Drug Polymorphism and its Importance on Drug Development Process

  • Polymorphism has been recognized to be a critical issue throughout the drug product development process. Most of solid phase drugs have polymorphism, which has generated a great deal of interest and the field has been evolving rapidly. Preferably, thermodynamically most stable form of a drug substance is selected to obtain consistent bioavailability over its shelf life and various storage conditions. Moreover, it has the lowest potential for conversion from one polymorphic form to another. However, metastable or amorphous forms may be used intentionally to induce faster dissolution rate for rapid drug absorption and higher efficacy. For pharmaceutical industry, polymorphism is one of the key activities in form selection process together with salt selection. This article introduces the main features in the investigation of solid form selection especially polymorphic behavior with thermodynamic backgrounds, physicochemical properties with solubility, dissolution, and mechanical properties, and characterization techniques for proper analysis. The final form can be recommended based on the physicochemical and biopharmaceutical properties and by the processability, scalability and safety considerations. Pharmaceutical scientists especially in charge of formulation need to be well aware of the above issues to assure product quality.

  • Decision frameworks for product safty policy are developed in theory and practice. Product characteristic approach and expected utility analysis are applied to situations involving risk and misinformation. Eight types of regulatory frameworks are explained and critiqued form practical purposes on behalf of consumer policy makers. Various international organizations and their roles are briefly reviewed.

  • In this paper, we obtain the general solution and the stability of the 2-dimensional vector variable quadratic functional equation f( x + y, z - w) + f(x - y, z + w) = 2f(x, z ) + 2f(y, ${\omega}$). The quadratic form f( x, y) = $ax^2$ + $by^2$ without cross product terms is a solution of the above functional equation.

  • In this paper, we construct a Sasakian manifold by the product of real line and Kählerian manifold with exact Kähler form. This result demonstrates the close relation between Sasakian and Kählerian manifold with exact Kähler form. We present an example and an open problem.

일반적인 큐잉네트워크에서의 체류시간분포의 근사화 (An approximation method for sojourn time distributions in general queueing netowkrs)

  • Even though sojourn time distributions are essential information in analyzing queueing networks, there are few methods to compute them accurately in non-product form queueing networks. In this study, we model the location process of a typical customer as a GMPH semi-Markov chain and develop computationally useful formula for the transition function and the first-passage time distribution in the GMPH semi-Markov chain. We use the formula to develop an effcient method for approximating sojourn time distributions in the non-product form queueing networks under quite general situation. We demonstrate its validity through numerical examples.

  • Functional form and dysfunctional form of Kano model are considered as customer need regarding attribute of product. Both functional and dysfunctional forms are: Like, Must-be Neutral, Live-with and Dislike. The answers of customer regarding a product of functional and dysfunctional forms have been applied for selection of customer needs regarding product attribute (Kano evaluation). Filling.up and returning the Questionnaires by the individuals are essential for determining Kano evaluation. But many Questionnaires have not been returned in that case. Moreover, many possible consumers could not get opportunity to fill-up questionnaire. These uncertain or unknown consumers' opinions are also essential for product development. The choices of Kano evaluations have been outlined by: Attractive, One-dimensional, Must-be, Indifferent and Reverse. In this study, choices of evaluation of unknown customer are considered uniform cumulative vector probability (scenario 1). This study is based on the Monte Carlo simulation method, concept of probability and Kano model. This model has also been tested for its soundness and found fairly consistent including existing Kano model (scenario 2) and case survey for headlight of bicycle (scenario 3).

  • This paper presents a conceptual approach to convert customer requirements expressed in ordinary language into a form of qualitative and quantitative functions for developing new products. This approach attempts to combine the concepts of the value engineering and the Kansei engineering. It emphasizes that customer require- ments should be interpreted and reflected on the design of new product. Specific are discussed for extracting subjective requirements and transforming them into qualitative and quantitative functions for product design. This approach is expected to provide the product designer with a systematic efficient tool for incorporating subjective requirements into a product design.

  • This study presents an 𝛼-Sasakian structure on the product manifold M1 × 𝛽(I), where M1 is a Kähler manifold with an exact 1-form, and 𝛽(I) is an open curve. It then defines a new type warped product metric to study the warped product of almost Hermitian manifolds with almost contact metric manifolds, contact metric manifolds, and K-contact manifolds.
