Formulation and In vitro Evaluation of Transdermal Drug Delivery System for Galantamine

  • The effects of different formulation variables including pressure sensitive adhesive (PSA), permeation enhancer, thickness of the matrix and loading amount of drug on the transdermal absorption of galantamine were investigated across the hairless mouse skin. The permeation profile of galantamine was different depending on the types of PSA, loading amount of drug, thickness of the matrix and type of enhancer used. Highest flux of galantamine was obtained from acrylic PSA but crystals were formed in the patch within 72 h. Among the PSAs screened, crystal formation was not observed only in the patches formulated in Styrene Butadiene Styrene (SBS) matrix. Permeation rate increased linearly as the concentration of galantamine in SBS matrix increased from 2.5 to 15% w/w. Among the enhancers screened, Brij$^{(R)}$ 30 provided highest flux of galantamine. Matrix thickness of 80 ${\mu}m$ was optimum for maintaining adhesiveness as well as consistently delivering galantamine for longer period of time.

  • UV-curable pressure sensitive adhesives were prepared by blending acrylic copolymer, copolymerized with butyl acrylate (BA), acrylic acid (AA) and vinyl acetate (VAc) by solution polymerization, triethyl amine (TEA) and trimethylolpropane triacrylate (TMPTA). The PSAs were evaluated by peel strength with varying contents of TMPTA and UV dose, and also glass transition temperature($T_g$) of PSAs were measured. When exposed on UV irradiation, the PSAs showed the decreased peel strength and increased $T_g$. And following UV irradiation, the PSAs did not leave any residue on wafer after peel off PSA.

  • Most of the materials used in various industrial fields and also in our daily life are multi-component materials or composite materials, and it is well known that there are many cases where adhesion between the constituents within the bonded systems plays an important role. There are various types of performance evaluation tests for the bonded materials, among which tests for evaluating the bond performance under various conditions may be regarded as the most interesting ones for those engaged in work related to adhesion. I have studied on the mechanism of adhesion form the rheological standpoint with my colleagues, including some students from Korea, and I am very happy to be able to have a talk on some of our research works. In Japan, the so-called "adhesives" are usually classified into two categories;adhesives and pressure sensitive adhesives (PSA). Adhesives are the materials which solidify after bonding and are after used as the structural adhesives because the adhesive strength is comparatively strong. On the other hand, the pressure sensitive adhesives never solidify and are used as PSA tapes, labels or decals. About the adhesives, we have examined the dependence of adhesive strength(shear, tensile, peel) upon both temperature and rate of deformation, and found out some empirical rules which are applicable to most of the adhesive systems. We have also developed a simplified theory of adhesion, which is deseribed in terms of mechanical equivalent mode1 and a few failure criteria. Although some of the common rules can be accounted for according to this theory, it must be pointed out that a fracture mechanical approach ms inevitable especially in the region where the meehanical relaxation time of the adhesive is extremely large [W. W. Lim and H. Mizumachi]. About the pressure sensitive adhesives, we have studied on the PSA performance (peel, tack, holding power) as a function of both the viscoelastic properties and surface chemical properties of the materials, and found out some rules, and again we have developed a theory which deseribes the mechanism. And in addition, we have studied on the miscibility between linear polymers and oligomers, because PSA is generally manufactured by blending gums and tackifier resins. Many phase diagrams have been found and some of them have been analyzed on thermodynamic basis, and it became evident that the miscibility is a very important factor in PSA [H. J. Kin and H. Mizumachi]. In this presentation, I want to emphasize the fact that the adhesion performance is closely related to the structure/property of the adhesives.adhesives.

  • In making emulsion-type pressure sensitive adhesive (PSA), environmentally friendly sorbitan nonionic surfactants having various chemical structures were prepared and used to find out the possibility of substituting the traditional nonyl-phenyl nonionic surfactant (NPE40) by comparing their adhesive properties. Results exhibited that the PSA used NSR10-series and Tween 60 showed better adhesive properties in initial ball tack, peel strength and holding power than those of NPE40. The reason for this is their particular chemical structure and the proper ratio of hydrophobic groups to the ethylene oxide groups.

  • To increase peel strength, low temperature properties and flowability of pressure sensitive adhesives(PSA) used in a heat shrinkable sheet, these properties were evaluated by changing each components in type and content. In this study, Isobutylene-co-Isoprene Rubber (IIR) which has good wetherability was selected as a base polymer. Instead of rosin ester, petroleum resin was selected as a tackifier because of superior peel strength. By decreasing petroleum resin contents, flowability of PSA was decreased. High molecular weight of polybutene was better than low molecular weight for the peel strength of PSA. Large particle size of carbon black showed better properties than small one in peel strength and brittleness temperature. By adding calcium carbonate, the cost of compound was able to be reduced. But it must be used with carbon black.

