상황버섯, 차가버섯, 느타리버섯 발효식초의 항산화와 항암활성 효과 (Antioxidant and Anticancer Effects of Fermentation Vinegars with Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus)

  • The antioxidant and anticancer activities of Phellinus linteus, Inonotus obliquus, and Pleurotus ostreatus according to extraction methods were measured. The contents of polyphenolic compounds were higher in fermented extracts than those in water extracts. The contents were 192.4mg/100 $m{\ell}$ for Inonotus obliquus vinegar, 186.9 mg/100 $m{\ell}$ for Phellinus linteus vinegar, and 156.5 mg/100 $m{\ell}$ for Pleurotus ostreatus vinegar, respectively. EDA(electron donating ability) in Phellinus linteus that was highest among mushrooms was 83.9% in the water extract and 96.8% in the fermented extract. The growth of stomach cancer cells, SNU-719 was inhibited 62.4%, 65.5%, and 53.0% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, respectively and also the growth of liver cancer cells, Hep3B was inhibited 67.1%, 68.3%, and 57.6% by Phellinus linteus vinegar, Inonotus obliquus vinegar, and Pleurotus ostreatus vinegar, while the growth of normal cell, DC2.4 was not affected. Even though Phellinus linteus and Inonotus obliquus showed higher inhibition in cancer cell growth, Pleurotus ostreatus can be efficiently used for antioxidant and anticancer activities due to their cheap price in the market.

느타리속(Pleurotus species) 균의 균사 배양을 위한 새로운 합성 배지 (New synthetic medium for growth of mycelium of Pleurotus species)

  • The nutritional requirements of mycelial growth for Pleurotus spp. were studied. As the carbon sources, soluble starch and dextrin, nitrogen sources, arginine and ammonium tartrate, and calcium sources, $CaCO_3$ enhanced mycelial growth. Optimum C/N ratio was 100 : 1. On the base of results from the experiment on nutritional requirements, the following receipe is suggested for growth of Pleurotus spp.; starch 15 g, arginine 3.484 g, ammonium tartrate 3.06 g, $CaCO_3$ 0.314 g, $KH_2PO_4$ 8.138 g, $K_2HPO_4$ 1.584 g, $MgSO_4{\cdot}7H_2O$ 1.5 g for P. sajor-caju and 2.5 g for P. ostreatus. $FeSO_4{\cdot}7H_2O$ 0.02 g, $ZnSO_4{\cdot}7H_2O$ 0.03 g, $MnSO_4{\cdot}7H_2O$ 0.02 g, optimum pH 6.0. This new synthetic medium is tentatively designated as Park's medium.

Genomic Differentiation Among Oyster Mushroom Cultivars Released in Korea by URP-PCR Fingerprinting

  • URP primers of 20 mer derived from repetitive sequence of rice were used to assess genetic variation of oyster mushroom consisting of 10 cultivars of Pleurotus ostreatus, two cultivars of P. florida and two cultivars of P. sajor-caju which were registered in Korea. URP2F and URP38F primers produced cultivar-specific PCR polymorphic bands in the Pleurotus species. UPGMA cluster analysis using the URP-PCR data showed that 14 Pleurotus cultivars are genetically clustered into large three groups. The URP-PCR data provided important information for more efficient breeding strategies of Pleurotus cultivars.

  • 리그닌 분해균인 느타리 버섯균(Pleurotus ostreatus) K-2946을 glucose-peptone-yeast(G-P-Y) 액체배지에서 배양 하여 망간퍼옥시데이즈를 생산하였다. 본 실험조건 하에서 리그닌 퍼옥시데이즈는 생산되지 않았다. 느타리버섯균 K-2946이 생산하는 망간 퍼옥시데이즈를 이온크로마토그래피, 겔크로마토그래피를 이용하여 정제하였다. 정제한 망간 퍼옥시데이즈의 분자량은 36400 Da이였고 pI는 3.95였다. 정제된 효소의 최적 pH는 5.0이었으며 최적온도는 $55^{\circ}C$이었다.

Characterization of an Anti-gout Xanthine Oxidase Inhibitor from Pleurotus ostreatus

  • We selected Pleurotus ostreatus from among several edible mushrooms because it has high anti-gout xanthine oxidase (XOD) inhibitory activity. The maximal amount of XOD inhibitor was extracted when the Pleurotus ostreatus fruiting body was treated with distilled water at $40^{\circ}C$ for 48 hr. The XOD inhibitor thus obtained was purified by Sephadex G-50 gel permeation chromatography, ultrafiltration, $C_{18}$ solid phase extraction chromatography and reverse-phase high-performance liquid chromatography with 3% of solid yield, and its XOD inhibitory activity was 0.9 mg/mL of $IC_{50}$. The purified XOD inhibitor was a tripeptide with the amino acid sequence phenylalanine-cysteine-histidine and a molecular weight of 441.3 Da. The XOD inhibitor-containing ultrafiltrates from Pleurotus ostreatus demonstrated dose-dependent anti-gout effects in a Sprague-Dawley rat model of potassium oxonate-induced gout, as shown by decreased serum urated levels at doses of 500 and 1,000 mg/kg, although the effect was not as great as that achieved with the commercial anti-gout agent, allopurinol when administered at a dose of 50 mg/kg.

