Efficient plant regeneration from immature embryo cultures of Jatropha curcas, a biodiesel plant

  • Jatropha curcas L. (Physic nut) is a commercially important non-edible oil seed crop known for its use as an alternate source of biodiesel. In order to investigate the morphogenic potential of immature embryo, explants from four developmental stages were cultured on medium supplemented with combinations of auxins and cytokinins. It was found that the size of embryo is critical for the establishment of callus. Immature embryos (1.1-1.5 cm) obtained from the fruits 6 weeks after pollination showed a good response of morphogenic callus induction (85.7%) and subsequent plant regeneration (70%) with the maximum number of plantlets (4.7/explant) on Murashige and Skoog's (MS) medium supplemented with IBA (0.5 $mg\;l^{-1}$) and BA (1.0 $mg\;l^{-1}$). The above medium when supplemented with growth adjuvants such as 100 $mg\;l^{-1}$ casein hydrolysate + 200 $mg\;l^{-1}$ L-glutamine + 8.0 $mg\;l^{-1}$ $CuSO_4$ resulted in an even higher frequency of callus induction (100%). Plant regeneration (90%) with the maximum number of plantlets (10/explant) was achieved on MS medium supplemented with 500 $mg\;l^{-1}$ polyvinyl pyrrolidone + 30 $mg\;l^{-1}$ citric acid + 1 $mg\;l^{-1}$ BA + 0.5 $mg\;l^{-1}$ Kn + 0.25 $mg\;l^{-1}$ IBA. It was observed that plantlet regeneration could occur either through organogenesis of morphogenic callus or via multiplication of pre-existing meristem in immature embryos. The age of immature embryos and addition of a combination of growth adjuvants to the culture medium appear to be critical for obtaining high regeneration rates. Well-developed shoots rooted on half-halfstrength MS medium supplemented with 0.5 $mg\;l^{-1}$ IBA and 342 $mg\;l^{-1}$ trehalose. The rooted plants after acclimatization were successfully transferred to the field in different agro-climatic zones in India. This protocol has been successfully evaluated on five elite lines of J. curcas.

The Meaning of Practice in Theory (이론(理論, Theoria)에 있어서의 실기의 의미)

  • What is 'Art Theory'? In the western sense, the term poses a vague ambiguity, and in the eastern, it is rather an abstract and metaphysical concept. As for etymology, theory is derived from theoria and theoria from theoros. It refers to an act of viewing or seeing, of course not in a metaphysical sense. Plato understood it as 'eide'. During the time of Plotinus, theoria encompassed gazing at every possible reality, and this gazing, that is theoria, is closely related to reality as aunit that theoriacan perceive. However, we tend to distinguish, as other scientists of dualism have done, studio art from theory since a pre-modern approach to art has been particularly tuned to studio practice, set apart from theory. Therefore, in studio classes, students are expected to learn the subject based on the foundational curriculum methods such as medium, genre, technique:, rather than bringing out their own interpretations and discussing theories. As a result, students have become artists, who are not able to understand their own art. Art professors who conduct class in studio are required to proceed with specific 'theories' as well as 'intellectual reflections'. In this respect, this thesis presents poiesis and an idea of 'acting out'. Although art history and aesthetic theory tend to view art as a finished product, actual art-making and related theories should not only be acknowledged as 'completion' (finition) but also be accompanied by theoretic interpretations of the act itself and process. Accordingly, it is to accept and appreciate art as finished result in view of current theory and aesthetics thus boils down to aisthesis. Likewise, poietics starts from a point where an artist is related to studio and examines the 'work process' that extends as far as to the exact end of work. Through the study of such relationship, it is possible that theory understands 'studio' and 'process', and an artist can grant an independent meaning to studio where s/he pours her/his heart out creating a work of art. Theory is a study on artistic discovery thus should be equipped with functions that can accommodate fortuity, imitation, thinking, culture, and surrounding.

Identification of bacterial contaminants in porcine semen and its removal (돼지 정액내의 오염 세균의 동정 및 오염된 세균의 제거)

  • Bacteriospermia is a frequent finding in fresh boar semen and can result in detrimental effects on semen quality and longevity. The objectives of this study was to evaluate types of bacterial contaminants in porcine fresh semen and the reducing effect of antibiotic and density gradient with percoll on the bacterial contaminants. Fresh semen was collected by gloved-hand method into a pre-warmed($37^{\circ}C$) thermostable bottle, and was inoculated onto blood agar and MacConkey agar, respectively. After incubated for 48 hour, 7.5% $CO_2$ at $37^{\circ}C$, bacterial colonies were selected and identified by Gram staining, oxidase test, catalase test and finally identified using API kits and Vitek system. Aerobic culture yielded a variety of bacteria from different genera. The most prevalent contaminant of fresh semen were Leclecia adecarboxylata, Acineobacter banmanni, Staphylococcus epidermidis, Staphylococcus cohni spp urealyticus, Proteus mirabilis. Most of identified bacteria were Gram(-) and non-pathogenic bacteria. It seems that bacterial contaminants in fresh semen were seem originated from multiple sources at the stud/farm, and were from animal and non-animal origins. Gentamicin treatment did not eliminate the bacterial contaminants completely but 3 step-density gradient with percoll completely removed the bacterial contaminants in fresh semen. Therefore, future study is necessary to prove that density gradient method with percoll can eliminate bacteria in fresh semen without significantly affecting sperm viability or function.