  • The pressure-sensitive adhesive(PSA) tapes are widely used as floor-sealing material in Korean constructive industry. However, the general PSA tapes can not seal the apertures completely. This is one of main reasons to cause imperfect finishing. In order to solve the problems, the asphalts were modified by FKM powder and SBS in this study. The physical properties of modified asphalts were examined by measuring the softening point, viscosity, tensile strength and adhesion strength of FKM and SBS modified asphalt. The softening point of SBS modified asphalt was increased greatly due to the formation of network structure in asphalt. Also, the change of adhesion strength of SBS modified asphalt with temperature was not so great comparing to those of pure asphalt and FKM modified asphalt. The best results of pressure-sensitive adhesion could be obtained by the SBS(linear) modified asphalt containing 6 wt.% of linear type SBS.

  • Sibutramine is a serotonin-norepinephrine reuptake inhibitor indicated for the management of obesity in conjunction with a reduced calorie diet. The oral administration of sibutramine is followed by its dose-related side effects. In this study, sibutramine was formulated into drug in adhesive (DIA) patches in an attempt to overcome these problems. The effects of different formulation variables including pressure-sensitive adhesive (PSA), loading amount of drug, thickness of matrix and enhancer on the skin permeation of the drug were evaluated using excised hairless mouse skin. In the acrylic adhesive with carboxyl functional group, low release of sibutramine was observed due to the strong interaction between carboxyl group of adhesive and amine group of sibutramine. The acrylic adhesive without functional group provided good adhesion force and allowed high drug loading. Changing drug load as well as thickness of the matrix was found to alter permeation rate. $Crovol^{(R)}$ PK40 and $Crovol^{(R)}$ A40, were found to be effective enhancers for sibutramine. The optimized patch contained 20% sibutramine, and 5% $Crovol^{(R)}$ A40 as permeation enhancer, in $80\;{\mu}m$ thick Duro-$Tak^{(R)}$ 87-9301 matrix.

Adhesive and Removable Characteristics of UV Curable Adhesive (자외선 경화형 점착제의 접착 및 재박리 특성)

  • UV curable pressure sensitive adhesive (PSA) was made by blending the di-functional or hexa-functional urethane-acrylate oligomer with synthesized acrylic PSA. The change of adhesive property and removability were investigated at various oligomer contents under each oligomer. As the content of oligomer increased, the peel strength was increased before UV irradiation, but it was decreased after UV irradiation. Also, it was observed that the peel strength increased with decrease of storage modulus (G'), and that loss factor (tan ${\delta}$) of the adhesive material affected the adhesive properties and removability.

The Effects of Crosslinking Agent and pH Controlling Agent on Adhesive Properties of Water Soluble Acrylic PSA (가교제 및 pH 조절제가 수용성 아크릴 점착제의 점착 물성에 미치는 효과)

  • In this paper, the effects of crosslinking agent and pH controlling agent on adhesive properties and water solubility of water soluble acrylic pressure sensitive adhesives were studied by adding the crosslinking agents or pH controlling agents after the polymerization. the tack, cohesive strength, and peel strength were measured to evaluate adhesive properties. the turbidity was also measured to evaluate water solubility of pressure sensitive adhesives. When crosslinking agent was added less than 0.2 wt%, tack increased and cohesive strength and peel strength decreased with increasing concentration. When crossinking agent was added more than 0.2 wt%, the results were opposited. When pH controlling agent was added, tack and water solubility decreased and peel strength and cohesive strength increased with increasing concentration. the influence of pH controlling agents on adhesive properties and water solubility of pressure sensitive adhesive increased in the order of lithium hydroxide, potassium hydroxide and sodium hydroxide.

Peel Stength of the Acrylic Copolymer and Pressure Sensitive Adhesives (아크릴계 점착제의 박리강도와 점착부여제)

  • The stability and performance (peel strength) of the acrylic copolymer and various modified rosin systems were investigated. The peel strength was measured over a wide range of scaling rates, and the influence of the viscoelasticity of the PSA(pressure sensitive adhesive) was considered. In the case of miscible systems, the peak of peel strength (PSA performance) over wide peel rates was changed and modified systematically with increasing glass transition temperature of the blends. The peak of the peel strength for blended systems shifts toward the lower rate side as glass transition temperature ($T_g$) of the blend increased. The influence of esterification of the rosin on performance and stability against deterioration was greatly modified by blending with rosin of glycerol ester and rosin pentaerythritol ester. The failure mode of the blend varies with the combination with acrylic copolymer and modified rosin, and cohesive failure was found at a lower peel rate while interfacial failure was found at a high peel rate. A few systems where a single Tg could be measured, despite the fact that two phases were observed microscopically, were detected.
