BIPHASIC CULTURE STRATEGY BASED ON HYPEROSMOTIC PRESSURE FOR IMPROVED HUMANIZED ANTIBODY PRODUCTION IN CHINESE HAMSTER OVARY CELL CULTURE

  • Hyperosmotic pressure increased specific antibody productivity ($q_{Ab}$) of recombinant CHO cells (SH2-0.32) while it depressed cell growth. Thus, the use of hyperosmolar medium did not increase the maximum antibody concentration substantially. To overcome this drawback, the feasibility of biphasic culture strategy was investigated. In the biphasic culture, cells were first cultivated in the standard medium with physiological osmolality(294 mOsm/kg) for cell growth. When cells reached the late exponential phase of growth, the spent standard medium was replaced with the fresh hyperosmolar medium (522 mOsm/kg) for antibody production. The ($q_{Ab}$) in growth phase with the standard medium was 2.1 ${\mu}g/10^6cell/day$ while the ($q_{Ab}$) in antibody production phase with the hyperosmolar medium (522 mOsm/kg) was 11.1 ${\mu}g/10^6cell/day$. Northern blot analysis showed a positive relationship between the relative contenet of Ig mRNA and ($q_{Ab}$), indicating that transcriptional regulation was involved in the response of rCHO cells to hyperosmotic pressure. Due to the enhanced ($q_{Ab}$) and increased cell concentration in biphasic culture, the maximum antibody concentration obtained in biphasic culture with 522 mOsm/kg medium exchange was 161% higher than that obtained in batch culture with the standard medium. Taken together, simple biphasic culture strategy based on hyperosmotic culture for improved foreign protein production from rCHO cells is effective in improving antibody production of rCHO cells.

  • This paper presents a numerical analysis of the delivery pressure wave in a 210-kW/cyl fuel injection pump (P. Corporation, Changwon-si, Korea) for medium-speed diesel engines using Ansys Fluent R15.0. Results obtained from experiment and from numerical analysis of the fuel delivery pressure wave were compared and found to be similar, thereby confirming the reliability of the numerical analysis of the delivery pressure wave in the fuel injection pump.

  • A pressure wave created by the photoacoustic effect is affected by the medium and by laser parameters. The effect of these parameters on the generated pressure wave can be seen by solving the photoacoustic wave equation. These solutions which are examined in the time domain and the frequency domain should be considered by researchers in acoustic sensor design. In particular, frequency domain analysis contains significant information for designing the sensor. The most important part of this information is the determination of the operating frequency of the sensor. In this work, the laser parameters to excite the medium, and the acoustic signal parameters created by the medium are analyzed. For the first time, we have obtained solutions for situations which have no frequency domain solutions in the literature. The main focal point in this work is that the frequency domain solutions of the acoustic wave equation are performed and the effects of the frequency analysis of the related parameters are shown comparatively from the viewpoint of using them in acoustic sensor designs.

A Study of Bulk Modulus of Beryl Using Water as a Pressure-Transmitting Medium (물을 압력 매개체로 이용한 녹주석의 체적탄성률 연구)

  • In-situ high-pressure and ex-situ high temperature-pressure experiments of natural beryl ($Be_3Al_2Si_6O_{18}$, P6/mcc) from two different localities (beryl-A and beryl-B) were studied using pure water as pressure transmitting medium. Compared to the previous study using a mixture of methanol:ethanol medium in 4 : 1 by volume, pressure- and temperature-induced chemical and structural changes under water medium are expected to be different. The derived bulk moduli are 111(7) GPa, $K{_0}^{\prime}=73(7)$; 110(9) GPa, $K{_0}^{\prime}=65(8)$ for beryl-A and beryl-B, respectively. We observe densifications in volume compression, which appear to be attributed to the phase transitions of water to ICE VI and ICE VII around 1.0 GPa and 2.5 GPa, respectively.

Assessment of Advanced Oxidation Processes using Low and Medium-Pressure Lamps with H2O2 for Reclamation of Biologically Treated Wastewater Effluents (하수 2차 처리수 재이용을 위한 저압 및 중압 고도산화시스템의 성능평가)

  • In the present study, the feasibility of $UV/H_2O_2$ systems was investigated using low and medium-pressure lamps on biologically treated wastewater effluents for secondary effluent reclamation. Two types of UV lamps were used as the light sources (a 39-W low-pressure mercury lamp and a 350-W medium-pressure mercury lamp). The results from these UV systems showed that the removal of organic compounds could be achieved in the contact time of longer than 30min (i.e., low UV doses). Efficiencies of color removal and disinfection were far better than those of organic matters measured as TOC, DOC and $TCOD_{cr}$. In the low-pressure lamp UV system, it has been found that DOC and color removals were 60.9 and 86.2% with 50mg/L of $H_2O_2$ and contact times of 30 minute, respectively. Whereas, with the medium-pressure lamp UV system, TOC, DOC and color removal were 27.1, 5.6 and 95% with 14.3mg/L of $H_2O_2$ and 14 minute of contact times, respectively. Both systems could be applied for the reclamation of secondary effluent treated with biological treatment processes.