A Study on Newborn Infection during Hospitalization (일 종합병원 신생아실 감염증에 관한 소고)

  • This study was conducted to evaluate the difference of staphylococcal colonization between lactating mothers and nursery stares. Samples were obtained from the hands and noses of the newborn(admitted to S Univ. hospital's nursery from 1991. 5. 23 to 6. 8), their mothers and nursery staffs. They were cultured, and then the presence/absence of pathogenic staphylococci in them were analyzed by Fisher exact probability test which led to these conclusion of significance as follows: 1. No differences were detected in pathogenic rate of nasal swab culture between breast~fed newborns and formula-fed ones. 2. Delivery type and hospitalization the ones from the brest-fed newborns show high rate of Sta. aureus coa(+). In case of either C-sec delivery or long (more than 4 days) hospitalization, formula-fed ones in case of NSVD or short (less than 3 days) hospitalization. 3. The ones from breast feeding mothers show higher sta. aureus coa(+) than those from nursery staffs, which was of no significance statistically. Considering nursery staffs only, however, the comparison of those with the data in April indicates that the pathogen rate is higher for hand than nose, and Sta aureus coa(+) for hand is far lower in June than in April, which was statistically significant. These results lead us to infer that newborn infection in hospitalization could be largely reduced by maintaining the ordinary hygienes. Such as the handwashing of mothers as well as nursery staffs (directly involved in newborn care) Thus the pre-enterance hand washing of a mother who visits the nursery only for breast feeding (without any other medical responsibility) should be done, like other medical agents, with $0.05\%$ chlorhexidine antiseptics rather than simple soap cleansing; the one 'that is worth emphasizing thoroughly.

  • To establish high frequency shoot formation from two cultivars (Cheongyangjaerae and Myungan) of boxthorn (Lycium chinense Mill.), hypocotyl segments with cotyledons from seedlings were used as explants. High frequency adventitious shoot formation (more than 80%) were obtained from hypocotyl segments with cotyledon on MS medium supplemented with 1.0 mg/L zeatin, when precultured for 3 weeks under dark conditions followed by transfer to light conditions. But there was no shoot induction in the explants cultured without preculture under dark conditions. Roots were induced from the shoots when transferred to rooting medium supplemented with 1.0 mg/L IAA for 4 weeks. Regenerated plantlets were grown to normal mature plants in soil.

Degradation of Polycyclic Aromatic Hydrocarbons by Selected White-rot Fungi and the Influence of Lignin Peroxidase

  • The white-rot fungi Phanerochaete chrysosporium ATCC 24725, Pleurotus ostreatus ATCC 32783, Lentinus edodes ATCC 24462, and Trametes versicolor ATCC 42530 were studied for their ability to degrade lignin, phenanthrene, and anthracene. Lignin in rice-straw was degraded by 14.4, 28.73, and 33.88% by P. chrysosporium, T. versicolor, and P. ostreatus, respectively. Approximately 12% and 83% of phenanthrene was degraded in 1 and 5 days, respectively, when the pre-grown mycelIium matrix of P. ostreatus. was incubated with 10 ppm of phenanthrene in modified Kirk's medium (nitrogen limited) at $25^{\circ}C$. Approximately 2%> and 61% of phenanthrene was degraded when the phenanthrene concentration was increased to 30 ppm. Similar trends were observed with phenanthrene using P. chrysosporium. Mycelial growth of T. versicolor was less inhibited at 30 ppm phenanthrene than for P. ostreatus and P. chrysosporium. Better degradation of phenanthrene by T. versicolor may be attributed to better mycelium growth. One hundred percent of 15 ppm anthracene was degraded in 10 days by both P. chrysosporium and T. versicolor. 40 ppm anthracene inhibited the mycelial growth of P. chrysosporium. lignin peroxidase activity, which was previously reported to be involved in initial phenanthrene oxidation, was also detected from the culture broth of the strains tested. The rates of lignin peroxidase production in the cultures were not consistent with the rate of PAH hydrolysis during incubation.

  • Citric acid is produced via submerged fermentation using yeasts. Among eight different strains of yeast, Candida zeylanoides was chosen as the strain for producing citric acid and optimized for various C/N ratios and effects of phosphate or Fe2+ ions in a clean carbon source medium (glucose: fructose, 1:1). The yield of citric acid was maximized at a C/N ratio of 40/1, a phosphate addition of 1.0 g/l, and an Fe2+ ion concentration of less than 50 mg/l, yielding up to 91 g/L in the broth with 18.5 g/l of isocitric acid in a six-day fermentation period using a pre-treated molasses medium. The yield of batch culture was 0.51 (Yp/s, g/g) in a 5 L-Jar fermenter.

