This study was carried out to establish the in vitro culture system of potato germplasms for minimizing the occurrence of variation and maximizing the culture period. We used osmoticum such as sorbitol or mannitol with sucrose in the absence of plant growth regulators. The growth of potato germplasms in the medium containing osmoticum was increased when the growth temperature was lowered. After six months storage in low temperature, plant heights of tetraploid was somewhat higher than those of diploid with the exception of stn-16 and the difference due to media was not observed. But after twelve months storage, survival rates of plants cultured in LSM 1(sucrose and sorbitol) was higher than those of plants cultured in LSM 2(sucrose and mannitol). The survival rate of stn-16, diploid wild species, was approximately 75% and it was considerably high. In Atlantic, tetraploid cultivated variety, every individual was survived.
A total of 40 virus infected tobacco plants (Nicotiana tabaccum L.) with various symptom types Were collected from Bucheon and Jeonju area by its symptoms were investigated on the incidence of tobacco mosaic virus (TMV), cucumber mosaic virus (CMV), alfalfa mosaic virus (AMV), potato virus X (PVX) and potato virus Y (PVY) by serological methods. van Slogteren's microprecipitin test was applied for the testing of PVX and PVY from infected plants and Ouchterlony agar double diffusion test was used for CMV, TMV and AMV. Results obtained are as follows: 1. TMV, CMV, AMV, PVX and PVY wcre found to occur on the tobacco plants growing in Korea. 2. The prevalence of each of these viruses among the 40 tobacco plants investigated was in the order of AMV: $(67.5\%)>CMV:(60.0\%)>TMY:(47.5\%)>PVY:(17.5\%)>PVX: (10.0\%).$ 3. In Burley variety, the percentage of infection by TMV was $15\%$, whereas it was as high as $80\%$ in Hicks variety. 4. Among the 40 tobacco plants investigated, $37.5\%$ showed infection with one kind of virus whereas the remaining $62.5\%$, revealed mixed infection with more than two different viruses.

We investigated the quality characteristics of sweet potatoes [Shinyulmi (SM), Yeonhwangmi (HM) and Yunmi (YM) variety] baked and subsequently freeze thawed. Baking was performed with a far infrared radiation oven at $250^{\circ}C$ for 40 minutes. Baked sweet potatoes were frozen at $-18^{\circ}C$ for 30 days, and thawed by a microwave oven for 3 minutes. The crude fat and protein content of HM were higher than the SM and YM varieties. Total sugar, reducing sugar, and free sugar content were increased after baking. Lightness, redness, and yellowness, as well as content of ${\beta}$-carotene, decreased after baking and freeze-thawing. Hardness, gumminess, and cohesiveness decreased during thawing after freezing, except for the YM variety. In a sensory evaluation, appearance, flavor, sweetness and overall acceptance were higher from baking and freeze-thawing the SM variety, instead of the HM and YM variety. From the results, the SM variety is preferred for the production of baked sweet potatoes.
In order to investigate the characteristics of various sweet potato starches, gelatinization temperatures, solution viscosity of starch separated from two cultivars of the dry type sweet potatoes(Yulmi and Gunmi), one cultivar of moist type sweet potato(Jinmi), and one cultivar of purple colored variety(Jami) were compared, and properties of the edible films prepared with the starches were determined. Under a differential scanning colorimetry(DSC), initial temperatures for starch gelatinization of the dry type sweet potatoes (Yulmi and Gunmi) were higher than that of the moist type sweet potato (Jinmi), and that of Jami was close to those of the dry type ones. The sweet potato starch solutions tested by a cone and plate viscometer, showed peudoplastic characteristics. The moist type sweet potato was the most viscous followed by Jami, Yulmi, and Gunmi among the tested starch solutions. Total color difference of the edible films prepared with different cultivars of sweet potatoes showed appreciable differences between cultivars, caused by differences in Hunterb values. Water Vapor Permeability (WVP) of sweet potato starch films also showed significant differences between cultivars. Films prepared with the dry type sweet potato, Gunmi, showed the lowest WVP value of $0.83{\times}10^{-9}\;g\;{\cdot}\;m/m^{2}\;{\cdot}\;s\;{\cdot}\;Pa$, followed by Jami, Yulmi, and Jinmi. Water solubility of the films did not show any significant differences between cultivars. Tensile strength of the dry type sweet potato and Jami, which ranged 14.18-18.75 MPa, were higher than that of the moist type sweet potato, which was 4.66 MPa. Elongation values of the films, which were 5-6%, indicated that sweet potato starch films were not so elastic.
The purple color in flesh of 'Jasim' potato (Solanum tuberosum L.) tubers is due to an accumulation of anthocyanin. The experiment was conducted to determine what is the key factor among cultural environments that affect the accumulation of anthocyanin in 'Jasim' potato tubers, and the effect of nitrogen fertilization levels on that. Through analysis of anthocyanin content of purple-fleshed tubers produced under the different cultural environments of three localities and two planting dates, we determined that lower temperature during 30days before harvesting resulted in higher anthocyanin accumulation of 'Jasim' potato tubers. In the experiment of different level of nitrogen fertilization, 'Jasim' potato tubers produced in highland (Daegualryeong) showed higher content of anthocyanin in the treatment of recommended level than in treatment of lower or higher level as compared with the recommended level. However, significant difference of anthocyanin content among different nitrogen fertilization levels was not observed in lowland (Gangnung). These results suggest that the selection of locality or cropping system and the optimal fertilization management are very important factors for production of higher pigmented tubers in purple-fleshed potato.

