This study was performed in Korea to get serological information for bovine herpesvirus 1 (BoHV-1), most commonly found in cattle. Antibodies against BoHV-1 were examined by targeting infectious bovine rhinotracheitis (IBR) in unvaccinated and vaccinated cattle, using viral neutralization (VN) test. In 2013, among 261 sera collected from IBR-unvaccinated herds, 7 sera (2.7%) were found seropositive and their VN titers were ranging from 1:4 to 1:32. Among 315 sera collected from IBR-vaccinated herds in large capacity farms, 303 sera (96.2%) were found to be seropositive for BoHV-1 and their VN titers were in the range of 1:4 to 1:2048. It was found that the IBR-vaccinated herds had higher levels of VN titer than IBR-unvaccinated herds. The results indicated that it may be due to heavy vaccination in vaccinated herds and no or a little infection in unvaccinated herds. At the end of the study it was concluded that although the seropositivity in IBR-unvaccinated herds was low, the monitoring of IBR should be continuously practiced to control and prevent the disease because of exportation of living cattle causing its nationwide outbreaks.
The distribution of mosquitoes collected in the Cheongju area from 2016-2018 were compared. The collection sites were established at a residential area, migratory bird sanctuary, and cowshed using black light (BL) and BG sentinel traps. Culex pipiens was the most collected species in the residential area, and Aedes vexans was the most collected species in the migratory bird sanctuary and cowshed in all years. The BG trap collected more individuals than did the BL trap in the residential area and migratory bird sanctuary. In total, 22,679 (10 species) mosquitoes were collected in 2016, and 6,502 (8 species) and 6,803 (9 species) mosquitoes were collected in 2017 and 2018, accounting for 3.49- and 3.33-fold decreases, respectively. The relationship between meteorological conditions and mosquito density, according to various variables, was not significant different. The Chaoyang virus was found in Aedes vexans and Culex pipiens collected at the cowshed in 2016 and 2018, respectively. Based on the results of this study, it is considered that continuous surveillance of mosquitoes should be performed for controlling mosquito populations and mediating diseases spread by this vector.
In 2005, average temperature was lower, and average rainfall was less than those of previous year. The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Bacterial blight, bacterial grain rot, and panicle disease of rice, black rot of pear, and white rot and bitter rot of apple were severe. Especially, brown rot of rice occurred four times higher than those of previous year. Panicle blight of rice increased about 3 times, compared with the previous year, presumed that the higher rainy days, rainfall and RH promoted spread of the fungal pathogens to panicles of rice. The diseases of rice leaf blast, sudden wilt syndrome, downy mildew and powdery mildew of chinese melon in plastic greenhouse, and virus diseases of hot pepper occurred distinctly less than those of the previous year. Another diseases surveyed occurred similar or less.
Although there were differences depending on strains and environmental conditions, virus infected oyster mushroom, Pleurotus florida showed slow growth on sawdust and rice straw substrates. Many harmful microorganisms occurred on the cultural bed of virus infected isolates. Pinhead formed too densely or too rarely sometimes. Stipes of the mushrooms were long slightly bent with small cap. The virus infected mushrooms formed branch on their stipes. The first pinheading days of the infeeted mushroom were later than that of healthy culture. The loss of fruit body yield was about 30% compared with that of virus free culture. Spores which contaminated by viruses damaged more seriously than the other source. The authors would like to call these symptoms as a new disease in oyster mushroom culture in the world.
Since 1980, burley tobacco plants grown mainly in the western hat of the Korea. have shown two new types of disease symptom. Both symptoms were found to be caused by two different PVY strains : the vein banding type by a PVY strain designated as PVY-VB and necrosis on leaf veins by a PVY strain designated as PVY-VN, Identification of the PVY strains was based on host range test. aphid( Myzus Persicae) transmission test, physical properties, serology, and observation of virus particle morphology. The virus particles were measured to be about 730 nm without any difference in shape or dimensions between the two strains. Both strains also gave a positive reaction to the PVY antiserum in SDS-agar gel double diffusion test. These strains, however, gave a negative reaction to the tobacco etch virus and tobacco vein mottling virus antisera.
In many organisms, trehalose has been Down as an energy source and a protectant against various environmental stresses such as desiccation, freezing, heat and osmotic pressure. Previously, we have isolated and characterized the genes encoding trehalose-6-phosphate synthase (ZrTPS1) and trehalose-6-phosphate phosphatase (ZrTPS2) from one of the most osmotolerant yeasts, Zygosaccharomyces rouxii. We have also generated transgenic plants by co-introduction of ZrTPS2 and ZrTPS2 into potato plant (ZrTPS2-2A-ZrTPS1 plant) in an attempt to metabolically engineer trehalose in the transgenic plant using the foot-and-mouth disease virus(FMDV) 2A system and to generate drought resistant crop plants. In this research, we assayed previously generated the ZrTPS2-2A-ZrTPS1 plant biofunctionally by drought treatment, and measured the amount of trehalose in the ZrTPS2-2A-ZrTPS1 transgenic plants. The ZrTPS2-2A-ZrTPS1 transgenic plant showed strong drought resistance in spite of little or no accumulation of transgenic in he transgenic plant compared with control plant.

