양송이 수확 후 배지로부터 식물생장촉진세균의 분리 및 생육특성 (Isolation and Characterization of Plant Growth Promoting Rhizobacteria from Waste Mushroom bed from Agaricus bisporus)

  • An auxin-producing bacteria (3YN11-02) was isolated from waste mushroom bed from Agaricus bisporus of Chungnam Buyeo-Gun area. The strain 3YN11-02 was identified as a novel species belongs to Rahnella aquatica by a chemotaxanomic and phylogenetic nalysis. The isolate was confirmed to produce indole-3-acetic acid (IAA) which is one of auxin hormone by TLC and HPLC analysis. When the concentration of IAA was assessed by performing HPLC quantity analysis, the maximal $290mg\;L^{-1}$ of IAA detected in ether fraction extracted from the culture filtrate which was cultured in R2A broth containing 0.1% tryptophan for 24 h at $35^{\circ}C$. The molecular weight of the main peak obtained by LC-mass analysis was correspondent well to 175, that of IAA. To investigate the growth promoting effect of crop, when the culture broth of R. aquatica 3YN11-02 was infected onto water culture and seed pot of mung bean, the adventitious root induction and root growth of mung bean were 2.0 times higher than control.

Heterologous Expression of the Hot Pepper ABA 8'-Hydroxylase in Escherichia coli for Phaseic Acid Production

  • The CYP707A family genes encoding ABA 8'-hydroxylase catabolize abscisic acid (ABA), a plant stress hormone that plays an important role in stress condition, such as drought, heat, cold and salinity. Phaseic acid (PA) is a catabolic product of ABA. Recent studies have shown that PA is important for the physiological functions in plants. It is also a neuroprotective molecule that protects against ischemic brain injury in mice. To obtain enzymes for the PA production, four CaCYP707A genes (CaCYP707A1, CaCYP707A2, CaCYP707A3 and CaCYP707A4) were isolated from hot pepper. They were heterologously expressed in Escherichia coli. Among them, CaCYP707A2 showed significantly higher expression levels in both the membrane fraction and the soluble fraction. Preferred redox partners were investigated to improve the efficiency of CaCYP707A2's catalytic reaction, and NADPH-cytochrome P450 reductase (CPR) from hot pepper (CaCPR) was preferred over other redox partners (i.e., rat CPR and ferredoxin reductase/ferredoxin). The production of 8'-hydroxy ABA and PA by ABA hydroxylation activity was confirmed in CaCYP707A2 from both membrane and soluble fractions. Therefore, CaCYP707A2 is the first identified plant CYP protein that is expressed a soluble form in cytosolic fraction having stable activity. Taken together, we propose a new CYP707A protein with industrial applications for PA production without additional modifications in E. coli heterologous expression.

Nitrate enhances the secondary growth of storage roots in Panax ginseng

  • Background: Nitrogen (N) is an essential macronutrient for plant growth and development. To support agricultural production and enhance crop yield, two major N sources, nitrate and ammonium, are applied as fertilizers to the soil. Although many studies have been conducted on N uptake and signal transduction, the molecular genetic mechanisms of N-mediated physiological roles, such as the secondary growth of storage roots, remain largely unknown. Methods: One-year-old P. ginseng seedlings treated with KNO3 were analyzed for the secondary growth of storage roots. The histological paraffin sections were subjected to bright and polarized light microscopic analysis. Genome-wide RNA-seq and network analysis were carried out to dissect the molecular mechanism of nitrate-mediated promotion of ginseng storage root thickening. Results: Here, we report the positive effects of nitrate on storage root secondary growth in Panax ginseng. Exogenous nitrate supply to ginseng seedlings significantly increased the root secondary growth. Histological analysis indicated that the enhancement of root secondary growth could be attributed to the increase in cambium stem cell activity and the subsequent differentiation of cambium-derived storage parenchymal cells. RNA-seq and gene set enrichment analysis (GSEA) revealed that the formation of a transcriptional network comprising auxin, brassinosteroid (BR)-, ethylene-, and jasmonic acid (JA)-related genes mainly contributed to the secondary growth of ginseng storage roots. In addition, increased proliferation of cambium stem cells by a N-rich source inhibited the accumulation of starch granules in storage parenchymal cells. Conclusion: Thus, through the integration of bioinformatic and histological tissue analyses, we demonstrate that nitrate assimilation and signaling pathways are integrated into key biological processes that promote the secondary growth of P. ginseng storage roots.

