  • Virion captured reverse transcription polymerase chain reaction (VC/RT-PCR) could detect plant virus quickly and accurately. In the VC/RT-PCR, no antibody is needed unlike immuno-captured RT-PCR (IC/RT-PCR) which had been improved method of RT-PCR for plant viruses, and virus nucleic acids can be obtained easily within 30minutes by property of polypropylene PCR tube which is hold and immobilized viral particles on its surface. For the virion capture of Tomato spotted wilt virus (TSWV), the extraction buffer was tested. The optimum macerating buffer for TSWV was 0.01M potassium phosphate buffer, pH 7.0, containing 0.5% sodium sulfite. The viral crude sap was incubated for 30 min at $4^{\circ}C$. The virions in the PCR tubes were washed two times with 0.01M PBS containing 0.05% Tween-20. The washed virions were treated at $95^{\circ}C$ immediately for 1 min containing RNase free water and chilled quickly in the ice. Disclosed virions' RNAs by heat treatment were used for RT-PCR. Dilution end point of $10^{-5}$ from plant's crude sap infected with TSWV showed relatively higher detection sensitivity for VC/RT-PCR. During multiple detection using two or more primers, interference was arisen by interactions between primer-primer and plant species. The result of multiplex RT-PCR was influenced by combinations of primers and the kind of plant, and the optimum extraction buffer for the multiplex detection by VC/RT-PCR should be developed.

  • Traffic causes more and more stress and injury to grasses on golf course green in Korea due to the increased playing. This study compared the effect of fertilizer component (Nitrate, Phosphate, Potassium) on turfgrass growth and quality golf course green under traffic stress during early winter. Experiment was conducted by using different fertilizer components for 2 months(Oct. 1 to Nov. 30, 2005). Turfgrass leaf color, leaf texture, density and traffic tolerance were evaluated visually, and the root length(cm) and tiller density(tiller/$cm^2$) were measured. Creeping bentgrass(Agrostis palustris cv. 'Seaside II') fromthe nursery of Incheon Grand Golf Club was used. Results of this study are as followings: 1). Turfgrass color was the best in A6(20-15-10) and A5(15-15-10) treatments in the N study. Leaf texture was not different among treatments. Turf quality and traffic tolerance were the highest In A5 and A6. Root length was the longest(15.8cm) in A6 (20-15-10). Root length increased with nitrogen levels. 2). Turf color of A9(5-7.5-5) and A10(15-22.5-15) was darkest in the comparison of P study. Leaf texture was the best in A4(10-15-10) and A9. Turf quality was the best in A10. A7(10-0-10). 3). In general, to recover turfgrass damage on the putting greens during low temperature period, fertilizer amount need to be increased; and nitrogen is better than phosphate and potassium for that purpose.

  • The investigations were conducted to find out the situation of fertilizer use and the contents of soil chemical components on summer vegetable crops at 791 farmers' upland fields located in the parts of Gangwon-do, Gyengsangbuk-do and Jeollabuk-do of alpine area. Major vegetable crops were potato, Chinese cabbage, radish, carrot, onion, and cabbage. From the location surroundings cultivated alpine vegetable crops, the orders were Gangwon-do>Gyeongsangbuk-do>Jeollabuk-do part in the sizes of a fie1d area and the height above sea level, and Jeollabuk-do>Gyeongsangbuk-do>Gangwon-do part in the slope degrees. The soil texture was of wide distribution on sandy loam soil for Gangwon-do(76%) and Jeollabuk-do part(64%), and 1oam(42%) and sandy loam soil(35%) for Gyeongsangbuk-do part. From the numbers of investigated fields, the distribution of slope degree was wider than those of height above sea level in relation to location surroundings. The upland soils of 785 fields cultivated vegetable crops were sampled at 0~15 cm of top soil before seeding or transplanting and analyzed. On an average, pH, organic matter, available phosphate and exchangeable potassium, calcium, magnesium of soil were 5.7, $27.6g\;kg^{-1}$, $765mg\;kg^{-1}$, $1.16cmol_c\;kg^{-1}$, $6.1cmol_c\;kg^{-1}$, and $1.6cmol_c\;kg^{-1}$, respectively. The average cation exchange capacity(CEC) of 120 sites in Gangwon-do part was $9.2cmol_c\;kg^{-1}$. The content of organic matter, exchangeable potassium, exchangeable calcium and exchangeable magnesium was higher, while that of available phosphate was lower with slope degrees. And the content of major chemical components in carrot soil was lower in comparison with other crop soils. The average levels of N, $P_2O_5$, $K_2O$, livestock manure and lime fertilizer of 785 Belds applied by farmers were 335, 198, 244, 12,680 and $1,750kg\;ha^{-1}$, respectively, for summer vegetable crops in alpine area. The average amounts of $N-P_2O_5-K_2O$ fertilizers applied by farmers in 785 Gelds of vegetable crops were higher 1.7~2.0-4.2~7.0-1.4~2.0 times on potato, 1.4~1.6-4.6~8.3-3.5~4.2 times on Chinese cabbage, and 1.2~1.3-4.2~7.2-3.0~3.61 times on radish than the rates of NPK fertilizers based on soil testing for each crop.

