Amaranths (Amaranthus sp.), an endemic American crop, are now grown widely across the world. This study used 14 simple sequence repeat (SSR) markers to analyze the genetic diversity of 74 amaranth accessions from the United States, with eight accessions from Australia as controls. One hundred twenty-two alleles, averaging eight alleles per locus, were observed. The average major allele frequency, expected heterozygosity, and polymorphism information content (PIC) were 0.44, 0.69, and 0.65, respectively. The structure analysis based on genetic distance classified 77 accessions (94%) into three clusters, while five accessions (6%) were admixtures. Among the three clusters, Cluster 3 had the highest allele number and PIC values, while Cluster 2 had the lowest. The lowest FST was between Clusters 1 and 3, indicating that these two clusters have higher gene flow between them compared to the others. This finding was reasonable because Cluster 2 included most of the Australian accessions. These results indicated satisfactory genetic diversity among U.S. amaranths. These findings can be used to design effective breeding programs involving different plant characteristics.
Echinochloa (L.) P. Beauv. includes some of the noxious weeds, causing a serious yield loss when they are dominant in the fields. Identification of the Echinochloa is very difficult because many interspecific and intraspecific forms of the species are found. However, it is important to identify the species exactly and to know the genetic diversity of the species for effective weed management. This study was conducted to identify and summarize the Echinochloa species by comparing the genetic variation and relationship among Korean Echinochloa species using SSR. The genetic diversity of 107 individuals, including seven species were assessed using five SSR markers. UPGMA dendrogram generated two clades (I and II) and clade II divided again into two subclades (II-1 and II-2) whereas the model based genetic structure proposed four subpopulations. The two subpopulations were corresponded to clades I and II-1 and the other two were arranged to clade II-2 of the UPGMA dendrogram. We have concluded that E. colona and E. glabrescens might have not distributed in Korea. The biological varieties, praticola and echinata, of E. crus-galli should be treated as E. crus-galli. Korean Echinochloa should be summarized with four species, i.e., E. oryzicola, E. crus-galli, E. esculenta, and E. oryzoides.

Halla horse is crossbreed between Jeju and Thoroughbred horses and is used for riding, racing and meat production. Thus, molecular genetic studies are needed to establish and preserve the industrially valuable Halla horses. This study aimed to analyses the genetic diversity and population structure through 12 microsatellite (MS) markers for Halla and putatively related 3 breeds (Jeju, Mongolian and Thoroughbred horses). On average, the number of alleles, observed heterozygosity (Hobs), expected heterozygosity (Hexp), and polymorphic information content (PIC) among all horses were 10, 0.767, 0.799, and 0.771, respectively. Neighbor-joining tree and STRUCTURE analysis showed that Halla horses were between Thoroughbred and Jeju horses, tend to more influenced by Thoroughbred horses. Therefore, these results could be considered for use as the basic genetic breed relationships resource among the horse breeds (Jeju, Mongolian, and Thoroughbred horses) related to the origins of the Halla horse.
The vir genes are antigenic genes and are considered to be possible vaccine targets. Since India is highly endemic to Plasmodium vivax, we sequenced 5 different vir genes and investigated DNA sequence variations in 93 single-clonal P. vivax isolates. High variability was observed in all the 5 vir genes; the vir 1/9 gene was highly diverged across Indian populations. The patterns of genetic diversity do not follow geographical locations, as geographically distant populations were found to be genetically similar. The results in general present complex genetic diversity patterns in India, requiring further in-depth population genetic and functional studies.

Natural habitats of the Korean long-tailed goral (Naemorhedus caudatus) have been fragmented by anthropogenic activities in South Korea in the last decades. Here, the individual identity, genetic variation, and population differentiation of the endangered species were examined via the multiple-tube approach using a non-invasive genotyping method. The average number of alleles was 3.16 alleles/locus for the total population. The Yanggu population (1.66) showed relatively lower average number of alleles than the Inje population (3.67). Of the total 19 alleles, only seven (36.8%) alleles were shared by the two populations. Using five polymorphic out of six loci, four and six different goral individuals from the captive Yanggu (n=24) and the wild Inje (n=28) population were identified, respectively. The allele distribution was not identical between the two populations (Fisher's exact test: P<0.01). A considerably low migration rate was detected between the two populations (no. of migrants after correction for size=0.294). Additionally, the F statistics results indicated significant population differentiation between them, however, quite low (FST=0.327, P<0.01). The posterior probabilities indicated that the two populations originated from a single panmictic population (P=0.959) and the assignment test results designated all individuals to both populations with nearly equal likelihood. These could be resulted from moderate population differentiation between the populations. No significant evidence supported recent population bottleneck in the total Korean goral population. This study could provide us with useful population genetic information for conservation and management of the endangered species.

