Proteome Approach as a Tool for the Efficient Separation of Seed Storage Proteins from Buckwheat

  • Two-dimensional electrophoresis (2-DE) was executed to separate the seed storage proteins from the buckwheat. The proteins extracted from the whole seed proteins were better separated and observed in the use of lysis buffer. Using this method, the highly reproducible isoelectric focusing (IEF) can be obtained from polyacrylamide gels, and IEF from the polyacrylamide gel at all the possible pH range (5.0-8.0) was more easily separated than IPG (immobilized pH gradient) gels. The polyacrylamide gels in the first dimension in 2-DE was used to separate and identify a number of whole seed proteins in the proteome analysis. In this new apparatus using 2-DE, 27cm in length of plate coated with polyacrylamide gel was used and the experiment was further investigated under the various conditions.

Saccharomyces 종의 등전점 전기영동에 의한 단백질 분획상 차이에 관한 연구 (Studies on the Differentiation of Protein Patterns from Saccharomyces species by Isoelectric Focusing in Polyacrylamide Gels)

  • The whole proteins from 10 different Saccharomyces species were separated by isoelectric focusing, which was carried out in pH gradient polyacrylamide gels with the carrier ampholytes of various pH ranges. About 25 protein bands were found in the gel using pH 3.0-10.0 carrier ampholytes. In gel using pH 4.0-7.0 carrier ampholytes, the protein band of pI 6.3 was found in Sacch. cerevisiae NCYC 478, ATCC 26787, Sacch. rosei and Sacch. uvarum, but it was absent in Sacch. cerevisiae ATCC 24903, ATCC 42949, ATCC 36029, Sacch. steineri var hara, Sacch. bayanus, and Sacch. diastaticus.

  • The neurotoxin of Clostridium botulinum type B was purified from a liquid culture. The purification steps consist of ammonium sulfate precipitation of whole culture, treatment of Polymin P(0.15%, v/v), gel filtration on Sephadex G-100 at pH5.6 and DEAE-Sephadex charomatography at pH8.0. The procedure recovered 17% of the toxin assayed in the starting culture. The toxin was homogeneous by sodium dodecyl sulfate(SDS)-polyacrylamide gel electrophoresis and had a molecular weight of 163,000. Subunits of 106,000 and 56,000 molecular weight were found when purified toxin was treated with a disulfide-reducing agent and electro phoresed on SDS-polyacrylamide gels.

  • The objective of the present study was to analyze genetic variation and characteristics in rainbow trout(Oncorhynchus mykiss) using amplified fragment length polymorphism(AFLP) method as molecular genetic technique, to evaluate the usefulness of AFLP as genetic markers, and to compared the efficiency of agarose and polyacrylamide sequencing gels. The amplified products were performed by agarose and sequencing gel electrophoresis to detect AFLP band patterns, respectively. Using 9 primer combinations, total of 141 AFLP bands were produced, 108 bands(82.4%) of which were polymorphic in agarose gels. In sequencing gels, total of 288 bands were generated, and 220 bands (76.4%) were polymorphic. The level of bandsharing(BS) ranged from 0.18 to 0.32 for the 9 primer combinations tested, with a mean of 0.24. Consequently, AFLP markers of these rainbow trout could be used as genetic information such as species identification, genetic relationship or analysis of genome structure, and selection aids for genetic improvement of economically importment traits in fish species.

Clostridium botulinum Type B 독소의 정제방법에 관한 연구 (Improved Procedure for Purification of Clostridium botulinum type B Toxin)

  • Clostidium botulinum type B가 생성하는 독소를 정제할 수 있는 방법을 연구하였다. 정제과정은 독소를 ammonium sulfate로 배양액에서 침전시켜 추출한후 Polymin P를 처리하여 핵산 및 기타 단백질을 최대한 제거한 후 Sephaex G-I00에서 gel fiItration을 시키고 DEAE-Sephadex로 이온교환 크로마토그래피를 시켰다. 이러한 과정으로 정제된 독소의 회수율은 17%였으며 SDS-polyacrylamide gel electrophoresis 결과 하나의 선을 나타내 동질성을 증명하였다. 정제된 독소의 분자량은 163,000이였으며 $\beta$-mercaptoethanol을 사용하여 환원시킨 결과 분자량 106,000과 56,000의 하위 단위체로 분리되었다.

  • The morphological hetergeneity of lipopolysaccharides(LPSs) and variation in the O-side chain lengths of LPSs of Salmonella pullorum, which was serially subcultured on the brain heart infusion agar containing $100{\mu}g/m{\ell}$ of acridine orange, was analyzed in Sephadex G-50 column chromatography and silver-stained polyacrylamide gels. The biochemical differences in LPS W and LPS A0150 were identified. Increases in the contents of O-polysaccharides of LPS A0150 than those of LPS W were reflected in the profiles of chromatography and silver-stained polyacrylamide gels. In summary, LPS molecules of S pullorum A0150 appeared to be enriched in the molecules with long O-polysaccharide chains than those of LPS W.

