The effect of acid rain on plant isozyme response was studied with the treatment of simulated acid rain to Pinus densiflora, P. nigida and P. koraiensis for 9 months. The isozyme pattern of $\alpha$-Esterase($\alpha$-Est), Peroxidase(POD) and Glutamate dehydrogenase(GDH) were observed in the control (pH 5.6) and simulated acid rain (pH 3.5) treatment. No changes of isozyme pattern in $\alpha$-Est was observed in P. densiflora after 9 month treatment of simulated acid rain, but, two new isozymes were activated in P. nigida in the same treatment. In P. koraiensis, two new isozyme were activated but five isozymes were not activated. P. densiflora did not show any difference in POD and GDH after the treatment of simulated acid rain. P. nigida showed activation of eight and two isozymes in POD and GDH, respectively. P. koraiensis showed inactivation of 4 isozymes in POD but showed no changes In GDH.
A die-back fungus isolated from Pinus koraiensis was identified as Cenangium ferruginosum. Mycelial growth of the fungus was the best on PDA among five media tested. Mycelial growths on agar media supplemented with extracts from coniferous trees were relatively better than those on media with extracts from non-coniferous trees. Optimum temperature and pH for the mycelial growth of C. ferruginosum on PDA were $10^{\circ}C$ and pH 5, respectively. When five-years-old P. koraiensis seedlings were artificially inoculated with the fungus for the pathogenicity test of this fungus, 80% of seedlings were infected. and the same symptoms and signs were developed as those of naturally-infected trees.

The purpose of this research was to evaluate the vapor permeability of nine different species of domestic softwood and the vapor permeability evaluation through the production of small wall structures for the developing applications, utilizing the vapor permeability of wood. In addition, the permeability evaluation was attempted by means of the production of a small wall structure injected with a waste material, bottom ash, as a moisture absorbent for improving the permeability. Consequently, the results of the vapor permeability evaluation by means of Sd value are as follows: (1) It was observed that Abies holiphylla, Picea jezoensis, Ginkgo biloba, Pinus koraiensis and Pinus rigida are permeable to moisture among 9 species of domestic softwood in Korea. (2) By means of this, semi-permeability efficiency was evaluated when producing a small wall structure. (3) Besides, improved effects of permeability were evaluated when producing a small wall structure inserted with bottom ash. As a result, it was confirmed that the Sd value of Pinus koraiensis turned out to be 1.63, which is superior to other 8 tree species.
This experiment was carried out to investigate the effects of autohydrolysis and extraction conditions on the separation of the chemical substances, the extractability of lignin by dioxane, and the yield of reducing sugars from cellulosic substrates by using a commercial cellulase derived from Trichoderma viride. Air-dried wood meals through 0.42mm (40 mesh) screen and retained on 0.25 mm (60 mesh) of Populus alba-glandulosa and Pinus koraiensis were autohydrolyzed with water at $180^{\circ}C$ for 30 and/or 60 minutes in a 6 liter stainless-steel digester with or without 2% 2-naphthol. The hydrothermally-treated wood meals were extracted the lignin with 100%, 90%, 75% and 50% dioxane solutions at $70^{\circ}C$ for 4 hours, respectively. The results obtained were as follows; 1) After autohydrolysis of Populus alba-glandulosa, the yield of wood meals decreased with lengthening the auto hydrolysis time from 30 minutes to 60 minutes and with 2% 2-naphthol addition. In case of Pinus koraiensis, the yield was not affected by 2%, 2-naphthol addition at the autohydrolysis in the digester. 2) After autohydrolysis and lignin extraction of Populus alba-glandulosa, the yield of wood meals decreased with lengthening the autohydrolysis time from 30 minutes to 60 minutes and with 2% 2-naphthol addition. Extraction of 50% dioxane solution was the best solvent for the yield among the solutions of 100%, 90%. 