Expression of Low Temperature Regulated Gene H28 in Solanum tuberosum L.

  • Leaf dise explants of Solanum tuberosum cultivar. Desiree and Atlantic, were infected with a Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic and low temperature regulated gene (H28) for transformation. regenerated multiple shoots were selected on a medium containing kanamycin and carbenieillin after exposure to Agrobacterium. Both PCR analysis of NPT Ⅱ, H28 genes and northern blot analysis indicated that the genes coding for the enzyme were successfully integrated into the potato genome and could be expressed in potato plants.

  • The phytoremediation of trinitrotoluene, nitroglycerine, pentaerytritoltetranitrate in plant tissue cultures of Solanum aviculare, Rheum palmatum and Populus simonii were studied. All above mentioned explosives were degradated to to less toxic products and finally mineralized or bound to the cell wall.

Effects of Several Additives on Plant Regeneration from Leaf Disc Culture of Solanum tuberosum L. (감자의 잎 절편 배양시 몇 가지 배지의 첨가물이 식물체 재분화에 미치는 영향)

  • The effects of several additives on plant regeneration were investigated from leaf disc culture of S. tuberosum cv. Atlantic which is known as poor in regeneration ability. The presence of 2 g/L casein hydrolysate significantly enhanced shoot regeneration. Addition of 10~20 $\mu$M $AgNO_3$, not only increased the frequency of shoot regeneration but also maintained the leaf disc green presumably by the inhibitory action of ethylene accumulation in vitro. Decrease of sucrose levels to below 3% significantly increased the degree of regeneration. The addition of CuSO$_4$had no effect on shoot regeneration.

  • In multicellular organisms, including humans, understanding expression specificity at the tissue level is essential for interpreting protein function, such as tissue differentiation. We developed a prediction approach via generated sequence features from overrepresented patterns in housekeeping (HK) and tissue-specific (TS) genes to classify TS expression in humans. Using TS domains and transcriptional factor binding sites (TFBSs), sequence characteristics were used as indices of expressed tissues in a Random Forest algorithm by scoring exclusive patterns considering the biological intuition; TFBSs regulate gene expression, and the domains reflect the functional specificity of a TS gene. Our proposed approach displayed better performance than previous attempts and was validated using computational and experimental methods.

  • Plant biotechnology including plant tissue culture is the most important science to rescue human beings from the crisis of biosphere of the earth which they will face in the 21st century. Global area other than Asia of trangenic crops increased more than 16 times from 1996 to 1998. In Asian countries, micropropagation using tissue culture techniques has been well developed and has contributed to agricultural production. Although researches on levels of laboratory and test field trial of transgenic crops have been performed actively in some Asian countries, areas of growing transgenic crops are still small in Asia countries except in China. In this paper, the status of plant biotechnology in global and Asian countries are reviewed and the future prospect of plant biotechnology in Asia and roles of Korean and Japanese plant scientists in it are discussed.

Detection of Potato Spindle Tuber Viroid Using RT-PCR Technique (RT-PCR 기법을 이용한 감자 걀쭉 바이로이드 (Potato Spindle Tuber Viroid)의 검정)

  • Potato spindle tuber viroid(PSTVd) RNAs were isolated from PSTVd-inoculated potato cv. Superioc and carried out RT-RCR with reverse transcriptase and PSTVd specifie primer pair desigened to amplify the 356 nucleotides of PSTVd genome. As a result, 356 nucleotides PCR products were amplified from PSTVd-inoculated potato cv. Superior. The 356 nucleotides DNA fragment was indeed the PSTVd geneby sequencing analysis. PSTVd could be successfully detected from infected leaf and tuber tissue of potato by using RT-PCR technique. Especially PSTVd was more effectively detected when both downstream and upstream primer were used than only downstream primer was used in RT reaction.

