Effect of Prepriming on Improving Germinability of Pelleted Carrot Seeds (Priming에 의한 당근 Pelleting 종자의 발아성 향상)

  • Pelleting the seeds often reduces seed germinability. In an effort to alleviate this problem, the seeds of four carrot cultivars were, before pelleting, either solid-matrix primed (SMP) with Micro Cel-E or osmotically-primed with polyehtylene glycol 8000. Some batches were imbibed in 100-ppm $GA_3$ solution. In laboratory tests, pretreatment of the seeds increased the precentages of germination by as much as 50% of the controls, indicating the increase of the germination speed by 1.9 to 4.6 days as determined by the $T_{50}$ values. In field tests conducted in the year of 2000 and 2001, however, effects of SMP were not observed. There was no significant difference between the SMP seeds and the controls of the percentages and $T_{50}$ values. The emergence of the seedling in the field was affected greatly by soil water concentration.

Effect of chlorhexidine application on the bond strength of resin core to axial dentin in endodontic cavity

  • Objectives: This study evaluated the influence of chlorhexidine (CHX) on the microtensile bonds strength (${\mu}TBS$) of resin core with two adhesive systems to dentin in endodontic cavities. Materials and Methods: Flat dentinal surfaces in 40 molar endodontic cavities were treated with self-etch adhesive system, Contax (DMG) and total-etch adhesive system, Adper Single Bond 2 (3M ESPE) after the following surface treatments: (1) Priming only (Contax), (2) CHX for 15 sec + rinsing + priming (Contax), (3) Etching with priming (Adper Single Bond 2), (4) Etching + CHX for 15 sec + rinsing + priming (Adper Single Bond 2). Resin composite build-ups were made with LuxaCore (DMG) using a bulk method and polymerized for 40 sec. For each condition, half of specimens were submitted to ${\mu}TBS$ after 24 hr storage and half of them were submitted to thermocycling of 10,000 cycles between $5^{\circ}C$ and $55^{\circ}C$ before testing. The data were analyzed using ANOVA and independent t-test at a significance level of 95%. Results: CHX pre-treatment did not affect the bond strength of specimens tested at the immediate testing period, regardless of dentin surface treatments. However, after 10,000 thermocycling, all groups showed reduced bond strength. The amount of reduction was greater in groups without CHX treatments than groups with CHX treatment. These characteristics were the same in both self-etch adhesive system and total-etch adhesive system. Conclusions: 2% CHX application for 15 sec proved to alleviate the decrease of bond strength of dentin bonding systems. No significant difference was shown in ${\mu}TBS$ between total-etching system and self-etching system.

Effect of Priming on Seed Germination and Seedling Growth of Lilium formolongi (Priming 처리(處理)가 신나팔나리의 종자발아(種子發芽) 및 초기생육(初期生育)에 미치는 영향(影響))

  • This experiment was conducted to improve germination rate and growth of Lilium formolongi 'Neosan No. 2' seeds by priming. Seeds were immersed in the solutions of 200 mM $Ca(NO_3)_2$, 200 mM $KNO_3$ for 3 days and 100 mg/L $GA_3$ for 1 day at $17{\pm}1^{\circ}C$. For chilling treatment, seeds were kept in the low temperature chamber of $2{\pm}1^{\circ}C$ for 30 days. Germination percentage of seeds treated with $Ca(NO_3)_2$ or $KNO_3$ was higher than that of seeds with other treatments and control seeds to 10 days after seeding, but final germination percentage was not significantly different among all treatments including control. Early growth of seedlings treated with $Ca(NO_3)_2$ and $KNO_3$ were better than that of seedling with other treatments.

Effect of Presowing Seed Treatments on Germination and Seedling Emergence in Taraxacum platycarpum (민들레 종자의 파종전 처리가 발아율 및 입모율에 미치는 영향)

