Transcriptome and Flower Color Related Gene Analysis in Angelica gigas Nakai Using RNA-Seq (RNA-seq을 이용한 참당귀의 전사체 분석과 꽃 색 관련 유전자 분석)

  • Angelica gigas Nakai (Korean danggui), a member of the Umbelliferae family, is a Korean traditional medicinal plant whose roots have been used for treating gynecological diseases. Transcriptomics is the study of the transcriptome, which is the complete set of RNA transcripts that are produced by the genome, using high-throughput methods, such as microarray analysis. In this study, transcriptome analysis of A.gigas Nakai was carried out. Transcriptome sequencing and assembly was carried out by using Illumina Hiseq 2500, Velvet and Oases. A total of 109,591,555 clean reads of A. gigas Nakai was obtained after trimming adaptors. The obtained reads were assembled with an average length of 1,154 bp, a maximum length of 13,166 bp, a minimum length of 200 pb, and N50 of 1,635 bp. Functional annotation and classification was performed using NCBI NR, InterprotScan, KOG, KEGG and GO. Candidate genes for phenylpropanoid biosynthesis were obtanied from A.gigas transcriptome and the genes and its proteins were confirmed through the NCBI homology BLAST searches, revealing high identity with other othologous genes and proteins from various plants pecies. In RNA sequencing analysis using an Illumina Next-Seq2500 sequencer, we identified a total 94,930 transcripts and annotated 71,281 transcripts, which provide basic information for further research in A.gigas Nakai. Our transcriptome data reveal that several differentially expressed genes related to flower color in A.gigas Nakai. The results of this research provide comprehensive information on the A.gigas Nakai genome and enhance our understanding of the flower color related gene pathways in this plant.

Antioxidative and Antimicrobial Qualities of Different Pare of the Dandelion Plant (Taraxacum officinale) from Different Habitats (민들레의 서식지와 부위별 항산화 및 항균 활성 연구)

  • To classify the physiological activity by habitat and part of Dandelion (Taraxacum officinale), antioxidative and antimicrobial activities were investigated from methanol extracts of Dandelion. Dandelion was taken from Uiryeong (in Kyungnam province) and from Seoul, and antioxidative and antimicrobial activities were measured for three different parts of the dandelion: the flower, leaf and root. Antioxidative activities of methanol extracts from Dandelion were examined by a DPPH test, and antimicrobial activities were examined against 5 microorganisms: Bacillus subtilis and Bacillus cereus as G(+) microorganisms, and Salmonella typhimurium, Staphylococcus aureus and Eschrichia coli as G(-) microorganisms. No significant differences were found in terms of antioxidative and antimicrobial activities between the dandelions in Uiryeong and the dandelions in Seoul. Antioxidative activity, however, was higher in the flower of the dandelion, irrespective of habitat. Further research with other physiological factors (besides antioxidative and antimicrobial activities) is necessary to shed light on dandelion physiological function and to enhance the utilization of dandelion.

  • The experiment was conducted to obtain basic information on breeding of flue-cured tobacco varieties. Nine cultivars and partial diallel set of 36 F1 hybrides were grown at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute in 1983. Estimated heritability in the narrow-sense ranged from 66.21% to 94.12% for yield, leaves per plant, days to flower, leaf weight, leaf width, leaf shape, nicotine content and reducing sugar content, while that for stalk height, leaf length, midrib weight and midrib width ranged from 28.12% to 56.25%. The genotypic correlations were positive among yield, leaf weight, leaf length, leaf width, midrib weight and midrib width in the middle leaves, days to flower, nicotine content and reducing correlated with leaf length than leaf width. At the path coefficient analysis of the effect of leaf and midrib characters upon yield according to stalk position, leaf weight, leaf width and midrib width in the middle leaves, and leaf weight, and leaf width in the top leaves showed high direct effects.

  • The present experiments were conducted to investigate some of the factors affecting the number of haploids derived from anther culture of Nicotiana tabacum. Anther - donor plants grown under controlled environment room at 3$0^{\circ}C$ yielded more haploid than room at 18, 25 and 26-22-18$^{\circ}C$ in anther culture. Donor plants starved of fertilizer yielded more haploids as compared to those of the well fed with fertilizer in anther culture. Pretreatment of exercised flower bud at 5$^{\circ}C$ was shown to be more effective in anther culture than pretreatment at 7 and 1$0^{\circ}C$, and the optimum temperature and period of pretreatment were 4 or 6 days at 5$^{\circ}C$.