Production of Baicalin, Baicalein, and Wogonin on Plant Tissue Culture of Scutellaria baicalensis (황금(Scutellaria baicalensis) 조직배양에 의한 Baicalin, Baicalein 및 Wogonin 생산)

  • Scutellaria baicalensis Georgi (SJ) is a perennial plant and its root has been used in oriental traditional medicine for treatment of fever, inflammation, diarrhea and anticancer effect, etc. In this study, plant tissue culture system for SJ was developed. Stem piece of younger plant was optimum explant for callus induction and growth on MS medium supplemented with NAA 1.0 ㎎/L plus BA 0.5 ㎎/L. Plantlet regeneration through callus culture was well on MS medium containing NAA 1.0 mg/L. SJ has been known biologically active substances such as baicalin, baiclein, and wogonin. This study was carried out to examine the effect of plant growth regulators for production of baicalin, baicalein, and wogonin through callus culture. The HPLC pattern of callus extract was identical to that of standard solution, it shows that the callus produced by tissue culture has the same flavonoids composition of SJ. Baicalin, baicalein, and wogonin production was 471.5~52.8 ㎍/g, 137.6~4.0 ㎍/g, and 16.6~1.3 ㎍/g, respectively, on MS media with nine different plant growth regulator combinations. This may indicate that plant tissue culture of SJ possible to produce the biologically active substances effectively

Characteristics in Tissue Cultured Plants of Chinese foxglove(Rehmannia glutinosa) (지황 조직배양주의 수량성과 성분함량 특성)

  • Chinese foxglove(Rehmannia glutinosa) is receiving much attention as one of the principal medicinal crops and the crud drug. This study was conducted to obtain the basic breeding information of Chinese foxglove derived from tissue culture. To compare plant characteristics between local variety and tissue cultured seedlings, plant growth and root yield has been investigated. In addition, catalpol and free sugar contents were also analized. The ability for the storage of root stock originated from tissue culture seedlings were better than that of local variety. The growth and root yield of in vitro propagated plants were superior to those of conventionally propagated plants. Root yield of 1-year-old and 2-year-old seedlings investigated 112% and 246% respectively than Seacheon local shows 384 kg/10 a. Although there were no significant different between tissue culture seedlings and conventionally propagated one, slight decline of component contents in tissue culture plants were still existed. Investigated sugar content among Seocheon local, 1-year-old tissue culture seedlings, and 2-year-old tissue culture seedlings were 1.86%, 1.21%, and 1.10% respectively. Catalpol content as one of standard materials indicates 0.46% in Seocheon local and 0.40% in tissue cultured ones.

Characterization of Pectate Lyase Produced by Erwinia rhapontici During Growth in Host Plant Tissue (Erwinia rhapontici가 기주식물 조직에서 생산한 Pectate Lyase의 특성)

  • Erwinia rhapontici causes soft-rot disease in a number of plants such as rhubarb, onion, hyacinth and garlic. Pectate lyase (Pel) depolymerizes pectin and other polygalacturonates, which is though to play a role in bacterial invasion of plants. Pel activity was not detected in E. rhapontici cultured in a minimal salts medium containing glycerol, polygalacturonate, or citrus pectin as a carbon source. However, when sterilized potato tuber and Chinese cabbage slices were added to minimal salts polygalacturonate (0.5%) medium, E. rhapontici produced pectate lyase enzyme. Also Pel activity was consistently detected from macerated potato tubers, Chinese cabbage leaves, lettuce leaves and celery petioles tissue. Pel in the extract of macerated Chinese cabbage caused by E. rhapontici strain 1, resulted in electrolyte loss, tissue maceration and cell death of potato tuber tissue. These results indicate that E. rhapontici produces pectate lyase only in the presence of non-diffusible plant components, and that this enzyme probably contributes to its pathogenicity.

High Frequency Regeneration of Plantlets from Seedling Explants of Asteracantha longifolia (L.) NEES

  • Plantlet regeneration in Asteracantha longifolia(L.) Nees (Acanthaceae), a medicinal herb has been achieved from seedling explants on basal MS medium. Three different seedling explants including node, internode and leaf segments on used. Of these three explant, leaf explants gave better response for both callus mediated organogenesis and direct multiple shoot induction. Number of explants showing differentiation of shout buds was higher on MS media supplemented with BA compared to kinetin. MS medium fortified with BA ($2.0mgl^{-1}$) and NAA ($0.5mgl^{-1}$) was found to be most suitable for both callus mediated organogenesis and elongation of shouts. The elongated shoots were successfully routed on MS medium fortified with NAA or IBA. Among them $0.1mgl^{-1}$ NAA or $0.2mgl^{-1}$ IBA provides better response for rhizogenesis. Regenerated plantlets were successfully established in soil where 85.4% or them developed into morphologically normal and fertile plants. RAPD profiling using four decamer primers confirmed the genetic uniformity of the regenerated plantlets and substantiated the efficacy and suitability of this protocol for in vitro propagation of A. longifolia.
