Pleurotus ostreatus, P. eryngii, and Flammulina velutipes are major edible mushrooms that account for over 89% of total mushroom production in Korea. Recently, Agrocybe cylindracea, Hypsizygus marmoreus, and Hericium erinaceu are increasingly being cultivated in mushroom farms. In Korea, the production of edible mushrooms was estimated to be 614,224 ton in 2013. Generally, about 5 kg of mushroom substrate is needed to produce 1 kg of mushroom, and consequently about 25 million tons of spent mushroom substrate (SMS) is produced each year in Korea. Because this massive amount of SMC is unsuitable for reuse in mushroom production, it is either used as garden fertilizer or deposited in landfills, which pollutes the environment. It is reasonably assumed that SMS includes different secondary metabolites and extracellular enzymes produced from mycelia on substrate. Three major groups of enzymes such as cellulases, xylanases, and lignin degrading enzymes are involved in breaking down mushroom substrates. Cellulase and xylanase have been used as the industrial enzymes involving the saccharification of biomass to produce biofuel. In addition, lignin degrading enzymes such as laccases have been used to decolorize the industrial synthetic dyes and remove environmental pollutions such as phenolic compounds. Basidiomycetes produce a large number of biologically active compounds that show antibacterial, antifungal, antiviral, cytotoxic or hallucinogenic activities. However, most previous researches have focused on therapeutics and less on the control of plant diseases. SMS can be considered as an easily available source of active compounds to protect plants from fungal and bacterial infections, helping alleviate the waste disposal problem in the mushroom industry and creating an environmentally friendly method to reduce plant pathogens. We describe extraction of lignocellulytic enzymes and antimicrobial substance from SMSs of different edible mushrooms and their potential applications.
Endophytic bacterial communities of tidal flat plants antagonistic to oomycete plant pathogens were studied by the isolation of 256 root colonizing endophytic bacteria from surface-disinfected root tissues of six plants ($Rosa$$rugosa$, $Suaeda$$maritima$, $Vitex$$rotundifolia$, $Carex$$scabrifolia$, $Glehnia$$littoralis$ and $Elymus$$mollis$) growing in a tidal flat area of Namhae Island, Korea. To understand the antagonistic potential, an $in$$vitro$ antagonistic assay was performed to characterize and identify strains that were antagonistic to the oomycete plant pathogens $Phytophthora$$capsici$ and $Pythium$$ultimum$ from the total population. Nine percent of the total number of isolated bacteria exhibited in vitro inhibitory activity against target plant pathogenic oomycetes. Taxonomic and phylogenetic placement of the antagonistic bacteria was investigated by analysis of the 16S rRNA gene sequences. The sequence analysis classified the antagonistic strains into four major classes of the domain bacteria ($Firmicutes$, ${\alpha}-Proteobacteria$, ${\gamma}-Proteobacteria$ and $Actinomycetes$) and 10 different genera. Further production of secondary metabolites, hydrolytic enzymes and plant growth promoting traits were determined for the putative new species of antagonistic endophytic bacteria. These new strains could not be identified as known species of ${\alpha}-Proteobacteria$, and so may represent novel bacterial taxa. The unexpected high antagonistic bacterial diversity associated with the tidal flat plants may be indicative of their importance in tidal flat plants as a promising source of novel antimicrobial compounds and biocontrol agents.
The rhizosphere is the active zone where plant roots communicate with the soil microbiome, each responding to the other's signals. The soil microbiome within the rhizosphere that is beneficial to plant growth and productivity is known as plant growth-promoting rhizobacteria (PGPR). PGPR take part in many pivotal plant processes, including plant growth, development, immunity, and productivity, by influencing acquisition and utilization of nutrient molecules, regulation of phytohormone biosynthesis, signaling, and response, and resistance to biotic- and abiotic-stresses. PGPR also produce secondary compounds and volatile organic compounds (VOCs) that elicit plant growth. Moreover, plant roots exude attractants that cause PGPR to aggregate in the rhizosphere zone for colonization, improving soil properties and protecting plants against pathogenic factors. The interactions between PGPR and plant roots in rhizosphere are essential and interdependent. Many studies have reported that PGPR function in multiple ways under the same or diverse conditions, directly and indirectly. This review focuses on the roles and strategies of PGPR in enhancing nutrient acquisition by nutrient fixation/solubilization/mineralization, inducing plant growth regulators/phytohormones, and promoting growth and development of root and shoot by affecting cell division, elongation, and differentiation. We also summarize the current knowledge of the effects of PGPR and the soil microbiota on plants.

