알라클로르와 펜디메탈린이 도라지의 발아 및 생육에 미치는 영향 (Effects of Alachlor and Pendimethalin on Germination and Growth of Balloon Flower (Platycodon grandiflorum))

  • Balloon flower (Platycodon grandiflorum) is one of minor crops in Korea. Only four ACCase inhibiting herbicides are registered for P. grandiflorum and the farmers usually use general but unregistered herbicides which can cause critical phytotoxicity. So, this study was conducted to investigate effects of alachlor and pendimethalin on germination and early growth of P. grandiflorum. To evaluate the effect of alachlor and pendimethalin, two herbicides were treated to the seeds in petri-dish and soil. In the petri-dish, alachlor completely inhibited the germination as well as pendimethalin inhibited the germination slightly but not significantly. In case of soil application, alachlor inhibited germination and plant height of P. grandiflorum significantly as well as pendimethalin inhibited plant height and root length significantly. We showed the possibility of phytotoxicity of alachlor and pendimethalin to P. grandiflorum and strongly suggest that two herbicides should not be used to the cultivation of P. grandiflorum. Unregistered herbicides should not be used for weed management and additional researches for screening of safe herbicide to minor crops should be conducted as soon as possible.

  • This study investigated the quality characteristics and antioxidant activities of Makgeolli (a traditional Korean rice wine) made with Etteum bell flower root (0.5, 1, 1.5, and 2% with steamed rice) during fermentation. The pH values of Makgeolli with Etteum bell flower root powder decreased after 3 days of fermentation and then increased after 5 days of fermentation. Sugar content decreased after 3 days of fermentation. Color evaluation showed the L values of these drinks decreased during fermentation, whereas a and b values both increased. The alcohol content of Makgeolli increased after fermentation, reaching a maximum concentration of 7.90-8.07% by the end of fermentation. Total phenolic compound contents and DPPH radical scavenging activities increased as the ratio of Etteum bell flower root increased. Sensory scores of Makgeolli fermented with 1.5% Etteum bell flower root ware greater than those of Makgeolli prepared by other treatments. Therefore, Makgeolli added with 1.5% Etteum bell flower root added considered to be the most suitable for manufacturing.

HPLC-ELSD법에 의한 길경의 platycodin D 정량분석 (Quantitative Analysis of Platycodin D from Platycodon grandiflorum by HPLC-ELSD)

  • 길경의 부탄올 분획으로부터 지표성분을 분리, 정제하기 위해 $SiO_2$ 컬럼크마토그래피를 실시한 결과 1종(화합물 1)의 단일 물질을 순수 분리하였으며 화합물 1의 구조는 $^1H$, $^{13}C$ NMR 등 분광학적 관측 결과 platycodin D로 확인되었다. Platycodin D의 HPLC 정량법을 검토한 결과, 검출기는 증기화광산란검출기(ELSD)를, 컬럼은 역상컬럼을 사용하여 길경 MeOH 추출액으로부터 효율적인 지표성분 정량이 가능하였다. 추출조건을 재검토한 결과 환류추출보다는 초음파 추출이 더 효율적이었으며 추출용매로는 80% MeOH이 다른 용매조건보다 추출효율이 우수하였다. 건조조건의 검토에서는 platycodin D의 함량이 양건이나 양건과 건조기 혼용건조 보다 건조기 단독 건조시 더 높았으며 특히 $60^{\circ}C$ 열풍건조에서 그함량이 0.083%로서 가장 높았다. 묽은 에탄올 엑스 함량도 건조기만을 이용한 열풍건조가 양건과 건조기 열풍건조를 혼용한 방법보다 4% 정도 더 높았다.

  • Present studies were carried out to produce tetraploid plants by colchicine treatment using seeds, seedlings and shoot tips of Platycodon grandiflorum in Campanulaceae. The most successful colchicine treatment for tetraploid production in P. grandiflorum was soaking treatment using 0.01 and 0.5% colchicine solution for 1 hour and 12 hours, respectively. Morphological characteristics of both diploid and tetraploid were similar, but tetraploid plants had more leaves. Compared to diploid, tetraploid had the larger stomata, but less number of stomata. Fresh weight of tetraploids was 20∼40% heavier than that of diploid.

