Comparative Studies on the Growth and Differentiation in Three Different Varieties of Hop (Hop의 생장과 분화에 대한 식품간의 차이에 관하여)

  • The present investigation dealt with observation and experiments concerning the growth and differentiation of hop plant, using the varieties of Cascade(C), Shinshuwase(Sh), and Hallertau(H). The results were as follows: (1) Life cycle of hop plants. The annual growth period of hop plant was devided chiefly into 3 phases, dormant, vegetative and reproductive. (2) Growth of main stem. The hop vine begun to grow in the middle of May and grow vigorousely in the middle and latter of June, then gradually decline or stops at the middle of July and the early of August. (3) Growth of lateral vines. By the statistical analysis, it is judged that the varieties of H and Sh were more grown than that of C. H and Sh were not significant, but H and Sh from C were significant in 5% level. (4) Fresh weight and water content of hop cone. Hop cone in fresh weight of C variety was higher than those of other two varieties and water content of hop cone was decreased with time elapse in three races together. (5) Growing point. Histological view of hop varities in each was different. C showed form of sweet potato, H showed form of round, and Sh showed form of ellipse. (6) Shape of the leaf. C and H were 3 lobes, but Sh is 3∼5 lobes. Generally, the color is dark green. (7) Hop cones. Hop cones are as follows.

Changes in Texture during the Boiling Process of Potatoes (감자의 증자중(蒸煮中) 텍스쳐의 변화(變化))

  • The texture changes of potato of the variety ‘Namjack’ were determined as a function of temperature and boiling time by a puncture test with Universal Testing Machine. Apart from texture measurement, heat penetration test and sensory evaluation were carried out. Textural softening during cocking could be expressed by the puncture work and was followed first order kinetics. The Z-value for texture changes was $19.9^{\circ}C$. The cook value in equivalent minutes at $100^{\circ}C$ to get a boiled potatoes was 7 min. Mechanically the boiled product could be regarded as cooked when the final puncture work reduced to the equilibrium value, approximately 6/100 of the initial value.

GUS Expression by CaMV 35S and Rice Act1 Promoters in Transgenic Rice

  • To determine the patterns and the levels of expression of the cauliflower mosaic virus (CaMV 35S) promoter and the rice actin 1 (Act1) promoter in rice, transgenic rice plants containing CaMV 35S-$\beta$-glucuronidase (GUS) and Act1-GUS constructs were generated and examined by fluorometric and histochemical analyses. The fluorometric analysis of stably transformed calluses showed that the activity of the rice Act1 promoter was stronger than that of the CaMV 35S promoter in rice cells. In a histochemcial study of the transgenic rices, it was shown that the GUS activity directed by the CaMV 35S promoter was localized mainly in parenchymal cells of vascular tissues of leaves and roots and mesophyll cells of leaves. These results are similar to those of potato, a dicot plant. In contrast, rice plant transformed with Act1-GUS fusion construct revealed strong GUS activity in parenchymal cells of vascular tissue, mesophyll cells, epidermal cells, bulliform cells, guard subsidiary cells of leaves and most cells of the root, suggesting that the rice Act1 promoter is more constitutive than the CaMV 35S promoter. It was also confirmed that in both types of transgenic rice little or no staining was localized in metaxylen tracheary elements of vascular tissue from leaves or roots. These results indicate that the rice Act1 promoter can be utilized more successfully for expression of a variety of foreign gene in rice than the CaMV 35S promoter.

Changes of Mating Type Distribution and Fungicide-resistance of Phytophthora infestans Collected from Gangwon Province (강원지역 감자 역병균 Phytophthora infestans의 교배형 및 약제저항성 변화)

  • Potato late blight caused by Phytophthora infestans was the most constrain disease at potato cultivation areas. The mating type distribution and fungicides response of P. infestans were investigated to elucidate the changes of pathogen from Gangwon province. On the fungal isolates in 2006, 58.7% were A1 mating type and 41.3% were A2 mating type. In 2007, A1 mating type isolates increased to 93.3% and A2 mating type isolates were collected from Jinbu areas as much as 6.7%. About 234 isolates analysed for metalaxyl response, the results was resistance 73.7%, intermediate 18.8% and sensitive 7.5% in 2006. And it was resistance 59.4%, intermediate 4.0% and sensitive 36.6% in 2007. It meant that mating type distribution and fungicide response were very different over the collection sites. Minimal inhibition concentration (MIC) of dimethomorph examined with 126 isolates in 2006 and 106 isolates in 2007. MIC over $1.0\;{\mu}g/ml$ was 56.3% in 2006 and it was 3.8% in 2007. The average $EC_{50}$ value of P. infestans was $0.37\;{\mu}g/ml$ in 2006, but it decreased to $0.12\;{\mu}g/ml$ in 2007. Fungicides response and pathogenesis of P. infestans should be monitored continuously to enhance the chemical efficacy at potato fields.