  • In order to obtain the basic information for the successful breeding of aromatic tobacco (Nicotiana tabacum L.), the heterosis, combining ability and gene distributions in 28 Fl hybrids by diallel crosses between Korea local and Oriental varieties were investigated. The yield were higher, stalk height, leaf length and leaf width were larger and days to flower was shorter for the Fl hybrids when compared to mid-parent. Estimates of general combining ability were significant for all characters investigated, and specific combining ability were significant for yield, stalk height, leaves Per Plant, leaf width and days to flower. In vr-wr graph, partial dominane was exhibited by all characters investigated.

  • Four known triterpenoids and one sterol glycoside were isolated from Viburnum opulus var. clavescens for. sterile flowers. By the spectral data analysis, we determined to be the structures of isolated compounds as 𝛼-amyrin (1), ursolic aldehyde (2), maslinic acid (3), ursolic acid (4) and 𝛽-sitosterol-3-O-glucoside (5). Among the isolated compounds, we revised 13C-NMR chemical shifts of ursolic aldehyde (2) using DEPT and HMBC spectra analysis. 𝛼-Amyrin (1), ursolic aldehyde (2), maslinic acid (3) and 𝛽-sitosterol-3-O-glucoside (5) were isolated for the first time from this plant.

  • Crop productivity would be determined by relative capacity of source and sink. The study was carried out to measure the effects of shading and flower bud pinching on growth and yield of Atractylodes japonica Koidz. Shading treatment after transplanting was done at 100%, 65, 45 or 25% level of natural light intensity but the pinching treatment was to remove all the inflorescences formed after June 10, July 10, August 10 or none. Growth and yield at early and late stages were periodically measured. At the early stage, shading mainly influenced fresh weight rather than morphological characters. At the late stage, however, severer shading decreased numbers of stems, leaves, roots, fraction and total fresh weight. 65% shading from July 1 to September 30 after skipped the emerging and early growth stages showed nearly same growth and morphological characters but more rhizome yield per unit area than non-shading. In flower bud pinching treatment, earlier pinching increased number of roots, fraction fresh weights per plant and rhizome yield per unit area.

A New Rose Cultivar 'Suryeo' with White-pink Flower Color (백·분홍색 절화장미 '수려' 육성)

  • A new rose cultivar was bred by crossing the red-yellow standard cultivar 'Konfetti' and pink standard cultivar 'Laser' at the Chungcheongbuk-do Agriculture Research and Extension Services. The cross was completed in 2003, after varietal characteristics was tested for 3 years from 2005 to 2007, 'Suryeo' was finally selected in 2007 and registered on the grant of plant variety rights in 2009. The cultivar was developed for a standard-type cut flower with white-pink petals. 'Suryeo' produced 149 stems/$m^2$ in a year and had 64 petals per flower. This new cultivar needed 48 days to flower and showed 12 days vase life. The length of cut stem was about 78 cm, and the fresh weight of stem was 50 g. This cultivar showed high resistance against powdery mildew.

Phenolic Compounds from the Flower Buds of Camellia japonica

  • Hot water extracts of Camellia japonica flower buds were found to have the higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity than the other solvent extracts. Five phenolic compounds were isolated and purified from the ethyl acetate soluble-neutral fraction of hot water extracts by Sephadex LH-20 column chromatography and octadecyl silane-high performance liquid chromatography using the guided assay of DPPH radical scavenging. Based on mass spectrometer and nuclear magnetic resonance, the isolated compounds were identified as p-hydroxybenzaldehyde (1), vanillin (2), dehydroxysynapyl alcohol (3), 7S,7'S,8R,8'R-icariol $A_2$ (4), and (-)-epicatechin (5). Four compounds (1-4) except for 5 were newly identified in this plant. Their DPPH radical scavenging activities as 50% scavenging concentration decreased in order to 5 $(20\;{\mu}M)>{\alpha}-tocopherol$ $(29\;{\mu}M)>4$ $(67\;{\mu}M)>3$ $(72\;{\mu}M)>1=2$ ($>250\;{\mu}M$). These results indicate that the antioxidant effect of the hot water extract of C. japonica flower buds may partially due to 5 isolated phenolic compounds.