Potent HAT Inhibitory Effect of Aqueous Extract from Bellflower (Platycodon grandiflorum) Roots on Androgen Receptor-mediated Transcriptional Regulation

  • Histone acetyltransferase (HAT) is a family of enzymes that regulate histone acetylation. Dysfunction of HAT plays a critical role in the development of cancer. Here we have screened the various plant extracts to find out the potent HAT inhibitors. The bellflower (Platycodon grandiflorum) root have exhibited approximately 30% of the inhibitory effects on HAT activity, especially p300 and CBP (CREB-binding protein) at the concentration of $100\;{\mu}g/mL$. The cell viability was decreased approximately 52% in LNCaP cell for 48 hr incubation. Furthermore, mRNA level of 3 androgen receptor target genes, PSA, NKX3.1, and TSC22 were decreased with bellflower root extract treatment ($100\;{\mu}g/mL$) in the presence of androgen. In ChIP assay, the acetylation of histone H3 and H4 in PSA promoter region was dramatically repressed by bellflower root treatment, but not TR target gene, Dl. Therefore, the potent HAT inhibitory effect of bellflower root led to the decreased transcription of AR target genes and prostate cancer cell growth with the repression of histone hyperacetylation.

풋거름작물에 따른 도라지의 생육 및 약용성분 변화 (Effect of Green Manure Crop on Growth and Medicinal Ingredients of Platycodon grandiflorum Radix)

  • 이 연구는 도라지의 유기재배를 위하여 3년근 이식 예정지에 풋거름 작물 재배에 따른 도라지 뿌리의 생육특성 및 사포닌 함량에 미치는 영향을 구명하고자 실시되었다. 생육특성에서는 기재배지에서의 뿌리의 직경 및 곁뿌리 수는 호밀 재배구에서 가장 굵거나 많았으며, 뿌리 생체중은 풋거름작물 재배구간 차이는 없었으나, 대조구에 비해 높게 나타났다. 도라지 뿌리의 무기성분 함량에서는 무기성분별 유의성이 있는 것으로 나타났으며 특히, 기재배지에서의 호밀 처리구에서 높은 함량이 나타났다. 사포닌 함량의 변화에서는 기재배지에서는 호밀 재배구에서 모든 사포닌 함량이 가장 높은 함량으로 나타났으며, 새재배지에서는 헤어리베치 재배구에서 높은 함량을 나타냈다. 대조구에 비해 풋거름작물 재배구의 기재배지와 새재배지 모든 재배구에서 사포닌의 함량이 높게 나타났다. 따라서 도라지 유기재배 시 풋거름작물에 따른 뿌리의 생육 특성 및 약리성의 변화 등의 유의적인 차이가 나타나는 것을 확인할 수 있었으며, 연차 간 재배 시 호밀 재배구에 생육 및 약리성이 우수한 것으로 나타났다.

다년생 도라지의 항암 및 면역활성 (Antitumor and Immunomodulatory Activities of the P. grandiflorum Cultivated for More Than 20 Years)

  • Platycodon radix is a dried root of Platycodon grandiflorum (P. grandiflorum) A. DC, a perennial grown on the hills and fields in Korea and Japan, or cultivated in various districts. Recently, P. grandiflorum (Changkil) has been successfully cultivated for more than 20 years and generally has been employed as folk remedy for adult diseases such as hyperlipidemia, hypertension and diabetes. We investigated various biological activities of the extracts from Changkil. When treated in vitro with B16-F1 mouse melanoma cell lines, it showed 100% laminin-binding inhibitory activities at the concentration over 0.125mg/ml. In in vivo test it showed 61.5% reduction of the solid tumor weight transplanted in mice and exhibited anticancer activity of 128% ILS against Sarcoma-180 ascites. It also increased the ratio of positive cells of natural killer cells in lymphocytic composition against Sarcoma-18O ascites and solid tumor transplanted in ICR mice when tests were carried out by FACScan method.