75% and 50% dioxane. In case of Pinus koraiensis, the yield was not affected by 2% 2-naphthol addition and the solution of 90% dioxane was the poorest solvent for the yield. 3) After autohydrolysis and lignin extraction of Populus alba-glandulosa, the Klason lignin content in cellulosic substrates for enzymatic hydrolysis decreased with lengthening the autohydrolysis time from 30 minutes to 60 minutes and with 2% 2-naphthol addition. Klason lignin content was the lowest after extraction by 90% or 75% dioxane solution. The content was also affected by interaction of the three factors-autohydrolysis time, 2% 2-naphthol addition and concentration of dioxane. In case of Pinus koraiensis, the Klason lignin content increased with 2% 2-naphthol addition but was not affected by the concentration of dioxane solution. 4) After autohydrolysis and lignin extraction of Populus alba-glandulosa, the extractable Klason lignin content by extraction increased with lengthening the auto hydrolysis time from 30 minutes to 60 minutes and with 2% 2-naphthol addition. The extractable lignin content was the highest after extraction by 90% or 75% dioxane solution. In case of Pinus koraiensis, the extractable lignin content increased with 2% 2-naphthol addition. Extractions by 100%, 90% and 50% dioxane solutions were more effective for the extraction of Klason lignin than by 75% dioxane solution. 5) After autohydrolysis and lignin extraction of Populus alba-glandulosa, the yield of reducing sugars increased with lengthening the autohydrolysis time from 30 minutes to 60 minutes but was not affected by 2% 2-naphthol addition and the concentration of dioxane. The yield of reducing sugars after enzymatic hydrolysis was slightly higher by extractions with 90%, 75% and 50% dioxane solutions than with 100% dioxane. In case of Pinus koraiensis, the yield of reducing sugars was not affected by 2% 2-naphthol addition but affected by the concentration of dioxane. The yield of reducing sugars was the highest in cellulosic substrates extracted by 100% dioxane solution.
This study was carried out to elucidate the relationship between wood anatomy and the water flow path in P. koraiensis and L. leptolepis. through the experiment of penetration of the dye solution. The experiment was performed by permeating 1% acid, alkali and direct solution into the xylem just after being cut. The results obtained were summarized as follows : 1. In P. koraiensis and L. leptolepis, the dye solution penetrated into sapwood and annual rings adjacent to cambial zone were only dyed according to ascent of tree height. 2. The penetrability of latewood was better than that of earlywood. 3. In P. koraiensis and L. leptolepis, the main water flow path in longitudinal direction was the trachied and that in transverse direction was ray trachied and ray parenchyma. Also, the dye solution was found in resin canal. 4. P. koraiensis was more permeable than L. leptolepis. 5. Among the acid, alkali and direct dye solution, the acid dye solution was the most permeable.
We investigated the effects of pollutants on two pine species (Pinus koraiensis and Pinus rigida) in an industrial region, using a physiological approach. We measured the growth and physiological parameters (photosynthetic activity and chemical contents) of the pine trees in relation to environmental pollutants. The concentrations of manganese (Mn), fluorine (F) and chlorine (CI) in needles of two pine species at the damaged site were significantly higher than those at the control site, and concentrations of essential elements (P) and chlorophyll in needles at the damaged site were significantly lower than at the control site. The light-saturated net photosynthesis $(P_{sat})$, apparent quantum yield $(\Phi)$, carboxylation efficiency (CE) of both pines at the damaged site were significantly lower than those at the control site. The length of shoots and survivorship of needles of two pines at the damaged site were significantly lower than those at the control site. Especially, at damaged site, growth of shoots and needle longevity of P. koraiensis are larger than those of P. rigida.
