Anti-inflammatory Activities of Cheongpyehwadam-tang

  • In oriental medicine, Cheongpyehwadam-tang (CHT) has long been used for the cure of inflammatory diseases in the lung and bronchus such as bronchitis, bronchial asthma, pneumonia and tuberculosis. It's use is currently further extended for the treatment of allergic asthma. To investigate the anti-inflammatory effects of CHT, we investigated the effects of CHT on the lipopolysaccharide (LPS)-induced nitric oxide (NO) and pro-inflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$) production, and on the level of inducible nitric oxide synthase (iNOS) and proinflammatory cytokines expression in murine macrophage RAW 264.7 cells. CHT alone did not affect NO or pro-inflammatory cytokines production. In contrast, CHT inhibited LPS-induced NO and proinflammatory cytokines and the levels of LPS-induced iNOS and proinflarnmatory cytokine mRNA in a dose-dependent manner. CHT also inhibited the nuclear factor-kappa B (NF-kB) activation. Taken together, these results suggested that CHT inhibits the production of NO and pro-inflammatory cytokines in RAW 264.7 cells through blockade of NF-kB activation.

Inhibitory Effects of Eutigosides Isolated from Eurya emarginata on the Inflammatory Mediators in RAW264.7 Cells

  • The anti-inflammatory activity of Eurya emarginata (Thumb) Makino, of which leaves have been traditionally used to treat ulcers or diuretic in Jeju Island, has been investigated in the present study. Through the phytochemical study from the methanol extract of E. emaginiata, eutigosides Band C were isolated as the active components. Sseveral inflammatory markers including TNF-$\alpha$, IL-1$\beta$, IL-6, NO, iNOS, and COX-2 were examined. Eutigosides Band C potentially inhibited production of pro-inflammatory cytokines (IL-6 and TNF-$\alpha$) in a dose-dependent manner. Additionally, the intracellular contents of iNOS protein were markedly decreased after treatment with eutigosides Band C. The inhibition of iNOS activity was correlated with the decrease in nitrite levels. These results suggest that eutigoside Band C from E. emarginata may have anti-inflammatory activity through the inhibition of pro-inflammatory cytokines (TNF-$\alpha$ and IL-6), iNOS and COX-2.

A Comparison of the Anti-inflammatory Activity of Surfactin A, B, C, and D from Bacillus subtilis

  • Natural surfactins are a mixture of isoforms that differ slightly in their physiological properties. In previous research, we obtained surfactin A, B, C, and D from the Bacillus subtilis complex BC1212. We found that surfactin C inhibited nitric oxide (NO)-production and suppressed the expression of pro-inflammatory cytokine mRNA, which was stimulated by $1{\mu}g/ml$ of lipopolysaccharide (LPS) in murine RAW264.7 cells. In order to compare the anti-inflammatory effects of surf actin isoforms, we examined the inhibition of LPS-induced NO production and the pro-inflammatory cytokine expression level. Surfactin C inhibited the LPS-induced NO production in murine macrophage RAW264.7 cells the most. In addition, surf actin C was superior to other surfactin's subtypes regarding inhibiting the expression of inducible nitric oxide synthase (iNOS) and monocyte chemoattractant protein 1 (MCP-1). Finally, the anti-inflammatory activity of surf actin C is the most potent, compared with surfactin A, B, and D.

Anti-Inflammatory Effect of Extracts from Ligustrum obtusifolium S. fruits in RAW 264.7 Macrophages (RAW 264.7 대식세포 내에서 남정목 열매 추출물의 항염증 효과)

  • Objectives This study was designed to investigate the anti-inflammatory effect of extracts from Ligustrum obtusifolium S. fruits(LOF) in RAW 264.7 Macrophages stimulated with lipopolysaccharide(LPS). Methods We examined productions of nitric oxide(NO), reactive oxygen species(ROS), inducible isoforms of NO synthase(iNOS), cyclooxygenase-2(COX-2) to investigate the anti-inflammatory effect of LOF extracts. In addition, we measured generation of pro-inflammatory cytokines(TNF-${\alpha}$, IL-6). Results Cell viability showed that LOF extracts had no cytotoxicity in Raw 264.7 cells. The treatment with LOF extracts significantly decreased the generation of NO and pro-inflammatory cytokines(TNF-${\alpha}$, IL-6) in LPS-stimulated macrophage cells. Furthermore LOF extracts inhibited intracellular ROS generation dose dependently and reduced the expression of iNOS, COX-2 proteins. Conclusions These results showed that the LOF extracts had an anti-inflammatory effect on LPS-stimulated Raw 264.7 cells. These findings provide scientific support for the use of this Ligustrum obtusifolium S. for inflammatory-related diseases.

Floridoside suppresses pro-inflammatory responses by blocking MAPK signaling in activated microglia

  • Inflammatory conditions mediated by activated microglia lead to chronic neuro-degenerative diseases such as Alzheimer's, Parkinson's, and Huntington's diseases. This study was conducted to determine the effect of floridoside isolated from marine red algae Laurencia undulata on LPS (100 ng/ml) activated inflammatory responses in BV-2 microglia cells. The results show that floridoside has the ability to suppress pro-inflammatory responses in microglia by markedly inhibiting the production of nitric oxide (NO) and reactive oxygen species (ROS). Moreover, floridoside down-regulated the protein and gene expression levels of iNOS and COX-2 by significantly blocking the phosphorylation of p38 and ERK in BV-2 cells. Collectively, these results indicate that floridoside has the potential to be developed as an active agent for the treatment of neuro-inflammation.