Black sea bream, Acanthopagrus schlegelii, is a commercially important fish in Korea. As a preliminary investigation into the effect of hatchery rearing for stock enhancement, we examined genetic diversity between wild and farmed black sea bream populations from Jeju using six microsatellite markers. High levels of polymorphism were observed between the two populations. A total of 87 different alleles were found at the loci, with some alleles being unique. Allelic variability ranged from 8 to 22 in the wild population and from 7 to 17 in the farmed one. Average observed and expected heterozygosities were estimated at 0.87 and 0.88 in the wild sample. The corresponding estimates were 0.83 and 0.86 in the farmed sample. Although a considerable loss of rare alleles was observed in the farmed sample, no statistically significant reductions were found in heterozygosity or allelic diversity in the farmed sample, compared with the wild one. Significant genetic heterogeneity was found between the wild and farmed populations. These results suggest that more intensive breeding practices for stock enhancement may have resulted in a further decrease of genetic diversity. Thus, it is necessary to monitor genetic variation in bloodstock, progeny, and target populations and control inbreeding in a commercial breeding program for conservation. This information may be useful for fisheries management and the aquaculture industry.

In this study, we investigated the developmental characteristics and genetic diversity of seven populations of two-spotted crickets (Gryllus bimaculatus De Geer, 1773 (Orthoptera: Gryllidae)) raised in South Korea. Regarding the developmental characteristics of the species, we observed no statistically significant difference in the weight of the nymphs in the six populations we tested. After molting, although weight differences were observed between the populations in each stage of the developmental period, the average weight for each developmental stage was constant. We also analyzed mitochondrial COI gene sequences (DNA barcoding region) of the reared crickets collected from five insect farms and two national insect rearing facilities and the resultant sequences were analyzed together with the 12 sequences from foreign countries specimens obtained from public data. We detected six haplotypes from 111 specimens, indicating a low intraspecific genetic distance (~1.8%). The most dominant haplotype was overwhelmingly haplotype 1, which was found in all South Korean specimens and four specimens from China, Indonesia, and Germany. These findings indicate that the low genetic diversity of South Korean specimens can be explained by the fact that the G. bimaculatus population imported for feed from Japan in the early 2000s became a maternal group that spread throughout cricket farms in South Korea. In order to breed healthy cricket strains, it is necessary to increase genetic diversity by importing them from other countries through appropriate quarantine procedures.
The seven selected primers OPA-02, OPA-04, OPA-18, OPD-07, OPD-08, OPD-15 and OPD-16 were used to generate unique shared loci to each species and shared loci by the two species. The hierarchical dendrogram indicates three main branches: cluster 1 (RORETZI 01~RORETZI 11) and cluster 2 (HILGENDORF 12~HILGENDORF 22) from two geographic populations of ascidians, Halocynthia roretzi and H. hilgendorfi. The shortest genetic distance displaying significant molecular difference was between individuals' HILGENDORF no. 14~HILGENDORF no. 19 (genetic distance =0.008). Ultimately, individual no. 02 of the RORETZI ascidian was most distantly related to HILGENDORF no. 21 (genetic distance=0.781). These results demonstrate that the H. roretzi population is genetically different from the H. hilgendorfi population. From what has been said above, the potential of PCR analysis to identify diagnostic markers for the identification of two ascidian populations has been demonstrated. Generally speaking, using a variety of decamer primers, this PCR method has been applied to identify specific markers particular to line, species and geographical population, as well as genetic diversity/polymorphism in diverse species of organisms.
한국 고유종이며 멸종위기에 처해 있는 어름치 Hemibarbus mylodon 3집단 (북한강, 남한강, 임진강)의 유전적 다양성 및 집단의 유전적 구조를 분석하기 위하여 AFLP분석을 실시하였다. 총 15 primer 쌍을 이용한 3집단의 AFLP 분석에서 795개의 bands가 생성되었으며, 집단 내 다형 band의 출현 빈도는 3개 집단에서 북한강 11.9%, 남한강 11.1%, 임진강 13.4%로 유사하게 나타났고, 평균 유사도는 96.6%로 나타났다. 평균 이형 접합율(0.033-0.040)과 유전적 다양도(0.036-0.043)는 매우 낮은 값을 보였다. 3개의 집단의 Pairwise distance 및 pairwise fixation index 역시 매우 낮은 값을 나타내어 본 연구에서 분석한 어름치 3개 집단은 유전적으로 매우 밀접한 근연관계를 나타내었다. 이러한 결과는 추후 본 종의 복원 및 관리에 필요한 기초자료로 활용될 수 있을 것이다.
마늘은 전 세계적으로 분포하는 다년생 초본이다. 마늘은 약리적, 경제적으로 중요한 작물이다. 야생종과 재배종의 유전관계를 ISSR 마커로 조사하였다. 또 ISSR 분석으로 이들 종의 유전적 다양도와 집단구조를 실시하였다. 한국의 세 야생 집단은 분리되어 있고 패치 분포를 보이지만 재배종에 비해 높은 유전적 다양성을 유지하고 있었다. ISS5R 마커로 야생종과 재배종의 계통관계는 잘 분리되었다. 비록 한국내 재배종 마늘이 산마늘에서 진화하였 다는 직접적 증거는 없지만 본 연구 결과 그런 가능성은 시사된다. 또한 야생종 산마늘 집단은 생식질 동정과 재배종 마늘의 진화과정에서 유익하게 쓰일 수 있다.