Characterization of a new variety Auricularia polytricha (Mont.) Sacc.'Geoni' (털목이 신품종 '건이')

  • Geoni, a new variety of Auricularia polytricha, was bred in JARES(Jeollanamdo Agricultural Research and Extension Services) in 2012. Geoni was selected through the monospore hybridization with JNM21008 and JNM21014 in 2010. Based on a performance test conducted from 2011 to 2012, the Geoni was selected from a line showing an excellent light brown pileus and strong pest resistance. Geoni has a favorable chewiness, light brown and smooth pileus. In addition, Geoni was rich in dietary fiber. MCM(mushroom complete medium), Malt and PDA(Potato Dextrose Agar) mediums were suitable for cultivating the Geoni. The number of effective stipes was 39 ea/0.9 kg and minor axis and major axis of pileus were 6.9 cm and 8.7 cm respectively. The yield of Geoni was 291 g/0.9 kg in plastic bag. Geoni was required 40~54 days for culture at $20^{\circ}C$ and 24days for the primordia and growth period, which is longer than that of the control(Pung-un). Somatic incompatibility was formed between parental strains and Geoni. Analysis of the genetic diversity of the new variety "Geoni"revealed a different profile from the parental strains when RAPD(random Amplified Polymorphic DNA) primers were used.

Breeding and characteristics of a low-temperature variety oak mushroom (Lentinula edodes) 'Sanjo 708 ho' (표고 중온성 품종 '산조708호' 육성 및 특성)

  • In 2010, a new variety for sawdust cultivation,was produced by monokaryotic-monokaryotic crossing between Sanjo 701ho and FMRI0995. The optimum temperature for Sanjo 708ho mycelial growth was $25^{\circ}C$; mycelial growth on potato dextrose agar was 51.6 mm over 7 days. The fruiting pattern was sporadic, and the optimal temperature range for fruiting was $5{\sim}20^{\circ}C$. Regarding fruiting body characteristics, the pileus was 65.8 mm in size, 16.1 mm in thickness, and hemispherical in shape. The stipe was 42.6 mm in and 19.6 mm in thickness. Mproductivity was good during thelow-temperature period from autumn to spring, with an average productivity of 310.7 g/bag.

Characteristics and breeding of a new variety Pleurotus eryngii, Gonji No. 8 (신품종 큰느타리버섯 '곤지8호' 육성 및 특성)

  • A new variety of Pleurotus eryngii which is named 'Gonji No. 8' was bred by mating monokaryotic strain isolated from E085D2 and a monokaryotic strain 'aerini No.3' obtained from the Mushroom Research Institute, Gyeonggi-Do A.R.E.S. The characteristics of the new variety 'Gonji No.8' is as follows. The optimum temperature for mycelium growth was from $23^{\circ}C$ to $26^{\circ}C$ on potato dextrose agar (PDA) medium. For the primodia formation and the growth of fruit bodies, the optimum temperature was from $14^{\circ}C$ to $16^{\circ}C$. The period of spawn running was around 30 days at $22^{\circ}C$ and the days taken after the removal of the spawn layer to initiate primodia was seven days. The hardness value of fruit body was $8,432{\pm}2,193$ $g/cm^2$, which was two times more than that of 'Keunneutari No.2'. The yield of 'Gonji No.8' was about 133 g per bottle(900cc) and it was same as 'Keunneutari No.2'.

