Laursen and Neill.

Diatoms have been used in examining water quality and environmental change in freshwater systems. Here, we analyzed molecular profiling of seasonal diatoms in the Han River, Korea, using the hypervariable region of 18S V1-V3 rRNA and pyrosequencing. Physicochemical data, such as temperature, DO, pH, and nutrients showed the typical seasonal pattern in a temperate region. In addition, cell counts and chlorophyll-a, were recorded at high levels in spring compared to other seasons, due to the diatom bloom. Metagenomic analysis showed a seasonal variation in the phytoplankton community composition, with diatoms as the most frequently detected in spring (83.8%) and winter (69.7%). Overall, diatom genera such as Stephanodiscus, Navicula, Cyclotella, and Discostella were the most frequent in the samples. However, a large number of unknown Thalassiosirales diatoms were found in spring (35.5%) and winter (36.3%). Our molecular profiling revealed a high number of diatom taxa compared to morphological observation. This is the first study of diatoms in the Han River using molecular approaches, providing a valuable reference for future study on diatoms-basis environmental molecular monitoring and ecology.
The goal of the present study was to investigate the antibacterial properties, enzyme production, and metabolic profiling of a new Ceratorhiza hydrophila strain isolated from the submerged aquatic plant Myriophyllum spicatum. Furthermore, the fungus' morphological characterization and DNA sequencing have been described. The fungus has been identified and submitted to the GenBank as Ceratorhiza hydrophila isolate EG19 and the fungus ID is MK387081. The enzyme analyses showed its ability to produce protease and cellulase enzymes. According to the CSLI standard, the ethyl acetate extract of C. hydrophila showed intermediate antibacterial activity against Streptococcus pneumonia, Micrococcus luteus, and Staphylococcus aureus. Metabolic profiling has been carried out using 700 MHz NMR spectroscopy. Based on the 1H and 1H-13C heteronuclear single quantum coherence (HSQC) NMR data and NMR databases, 23 compounds have been identified. The identified metabolites include 31% amino acids, 9% sugars, 9% amines, 4% sugar alcohols, and 4% alkaloids. This is the first report for the metabolic characterization of C. hydrophila, which gave preliminary information about the fungus. It is expected that our findings not only will pave the way to other perspectives in enormous applications using C. hydrophila as a new promising source of antimicrobial agents and essential metabolites, but also it will be valuable in the classification and chemotaxonomy of the species.
Recently, the technologies of DNA sequence variation and gene expression profiling have been used widely as approaches in the expertise of genome biology and genetics. The application to genome study has been particularly developed with the introduction of the nextgeneration DNA sequencer (NGS) Roche/454 and Illumina/ Solexa systems, along with bioinformation analysis technologies of whole-genome $de$$novo$ assembly, expression profiling, DNA variation discovery, and genotyping. Both massive whole-genome shotgun paired-end sequencing and mate paired-end sequencing data are important steps for constructing $de$$novo$ assembly of novel genome sequencing data. It is necessary to have DNA sequence information from a multiplatform NGS with at least $2{\times}$ and $30{\times}$ depth sequence of genome coverage using Roche/454 and Illumina/Solexa, respectively, for effective an way of de novo assembly. Massive shortlength reading data from the Illumina/Solexa system is enough to discover DNA variation, resulting in reducing the cost of DNA sequencing. Whole-genome expression profile data are useful to approach genome system biology with quantification of expressed RNAs from a wholegenome transcriptome, depending on the tissue samples. The hybrid mRNA sequences from Rohce/454 and Illumina/Solexa are more powerful to find novel genes through $de$$novo$ assembly in any whole-genome sequenced species. The $20{\times}$ and $50{\times}$ coverage of the estimated transcriptome sequences using Roche/454 and Illumina/Solexa, respectively, is effective to create novel expressed reference sequences. However, only an average $30{\times}$ coverage of a transcriptome with short read sequences of Illumina/Solexa is enough to check expression quantification, compared to the reference expressed sequence tag sequence.

This study aims to explore a new technique making complementary linkage between controlled vocabularies and uncontrolled vocabularies for analyzing a research domain. Co-word analysis can be largely divided into two based on the types of vocabulary used: controlled and uncontrolled. In the case of using controlled vocabulary, data sparseness and indexer effect are inherent drawbacks. On the other case, word selection by the author's perspective and word ambiguity. To complement each other, we suggest a descriptor profiling that represents descriptors(controlled vocabulary) as the co-occurrence with words from the text(uncontrolled vocabulary). Applying the profiling to the domain of information science implies that this method can complement each other by reducing the inherent shortcoming of the controlled and uncontrolled vocabulary.
Misidentification of cultured cell lines results in the generation of erroneous scientific data. Hence, it is very important to identify and eliminate cell lines with a different origin from that being claimed. Various methods, such as karyotyping and isozyme analysis, can be used to detect inter-species misidentification. However, these methods have proved of little value for identifying intra-species misidentification, and it will only be through the development and application of molecular biological approaches that this will become practical. Recently, the profiling of microsatellite variants has been validated as a means of detecting gene polymorphisms and has proved to be a simple and reliable method for identifying individual cell lines. Currently, the human cell lines provided by cell banks around the world are routinely authenticated by microsatellite polymorphism profiling. Unfortunately, this practice has not been widely adopted for mouse cells lines. Here we show that the profiling of microsatellite variants can be also applied to distinguish the commonly used mouse inbred strains and to determine the strain of origin of cultured cell lines. We found that approximately 4.2% of mouse cell lines have been misidentified; this is a similar rate of misidentification as detected in human cell lines. Although this approach cannot detect intra-strain misidentification, the profiling of microsatellite variants should be routinely carried out for all mouse cell lines to eliminate inter-strain misidentification.

Compared to conventional high resolution acoustic profiling, ultra high resolution shallow acoustic profiling using parametric echo sounder is limited in penetration, yet it provides resolution suitable for detailed seabed investigation in the shallow waters. The parametric sub-bottom profiler system provides not only the exact determination of water depth, but also the detailed information about sediment layers and sub-bottom structures. Possible applications include dredging project, search of buried pipeline, ship wrecks, and other artificial objects through the detailed mapping of thickness and structure of the upper sedimentary layers. In this study, contaminated sediments were discriminated by the correlation of ultra high resolution profiles with geologic data. In addition, the burial depth of the submarine cable was measured by the interpretation of acoustic anomalies in the profiles.

This study is to establish the varietal discrimination based on DNA profiling of different varieties of rice. We examined the genetic distance among Korean rice varieties using allele frequencies and a genetic diversity analysis with Simple Sequence Repeats (SSRs) markers. The analysis of the genetic diversity and genetic relationships of 243 Korean rice varieties was varied out using 20 SSRs markers. A total of 268 alleles were detected, ranging from 6 to 32, with an average of 13.45 alleles per locus, and and average of gene diversity (GD) of 0.556. Seven SSR markers were selected as key markers for discrimination among the Korean rice varieties. Concerning the results, 243 varieties (100%) were discriminated among by using acrylamide gel and fragment analyzer-based markers. In conclusion, this study provides useful basic data that can be utilized concerning Korean rice varieties breeding and development. In addition, we will have to manage and conserve as a valuable genetic resource, without losing the diversity of Korean rice varieties.