Temperature Effect on the Growth Parameters of Rice during Vegetative Period

  • Temperature is a crucial environmental factor for rice cultivation due to the climate change and can influence the rice growth and development. Therefore, the effect of temperature on plant growth characters was examined during the vegetative stage. Plants were grown under three different temperatures: 23℃/13℃ for 18℃, 26℃/16℃ for 21℃ and 29℃/19℃ for 24℃ in the phytotron. The temperature was treated after transplanting and ended in early panicle initiation stage. Heading date of the two varieties were strongly affected by the temperature and were delayed in the 18℃. The plant height in the 18℃ was 21 % shorter than the 21℃ and 24℃ and the tiller and leaf number were increased in the 18℃. All the growth rates of the characters were the slowest in 18℃. The stem dry weight was significantly increased in 18℃. Nitrogen content was increased in the leaves of 18℃ whereas available phosphate and potassium content was found to be increased in the stems of 21℃ and 24℃. OsNRT 2.1 was overexpressed in the leaves and stems of 18℃ and OsNRT2.3a could be expressed in 18℃ and 21℃ temperatures whereas more expressed in 21℃. OsPT1 and OsPT6 could be expressed in the leaf of 18℃ and 24℃ but could be expressed in the stem of 18℃. OsHAK1 and OsHAK5 could be overexpressed in the leaves and stems of 18℃. For hormone, OsCKX2 gene was found to be up regulated in the leaves of 18℃ and OsIAA1 gene could be expressed more in the stem of 24℃.

  • The Hairy labrador tea (Ledum palustre L. var. diversipilosum Nakai) is a evergreen small shrub, belonging to the Ericaceae and mainly distributed along the alpine areas of Hamgyeong-do in North Korea. Through seed physiological research on North Korean plants, we intend to obtain basic data for the development of mass propagation methods and use them for vegetation restoration. The internal and external morphological characters of the seed were observed using scanning electron microscopy or stereoscopy, and full seeds were selected through X-ray test. Seeds were cultured on a growth condition at 15/6℃ and 25/15℃ to examine the germination response according to temperature, and the number of germinated individuals was examined every 24 hours to calculate germination percentage and mean germination time. Low-temperature treatment (4℃) and hormone treatment (GA3) were performed to investigate the effect of dormancy breaking, and analysis of variance (ANOVA) was performed on the experimental results using SAS 9.4. The seeds of the Hairy labrador tea were elliptical-narrow in shape, and the epidermal cell wall pattern was reticulated. The final germination rate (FGP) was the highest at 58.9% under the condition of low temperature treatment at 25/15℃. The average number of days to germination (MGT) was the highest at 25.1 days without treatment at 15/6℃, and the average of the last germination days was confirmed to be about 13 days. As a result of low temperature treatment, it was confirmed that the final germination rate (FGP) increased by 43.3%. The significance of the final germination rate (FGP) was verified according to the pretreatment at 25/15℃.

Ethanol Extracts from the Roots of Reed Prevent Skin Hyperpigmentation, Wrinkle Formation and Dryness

  • The roots of reed (Phragmites australis) were used in traditional medicine to treat respiratory problems, including symptoms such as high fever and cough. In this study, we identified the active ingredient from 70% EtOH reed root extract, and evaluated the whitening, wrinkle improvement and moisturizing effects. The content of p-coumaric acid, the active ingredient of the roots of P. australis, was slightly lower in 70% EtOH extract than in 100% EtOH extract. However, 70% EtOH reed root extract showed similar or higher effect in reducing power, DPPH, hydrogen peroxide scavenging, and nitric oxide scavenging activity compared to 100% EtOH extract. Moreover, 70% EtOH reed root extract markedly inhibited melanogenesis in B16F10 cells treated with α-melanocyte-stimulating hormone. 70% EtOH reed root extract significantly inhibited the mRNA expression of matrix metalloproteinase-1 (MMP-1) and reduced elastase activity in HDF human dermal fibroblasts. In addition, 70% EtOH reed root extract ameliorated hyaluronic acid synthase-2 (HAS-2) expression induced by ultraviolet B (UVB) stimulation in HaCaT keratinocytes. The results of this study suggest that 70% EtOH reed root extract has potential as a functional cosmetic material related to whitening, wrinkle improvement, and moisturizing.

