Negative evidence on the transgenerational inheritance of defense priming in Arabidopsis thaliana

  • Defense priming allows plants to enhance their immune responses to subsequent pathogen challenges. Recent reports suggested that acquired resistances in parental generation can be inherited into descendants. Although epigenetic mechanisms are plausible tools enabling the transmission of information or phenotypic traits induced by environmental cues across generations, the mechanism for the transgenerational inheritance of defense priming in plants has yet to be elucidated. With the initial aim to elucidate an epigenetic mechanism for the defense priming in plants, we reassessed the transgenerational inheritance of plant defense, however, could not observe any evidence supporting it. By using the same dipping method with previous reports, Arabidopsis was exposed repeatedly to Pseudomonas syringae pv tomato DC3000 (Pst DC3000) during vegetative or reproductive stages. Irrespective of the developmental stages of parental plants that received pathogen infection, the descendants did not exhibit primed resistance phenotypes, defense marker gene (PR1) expression, or elevated histone acetylation within PR1 chromatin. In assays using the pressure-infiltration method for infection, we obtained the same results as above. Thus, our results suggest that the previous observations on the transgenerational inheritance of defense priming in plants should be more extensively and carefully reassessed.

The Calmodulin-Binding Transcription Factor OsCBT Suppresses Defense Responses to Pathogens in Rice

  • We previously isolated the OsCBT gene, which encodes a calmodulin (CaM)-binding protein, from a rice expression library constructed from fungal elicitor-treated rice suspension cells. In order to understand the function of OsCBT in rice, we isolated and characterized a T-DNA insertion mutant allele named oscbt-1. The oscbt-1 mutant exhibits reduced levels of OsCBT transcripts and no significant morphological changes compared to wild-type plant although the growth of the mutant is stunted. However, oscbt-1 mutants showed significant resistance to two major rice pathogens. The growth of the rice blast fungus Magnaporthe grisea, as well as the bacterial pathogen Xanthomonas oryzae pv. oryzae was significantly suppressed in oscbt-1 plants. Histochemical analysis indicated that the hypersensitive-response was induced in the oscbt-1 mutant in response to compatible strains of fungal pathogens. OsCBT expression was induced upon challenge with fungal elicitor. We also observed significant increase in the level of pathogenesis-related genes in the oscbt-1 mutant even under pathogen-free condition. Taken together, the results support an idea that OsCBT might act as a negative regulator on plant defense.

Involvement of the OsMKK4-OsMPK1 Cascade and its Downstream Transcription Factor OsWRKY53 in the Wounding Response in Rice

  • Plant has possessed diverse stress signals from outside and maintained its fitness. Out of such plant responses, it is well known that mitogen-activated protein kinase (MAPK) cascade plays important role in wounding and pathogen attack in most dicot plants. However, little is understood about its role in wounding response for the economically important monocot rice plant. In this study, therefore, the involvement of MAPK was investigated to understand the wounding signaling pathway in rice. The OsMPK1 was rapidly activated by wounding within 10 min, and OsMPK1 was also activated by challenge of rice blast fungus. Further analysis revealed that OsMKK4, the upstream kinase of OsMPK1, phosphorylated OsMPK1 by wounding in vivo. Furthermore, OsMPK1 directly interacted with a rice defense-related transcription factor OsWRKY53. To understand a functional link between MAPK and its target transcription factor, we showed that OsMPK1 activated by the constitutively active mutant $OsMKK4^{DD}$ phosphorylated OsWRKY53 in vitro. Taken together, components involving in the wounding signaling pathway, OsMKK4-OsMPK1-OsWRKY53, can be important players in regulating crosstalk between abiotic stress and biotic stress.

산화제 공급 방법에 따른 메탄올-산소 촉매연소기 특성 (Characteristics of Methanol-O2 Catalytic Burner according to Oxidant Supply Method)

  • Recently, a fuel reforming plant for supplying high purity hydrogen has been studied to increase the operation time of underwater weapon systems. Since steam reforming is an endothermic reaction, it is necessary to continuously supply heat to the reactor. A fuel reforming plant needs a methanol-O2 catalytic burner to obtain heat and supply heat to the reformer. In this study, two types of designs of a catalytic burner are presented and the results are analyzed through the experiments. The design of the catalytic burner is divided into that the O2 supply direction is perpendicular to the methanol flow direction (Design 1) and the same as the methanol flow direction (Design 2). In case of Design 1, backfire and flame combustion occurred in the mixing space in front of the catalyst, and in the absence of the mixing space, combustion reaction occurred only in a part of the catalyst. For above reasons, Design 1 could not increase the exhaust gas temperature to 750℃. In Design 2, no flashback and flame combustion were observed, the exhaust gas could be maintained up to 750℃. However, the O2 distributor was exposed to high temperatures, resulting in thermal damage.

사이버공격에 대비한 심층방호 특성을 만족하는 단방향 광통신 구조 설계 (Design of Uni-directional Optical Communication Structure Satisfying Defense-In-Depth Characteristics against Cyber Attack)

  • 원전 계측제어계통의 제어기기에 대한 사이버공격은 원자로 정지 또는 방사선 누출 등의 심각한 재앙을 유발할 가능성이 있다. 그러므로 원전 규제기관 및 산업표준의 요건에 따라 심층방호 특성을 고려한 사이버보안 대책설계가 원전 계측제어계통 설계에 요구된다. 본 논문에서는 사이버공격에 대한 원전 계측제어계통의 심층방호 대처 설계시 필수적으로 요구되는 단방향통신 구조를 구현하기 위한 설계방법을 제안한다. 제안한 단방향 통신 구조 구현방법은 통신보드 내에 구현되었다. 단방향 통신을 지원하는 통신보드는 시험을 통하여 심층방호 설계에 필요한 단방향 통신 구조를 제공하며, 사이버공격에 강인한 특성을 갖는 것으로 분석되었다. 이러한 사이버공격에 대비한 심층방호 대처 설계 및 단방향 통신을 지원하는 통신구조는 일체형원자로(SMART) 계측제어계통 설계에 적용되었다.