  • The blueberry farming requires the soil condition of well-drainage, pH of 4.5 to 5.2, and high in organic matters for stable growth and development. Most of soil type of cultivated land in Korea, however, belongs to alkaline soils with low organic matter content and poor drainage. Therefore, the blueberry farmers use peat moss heavily to improve the soil condition, but the guideline on the effective and economic ratio of peat moss is not established yet. This study was performed to determine the cost effective peat moss ratio for amending soils, and to investigate the feasibility of using sawdust and coco peat as soil amendments. Peat moss, coco peat and sawdust are mixed with soil at the ratio of 0, 12.5, 50 and 100% (v/v). Among 3 organic materials with various mixture ratios, the pH of soil was the lowest in 100% peat moss and sawdust mixtures (pH 3.67 and pH 3.73, respectively), followed by pH 5.30 at 50% peat moss. The soil organic matter content are directly proportional to the mixture ratios in all three organic materials and the same trend was observed in the variation of content of exchangeable potassium in the coco peat treatments. On the contrary, the content of available phosphate, exchangeable calcium and magnesium decreased with increasing the ratio of organic materials. The nitrogen content in the leaves decreased as increasing the ratio of peat moss and coco peat in soil, but not of sawdust. The content of phosphate decreased but potassium increased as the ratio of sawdust and coco peat increased. There was no clear difference in the contents of magnesium and calcium among 3 organic materials. The plant height, stem diameter and dry weight of blueberry plants were the highest in 50 % peat moss, followed by 12.5% peat moss and 12.5% coco peat. The plants in 100% peat moss showed very poor growth. It can be concluded that peatmoss, when applied and managed appropriately, will be a good material for improving soil condition as well as securing desirable growth for blueberry. Upon coupling economic aspect, the optimum mixing ratio of peatmoss for blueberry farming is approximately 25-50%.

  • Monitoring of the dynamic changes at paddy rice agriculture is very important for agricultural sustainability. Field monitoring was performed to evaluate the soil chemical properties of 260 paddy soil samples every four years from 1999 to 2007 in Gyeongnam Province. Soil chemical properties such as pH, organic matter, available phosphate, silicate, exchangeable potassium, calcium, and magnesium contents were analyzed. The contents of exchangeable cations, and available silicate were significantly increased in 2007 compared to 1999. The chemical contents of organic matter, exchangeable potassium, and magnesium were significantly increased in acid sulfate soil, and silty clay loam compared to those of other soil types, and textures. Especially, content of organic matter was significantly increased in hill area compared to other soil topographies, while exchangeable potassium was significantly decreased. Principle component analysis (PCA) of chemical properties in paddy soils was obtained with eigenvalues > 1 summing 39.1%of variance for PC1, 20.4%of variance for PC2, and 59.5%of the total variance in the all of soil chemical properties. Therefore, principal component analysis is more effective for monitoring from chemical properties of paddy soil.

