This study was performed to investigate the effects of Korean native plant diets on lipid and cadmium (Cd) metabolisms and also antioxidative enzyme activities in rats. Seventy male Sprague-Dawley rats weighing 210.3$\pm$2.7g were blocked into ten groups according to body weight. Rats were raised for four weeks with diets containing 0 or 0.04%(w/w) cadmium chloride and 5%(w/w) plant powder-Ssook(Artemisia princeps var. orientalis HARA), Chamchwi(Aster scaber THUNB), Gomchwi(Lingularia fischeri TUBCZ). or Solbeerum (Portulaca oleracea LINNE). Food intake, weight gain, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significantly lower in Cd-exposed groups, food efficiency ratio, and weights of liver, kidney, spleen and epididymal fat were significanlty lower in Cd-exposed groups. Plasma total lipid level, liver total lipid, cholesterol and triglycerde concentrations, and fecal total lipid, cholesterol and triglyceride excretions were decreased by Cd administration. Total lipid and triglyceride concentration in plasma and liver were lower in Chamchwi groups together with increased fecal total lipid and triglyceride excretions. Cholesterol concentration in plasma and liver were lower in Gomchwi groups with increased fecal cholesterol excretions. Activities of red-blood-cell superoxide dismutase(SOD), and liver catalase, SOD and glutathione peroxidase (GSH-px) were decreased significantly by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentrations of plasma and liver were increased by Cd administration. Thiobarbituric acid reactive substance(TBARS) concentration of plasma and liver were increased by Cd administration. Administration of plant diets decreased plasma and liver catalase, SOD and GSH-px activities in Cd-free diet groups and increased them in Cd-administered groups. Plasma and liver TBARS concentrations were decreased in animals fed plant diets, with Chamchwi showing the most effective antioxidative capacity. The concentration of Cd accumulated in blood and tissue decreased in Cd-exposed groups fed plant diets. Liver Cd concentration were lower in the Chamchwi and Gomchwi groups, and kidney Cd levels were lower in the Ssook and Chamchwi groups. Among them, Chamchwi was the most effective in Cd detoxification. In conclusion, Chamchwi and Gomchwi were effective in lowering tissue lipid levels, with Chamchwi having the greatest antioxidative and detoxifying effects.
To induce adventitious buds, hypocotyl and cotyledonary explants from 7 to 10 day-old seedlings of lettuce (Lactuca sativa L.: two Japanese cultivars of crisphead lettuce and four Korean cultivars of leaf lettuce) were cultured or Murashige and Skoog (MS) and Schenk and Hildebrandt (SH) media supplemented with BA and NAA in the light for five weeks. Cotyledonary explants produced adventitious shoots at greater frequencies than hypocotyl explants. MS medium was more favorable to adventitious shoot formation than HS medium. Combination of 0.5 mg/L BA and 0.1 to 1 mg/L HPh in MS medium led to the greatest frequency (86%) in adventitious shoot formation. Creator than 95% of shoots excised from explants were rooted when cultured on MS basal medium.

Mesenchymal stem cells (MSCs) are useful candidates for tissue engineering and cell therapy. Physiological cell environment not only connects cells to each other, but also connects cells to the extracellular matrix that provide mechanical support, thus exposing the entire cell surface and activating signaling pathways. Hydrogel is a polymeric material that swells in water and maintains a distinct 3-dimensional (3D) network structure by cross linking. In this study, we investigated the optimized cellular function for canine adipose tissue-derived MSCs (cAD-MSCs) using hydrogel. We observed that the expression levels of Ki67 and proliferating cell nuclear antigen, which are involved in cell proliferation and stemness, were increased in transwell-hydrogel (3D-TN) compared to the transwell-normal (TN). Also, transforming growth factor-${\beta}1$ and SOX9, which are typical bone morphogenesis-inducing factors, were increased in 3D-TN compared to the TN. Collagen type II alpha 1, which is a chondrocyte-specific marker, was increased in 3D-TN compared to the TN. Osteocalcin, which is a osteocyte-specific marker, was increased in 3D-TN compared to the TN. Collectively, preconditioning cAD-MSCs via 3D culture systems can enhance inherent secretory properties that may improve the potency and efficacy of MSCs-based therapies for bone regeneration process.