One hundred and ninety two isolates of Armillaria were obtained from mycelial fans on infected hosts, rhizomorphs, and single basidiospores or trauma tissue of fruiting bodies. Mating tests showed that two of these isolates were A. mellea, eight were A. tabescens, 20 were A. ostoyae, and 162 were A. gallica. Armillaria ostoyae was mainly isolated from Pinus koraiensis and Qurecus spp., A. tabescens from fruiting bodies on Pinus densiflora and Qurecus spp., and A. gallica from many tree species but not Pinus koraiensis. Armillaria mellea, A. gallica, A. ostoyae and A. tabescens showed distinct protein banding patterns. Mycelial growth and rhizomorph formation was good on basal medium with ethanol added. A. gallica and A. mellea formed many rhizomorphs, but A. ostoyae did not. A. gallica showed the best rhizomorph formation on media with tannic acid and ethanol, but a. mellea formed the most rhizomorphs on gallic acid. Rhizomorphs showed monopodial branching for A. gallica and dichotomous branching for A. ostoyae. Fruiting bodies. formed in the laboratory on sawdust media most abundantly by A. tabescens. In nature, fruit body formation by A. tabescens was from early to mid August. A. ostoyae and A. gallica fruit bodies were formed from early August to late October. While there are common names in Korea for A. mellea and A. tabescens, such as mulberry mushroom relative, no common names are available for A. gallica and A. ostoyae. Therefore, we refer to a. gallica as the Gastrodia mushroom because it has been used to produce Gastrodia and A. ostoyae as the Korean pine mushroom because it is frequently found as mushrooms on Korean pine.
This study was carried out to investigate the dimensional stability of three softwoods (Pinus densiflora S. et Z., Pinus koraiensis S. et Z., Larix kaempferi CARR.) and two hardwoods(Quercus mongolica F., Quercus variabilis BL.) treated with polyethylene glycol(PEG). Existence of PEG in wood was examined by an X-ray diffraction method. Shrinkage of the sample woods decreased with increasing concentration of PEG. The dimensional stability of five species had improved highly in thirty percent solutions of PEG with molecular weight 1000~4000, and the dimension of PEG-treated softwoods was more stable than that of PEG-treated hardwoods. Weight gain and bulking effect of the woods treated with PEG increased with increasing concentration of PEG. X-ray diffractograms of Pinus koraiensis wood treated with PEG showed two peaks derived from PEG crystal at 2𝜃 = 19° and 2𝜃 = 23°. However, the diffractograms of Quercus mongolica wood did not present any peaks caused by PEG crystal. From the result of X-ray diffraction, it was revealed that the content of PEG in Pinus koraiensis wood was higher than that in Quercus mongolica wood.
Radial and vertical variations of relative crystallinity and crystallite width of cellulose within stems of three softwoods (Pinus densiflora S. et Z., P. koraiensis S. et Z, P. rigida Mill.) grown in Korea were examined by an X-ray diffraction method. The mean of relative crystallinity was 61.7% in P. koraiensis, 60.6% in P. densiflora and 49.4% in P. rigida. The degree of crystallinity in earlywood and latewood increased with the age from pith to about 10~15 years, and then remained almost constant value. The relative crystallinitiy of latewood was slightly higher than that of earlywood. The relative crystallinity in P. densiflora was a little lower at the base of stem, but no significant difference by height was shown in P. koraiensis and P. rigida. The crystallite widths in the stems were 2.8 to 3.0 nm, but were not significantly different in earlywood and latewood by height. In conclusion, the relative crystallinity appeared to be a useful index for separating juvenile wood from adult wood in the softwoods of P. densiflora, P. koraiensis, and P. rigida grown in Korea.
A metabolomic study was conducted to identify key metabolic components, which are correlated with the growth of 4-year-old Pinus koraiensis seedlings harvested at actively height growing season (May 18th). Among 105 individual metabolites identified by GC/MS analysis, alanine, threonine, oleic acid, and butanoic acids were negatively correlated with both height and weight of 4-year-old seedlings, while malic acid, xylose, glucose, d-turanose and inositol had positive correlation with various growth parameters. During the actively growing season, the concentrations of both amino acids and organic acids in the main stem of Superior seedling group were lower but the photosynthates such as mono-saccharide and sucrose were higher than in other seedling groups such as Intermediate and Inferior. Interestingly, d-turanose, an analogue of sucrose that is not metabolized in higher plants but used as carbon source by many organisms including numerous species of bacteria and fungi, showed the highest correlation (r=0.896, p<0.001) with height of 4-year-old seedlings, indicating that possible interaction with mycorrhizal organisms. Therefore we suggest that several metabolites selected in this study may be used as metabolic markers for complex traits in P. koraiensis.