버섯 천연조미료 제조에 관한 연구 (Development of natural Seasoning Based on Mushroom)

  • This study was conducted to provide basic data for the development of natural seasoning. The quality chatacteristics of water exacts from Pleurotus ostreatus and Lentinus edodes were determind. Also, savory taste, fragrance, color and overall preference was evaluated. The preference test on the seasonings prepared from Pleurotus ostreatus and Lentinus edodes with the addition of sea tangle was also performed. The crude protein content of dried, Lentinus edodes and Pleurotur ostreatus was 20.4% and 30.4% respectively. The total organic acid contents were higher in Plourotus ostreatus than in Lentinus edodes extracts. The total free amino acid adn total nucleotide contents were higher in Pleurotus ostreatus than in Lentinus edodes extracts. Three nucleotide, adenosine monophosphate(AMP) , guanosine monophosphate(GMP), xanthine monophosphate(XMP) were found in mushroom seasonings while inosine monophosphate(IMP) was not detected. Pleurotus ostreatus extract showed higher score in savory taste and overall acceptability than Lentinus edodes extract, Pleurotur ostreatus seasoning added with 1% sea tangle and Lentinus edodes seasoning added with 1.5% sea tangle were rated as high in overall quality In a sensory test of foods cooked with mushroom seasoning. It was shown that panels in their twenties well used to the taste of monosodium glutamate while the people in their thirties preferred the specific flavor of the mushroom seasoning.

Isolation, Regeneration and PEG-Induced Fusion of Protoplasts of Pleurotus pul-monarius and Pleurotus florida

  • Inter-specific hybridization between Pleurotus pulmonarius and P. florida was attempted through PEG-induced protoplast fusion to select a fusant. The protocol for protoplast release, regeneration and fusion in these two Pleurotus species was standardized using the variables controlling the process. The mixture of mycolytic enzymes, i.e. commercial cellulase, crude chitinase and pectinase, KCl (0.6 M) as osmotic stabilizer, pH 6 of the phosphate buffer and an incubation time of 3 hours resulted in the maximum release of protoplasts from 3-day-old mycelia of P. florida ($5.3{\sim}5.75{\times}10^{7}$ protoplasts/g) and P. pulmonarius ($5.6{\sim}6{\times}10^{7}$ protoplasts/g). The isolated protoplasts of P. florida regenerated mycelium with 3.3% regeneration efficiency while P. pulmonarius showed 4.1% efficiency of regeneration. Polyethyleneglycol (PEG)-induced fusion of protoplasts of these two species resulted in 0.28% fusion frequency. The fusant produced fruiting bodies on paddy straw but required a lower temperature of crop running ($24{\pm}2^{\circ}C$) than its parents which could fruit at $28{\pm}2^{\circ}C$. The stable fusant strain was selected by testing for the selected biochemical markers i.e. Carbendazim tolerance and utilization of the lignin degradation product, vanillin.

Mycelial Yield of Pleurotus ostreatus Using Thinned Apple, Pear, and Peach on Submerged Culture

  • The effect of thinned fruits, apple, pear and peach, on the mycelial growth of mushrooms was investigated. The growth of mycelia with the addition of thinned fruit was clearly better than that in the control for all the tested mushrooms. The growth rate of Pleurotus ostreatus was faster than any other mushroom. The optimal concentrations of thinned apple, pear, and peach in a solid culture were 1.0%, 1.0%, and 3%, respectively, while in a liquid culture the optimal concentrations were 5,0%, 3.0%, and 5.0%, respectively. When Pleurotus ostreatus was incubated in a 20-L pilot scale fermenter with 10 L of a liquid medium containing 3% thinned fruit at 25$^{\circ}C$ and 6 vvm for 10 days, the mass-production of mycelia was 74.2 g/10 L (apple), 96.2 g/10 L (pear), and 86.3 g/10 L (peach). The mycelial yield of Pleurotus ostreatus in a medium containing thinned fruit was 2 ∼ 3 times higher than that in the control.

느타리버섯 균사체 액체배양으로부터 항보체 다당 생산에 관한 연구 (Studies on the anti-complementary polysaccharides produced from submerged mycelial culture of Pleurotus sajor-caju)

  • Pleurotus sajor-caju를 각각 다른 영양원과 배양조건에 따라 배양했을 때 항보체 활성 다당 생산에 미치는 영향을 검토하였다. 그 결과 탄소원으로는 배양액에서는 lactose를 탄소원으로, 균사체로 부터의 열수 추출시에는 탄소원을 maltose로 사용했을 때 가장 높은 항보체 활성 다당을 얻을 수 있었다. 질소원으로는 ammonium phosphate(monobasic), amino acid로는 serine을 배양원으로 하였을 때 활성이 최대치를 나타내었다. 온도조건에서는 $25^{\circ}C$에서 최대활성을 나타내었으며 생산된 다당의 조성을 분석한 결과 Pleurotus sajor-caju의 배양액에서 분리한 다당은 peptidoglycan으로 나타났다.

느타리버섯 배지개발을 위한 폐면포 이용에 관한 연구 (Studies on the Media Development of Pleurotus ostreatus by Waste Cotton Stuff)

  • 느타리버섯의 볏짚 배지 대용으로 산업폐기물인 폐면포(메리야스 조각)를 이용하여 배지의 가능성이 인정된 그 결과는 다음과 같다. 균사 생장 및 수량성은 볏짚 배지에 비하여 폐면포에 미강 20%를 첨가한 배지에서 균사활착이 양호하였고 수량도 $47.5\;kg/3.3\;m^2$으로 11%가 증수되었다. 느타리버섯의 영양성분은 볏짚 배지보다 폐면포 배지에서 생성된 자실체가 조단백질이 많았고 조회분은 적었으나 조지방과 조섬유는 같은 함량이었다. 느타리버섯의 아미노산 함량은 Glutamic acid가 많았고, Cystine이 가장 적었으며 볏짚 배지보다 폐면포 배지에서 생성된 자실체가 총 아미노산과 필수아미노산 함량이 많았다.