  • In the process of oyster mushroom production, loading work of culture medium needs the most intensive labor power and cost. Therefore, the development of culture medium loading machine causes to reduce the manpower and cost. The main objectives of this study are identify cultivating environment, physical properties of culture medium and to make an element design of culture medium loading machine. The results are summarized as follows: 1. The moisture content and density of popularly used culture medium were 70%(w.b), $26\;kg/m^3$, respectively. 2. Pressure of the blower increased as the impeller speed increased, and the opening ratio of pressure controller decreased. 3. Recommendable c1earance(${\delta}$) between an impeller plate and a blower case was 25 mm at an impeller speed of 3183 rpm 4. Discharge device of type B with a hopper and suit was better than type A with a hopper.

Synthesis of Silica Aerogel at Ambient Pressure and Characterization (II) (실리카 에어로겔의 상압합성 및 특성연구(II))

  • Low-density silica gel monolith was synthesized at ambient pressure by surface modification with TMCS and sub-sequent heat treatment. The mechanical thermal and optical properties of gel were studied. Compressive strength and modulus of compression of 350$^{\circ}C$-heated gel with the density of 0.24g/cm3 were 250kPa and 0.2MPa respectively. The thermal conductivity of silica gels synthesized at ambient pressure exhibited 0.02 W/m$.$K for the density of 0.24g/cm3 which is similar to that of the gel via supercritical drying and their main thermal transfer mechanism is considered to be solid and radiation conduction at room temperature. Ambient-dried silica gels were transparent blue showing about 60% of transmittance in the wavelength of 1500-2100nm and typical absorption bands of existing bonds under heat treatment at 350$^{\circ}C$. Medium scale monolity(${\Phi}$=50mm) at ambient pressure could be successfully prepared through total 5-month process period.

Fracture Characteristics of D 3507 and D 3631 City Gas Steel Pipes (도시가스용 D3507 및 D3631 강관의 파괴특성)

  • It is noted that KS D 3507 pipe steel has several problems when it is used as a city gas pipe at medium pressure. So new pipe steel, KS D 3631, was developed in order to be used as a pipe for medium and low pressure and now it is being substituted for D 3507. In this study, several mechanical tests, such as tensile test, microhardness test, and Charpy impact test were conducted to get material properties of D 3507 and D 3631 pipe steels. And also microstructures at the weld and heat affected zones were observed for the two materials. From the Charpy test results $K_{IC}/$ was estimated for the upper and lower shelf and the critical crack length is calculated for supposed axial semi-elliptical surface cracks. And the burst pressure is estimated as a function of wear depth for a defective D 3631 pipe by using the finite element method. The burst pressure is also calculated for pipes with an axial crack by using the published equations.

  • In studies on an alternative insulating gas of $SF_6$ gas, the section of the alternative gas and an insulation technique to improve its low dielectric strength have been reported, but very few attempts have been made at the dependence of a gas pressure and a gap as well as the medium effect in the alternative gas. The purpose of this paper is to analyze the dependence of the gas pressure and the gap at surface flashover voltage in dry air. The dependence is analyzed based on the medium effect. The medium effect by the gas pressure and the gap can be explained by surface roughness of a solid dielectric and an electrode as well as an electric field which decreases due to the correlation between the collision ionization coefficient and the gap, respectively. In addition, an insulation technique which can fabricate a compact eco-friendly gas insulated switchgear is proposed by the results of this paper.

Effect of Health Services on Workers with Hypertension in Small and Medium Sized Enterprises (중소규모 사업장에서의 고혈압 근로자에 대한 보건관리 효과)

  • The purpose of this study was to evaluate the effect of health services on workers with hypertension in small and medium sized enterprises (20-300 employees). One hundred seventy three workers among 296 workers with hypertension were responded to the questionnaire on general and occupational characteristics, health habits and anti-hypertensive medications. Blood pressure was obtained while seated using a calibrated standard manometer before and after 4 months health services. Statistical analysis was performed using $X^2$-test and logistic regression analysis. The results were as follows ; 1. No significant differences were noted in variables of smoking, glucose intake and sleeping time, while significant differences were showed in variables of exercise time, frequency and amount of drinking and intake of salt, protain, lipid, calcium, vegetable and fruits between groups which was provided and not provided health services. 2. The rate of anti-hypertensive medication was increased and the blood pressure was decreased significantly in group which was provided health services compared with those of group which was not provided. 3. With the logistic regression analysis, significant variables affecting blood pressure change were smoking and anti-hypertensive medication. The results of this study suggested that health services on workers with hypertension in small and medium sized enterprises was effective, and the role of health professionals for health promotion movement in enterprises was important.