Plants are inherited spatial and temporal coordination systems in their growth and differentiation processes which are precisely governed by the two interlocked control systems; autogenous and environmental. Looking into the overall course of plant development from molecular to organismal level, it can be comparable to a concerto for plant hormones, environmental stimuli and plant genomic orchestra conducted by an unidentified virtuoso. Some of the recent significant attempts to puzzle out the mystery of the life processes of plant development are briefly reviewed. The revolutionary advances in understanding the mystic processes are contemporarily achieved by the application of various molecular techniques. The characterization of plant genomes is now attained through recombinant DNA approaches, and the sensitive detection of specific gene products during the plant development is perimitted by the immunochemical procedures. However, along with the recognition of underlying molecular events such as developmental changes in gene expression and hormone-receptor interrelation associated with tissue sensitivity to hormones, more emphasis should be placed upon the physiological approaches of organismal level for the understanding the correlative systems of the developmental processes of plants as intact eukaryotic organisms.
The effect of phosphorus nutrition on the content of deoxyribonucleic acid(DNA), ribonucleic acid(RNA), crude protein and plant growth of soybean plant(Glycine max, Merr.) was studied. Yields of the above-and under-ground parts of the soybean plant in terms of dry weight, the amounts of crude protein, RNA and DNA continued to increase with increasing phosphorus supply. The amounts of RNA and crude protein were highest in the leaf tissues where most intensive growth was taking place. The relationships among DNA, RNA, crude protein and plant growth appeared to consist of the central dogma which has immortalized, while DNA in plant tissue was subject to charges cuased by external environmental facters such as phosphorus nutrition.
Grapevine leafroll-associated 3 closterovirus (GLRaV-3) is phloem limited virus, and one of the most severe viral diseases found in Korea. However, nonhomogenous distribution and low concentration and seasonal variations of GLRaV-3 in grapevines remain as main problems which prevent the introduction and molecular biology or serology experiments. Virus-infected plantlet in vitro was obtained from node tissue cuttings, which was GLRaV-3 infected 'kyoho' vines. The amount of purified virus was highest in vitro plantlet. Moreover, viruses seem to be relatively homogeneously distributed in all organs including leaf, stem and callus derived from in vitro plantlets. RT-PCR detection using in vitro plantlet tissue as template was most effective. When comparing ELISA to RT-PCR, RT-PCR detection was 1,000 times as effective as ELISA. These results can be explained by improved quality such as tenderness or less tannins in plantlet in vitro. In conclusion, until infected herbaceous host will be available, tissue culture can be usefully adopted as a technique for a good source of GLRaV-3 closterovirus for further studies.

Nerine was originated from South Africa and around 30 kinds of species are distributed in worldwide. In present study we have characterized the condition of storage and growing conditions on the bulb propagation of Nerine by the tissue culture. Result indicated that the percentage of decrement, decay and sprouting of bulblets in the tissue culture was high at the higher temperature compare to lower temperature. The optimum conditions for storage and time were 12 weeks at $15^{\circ}C$ which were having 86.4% sprouting rate. The sprouting rate at different temperature and acclimation time, using bulblets in the tissue culture, were optimum at early June when temperature is relatively higher($22^{\circ}C$) that facilitate to breaking the dormancy. Growth characteristics and capacity of photosynthesis of Nerine were better at red and blue mixed lightness.

The objective of this study was to carry out treatment of various plant hormones in order to determine morphological and genetic variation degree of tissue-cultured strawberry. The cultivar used in this experiment was 'Goha' and 'Seolhyang', the plant hormones used for experiment were benzyladenine (BA), N-(2-Chloro-4 pyridyl)-N'-phenylurea (CPPU) and thidiazuron (TDZ), and the concentrations were 0.5, 1.0, 2.0, $4.0mg{\cdot}L^{-1}$ with each hormone. The BA treatment of the proliferation efficiency of tissue-cultured strawberry 'Goha' and 'Seolhyang' was the highest. When processing BA, CPPU and TDZ, morphological variation and genetic variation happened in strawberry 'Goha' and 'Seolhyang', especially, the variations appeared highly in CPPU treatment. The genetic variation in 'Goha' appeared at the concentration more than BA $0.5mg{\cdot}L^{-1}$ as 1.1%, appeared at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 15.3%, and at the concentration of TDZ $2.0mg{\cdot}L^{-1}$ as 1.2%. The genetic variation in 'Seolhyang' appeared at the concentration of BA $4.0mg{\cdot}L^{-1}$ as 2.3%, and at the concentration of CPPU $0.5mg{\cdot}L^{-1}$ as 14.3%. Therefore, CPPU should not be treated during strawberry tissue culture, and BA and TDZ should be treated at low concentration.
Oilseed rape (Brassica napus L.) is an important crop due to its high oil content in the seed. Recently, the demand for the improvement of crop for biodisel energy source is increased as oil prices in the world has increased dramatically. Until now, oilseed rape breeding was carried out by cross-hybridization between different varieties and related germplasms. However, like as many other crops, the application of tissue culture and gene transformation systems has been introduced into oilseed rape breeding program including the development of transgenic canola plants. In this study, we reviewed a history of tissue culture and genetic transformation research in oilseed rape plants and indicated some important aspects for the production of transgenic oilseed rape plants.
Plant tissue culture is a technique that has invariably been used for various purposes such as obtaining transgenic plants for crop improvement or functional analysis of genes. However, this process can be associated with a variety of genetic and epigenetic instabilities in regenerated plants, termed as somaclonal variation. In this study, we investigated mutation spectrum, chromosomal distributions of nucleotide substitution types of single-nucleotide polymorphisms (SNPs) and insertions/deletions (InDels) by whole genome re-sequencing between Dongjin and Nipponbare along with regenerated plants of Dongjin from different induction periods. Results indicated that molecular spectrum of mutations in regenerated rice against Dongjin genome ranged from $9.14{\times}10^{-5}$ to $1.37{\times}10^{-4}$ during one- to three-month callus inductions, while natural mutation rate between Dongjin and Nipponbare genomes was $6.97{\times}10^{-4}$. Non-random chromosome distribution of SNP and InDel was observed in both regenerants and Dongjin genomes, with the highest densities on chromosome 11. The transition to transversion ratio was 2.25 in common SNPs of regenerants against Dongjin genome with the highest C/T transition frequency, which was similar to that of Dongjin against Nipponbare genome.