To determine the concentrations and the mass flow of selected 10 perfluorinated compounds (PFCs), a field study was conducted in a wastewater treatment plant. Raw influent, primary influent, primary effluent, aeration tank effluent, secondary effluent, final effluent, dehydration liquor, primary sludge, thickened sludge, final sludge were collected over 3 days in the summer and the winter respectively. Collected samples were equally mixed and then served as an analytical sample. Total 10 compounds were analyzed. In terms of treated water, the concentration of perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) were in range of N.D.~26.29 ng/L and N.D.~38.15 ng/L respectively. Perfluorononanoate (PFNA) and perfluorohexanesulfonate (PFHxS) were ranged from N.D. to 36.79 ng/L and from N.D. to 24.36 ng/L. In terms of sludges, a concentration of PFOS, PFOA, and perfluorodecanesulfonate (PFDS) were detected from 6.82 to 59.37 ng/g, from 0.13 to 0.37 ng/g, from N.D. to 0.83 ng/g respectively. Mass loading for PFCs increased during wastewater treatment except for PFNA. The observed increase in mass flow of PFCs may have resulted from biodegradation of precursor compounds.
Plants are the promising reservoirs for natural products with their diverse secondary metabolites. Many invasive plants have been introduced in Korea, which adversely affect on the native ecosystem but holds difficulty removing them due to their proliferation. In this study, we evaluated disease control efficacy of methanol extracts from four invasive plant species against 7 representative crop pathogens. Methanol extract of Phytolacca americana effectively suppressed rice blast, tomato gray mold, and tomato late blight in a dose dependent manner. The methanol extract of Amorpha fruticosa also exhibited potent antifungal activity against pepper anthracnose in a concentration dependent way. These data suggest that the extracts of P. americana and A. fruticosa can be developed as plant disease protection agents against rice blast, tomato gray mold, tomato late blight, and pepper anthracnose. Furthermore, more extensive research will be required to identify and isolate active compounds from problematic invasive plant species to develop valuable agrochemicals.
Several genetic strategies have been proposed for the successful transformation and expression of microbial transgenes in model and crop plants. Here, we bring into focus the prominent applications of microbial transgenes in plants for the development of disease resistance; mitigation of stress conditions; augmentation of food quality; and use of plants as "bioreactors" for the production of recombinant proteins, industrially important enzymes, vaccines, antimicrobial compounds, and other valuable secondary metabolites. We discuss the applicable and cost-effective approaches of transgenesis in different plants, as well as the limitations thereof. We subsequently present the contemporary developments in targeted genome editing systems that have facilitated the process of genetic modification and manifested stable and consumer-friendly, genetically modified plants and their products. Finally, this article presents the different approaches and demonstrates the introduction and expression of microbial transgenes for the improvement of plant resistance to pathogens and abiotic stress conditions and the production of valuable compounds, together with the promising research progress in targeted genome editing technology. We include a special discussion on the highly efficient CRISPR-Cas system helpful in microbial transgene editing in plants.
Eurycoma longifolia is an important rare medicinal plant that contains valuable bioactive compounds. In the present study, cell suspension culture of E. longifolia was established for the production of biomass and phenolic compounds. Various medium parameters, such as concentration of auxin, salt strength of the medium, and sucrose and nitrogen concentrations, were optimized for the production of biomass at the flask-scale level. Full strength Murashige and Skoog (MS) medium supplemented with $3.0mg{\cdot}L^{-1}$ naphthaleneacetic acid (NAA), 3% (w/v) sucrose, 0:60 $NH{_4}^+:NO{_3}^-$ was found suitable for biomass accumulation. Based on the optimized flask-scale parameters, cell suspension cultures were established in balloon-type bubble bioreactors, and bioprocess parameters such as inoculum density and aeration rate were optimized. Inoculum density of $50g{\cdot}L^{-1}$ and increasing aeration rate from 0.05 to 0.3 vvm, with increases every 7 days, were suitable for the accumulation of both biomass and phenolic compounds. With the optimized conditions, $14.70g{\cdot}L^{-1}$ dry biomass, $10.33mg{\cdot}g^{-1}$ DW of phenolics and $3.89mg{\cdot}g^{-1}$ DW of flavonoids could be achieved. Phenolics isolated from the cell biomass showed optimal free radical scavenging activity.