In July 2020, total RNA was extracted from passion fruit (Passiflora edulis) leaves showing virus symptoms such as chlorotic spots and vein banding in Haenam, South Korea. Cucumber mosaic virus (CMV)-HN2 was identified through reverse transcription polymerase chain reaction and sequencing analysis. To confirm the biological characteristics of the CMV infecting passion fruit, 10 indicator plants were inoculated with CMV-HN2, and the results showed a typical CMV symptoms. Phylogenetic analysis based on the amino acid of the coat protein (CP) of CMVs revealed that the CMV passion fruit isolates belonged to subgroup I, among which CMV-HN2 belonged to subgroup IA. Additionally, CMVs isolated from passion fruit in Korea have amino acid sequence variation between the subgroup. Among them, CMV-HN2 had four to eight amino acid differences in CP from other CMV isolates from passion fruit. These results confirm the presence of genetic diversity in the CPs of passion fruit CMV isolates.
Chinese yam (Dioscorea opposita cv. Jang-Ma) plants showing necrotic mosaic symptom were collected from their growing fields in Andong, Euisong, Gunwi and Daegu, Korea. Direct negative stainning method by electron microscope showed filamentous particles of about 660 nm in length. Immunosorbent electron microscopy (ISEM) revealed filamentous particles of 660nm decorated with antiserum of Chinese yam necrotic mosaic virus (ChYNMV). The virues purified partially were used to isolate viral RNA as template for RT-PCR to amplify about 1.2 kbp of 3'-terminal region (coat protein, 3'-UTR) with ChYNMV specific and oligo-dT primers. Amino acids sequences of amplified CP genes revealed that the viruses shared 97.9% similarity with ChYNMV (AB044386) wh ich was known as the member of Macluravirus. So the viruses from Chinese yam (D. opposita cv. Jang-Ma) plants were identified as ChYNMV. Comparing the CP amion acid sequences of ChYNMV strains with other macluraviruses such as Cardamon mosaic virus (CdMV), Narcissus latent virus (NLV) and Maclura mosaic virus (MacMV) revealed that N-terminal was the most varialbe region and conserved regions were present within the genus Macluravirus.
Incidence of virus diseases in red pepper of open field in Yeongyang-Gun, Gyeongbuk Province was investigated by reverse transcription-polymerase chain reaction in 2012-2016. The infection rate of Cucumber mosaic virus (CMV), Broad bean wilt virus 2 (BBWV2), Pepper mottle virus, Potato virus Y, Pepper mild mottle virus and Tomato spotted wilt virus was 46.1%, 41.5%, 2.0%, 2.0%, 4.4% and 0.1% respectively. Incidence rate of single and mixed infection was 31.2% and 62.6%. Most of single infections were CMV and BBWV2. Among mixed infections, the incidence rate of CMV+BBWV2 mixed infection was the highest as 49.3% and most of mixed infections of triplex and tetraplex included CMV+BBWV2 mixed infection. CMV single infection caused mosaic, chlorosis, yellowing and vein necrosis and BBWV2 single infection induced cholosis and mosaic. CMV+BBWV2 mixed infection caused severe mosaic with chlorosis or malformation.
Cucumber mosaic virus(CMV) and Broad bean wilt virus 2(BBWV2) were isolated from Gentiana scabra plants showing typical mosaic and yellowing symptoms. When the inoculum of mosaic symptom propagated in Nicotiana benthamiana was inoculated to primary leaves of Vigna sinensis, the local lesions of different types was developed. Type one produced a small necrotic spot(SNS) of pinpoint type, while the other one showed a large necrotic spot(LNS) of halo type. LNS on primary leaves of V. sinensis was also induced by inoculum from yellowing symptom on G scabra. Single lesion from SNS induced a typical mosaic symptom on N. Benthamiana. On the other hand, LNS produced a chlorotic ring symptom on inoculated leaves and mosaic plus necrotic ringspot on upper leaves of N. benthamiana. An isolate of CMV from SNS and BBWV2 from LNS were detected by using dsRNA analysis, RT-PCR and agar gel double-diffusion test. Thus, our results should provide a tool of a simple method for discrimination from mixed infected plants by CMV and BBWV2.
