A 2014 nationwide survey in radish fields investigated the distribution of common viruses and possible emerging viruses. Radish leaves with virus-like symptoms were collected and 108 samples assayed by RT-PCR using specific primers for Radish mosaic virus (RaMV), Cucumber mosaic virus (CMV), and Turnip mosaic virus (TuMV); 47 samples were TuMV positive, and RaMV and CMV were detected in 3 and 2 samples, respectively. No samples showed double infection of TuMV/RaMV, or RaMV/CMV, but two double infections of TuMV/CMV were detected. TuMV isolates were sorted by symptom severity, and three isolates (R007-mild; R041 and R065-severe) selected for BLAST and phylogenetic analysis, which indicated that the coat protein (CP) of these isolates (R007, R041, and R065) have approx. 98-99% homology to a previously reported TuMV isolate. RaMV CP showed approx. 99% homology to a previously reported isolate, and the CMV CP is identical to a previously reported Korean isolate (GenBank : GU327368). Three isolates of TuMV showing different pathogenicity (degree of symptom severity) will be valuable to study determinants of pathogenicity.

The incidence factors of Rice stripe virus (RSV) were analyzed by studying the population density and the viruliferous insect rate (VIR) of small brown planthopper (SBPH), the incidence of stripe disease, alternate host, and susceptible cultivar in Chungnam Province. The population of overwintering SBPH had been decreasing, but the VIR of overwintering SBPH had not been differing for three years, 2008 to 2010. No RSV was detected in the natural host plants, such as short awn, annual bluegrass, and barley. In 2009, relatively large population of SBPH with the VIR of 5.4% migrated from China. However, there was no evidence relating of migration large amount of SBPH from China in 2008 and 2010. Also the infection rate of RSV in rice was less than 1% in these periods. The cultivation area of the susceptible varieties had steadily decreased from 41% to 19% from 2007 to 2009. Therefore, the reduction factors of rice stripe disease in Chungnam Province with higher influx of inoculum could be with an appropriate forecasting and chemical control, cultivation of resistant varieties, changes in the cropping system, and the low winter-spring temperature.
Throughout the years 2008 to 2010, we analyzed approximately two thousand oriental melon samples collected from Seongju, using electron microscopy and testing by RT-PCR using primers specific for eight cucurbit-infecting viruses. Data from RT-PCR indicated that Cucumber green mottle mosaic virus (CGMMV), Watermelon mosaic virus 2 (WMV2) and Zucchini yellow mosaic virus (ZYMV) were present and the other viruses were not detected. Among them, CGMMV and WMV2 were the most prevalent pathogens. CGMMV was thought to infect oriental melon from the early growing season, and reached nearly 100% in the later of growing period. Otherwise, WMV2 emerged from June, several months later compared to CGMMV. CGMMV was detected from all aerial parts of the oriental melon including seeds, but not from the roots of the grafted pumpkin rootstock. Seed of two out of five commercial varieties were shown to be CGMMV positive. Nine varieties of pumpkins used as rootstocks were not infected with CGMMV. When the seedlings of grafted oriental melon were transplanted into pots mixed with the oriental melon debris infected with CGMMV, they were not infected by CGMMV. Cutting of pruning shear and the contact of tendrils contributed 48% and 30% to the transmission of the virus, respectively.
Two attenuated Cucumber mosaic virus (CMV) isolates, Paf-CMV and Rs2-CMV that had been selected from CMV isolates associated with satellite RNA (satRNA) were tested for cross-protection effect in pepper plants. The viruses selected as attenuated strains appeared to be identical serologically and physically to the challenge virus (Mf-CMV), but they were lower in the dilution end-point of infectivity of crude sap than Mf-CMV When symptoms were observed in several indicator plants after inoculation, Paf-CMV and Rs2-CMV were symptomless or showed mild mosaic symptoms while another satRNA isolate Ap-CMV developed severe mosaic symptoms on the leaves as Mf-CMV. The nucleotide sequences of the satRNAs were determined by sequencing full-length cDNA clones. Paf-, Rs2- and Ap-satRNAs were 386, 335, and 347 nucleotides long, respectively, The sequences were then compared with the other known Y-satRNA, revealing that nucleotide sequences of the satRNAs consisted of 5'- and 3'-terminal conserved regions. However variations occurred on the middle regions of the sequences, especially those related to symptom interference, showing significant differences between Paf-satRNA and other isolates. Infectious transcripts of Paf-satRNA and Rs2-satRNA induced mild mosaic symptoms in pepper plants when supported by genomic RNAs of Mf-CMV. Under greenhouse conditions, Paf-CMV and Rs2-CMV were tested for cross-protection effect in pepper and tobacco (Nicotiana tabacum cv, Xanthi nc) plants against Mf-CMV. No symptoms were developed on the plants vaccinated with Paf-CMV until 3 weeks after inoculation with the virulent strain; however another attenuated isolate, Rs2-CMV, showed less effectiveness in cross-protection. Depending on the concentration of the challenged virus, symptoms sometimes appeared later in the upper leaves. However, in plants challenged with low concentrations (below 0.2 mg/ml) of the challenge inoculum, symptoms caused by the virulent strain did not develop on the plants vaccinated with Paf-CMV. In the field experiments, the number of pepper plants with severe mosaic symptoms in the control plots was progressively increased after transplanting and reached approximately 50% after 50 days. On the other hand, the incidence of mosaic disease appeared very low on the plants that had received the protective inoculation with Paf-CMV.
In this study, we developed a cost and time saving one-step multiplex RT-PCR for the simultaneous detection and differentiation of swine influenza viruses (SIV) and 2009 pandemic influenza H1N1 virus (pH1N1). The one-step multiplex RT-PCR using four sets of primer was confirmed to be capable of detection of all SIV subtypes and differential diagnosis of major SIV subtype H1, H3 and pH1N1 on individual or mixed viral culture samples. The sensitivity of the multiplex RT-PCR was determined to be at least $2^{-6}$$HA/25{\mu}L$ of the presented SIVs, providing sufficient efficacy for a routine SIV monitoring in diagnostic laboratories. In addition, compared with the conventional RT-PCR methods that cannot avoid the carryover DNA contamination, the developed RT-PCR applied with the uracil DNA glycosylase (UNG) system was proven to prevent a false positive reaction by carryover contamination of the pre-amplified DNA. In conclusion, the one-step RT-PCR with UNG system could be applicable to detect and differentiate of SIV from the viral cultures without worry of carryover DNA contamination in clinical laboratories.
Tomato spotted wilt virus (TSWV) was occurred at 8 areas including Naju, Suncheon, Younggwang, Youngam, and Shinan in Jeonnam province and the crops of Younggwang were severely damaged by TSWV. The hot pepper (Capsicum annuum), bell pepper (Capsicum annuum v ar. angulosum) and tomato (Solanum lycopersicon) in greenhouse and hot pepper in open field were infected by TSWV. Especially, hot pepper was severely damaged by TSWV infection. The survey data indicated that 1.1-30% in the nursery field at Naju, Suncheon, and Jangheung were infected by TSWV. Plants were infected by TSWV from early June to August. However, TSWV-infected seedlings from nursery fields showed the disease symptoms from May after transplanting. In pepper greenhouses, Frankliniella occidentalis was more dominant insect vector than Frankliniella intonsa. But in open field, the population of insect vector was opposed to greenhouse. In addition, the removal of weeds was able to delay the incidence of TSWV via side-window of greenhouse in Winter. Taken together, the control of weed and insect vector nearby side-window of greenhouse is important to prevent TSWV infection of plants.
Leaves of twenties radish (Raphanus sativus L.) inbred lines were mechanically inoculated with Turnip mosaic virus (TuMV) strain HY to evaluate TuMV resistance of the radish inbred lines. The inoculated radish plants were incubated at $22^{\circ}C{\pm}3^{\circ}C$ and resistance assessment was examined using symptom development for 4 weeks. Based on the reactions of differential radish inbred lines, 16 radish lines were produced mild mosaic, mottling, mosaic and severe mosaic symptoms by TuMV infection. These results were confirmed by RT-PCR analysis of TuMV coat protein gene, suggesting that TuMV is responsible for the disease symptoms. Four resistant radish lines did not induce systemic mosaic symptoms on upper leaves and chlorosis in stem tissues for 4 weeks, showing they were symptomless by 8 weeks. Further examination of TuMV infection in the 4 radish lines showed no TuMV infection in all systemic leaves. These results suggest that the 4 radish lines are highly resistant to TuMV.