Bacillus velezensis is an important plant growth-promoting rhizobacterium with immense potential in agriculture development. In the present study, Bacillus velezensis Lle-9 was isolated from the bulbs of Lilium leucanthum. The isolated strain showed antifungal activities against plant pathogens like Botryosphaeria dothidea, Fusarium oxysporum, Botrytis cinerea and Fusarium fujikuroi. The highest percentage of growth inhibition i.e., 68.56±2.35% was observed against Fusarium oxysporum followed by 63.12 ± 2.83%, 61.67 ± 3.39% and 55.82 ± 2.76% against Botrytis cinerea, Botryosphaeria dothidea, and Fusarium fujikuroi, respectively. The ethyl acetate fraction revealed a number of bioactive compounds and several were identified as antimicrobial agents such as diketopiperazines, cyclo-peptides, linear peptides, latrunculin A, 5α-hydroxy-6-ketocholesterol, (R)-S-lactoylglutathione, triamterene, rubiadin, moxifloxacin, 9-hydroxy-5Z,7E,11Z,14Z-eicosatetraenoic acid, D-erythro-C18-Sphingosine, citrinin, and 2-arachidonoyllysophosphatidylcholine. The presence of these antimicrobial compounds in the bacterial culture might have contributed to the antifungal activities of the isolated B. velezensis Lle-9. The strain showed plant growth-promoting traits such as production of organic acids, ACC deaminase, indole-3-acetic acid (IAA), siderophores, and nitrogen fixation and phosphate solubilization. IAA production was accelerated with application of exogenous tryptophan concentrations in the medium. Further, the lily plants upon inoculation with Lle-9 exhibited improved vegetative growth, more flowering shoots and longer roots than control plants under greenhouse condition. The isolated B. velezensis strain Lle-9 possessed broad-spectrum antifungal activities and multiple plant growth-promoting traits and thus may play an important role in promoting sustainable agriculture. This strain could be developed and applied in field experiments in order to promote plant growth and control disease pathogens.
This paper describes an investigation to determine whether the activated sludge (AS) process could be used for the treatment of wastewater at the Union Carbide Coporation (UCC) plant in Seadrift, Texas. This plant presently utilizes a waste stabilization pond (WSP) system for treatment of the wastewater. The treatment system consists of an in-plant primary WSP and two off-plant WSPs (secondary and tertiary WSPs), run in series. The total hydraulic detention time of the WSP system is approximately 150 days. Several laboratory-based treatability studies have been conducted to evaluate the performace of the WSP system and the degradability of specific chemical compounds. From an additional study, it was determined that the WSP system was stressed and occasionally operating near the limit of its treatment capacity. The existing primary WSP plays an important role in the overall treatmemt system, because it not only functions as a pH and organic-strength equalization basin, but also serves as a "preconditioning" basin by fermenting high strength organic wastes to volatile organic acids for subsequent degradation in the escondary WSP. However, in view of pending RCRA legislatin conerning the "proposed organic toxicity characteristics limits" (40 CFR Part261: Federal Register, July, 1988), it is possible that the primary WSP will have to be abandoned in favor of alternative treatment options. Therefore the main purpose of this study was to perform activated sludge treatability evaluations for the development of an alternative to the existing primary WSP treatment ststem. In addition, another purpose was to determine the degradability of bis(2-chloroethyl)ether (Chlorex or CX) and benzene(BZ) in the activated sludge process. The presence of these two chemicals in the wastewater of the plant prompted the question of whatedether they could be degraded in an activated sludge system.
The limited effectiveness of current plant disease management treatments necessitates the development of new methods for controlling diseases using beneficial microbes. Demanding sustainable agriculture is increasingly highlighted as a biocontrol approach, particularly Streptomyces species known to produce a variety of antibiotic compounds and secondary metabolites. The Streptomyces globisporus SP6C4 strain and Streptomyces sp. S8 have been reported as potent antifungal agents and are gaining attention for improving crop growth in sustainable agriculture. In this study, we investigated the use of Streptomyces species formulations to enhance bacterial growth with nitrogen sources. Specifically, the addition of L-glutamic acid and L-cysteine resulted in earlier sporulation and bacterial growth in Streptomyces strains, respectively. This approach could expand the range of fermentation techniques in agriculture and be useful for controlling plant growth-promoting bacteria.