  • Several presowing seed treatments have been used to facilitate the seed germination and seedling emergence. The study was performed to design a model of presowing seed treatments in Taraxacum platycarpum. Germination and seedling growth responses to temperature, drying, chilling, $GA_3$ and priming were investigated to know the optimum germination conditions and presowing treatment. The optimum temperature condition for germination and seedling growth were $15{\sim}20^{\circ}C$ at dark condition. The seeds exposed to infra red light, however, did not germinate. The moisture content of seeds dried at $35^{\circ}C$ for 3.5 hours was returned to the level of unimbibed seeds. The germination of seeds dried under dark condition was better than those dried under infra red light. Prechilling treatment imposed for 6 weeks showed higher germination rate than $GA_3$ treatment or priming. In field test, the highest field emergence was observed in the seeds dried after prechilled for 6 weeks and followed by drying followed by $GA_3$ > low temperature > $KNO_3$ priming. Imbibition with 0.1 mM $GA_3$ solution for 2 hours then drying at 35 for 3.5 hour under dark condition was effective to the practical presowing treatment for Taraxacum seeds.

Priming Effect of Endotoxin in Human Alveolar Macrophage (사람 폐포대식세포에서 내독소의 Priming 효과)

  • Background: Endotoxin or lipopolysaccharide(LPS) can prime phagocytic cells such as polymorphonuclear leukocytes, monocytes or animal peritoneal macrophages to generate increased amounts of secretory products such as oxygen free radicals and tumor necrosis factor, which play an important role in developing adult respiratory distress syndrome in gram negative sepsis. Human alveolar macrophages(HAM) are continuously exposed to various stimuli inhaled into the alveoli, and the response to LPS might be different in HAM. Therefore, we investigated the effect of LPS pre-exposure on HAM adhered to plastic surface and A549 cell(type II human alveolar epithelial cell line) monolayer. Methods: HAM were isolated from bronchoalveolar lavage fluid from normal lung of the patients with localized lung cancer and esophageal cancer. LPS was exposed to HAM for 2hrs before or after adherence to plastic surface of 24-well Linbro plate and A549 cell monolayer. And then HAM was stimulated with PMA(phorbol myristate acetate) or fMLP(N-formyl-methionylleucyl-phenylalanine). The amount of hydrogen peroxide($H_2O_2$) production in the supernatant was measured on the principle of peroxidase-dependent oxidation of phenol red by hydrogen peroxide. Results: LPS pre-exposure could not enhance $H_2O_2$ production in neither HAM adhered to plastic surface nor one to A549 cell monolayer. But LPS even in the absence of PMA or fMLP stimulation directly increased $H_2O_2$ release in HAM if added after the adherence to A549 cell monolayer. Conclusion: Endotoxin does not prime HAM, but may directly activate HAM adhered to alveolar epithelial cells. Further investagation will be necessary.

The Effect of Incidental Semantic Activation on Hypothesis Generation: Exclusive vs Compatible Hypotheses (우연적 의미 활성화가 가설 생성에 미치는 영향: 가설 유형에 따른 차이)

  • Previous studies on the effect of incidental semantic priming on judgment, have focused mainly on mutually exclusive hypotheses. However, the present study explored whether incidental semantic activation affects diagnostic inference depending on the type of the hypothesis: mutually exclusive hypotheses vs compatible hypotheses. In Experiment 1, in case of mutually exclusive hypotheses, the final hypothesis was selected according to the incidental semantic priming, but there was no difference in the number of generated hypothesis in comparison with the control. However, for compatible hypotheses (i.e., both hypotheses can be true), the semantic priming affected the number of generated hypotheses, but not the selection of the final hypothesis. The same pattern of results was observed even when the cognitive load was increased. In Experiment 2, we found a boundary condition of incidental semantic activation on diagnostic inference. When cues related to each of the hypotheses were presented simultaneously, the incidental semantic effect disappeared. These results suggest that people consider all possible cues when making diagnostic inference in daily life. In light of these findings, further research on hypothesis generation/evaluation should take the type of hypothesis into account.

Effect of Controlled Hydration on Germination of Tobacco Seeds

  • Controlled hydration treatment of tobacco seeds enhanced seed performance greatly without additional materials associated with conventional osmotic or solid matrix priming technique. The seeds were hydrated by adding water to a level from 10 to 60% by 5% increments and incubated for 8 days at $25^{\circ}C$. After the treatment, the seeds were dried to the original seed moisture content under $20^{\circ}C$ for 72 hours. The moisture content of tobacco seeds equivalent to 35% by the hydration treatment gave the greatest improvement in germination rate and speed compared to untreated or polyethylene glycol (PEG) primed seeds, especially at low temperature of $15^{\circ}C$.