Jayoung was bred within the potato breeding program of Highland Agriculture Research Center (HARC), National Institute of Crop Science. It was originated from a cross made between Atlantic as a female and AG34314 as a male parent in 2003. Jayoung was evaluated as a clone number Daegwan 1-104 for the growth and tuber characteristics, yielding ability, and resistance to major potato diseases in the regional yield trials from 2006 to 2007. Finally, this clone was renamed as Jayoung and was presented to Korean Seed & Variety Service in 2007 for registration as a new potato cultivar. Jayoung has a oval shape, shallow eye depth, dark purple skinned and fleshed tuber characteristics. Its mean tuber yields from the regional yield trials was 37.3 MT/ha, and it showed the highest yield in spring cropping. Jayoung showed high resistance to both common scab (Streptomyces scabies) and potato leaf roll virus (PLRV), however it was susceptible to late blight (Phytophthora infestans). The dry matter content of Jayoung was fairy high 20.3%, therefore it is suitable for both chip processing and table usage. Its anthocyanin content was the highest in autumn cropping as 43.7 mg/100 g fresh weight and its mean anthocyanin content was 4.7 times higher than that of Jasim.

This study was conducted to examine the basis for the information to select the suitable potato varieties grown in new reclaimed land. The potatoes of five varieties were planted in the port with 4 different electrical conductivities of saturated extracts of soil taken the Saemangeum reclamation area, which was made of non-treatment salt and three concentrations of salt treatment, 1.6 dS $m^{-1}$, 3.2 dS $m^{-1}$, 4.8 dS $m^{-1}$, respectively. All of the potato varieties were uniformly emerged without missing plant in all treatment groups, even 4.8 dS $m^{-1}$ treatment group. According to the salt concentration of soil, required date to the emergence of the potato comparing to non-treatment salt was delayed 3-4 days in 1.6 dS $m^{-1}$, 6-10 days in 3.2 dS $m^{-1}$, 7-13 days in 4.8 dS $m^{-1}$, respectively, and the number of its branch decreased by 14-58% comparing to non-treatment salt depending on varieties. Since the increase of the salt concentration of the soil was more serious the decrease of the number of its branch, but plant height tended to increase when branch number per plant was small, which was depending on more number of its branch than salt concentration. Fresh tuber yield of potato comparing to non-treatment salt were decrease 33.7% in 1.6 dS $m^{-1}$, 59.5% in 3.2 dS $m^{-1}$, 79.3% 7-13 days in 4.8 dS $m^{-1}$, respectively. The threshold EC starting the growth inhibition of fresh weight decreased was 1.2 dS $m^{-1}$ for Chudong, 1.8 $m^{-1}$ for Chubeak, 1.9 $m^{-1}$ for Chugang and Chuyeong, and 2.0 $m^{-1}$ for Sumi, and EC which decreased 50% of dry weight index was 2.4 dS $m^{-1}$ for Chubaek, 2.45 dS $m^{-1}$ for Chudong, 2.81 dS $m^{-1}$ for Chugang, 3.03 dS $m^{-1}$ for Chuyeong, and 3.29 dS $m^{-1}$ for Sumi. The present results suggest that Sumi is considered to the suitable potato variety grown on saline soils.
Potato diseases, especially mosaics and leaf roll, appear to reduce potato yield in Korea more than any other factor. A seed potato certification program was established at the Alpine Experiment Station (AES) in 1961 to produce high quality seed potatoes for distribution to Korean farmers. The present program for production of certified seed of Namjak (Irish Cobbler), the only variety recommended for spring plantings, is outlined. In 1976, approximately 10,000 MT of certified grade Namjak seed was produced by members of two Seed Producers Cooperatives in the Daekwanryeong area for distribution by the Office of Seed Production and Distribution (OSPD). The seed was inspected and certified by officers of the National Agricultural Products Inspection Office (NAPIO). Although the quality of the certified seed is far superior to that used by many farmers, the supply planted less than 1/5 of the 1977 potato crop. Certified seed of Shimabara, the variety recommended for autumn plantings, is not produced in Korea. The yield response of virus infected seed to improved cultural practices is poor. Therefore, an increase in potato acreage and yields appears to be possible only if more good quality seed is used by Korean farmers. A two or three fold increase in seed supply would be desirable. The volume of seed could be increased by expanding the production area and by improving yield in seed fields. More land is available in the alpine area and good seed potatoes could be grown in other parts of Korea. Planting better quality seeds and using better cultural pracitces would improve seed yields. Several techniques could be used to improve the quality of elite seed produced at AES. Changes in seed potato certification program should be made so that healthy seed stocks of new varieties can be released rapidly.
