An RNA Mapping Strategy to Identify Ribozyme-Accessible Sites on the Catalytic Subunit of Mouse Telomerase

  • Telomerase reverse transcriptase (TERT) is an enzymatic ribonucleoprotein that prolongs the replicative life span of cells by maintaining protective structures at the ends of eukaryotic chromosomes. Telomerase activity is highly up-regulated in 85-90% of human cancers, and is predominately regulated by hTERT expression. In contrast, most normal somatic tissues in humans express low or undetectable levels of telomerase activity. This expression profile identifies TERT as a potential anticancer target. By using an RNA mapping strategy based on a trans-splicing ribozyme library, we identified the regions of mouse TERT (mTERT) RNA that were accessible to ribozymes. We found that particularly accessible sites were present downstream of the AUG start codon. This mTERTspecific ribozyme will be useful for validation of the RNA replacement as cancer gene therapy approach in mouse model with syngeneic tumors.

Accurate Metabolic Flux Analysis through Data Reconciliation of Isotope Balance-Based Data

  • Various techniques and strategies have been developed for the identification of intracellular metabolic conditions, and among them, isotope balance-based flux analysis with gas chromatography/mass spectrometry (GC/ MS) has recently become popular. Even though isotope balance-based flux analysis allows a more accurate estimation of intracellular fluxes, its application has been restricted to relatively small metabolic systems because of the limited number of measurable metabolites. In this paper, a strategy for incorporating isotope balance-based flux data obtained for a small network into metabolic flux analysis was examined as a feasible alternative allowing more accurate quantification of intracellular flux distribution in a large metabolic system. To impose GC/MS based data into a large metabolic network and obtain optimum flux distribution profile, data reconciliation procedure was applied. As a result, metabolic flux values of 308 intracellular reactions could be estimated from 29 GC/ MS based fluxes with higher accuracy.

The Future of Republic of Korea Navy : Toward a Korean Way of Naval Strategy (미래 대한민국 해군력 역할과 발전)

  • This study is an attempt to look into the future role of the ROKN and to provide a strategic way forward with a special focus on naval strategic concept and force planning. To accomplish this goal, this research takes four sequential steps for analysis: 1) assessing the role and utility of naval power of ROKN since its foundation back in 1945; 2) forecasting features of various maritime threats to influence the security of Korea in the future directly or indirectly; 3) identifying the roles to be undertaken by future ROKN; and 4) recommending Korean way of naval force planning and the operational concept of naval power. This study seeks to show that ROKN needs comprehensive role to better serve the nation with respect to national security, national prosperity and development, and future battle-space management. To safeguard the national security of Korea, it suggests three roles: 1) national guard for the peaceful unification; 2) protector of the maritime sovereignty; and 3) suppressor to maritime threats. Three more roles are highlighted for national prosperity: 1) escort of the national economy; 2) guardian for national maritime activities; and 3) contributor to the world peace. These roles need to be closely connected with the role for the battle-space management. This paper addresses the need for a dramatic shift of the central operational domain from land to maritime in the future. This will eventually offer future ROKN a leading role for developing strategic concept and force planning rather than merely a supporting one. This study finally suggests 'balanced' strategy both in concept development and force planning. A balanced force planning is a 'must' rather than an 'option' when considering a division of function between Task Fleets and Area Fleets, constructing cutting-edge conventional forces such as Aegis destroyer, CVs, or submarines, and the mix of high-profile platform and low-profile when composing future fleets. A 'balance' is also needed in operational concept. The fleet should be prepared to fulfill its missions based on two different types of force operation i.e., coercive or cooperative application of the utility of naval force. The findings and recommendations of the study are relevant today, and will be increasingly important in the future to achieve various political goals required by enhancing the utility of naval power.