  • Livestock manure compost (LC) generally contains high content of phosphorus, therefore can be a substitute for phosphorus fertilizers. In this experiment of the cultivation of lettuce in green house, the possibility of LC as a subsitute for phosphorus fertilizer was investigated and the fertilizer efficiency of nitrogen and potassium in LC as compared with chemical N fertilizer (urea) and K fertilizer (potassium chloride) was examined. In proportion to the increase in the application rate of nitrogen fertilizer, soil pH declined, whereas EC and $NO_3$-N content became higher. The application of LC appeared to increase the soil content of organic matter, available phosphate, exchangeable calcium, magnesium and sodium more than that of chemical fertilizer. Supplementation of the K fertilizer by the lack amount from the application of LC resulted in the same exchangeable potassium content in soil with NPK plot in which N, P and K fertilizers were applied by the amount of soil test recommendation. The relationship between soil $NO_3$-N content and nitrogen application rate from fertilizer and compost showed as y=0.57717a+0.19760b+74.65 ($R^2$=0.6347) in which y is the soil $NO_3$-N content (mg $kg^{-1}$), a is nitrogen application rate from fertilizer and b is nitrogen application rate from compost (kg $ha^{-1}$), respectively. From this equation, the supply ability of $NO_3$-N into soil of LC exhibited about 34% (pig manure compost 37.0, chicken manure compost 34.7, cattle manure compost 23.3) of nitrogen fertilizer (urea).

  • Chemical properties of the soils were surveyed in the field of vinyl houses concentrated in the area of Bunpyong-dong and Shinchon-dong of Cheongju, Chilgeum-dong of Chungju and Gageum-myeon of Jungweon-gun. Chungcheongbuk-do province. 1. Content of chemical component of the soil in the vinyl house was higher than in open field. In more than half of the vinyl house soils surveyed, electric conductivity was more than 4.0 mmhos/cm and available phosphate was more than 1,000ppm. 2. Contents of availble phosphate and exchangeable potassium were increased with years of cultivation and their content of accumulation in soil were in the order of Bunpyong-dong>Shinchon-dong>Gageum-myeon>Chilgeum-dong. While their mobility was comparatively low. 3. $NO_3$-N content was remarkably higher in vinyl house soil and the older in cultivation made $NO_3$-N content higher same as the case of available phosphate content. However easy leaching of $NO_3$-N through soil profile is expected due to the fact that $NO_3$-N content was rapidly decreased by removal of polyethylene film cover from the frame of house after harvest of crops. 4. It is a tendency that various chemical contents of vinyl house soils wer higher in 1992 than in 1988~1990, especially much higher from the area of Bunpyong-dong and Shinchon-dong of Cheongju. 5. Salt accumulation in vinyl house soil has been increased with continuing cultivation. Therefore amount of fertilizer application should be controlled in order to avoid salt toxicity, quality deterioration for crop and salt contamination of ground water.

  • Japanese red pines growing along the Taebaek mountains have been called "Kangsong" and considered to be superior in growth and wood quality. An attempt was made to determine whether their boundaries for planting may be expanded by testing their early growth at eight experimental plantations of the Republic of Korea. Seeds were collected from the six different natural populations including Uljin in Kyongbuk province. For the provenance test, they were planted in eight different regions including Taean in the spring of 1987. Experimental planting was a randomized complete block design with 3 replications. Height growth was measured at the ages of 2, 4, and 6 after planting, and the diameter at root collar at the age of 6 after planting. Significant variation in height growth was observed among the plantations. The height growth measured at the age of 6 after planting showed a positive correlation with the latitude of the test plantation, and the contents of phosphate, potassium and calcium in the soil. A negative correlation, however, was observed between the longitude of the test plantation and height growth. Ponghwa provenance appeared to be the best among the provenances in that the trees from the area grew 16% better in volume growth than the average of all the trees tested in the study, while the worst one was Kyongju from which trees grew 11% less than the average.