Development of PVY resistant flue-cured $F_1$ hybrid variety 'KF120'

  • Potato Virus Y (PVY), PVY-vein necrosis strain, causes severe damage at growth, yield and leaf quality on flue-cured tobacco in Korea. The development of PVY resistant flue-cured varieties without quality deterioration is therefore urgently desired. The flue-cured tobacco, KF120 (Korea Flue-cured 120), was a male-sterile (ms) $F_1$ hybrid derived from the cross between msKF117 and KF0007-7. msKF117 was developed from the cross of NC82 with N. africana and KF0007-7 was developed from the cross of KF117 with NC82. The agronomic characteristics and disease resistance of KF120 was evaluated during 2006-2007 field performance test. It showed better growth characteristics and yield performance than standard cultivar KF109. It had 2 more leaves per plant, flowered 2 days later than KF109. The yield of cured leaf of KF120 was increased by about 5% compared to KF109. The chemical composition and physical properties of the cured leaf of KF120 were as much acceptable as those of KF109. KF120 showed high resistance to PVY compared to KF109. It showed a similar mode of resistance to bacterial wilt and black shank as was found in KF109.

Gamma-ray Irradiation on Radio Sensitivity in Yacon (Samallanthus sonchifolius (Poepp. & Endl.) H. Robinson) Breeding (돌연변이 육종을 위한 야콘의 최적 감마선 조사량)

  • Yacon [Samallanthus sonchifolius (Poepp. & Endl.) H. Robinson], a member of Compositae plants, has sweet taste and crisp texture. Unlike other Andean root crops such as potato and sweet potato, the cultivation area of yacon has increased recently, since it is known to have large content of fructooligosaccharides (FOS). Since there are no yacon varieties bred in Korea, we have been trying to create new genetic resources using gamma-ray. The optimal gamma-ray dosage for mutation breeding in yacon was investigated. Crown bud and green bud of yacon were exposed to doses of gamma rays from 20 Gy to 80 Gy, and subsequently planted in a greenhouse. After 50 days of sowing, the survival rates and growth decreased rapidly at doses above 40 Gy, while all of crown bud individuals died above 60 Gy. The median lethal dose (LD50) of crown bud and green bud was 22.4 and 36.6 Gy, and the median reduction doses (RD50) for plant height, fresh weights, and tuberous root weight were 20-40 Gy, respectively. A dose of 20-40 Gy was found to be optimal for mutation breeding in yacon. Considering the growth factors, the optimum doses were determined to be within the range of 20-40 Gy for the selection of useful mutant lines. M2-M3 mutant lines were obtained from 20-60 Gy gamma-ray-irradiated M1 plants through clonal propagation. These mutant lines will be used for the development of a new variety of yacon plant with high FOS and no crack tuberous root.

Characterization of a new commercial strain 'Goni' by intra-specific hyphal anastomosis in Pleurotus ostreatus (계통간 교잡에 의한 백색느타리 품종 '고니'의 육성 및 그 특성)

  • To develop new white variety of Oyster mushroom, all white varieties which have been collected and kept in the lab were revived and screened their cultural characteristics. 84 intra-specific Oyster mushroom hybrids between the white-colored mutants Suhan and Wonhyeong were developed using hyphal anastomosis technique in 2007. The Po2007-63 ($2842-7{\times}0205-7$) was shown the best cultural characteristics, selected to be a new variety and named as 'Goni'. The new commercial strain, 'Goni' has white pilei and grows well under spring and autumn conditions in Korea. The fruiting bodies of 'Goni' are of an excellent quality in that not only the stipe is thick and long but also the pileus is small and hard. The optimum temperatures for mycelial growth and fruiting body development were $25-30^{\circ}C$ and $10-16^{\circ}C$, respectively. Time period required for the initiation of the first fruiting body is about 3 to 5 days depending on the temperatures. The shape of fruiting body is thin funnel shape. Fruiting body production per bottle was about $91{\pm}13$ g which is almost 97% quantity compared to that of other variety 'Miso'. Relatively low temperature incubation ($11^{\circ}C$) resulted in the development of better quality of 'Goni' mushrooms. When two different media including potato dextrose medium and mushroom complete medium were compared, the growth of mushroom were much faster in mushroom complete medium at $20-25^{\circ}C$, but not at $25^{\circ}C$. Similar results were observed with other variety 'Miso'. Analysis of the genetic characteristics of the new commercial strain 'Goni' showed a major DNA profile as that of the parental Suhan when primer URP 1 was used, but different to 'Miso' that was used as a control. When screens were performed with primer URP 2, DNA patterns were similar both to that of the parents and 'Miso'. This new variety of the white Oyster mushroom has a clean and fresh image that corresponds well to "health food". We therefore expect that this new strain will satisfy the consumers demand for variety and excellent mushrooms.