Profiling Approach for the Choice between Speculation and Postponement Strategy in Supply Chain Management (공급사슬관리의 예측전략과 지연전략 선택을 위한 프로파일링 접근법)

  • Purpose - The postponement strategy, which delays the form, place, and production of products as late as possible, has been widely considered as a competitive supply chain management scheme in an era of mass customization and modular manufacturing. An interesting business phenomenon is that not all manufacturing/logistics firms choose the postponement strategy. Given that postponement is a counter-measure to speculation, which has some advantages under certain environments, the current imprudent inclination toward the postponement strategy may cause firms to lose the potential of the speculation strategy, an alternative strategy in supply chain management. Building on the logistics and manufacturing literature, this study examines characteristics of two contrasting strategies, postponement and speculation, and major factors favoring each strategy. Research design, data, and methodology - We apply the profiling approach to two business cases, HP printer and LG mobile phone. The profiling approach is a method of choosing a particular strategy aligned with environmental factors. While various approaches have been used to check the fit between a business strategy and environmental factors, the literature on manufacturing strategy and logistics has commonly adopted the profiling approach. Major factors used in profiling variables are derived from the literature. Two samples, HP printer and LG mobile phone, are selected, because they represent major characteristics appropriate for each strategy. The profiling is based on data from semi-organized interviews with managers. Results - The profiling approach shows that the postponement strategy is a suitable one for HP printers. Most factors, such as product life cycle, large production volume, low-price, product value, and monetary density, support delaying end products until as late as possible. Despite some exceptions, such as delivery time and economy of scale, our analysis states that the overall profile of HP printer is favorable for the postponement strategy. On the other hand, LG mobile phone may adapt the speculation strategy. Although it has large production volume and low delivery frequency, most characteristics support the speculation strategy for this product. An interesting finding is that, despite common perception that advanced technology products such as mobile telephones favor the postponement strategy, profiling proposes the speculation strategy for this product. Conclusions - Our analysis shows that speculation is not the universal option for supply chain management, and that, when choosing a specific strategy, one should consider many factors simultaneously. A major implication of our work is to emphasize the role of environmental factors such as supply chain variables in choosing an inventory strategy, and the importance of fit rather than solely strategic orientation. A theoretical contribution is to demonstrate the benefit of the simultaneous consideration of business variables in choosing specific strategies. For practitioners, our work leads us to consider the existence and the potential of speculation as a counter-measure to postponement. In addition, the comprehensive framework in this research may be instantly used in examining a practical strategy.

  • Purpose - It is chaotic when doing a business without legal patterns and rules; individuals who make legal decisions without legal consideration are often protecting their interests and forgetting others. This study aims to suggest key solutions how companies can make better decision based on legal considerations through investigation comprehensive literature analysis. Research design, Data, and methodology - This research conducted qualitative textual method and a technique called 'Qualitative Comparative Analysis' (QCA) can be used to understand better why certain things change while others do not. In tough situations, QCA is a strategy for comparing several occurrences. Result - Total six considerations were founded by the QCA for better corporate decision. Based on these considerations, all stakeholders, shareholders, and every employee should nominate and vote on one person to be their leader in the organization; fair practices in choosing the governor of the organization through legal binding will bring peace and order to the company. Conclusion - It was time consuming to go through every detailed material that entails legal consideration in making corporate decision. The concept of same profile in the research is critical whereby many authors are using same concept to write their articles and books. Using pure concept from one source limits the research and gives inadequate information.

  • Product recommender system is one of the most popular techniques for customer relationship management. In addition, collaborative filtering (CF) has been known to be one of the most successful recommendation techniques in product recommender systems. However, CF has some limitations such as sparsity and scalability problems. This study proposes hybrid cluster analysis and case-based reasoning (CBR) to address these problems. CBR may relieve the sparsity problem because it recommends products using customer profile and transaction data, but it may still give rise to scalability problem. Thus, this study uses cluster analysis to reduce search space prior to CBR for scalability Problem. For cluster analysis, this study employs hybrid genetic and K-Means algorithms to avoid possibility of convergence in local minima of typical cluster analyses. This study also develops a Web-based prototype system to test the superiority of the proposed model.