  • Chrysin, a natural flavone compound contained in plants. has anti-inflammatory activity. Its anti-inflammatory effect has been previously explained in part by the suppression of promoter activities of inducible pro-inflammatory enzymes (cyclooxygenase-2 (COX-2) and inducible nitrogen synthase (iNOS)). Nitrate production triggered by the activation of lipopolysaccharides (LPS) was most highly suppressed by the treatment of chrysin, follwed by 5-hydroxy-7-methoxyflavone (Ch-2), 5,7-diacetylflavone (Ch-4) in cultured Raw 264.7 cells.   • Toona sinensis (TS) leaf is known to antinociceptive, antioxidative stress and skin moisturizing effects. Acnes vulgaris is a chronic skin disease with various symptoms including itchiness, pain and interruption of normal skin function. Propionibacterium acnes (P. acnes) is a major factor in the occurrence of inflammatory acnes. This study evaluated the antioxidant and anti-inflammation effects by TS extract from adventitious shoots. TS extract showed anti-inflammatory activities by suppression of pro-inflammation mediators (iNOS and COX-2) in LPS-stimulated RAW264.7 cells. TS extract also has anti-inflammatory activities by inhibiting the secretion of pro-inflammatory cytokines on P. acnes-stimulated HaCaT cells. These effects were regulated by MAPK signaling pathway. Therefore, we suggest that TS extract from adventitious shoots might have applications as a medicine for treating P. acnes-induced skin diseases.

  • This study describes a preliminary evaluation of the anti-inflammatory activity of Castanopsis cuspidata extracts. C. cuspidata was extracted using 80% ethanol and then fractionated sequentially with n-hexane, dichloromethane, ethylacetate, and butanol. To screen for anti-inflammatory agents effectively, we first examined the inhibitory effect of the C. cuspidata extracts on the production of pro-inflammatory factors and cytokines stimulated with lipopolysaccharide. In addition, we examined the inhibitory effect of C. cuspidata extracts on pro-inflammatory mediators (NO, iNOS, COX-2) in murine macrophage RAW 264.7 cells. The amounts of protein levels were determined by immunoblotting. Of the sequential solvent fractions of C. cuspidata, the n-hexane, dichloromethane and ethylacetate fractions inhibited the mRNA expression of pro-inflammatory cytokines (IL-$1{\beta}$ and IL-6), production of NO, and the protein level of iNOS and COX-2. These results suggest that C. cuspidata may have significant effects on inflammatory factors and may be provided as a possible anti-inflammatory therapeutic plant.

Suppression of Lipopolysaccharide-Induced Inflammatory and Oxidative Response by 5-Aminolevulinic Acid in RAW 264.7 Macrophages and Zebrafish Larvae

  • In this study, we investigated the inhibitory effect of 5-aminolevulinic acid (ALA), a heme precursor, on inflammatory and oxidative stress activated by lipopolysaccharide (LPS) in RAW 264.7 macrophages by estimating nitric oxide (NO), prostaglandin E2 (PGE2), cytokines, and reactive oxygen species (ROS). We also evaluated the molecular mechanisms through analysis of the expression of their regulatory genes, and further evaluated the anti-inflammatory and antioxidant efficacy of ALA against LPS in the zebrafish model. Our results indicated that ALA treatment significantly attenuated the LPS-induced release of pro-inflammatory mediators including NO and PGE2, which was associated with decreased inducible NO synthase and cyclooxygenase-2 expression. ALA also inhibited the LPS-induced expression of pro-inflammatory cytokines, such as tumor necrosis factor (TNF)-α, interleukin (IL)-1β, and IL-6, reducing their extracellular secretion. Additionally, ALA abolished ROS generation, improved the mitochondrial mass, and enhanced the expression of heme oxygenase-1 (HO-1) and the activation of nuclear translocation of nuclear factor-E2-related factor 2 (Nrf2) in LPS-stimulated RAW 264.7 macrophages. However, zinc protoporphyrin, a specific inhibitor of HO-1, reversed the ALA-mediated inhibition of pro-inflammatory cytokines production and activation of mitochondrial function in LPS-treated RAW 264.7 macrophages. Furthermore, ALA significantly abolished the expression of LPS-induced pro-inflammatory mediators and cytokines, and showed strong protective effects against NO and ROS production in zebrafish larvae. In conclusion, our findings suggest that ALA exerts LPS-induced anti-inflammatory and antioxidant effects by upregulating the Nrf2/HO-1 signaling pathway, and that ALA can be a potential functional agent to prevent inflammatory and oxidative damage.

Characteristics of immunomodulation by a Lactobacillus sakei proBio65 isolated from Kimchi (김치에서 분리한 Lactobacillus sakei proBio65의 면역 조절 특성)

  • We isolated and identified a novel probiotic strain, Lactobacillus sakei proBio65 from Kimchi. To determine whether L. sakei proBio65 has an immunomodulatory effect, we investigated cells via an in vitro screening system which co-cultured freshly isolated mesenteric lymphocyte with probiotics. A significant increase of $Foxp3^+$ transcription regulatory factor expression was observed, followed by an increase in anti-inflammatory cytokines transcription regulatory factor. L. sakei proBio65 exhibited high levels of the IL-10/IL-12 production ratio and enhanced Foxp3 expression in vitro. L. sakei proBio65 may thus be therapeutically useful for the modulation of inflammatory immune disorders.