Vegetarian Diets and Estrogen Metabolism in Korean Premenopausal Women

  • It has been suggested that dietary fat increases the risk of breast cancer by elevating serum estrogen concentrations. However, studies on a relationship between fat intake and breast cancer risk have shown contradictory results, possibly because the levels of fat intake in study populations were too high to observe differences. Also, the effect of other dietary factors may present. The present study was performed to investigate the relationship between diet and estrogen metabolism in premenopausal women whose usual fat intake is relatively low compared to their western counterparts. Twenty lacto-ovo vegetarians (LOV) and twenty omnivores participated in the study. Three day food records including a Saturday or a Sunday were used to estimate nutrient intake. Serum lipids, estradiol, sex-hormone binding globulin, and urinary estradiol were measured. Study results showed $24.8\%$ and $20.9\%$ of energy intake were provided from fat in omnivorous and LOV subjects, respectively. Serum and urinary levels of estradiol were two times higher in omnivores. Fat intake was not related to either serum estradiol nor urinary estradiol when the Spearman correlation coefficient analysis was performed. Carbohydrate, total dietary fiber and soluble dietary fiber intakes were negatively related to serum estradiol concentration. Legumes, vegetables and fruit consumption showed significantly negative relationships with both serum and urinary estradiol concentrations. These results indicate lower estrogen availability may be associated with plant food­based diets in premenopausal women whose usual diets contain less than $25\%$ of energy as fat.

Classification and Expression Profiling of Putative R2R3 MYB Genes in Rice

  • MYB genes, comprising group of related genes found in animal, plant, and fungal genomes, encode common DNA-binding domains composed of one to four repeat motifs. MYB genes containing two repeats (R2R3) constitute largest MYB gene family in plants. R2R3 MYB genes play important roles in regulation of secondary metabolism, control of cell shape, disease resistance, and hormone response. Eight-four R2R3 MYB genes were retrieved from rice genome for functional characterization of MYB genes. Analysis of MYB domains revealed each MYB domain contains three ${\alpha}$-helices with regularly spaced tryptophan residues. R2R3 MYB genes were divided into four subfamilies based on phylogenic analysis result. Real-time PCR analysis of 34 MYB genes revealed 12 MYB genes were highly expressed in seeds than in leaves, whereas 4 genes were highly expressed in leaves.

Identification of Phenylethyl Alcohol and Other Volatile Flavor Compounds from Yeasts, Pichia farinosa SKM-l, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59

  • Three strains of yeasts, Pichia farinosa SKM-1, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59, produced volatile flavor compounds during fermentation. To investigate these volatile flavor compounds, the liquid culture broth of the three yeast strains were extracted with methylene chloride, and then GC and GC-MS analyses were conducted. Flavor analyses revealed that 5, 12, and 15 kinds of volatile compounds were isolated, and 4, 8, and 11 volatile flavor compounds were identified, respectively. Phenylethyl alcohol was identified with the common volatile flavor compound of Pichia farinosa SKM-1, Pichia anomala SKM-T, and Galactomyces geotrichum SJM-59. 1H-indole-3-ethanol, a precursor of plant growth hormone, was identified from Pichia anomala SKM-T.

Production of Indole-3-Acetic Acid by the Cyanobacterium Arthrospira platensis Strain MMG-9

  • The filamentous cyanobacterium Arthrospira platensis strain MMG-9 was isolated from a rice field. The ability of this strain to synthesize the bioactive compound indole-3-acetic acid (IAA) was demonstrated. IAA was extracted from the culture of A. platensis strain MMG-9 and its identity was confirmed by thin-layer chromatography (TLC) as well as by high-performance liquid chromatography (HPLC). The IAA precursor L-tryptophan was required for IAA biosynthesis. Released IAA increased with the increase of the initial concentration of L-tryptophan in the medium and with the incubation time. A. platensis strain MMG-9 accumulated more IAA than it released into the medium. The bioactivity of the secreted IAA was shown by its effect on the formation of roots by Pisum sativum. There was a significant positive effect of the supernatant of cultures of A. platensis strain MMG-9 on the number of lateral roots of P. sativum, whereas a negative effect on root length was observed.