Egg yolk immunoglobulin (IgY) is much widely used in medical fields, but its use in serology of plant viruses is much limited. We produced an IgY against pepper mild mottle virus (PMMoV) and applied it to several serological tests. Polyclonal antibodies were obtained from the egg yolk of chicken immunized with a total of 2mg of purified PMMoV over 2 months. The titers of antibodies were measured with the ring-test over six months after the first injection. The highest.titers of IgY was 1/2,560 at 2 months after the first injection. Approximately 60-80 mg of IgY were obtained from one egg yolk. Using the IgY, 1ng/ml of purified PMMoV was detected with the indirect ELISA. Gelrite gel double diffusion test, ELISA and tissue immuno-binding assay employing IgY gave similar sensitivity and specificity to those of IgG developed in rabbit. Therefore, the IgY which can be obtained in large quantities from a chicken, might be useful for the antibody production and the serology of plant viruses.

Thrips infest hot peppers (Capsicum annuum) cultivating in open fields and give serious economic damages. This study reports their yearly occurrence from transplanting to harvest at three different places in Andong, an intense hot pepper-cultivating area. Two main occurrence peaks were detected in June and September. Two dominant thrips were the flower flowers, Frankliniella occidentalis and F. intonsa, which comprised of over 87% of the total occurring thrips. Other thrips did not follow the occurrence peaks of the two dominant species. Tomato spotted wilt virus (TSWV) was detected from the dominant two species, in which higher viruliferous rate was recorded in F. intonsa. Although the hot peppers were the resistant varieties against TSWV, some of them cultivating in the fields exhibited a characteristic disease symptom infected by the virus over the growing seasons. TSWV was isolated from the viruliferous thrips and assessed in NSs sequences encoded in S RNA segment of the virus. Compared to the known resistance breaking (RB) strains, the TSWV isolated from the viruliferous thrips in Andong did not show any RB mutations.