Evaluation of Pork Myofibrillar Protein Gel with Pork Skin Gelatin on Rheological Properties at Different Salt Concentrations

  • This study was performed to evaluate the physicochemical properties of myofibrillar protein (MP) gels containing pork skin gelatin at different salt concentrations. MP gels were prepared to the different salt levels (0.15, 0.30, and 0.45 M) with or without 1.0% of pork skin gelatin. Cooking yield (CY), gel strength, shear stress were measured to determine the physical properties, and SDS-polyacrylamide gel electrophoresis, scanning electron microscopy, fourier transform infrared spectroscopy, sulfhydryl group and protein surface hydrophobicity was performed to figure out the structural changes among the proteins. The addition of gelatin into MP increased CYs and shear stress. MP at 0.45 M salt level had the highest CY and shear stress, as compared to MPs at lower salt concentrations. As the salt concentration of MP gels increased, the microstructure became the compact and wet structures, and decreased the amount of ${\alpha}-helix$/unordered structures and ${\beta}-sheet$. MP with gelatin showed a decreased amount of ${\alpha}-helix$/unordered structures and ${\beta}-sheet$ compared to MP without gelatin. The addition of gelatin to MP did not affect the sulfhydryl group, but the sulfhydryl group decreased as increased salt levels. MP mixtures containing gelatin showed a higher hydrophobicity value than those without gelatin, regardless of salt concentration. Based on these results, the addition of gelatin increased viscosity of raw meat batter and CY of MP gels for the application to low salt meat products.

감자 걀쪽바이로드(PSTV) RNA 분자의 요소농도기울기겔에서 전기영동적 이동성에 관하여 (Electrophoretic Mobilities of the Potato Spindle Tuber Viroid RNA Molecules in the Urea-Gradient Gels)

  • 바이로이드 RNA 분자를 포함한 저분자량의 식물성 RNA 분자에 대해서 0M부터 8M 까지의 요소구배를 가진 폴리아크릴아미드켈을 이용하여 전기영동적 이동성을 조사하였다. 막대모양의 바이로이드 RNA 분자는 요소구배 겔에서 전반적으로 다른 저분량의 식물성 RNA들과 마찬가지로 요소구배에 수직인 방향으로 비슷한 수준의 전기영동적 이동성을 보여주었다. 이와 달리 고리모양의 바이로이드 RNA 분자는 요소구배 겔에서 고농도의 요소구배 부분에서 보다도 저 농도의 요소구배 부분에서 더 빠르게 이동하였다. 따라서 고리모양의 바이로이드 RNA 분자는 요소구배 겔의 중간부분에서 막대모양의 바이로이드 분자의 밴드를 가로 지르는 기울기를 가진 특이한 전기영동적 이동을 나타내었다. 결과적으로 고리모양의 바이로이드 RNA 분자의 전기영동적 이동성은 0M 8M의 요소구배를 가지는 바이로이드 RNA 분자의 변성겔에서 주로 요소의 농도에 따라서 영향을 받는 것으로 보인다.

  • 건조분말상태의 인삼 및 유사 숙근성 식물간의 구별방법을 확립하기 위하여 단백질 추출과 분리방법을 달리하여 전기영동법에 의해 각 식물의 단백질 pattern 및 분자량을 추정하였던바 그 결과를 요약하면 다음과 같다. 1. 단백질분리는 증류수와 phosphate buffer로 추출하였을 때 가장 잘 되었으며 비교하기가 용이하였다. 2. 단백질추출방법에 따라 각 식물의 minor band는 약관의 차이가 있었으나 major band는 차이가 없었다. 3. 각 식물을 구별할 수 있는 band는 인삼이 45,000 및 66,000 Kd이며 사삼은 32,000∼39,000kd이고 도라지는 39,000Kd에서 double band가 특징적인 band로서 각 식물을 구별할 수 있는 단백질이었다.

산성 pH가 박테리오로돕신의 분광학적 성질에 미치는 효과 (The Effect of Acidic pH on the Spectral Properties of Bacteriorhodopsin)

  • Halobacterium halobium으로부터 분리 정제된 퍼플멤브레인을 7.5% 폴리아크릴아미드겔에 혼합시켰다. 이 겔을 사용하여 pH 변화에 따른 흡수 스펙트라와 원편광 이색성스펙트라를 얻었다. pH 7.0에서 보여준 이들 스펙트라의 성질들은 수용액내에 부유하고 있는 퍼플멤브레인으로부터 얻은 것들과 동일하였다. pH 2.7에서는 최대흡광도를 605nm에서 나타내었으며 pH 0.8에서는 565nm에서 보여주었다. 광에 노출되지 않았던 겔의 경우는 광에 노출되었던 겔과는 달리 등흡광점을 보여주었다. pH 2.7과 pH 0.8에서 측정한 원편광 이색성스펙트라는 pH 7.0에서 보여준 bilobed형을 유지하였으며 UV영역에서 분자편광도나 스펙트라의 모양도 pH에 따라 큰 영향을 받지 않았다. pH 2.7에서 생성된 bR605acid 가 퍼플멤브레인의 정상 광화학 순환기 중간 생성물인 $O^{640}$와 유사한 성질을 가진종이 아닐까 추측된다.