금속연료인 과립화붕소의 입도에 따른 연료과농 고체 추진제 특성 연구 (A Study of Fuel-rich Solid Propellant Characteristic for Boron-bead Particle Size)

  • 공기흡입형 추진기관의 가스발생기에 사용하기 위한 연료과농(Fuel-rich) 고체 추진제의 특성에 대한 연구를 수행하였다. 일반적인 고체 추진제는 평균 60%이상의 산화제를 포함하는 데, 연료과농 고체 추진제를 개발히기 위해 산화제 함량을 30%내외로 낮추고 매우 높은 부피당 열량을 가지는 비정질 붕소를 입자화(Bead)하여 금속연료 함량을 증가시켜 고체 추진제의 제조 공정성 및 연소속도 특성을 확인하였다. 과립화붕소의 입도가 작을수록 추진제 제조 공정에서 초기 점도가 높아지고, 입도가 클수록 연소속도 및 압력지수가 증가하는 것을 확인할 수 있었다.

Overexpression of a Pathogenesis-Related Protein 10 Enhances Biotic and Abiotic Stress Tolerance in Rice

  • Pathogenesis-related proteins play multiple roles in plant development and biotic and abiotic stress tolerance. Here, we characterize a rice defense related gene named "jasmonic acid inducible pathogenesis-related class 10" (JIOsPR10) to gain an insight into its functional properties. Semi-quantitative RT-PCR analysis showed up-regulation of JIOsPR10 under salt and drought stress conditions. Constitutive over-expression JIOsPR10 in rice promoted shoot and root development in transgenic plants, however, their productivity was unaltered. Further experiments exhibited that the transgenic plants showed reduced susceptibility to rice blast fungus, and enhanced salt and drought stress tolerance as compared to the wild type. A comparative proteomic profiling of wild type and transgenic plants showed that overexpression of JIOsPR10 led to the differential modulation of several proteins mainly related with oxidative stresses, carbohydrate metabolism, and plant defense. Taken together, our findings suggest that JIOsPR10 plays important roles in biotic and abiotic stresses tolerance probably by activation of stress related proteins.

Role of RIN4 in Regulating PAMP-Triggered Immunity and Effector-Triggered Immunity: Current Status and Future Perspectives

  • As sessile organisms, plants have developed sophisticated system to defend themselves against microbial attack. Since plants do not have specialized immune cells, all plant cells appear to have the innate ability to recognize pathogens and turn on an appropriate defense response. The plant innate immune system has two major branches: PAMPs (pathogen associated molecular patterns)-triggered immunity (PTI) and effector-triggered immunity (ETI). The ability to discriminate between self and non-self is a fundamental feature of living organisms, and it is a prerequisite for the activation of plant defenses specific to microbial infection. Arabidopsis cells express receptors that detect extracellular molecules or structures of the microbes, which are called collectively PAMPs and activate PTI. However, nucleotidebinding site leucine-rich repeats (NB-LRR) proteins mediated ETI is induced by direct or indirect recognition of effector molecules encoded by avr genes. In Arabidopsis, plasmamembrane localized multifunctional protein RIN4 (RPM1-interacting protein 4) plays important role in both PTI and ETI. Previous studies have suggested that RIN4 functions as a negative regulator of PTI. In addition, many different bacterial effector proteins modify RIN4 to destabilize plant immunity and several NB-LRR proteins, including RPM1 (resistance to Pseudomonas syringae pv. maculicola 1), RPS2 (resistance to P. syringae 2) guard RIN4. This review summarizes the current studies that have described signaling mechanism of RIN4 function, modification of RIN4 by bacterial effectors and different interacting partner of RIN4 in defense related pathway. In addition, the emerging role of the RIN4 in plant physiology and intercellular signaling as it presents in exosomes will be discussed.

독성을 제거한 재조합 리신 백신 개발 및 효능평가 (Development of Non-toxic Recombinant Ricin Vaccine and Evaluation of Vaccine Efficacy)

  • Ricin is a highly toxic protein which is produced in the seeds of the castor oil plant. Ricin toxin A chain has ribosomal RNA N-glycosylase activity that irreversibly hydrolyses the N-glycosidic bond of the adenine residue at position 4324 within the 28S rRNA. In this study, we developed non-toxic recombinant ricin vaccine(R51) in E. coli expression system, and evaluated efficacy of the R51 according to adjuvants. When the R51 was administered using aluminum hydroxide as an adjuvant, the vaccine efficacy was higher than that of TLR agonists or aluminum phosphate. Because it is time-consuming to administer the vaccine three times at three-week intervals, we investigated the survival rate and antibody titer of mice according to the change of time interval of vaccination. Interestingly, there was no difference in survival rate and antibody titer when R51 was administered at 0, 1, and 3 weeks or 0, 2, and 4 weeks compared to when administered at 0, 3, and 6 weeks. Therefore, the developed R51 vaccine is promising to protect soldiers from Ricin attack.