The Correlation of Sensory Processing Type, Learning Styles and Learning Strategies for University Students (대학생의 감각처리 유형과 학습유형, 학습전략의 상관관계)

  • Objective : The purpose of this study is to investigate correlation of sensory processing patterns, learning styles and learning strategies for university students. Methods : Participants of this study are 115 students from K university in Busan, South Korea. Measurements are Adolescent/Adult Sensory Profile (AASP) for sensory processing patterns, the Study Process Questionnaire (SPQ) for learning styles, and the Motivated Strategies for Learning Questionnaire (MSLQ) for learning strategies. The data collected was analyzed by SPSS/WIN 20.0 for chisuare test and Pearson corelation coefficient. Results : For sensory processing patterns and learning styles, there were correlation between low registration type and surface type of learning (p=0.03), and between sensory seeking type and deep type of learning (p=0.02). For sensory processing patterns and learning strategies, sensory seeking type was correlated with organized learning strategy (p=0.00), and sensory sensitivity type was correlated with organizational learning strategy (p=0.03) and meta-cognitive learning strategy (p=0.00). Conclusion : This study found that there is correlation between sensory processing patterns, learning styles and learning strategies with implying learning styles and learning strategies can be different depends on sensory procession pattern. The results of this study can be used as a basic data to select learning type and learning strategy appropriate for an individual based on his or her sensory processing patterns.

  • Snowboarding has become one of the premier alpine sports. The past decade has seen the popularity of snowboarding increase dramatically and the recent Winter Olympic Game showcased the strong visual appeal of the sport and the youth-oriented lifestyle and culture that accompanies it. The injury profile of the sport has also undergone change along with technological advances in boot and binding systems and the changing demographics of the sports participants. Central to the development of injury-prevention strategies is knowledge of the profile of injuries that occur, understanding those who are at particular risk and, if possible, the biomechanical factors involved in each injury type. Snowboarding was initially considered a dangerous, uncontrolled, alpine sport - an opinion based on little or no scientific evidence. That evidence has rapidly grown over the past decade and we now know that snowboard injury rates are no different to those in skiing; however, the injury profile is different. The purpose of this review is to give some perspective to the current snowboard injury literature. It discusses not only the demographic profile of those injured and the type of injuries that occur, but also gives some insight into the progress that has occurred in determining the impact of specific prevention strategies, such as splints to prevent injuries to the wrist/forearm. As the literature indicates, however, some things will not change, e.g. injuries are more likely to occur in beginners and lessons need to be reinforced as a fundamental aspect of any injury-prevention strategy.

Lifetime Assignment Schemes for Dynamic Binding Update in Mobile IPv6 (Mobile IPv6 환경에서 동적 바인딩 갱신을 위한 라이프타임 할당 기법)

  • Frequent occurrence of binding update messages may incur high overhead in Mobile IP supporting users mobility. Thus, it needs to develop algorithms to deal with the situation. In this paper, we propose new lifetime assignment schemes for dynamic binding update considering the locality property related with mobile node's movement. Each mobile node maintains a profile which is based on log containing useful information about its visiting subnets. That is, it determines dynamic binding update lifetime for currently visiting subnet by computing past mean resident time recorded in the profile. In addition, we note that the resident time depends on the time when each node enters a subnet and thus, we devise another lifetime assignment algorithm. Extensive experiments are made to compare our schemes with existing Mobile IPv6 where major facts for performance comparison are both the number of binding update messages and the number of binding request messages. From the results, we come to know that our schemes obtain highly considerable performance improvements in terms of communication cost by decreasing the number of those messages.

A Function-characteristic Aware Thread-mapping Strategy for an SEDA-based Message Processor in Multi-core Environments (멀티코어 환경에서 SEDA 기반 메시지 처리기의 수행함수 특성을 고려한 쓰레드 매핑 기법)

  • A message processor is server software that receives various message formats from clients, creates the corresponding threads to process them, and lastly delivers the results to the destination. Considering that each function of an SEDA-based message processor has its own characteristics such as CPU-bound or IO-bound, this paper proposes a thread-mapping strategy called "FC-TM" (function-characteristic aware thread mapping) that schedules the threads to the cores based on the function characteristics in multi-core environments. This paper assumes that message-processor functions are static in the sense that they are pre-defined when the message processor is built; therefore, we profile each function in advance and map each thread to a core using the information in order to maximize the throughput. The benchmarking results show that the throughput increased by up to a maximum of 72 % compared with the previous studies when the ratio of the IO-bound functions to the CPU-bound functions exceeds a certain percentage.