길경 수용액 추출물에 의한 인체 폐암세포의 성장억제 기전 연구 (Cell Cycle Arrest of Human Lung Carcinoma A549 Cells by an Aqueous Extract from the Roots of Platycodon grandiflorum)

  • Platycodi Radix, the root of Platycodon grandiflorum, commonly known as Doraji, is used as a traditional oriental medicine. Extracts from the roots of P. grandiflorum have been reported to have wide ranging health benefits. We investigated the effects of an aqueous extract from the roots of P. grandiflorum (AEPG) on the cell proliferation of human lung carcinoma A549 cells in order to understand its anti-proliferative mechanism. AEPG treatment resulted in the inhibition of cell proliferation in a concentration-dependent manner. This anti-proliferative effect of A549 cells by AEPG treatment was associated with morphological changes such as membrane shrinking, cell rounding up and inhibition of cell migration. DNA flow cytometric histograms showed that populations of both Sand G2/M phase of the cell cycle were increased by AEPG treatment in a concentration-dependent manner. AEPG treatment induced a marked accumulation of tumor suppressor p53 and a concomitant induction of cyclin-dependent kinase (Cdk) inhibitor p21 and p27. In addition, SSS treatment resulted in down-regulation of Cdk2 and Cdk4 expression. The present results indicated that AEPG-induced inhibition of lung cancer cell proliferation is associated with the blockage of S to G2/M phase progression the induction of apoptosis. Taken together, these findings suggest that P. grandiflorum has strong potential for development as an agent for prevention against human lung cancer.

구증구포 도라지 Ethyl Acetate 분획물의 예쁜 꼬마선충 내의 항산화 효과 (Antioxidant Activity of Ethyl Acetate Fraction of the Guzeunggupo-procossed Platycodon grandiflorum A. De Candolle Roots in Caenorhabditis elegans)

  • Through Caenorhabditis elegans model system, the antioxidant activity of methanol extract of the guzeunggupo-processed Platycodon grandiflorum A. De Candolle (Campanulaceae) roots was calculated. Between the methanol extracts of guzeunggupo-processed and non-processed P. grandiflorum roots, the processed P. grandiflorum root showed higher DPPH radical scavenging effect than the non-processed one. The ethyl acetate soluble fraction of the methanol extract of the guzeunggupo-processed P. grandiflorum showed the best DPPH radical scavenging activity. The ethyl acetate fraction of the processed sample was measured for the activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance by using C. elegans along with reactive oxygen species level. In addition, to confirm the regulation of the stress response gene is responsible for the increased stress tolerance of C. elegans treated by the ethyl acetate fraction of the processed sample, SOD-3 expression was measured using a transgenic strain (CF1553). Consequently, the ethyl acetate fraction of the processed sample, increased SOD and catalase activities, and decreased ROS accumulation in a dose-dependent manner. Furthermore, the ethyl acetate fraction of the processed sample-treated CF1553 worm showed higher SOD-3::GFP intensity than the control worm.

2배체와 4배체 도라지의 단백질 발현양상 비교 분석 (Proteomic Responses of Diploid and Tetraploid Roots in Platycodon grandiflorum)

  • 본 연구는 2배체와 4배체 도라지 단백질 발현지도 제작과 도라지의 생리활성에 관여하는 메카니즘을 분자적 수준에서 해석하기 위한 기초자료를 얻고자 실시하였다. 2배체 및 4배체 도라지의 단백질의 발현 양상은 배수성에 따른 특이성은 관찰 할 수 없었으며, 모두 분자량 15~100 kDa 크기, pH 4.0~8.0의 범위에 분포하는 것으로 나타났다. 동정된 39개의 단백질 중 2배체에 비해 4배체에서 2개의 단백질이 up-regulated 되었고, 37개의 단백질이 down-regulated 되었다. 단백질을 기능별로 분류한 결과, 산화환원효소의 활성(oxidoreductase activity)기능을 갖는 단백질이 23.7%의 비율로 가장 많았고 다음은 nucleotide binding 기능의 단백질이 15.8%의 비율로 높았다.