The Effect of Carbon Sources on Nisin Z Biosynthesis in Lactococcus lactis subsp. lactis A164

  • The effect of carbon sources on nisin Z biosynthesis in Lactococcus lactis subsp. lactis A164 was studied in batch culture using M17 broth containing different carbon sources. Among the eleven carbon sources tested, glucose, sucrose, and lactose were suitable carbon sources for cell growth of L. lactis A164. In particular, cells grown on lactose produced at least 3-fold greater amount of nisin Z than those on other carbon sources. Galactose resulted in less amount of cell mass than did sucrose or glucose, but gave a higher level of nisin Z activity. Northern blot analysis revealed. that lactose increased the transcription of the nisZ pre-peptide gene. Although galactose was less efficient than lactose, it increased the transcription of nisZ along with a higher level of nisin Z than did sucrose and glucose. These results suggest that the increased nisin Z production is correlated with the induction of nisZ by lactose and galactose. Among all the carbon sources tested, no remarkable differences were observed in nisRK and nisFEG transcripts, indicating that the lactose- or galactose-mediated induction is unique to the nisZ promoter.

Taurine Transporter Activity in the Human Colon Carcinoma cell Line(HT-29) is Decreased during Cell Differentiation (인체 소장상피세포주(HT-29)의 분화단계에 따른 타우린수송체 활성의 변화)

  • Previous studies on the effect of age on intestinal taurine transport in animals have invariably shown a decline in the activity of the transport system with increasing age. In the present study changes in taurine transporter activity were observed during cell differentiation in the human colon carcinoma cell line HT-29 This cell line exhibits various enterocytic characteristics when differentiated and therefore has frequently been used to study the characteristcs and regulation of nutrient and drug absorption in the small intestine at the cellular level. Pre-treatment of the cells with $\beta$-alanine(10mM) reduced the taurine transport activity to 33% of the value for the control cells(p<0.05) which implies that taurine and $\beta$-alanine share a common $\beta$-amino acid transport system for their celluar uptake in the HI-29 was continued until 21 days post seeding. Kinetic studies of the taurine transporter were conducted in the HT-29 cell line with varying taurine concentration(5-60$\mu$M) in the uptake medium Both Vmax and the Michaelis-Menten constant(Km) of taurine transporter were decreased as differentiation of the HT-29 cell line was progressed ; Vmax of the taurine transporter in cells incubated for 4, 14 and 21 days post seeding was 2.79$\pm$3.4m 16.89$\pm$1.74, and 0.85$\pm$0.08 and 0.32$\pm$0.01nmol.mg protein-1 .30min-1 respectively(p<0.001) and Km was 42.3$\pm$3.4, 16.89$\pm$1.74, and 11.2$\pm$3.0$\mu$M respectively (p<0.01) These results indicate that the activity of sodium dependent active taurine transport system in the HT-29 cell line is decreased as confluent cells are differentiated. This phenomenon in cell culture system corresponds well with the earlier observation of lower intestinal taurine transport activity in suckling rats compared to that in adult animals although direct relationship of cell differentiation with in vivo aging process needs further verification.

Fatty Acid Compositions of Cultured Oyster (Crassostrea gigas) from Korean and Japanese Spats

  • Changes in lipid content, lipid class and fatty acid compositions of the cultured oysters in shallow-water, Bukman bay (Tongyeong, Korea), using both Korean and Japanese spats were investigated. The content of non-polar lipid (NL) comprised approximately $60- 80\%$ of total lipid (TL) in the cultured oysters. There was a positive correlation between NL content and meat weight, y=0.287lx-15.309 (r=0.834l, p<0.001). The prominent fatty acids of the oysters were 16:0, 20:5n-3 (eicosapentaenoic acid, EPA), 22:6n-3 (docosahexaenoic acid, DHA), 18:0, l8:1n-7, l8:1n-9, l6:1n-7, 14:0 and l6:4n-3. During the growth of the oysters, l6:4n-3 showed the highest coefficient of variation, accounting for $41.8\%$ for the Korean oyster and $32.3\%$ for the Japanese one, respectively. Both oysters showed low level of n-3 fatty acids such as DHA and EPA and high level of n-6 fatty acid, 20:4n-6, in the spawning period (August). During growth of the oysters, both EPA and DHA were the richest fatty acids in the harvest period (December, 314 mg/100g sample) and in the pre-spawning period (July, 237-247 mg/100g sample), respectively. Consequently, the cultured oyster with Japanese spat contained approximately two times more n-3 fatty acids per oyster individual than those with Korean one in the harvest season.
