Physiological Response of a White Mutant of Ganoderma lucidurn Induced by Light and Temperature (영지버섯 백색변이주의 광 및 온도에 의한 생리적 반응)

  • White mutant of Ganoderma lucidum(G4142) induced the non-basediocarpous basidiospores(NBB) from the aerial mycelia on agar media by the light illumination. Light was found to be necessary for NBB formation, but it also inhibited the growth of mycelium. The best sporulation was obtained at the periodic exposure of 16 hour light and 8 hour dark. Blue and yellow light were the most effective on sporulation, however, near UV and red light did not induce any spores. Effective light intensity for NBB bearing was about 1,000 lux as white light. Even after 16 days of culture, this strain did not form the pinhead nor chlamydospore. Optimum temperature for the mycelial growth and NBB formation were 30$\circ $C. Ganoderma lucidum G4142 exhibited the formation of stroma after five days of incubation at 30$\circ $C.

Studies on the development of mushroom media for bottle culture in new Pleurotus ostreatus 'Miso' (신품종 '미소' 느타리버섯 병재배 배지개발)

  • This study was carried out to develop the best medium of new oyster mushroom for bottle culture. The new oyster mushroom cv. Miso is a Pleurotus ostreatus developed at the Chungnam Agricultural Research & Extension Services. For the bottle culture media, poplar sawdust+beet pulp+cottonseed meal (5:3:2), poplar sawdust+wheat husk meal (8:2), poplar sawdust+beet pulp+cottonseed meal (6:2:2), poplar sawdust+beet pulp+cottonseed meal (4:4:2), and poplar sawdust+beet pulp+wheat husk meal (7:1:2) were used in 850cc PP bottle. The pH was 5~6 and the C/N ratio 19.7~28.3 in bottle culture media. The time of pinhead formation was 5 to 6 days. For the fruiting body formation after inoculation took 29~31 days. The yield of fruiting body of poplar sawdust+beet pulp+wheat husk meal (7:1:2) medium was the highest at 110.4g/bottle compared to other media. Therefore, such cultivation medium would be appropriate for the commercial production of bottle culture in the new oyster mushroom 'Miso'.

Fruit-body Production of Auricularia auricula-judae by Sawdust Cultivation (톱밥재배에 의한 목이의 자실체 생산)

  • Auricularia auricula-judae is an edible mushroom, which is known as wood ear, free ear, black ear mushroom, and free jelly fish. Present experiments were conducted to determine the possibility of artificial cultivation with oak sawdust of A. auricula-judae. The duration of mycelial growth and days of pinhead formation on oak sawdust bag (1.4 kg) were 29~32 days and 13~17 days, respectively. The yield of fresh fruit-body was 275~350 g.

  • Ganoderma neo-japonicum, which is also known as black lingshi mushroom and medicinal mushroom. Present experiments were conducted to determine the possibility of artificial culture with oak sawdust of G. neo-japonicum. The duration of mycelial growth and days of pinhead formation of oak sawdust bag (2.4 kg) were 28~35 days and 25~29 days, respectively. The yield of mushroom fresh fruitbody was 135~157 g.

Artificial Cultivation Characteristics and Bioactive Effects of Novel Tropicoporus linteus (Syn. Phellinus linteus) Strains HN00K9 and HN6036 in Korea

  • Phellinus strains were collected from different areas in Korea. Of them, the fast mycelial growing strains were artificially cultivated on the oak logs to produce fruiting body. The varieties, Phellinus linteus ASI26099 (Korea Sanghwang) and P. baumii PBJS (Jangsoo Sanghwang) were grown under the same conditions as controls. Their cultivating characteristics including mycelial colonization, pinhead formation, and fruiting body formation rate were investigated on the logs. Basidiocarps of Phellinus strains HN00K9, HN6036, and ASI26099 were concentrically zonate and shallowly sulcate, and dark chestnut showing typical characteristics of Tropicoporus linteus (synonyum: P. linteus, Inonotus linteus, polyporus linteus), which is distinguishably different to PBJS. HN00K9 showed the highest yield of fruiting body among the mushroom strains. The β-glucan content in fruiting bodies of HN00K9 was 20% higher than those of other strains. Bioactive effects of polysaccharide samples from fruiting bodies of Phellinus strains, HN00K9, HN6036, ASI26099, and PBJS were assessed on cell viability and cytokine (IL-6 and TNF-α) inhibition and finally on anticancer to different human cancer cells.

  • This study were carried out to evaluate the effect of quantities of substrate on the yields for 2 years $(1993{\sim}1994)$. The substrates are cotton waste, rice straw and rice hull. It were revealed that cotton waste mixed rice straw was the best yield of fruit body. It gave more increased $(10{\sim}22%)$ than other substrates $(7{\sim}15%)$ when increasing the quantities of substrate. And cotton waste mixed with rice hull was most fast for first pinhead formation.

Optimum $CO_2$ Concentration for Fruit-body Formation and Yield of Pleurotus ferulae Mushroom in the Growing Facilitiy for Bottle Cultivation (재배사내 $CO_2$ 농도가 아위느타리버섯의 생육 및 수량에 미치는 영향)

  • The effects of $CO_2$ concentration on fruit-body formation and yield of Pleurotus ferulae (KME65003) mushroom were examined in the growing facilities for bottle cultivation. The $CO_2$ concentration levels in the growing facilities were 500 ppm, 1000 ppm, 1500 ppm and 2000 ppm of $CO_2$, controlled by different ventilation amount. Yield of fruit body was highest of 102.4 g/bottle at 1000 ppm treatment and lowest of 75.1 g/bottle at 2000 ppm. As the $CO_2$ concentration increased up to 2000 ppm, the first pinhead formation and fruit-body growing period took longer, so total growing period took 16 days at 500 ppm, but 23 days at 2000 ppm. The number of pinhead formation was highest of 12.2/bottle at 1500 ppm and valid stipes was highest of 2.8/bottle at 1000 ppm. Fruit body characteristics such as pileus and stipe diameter, and stipe length were not significant at different $CO_2$ concentration. The fruit body ratio of 20~50 g range among the whole fruit body which was classified by weight was highest of 60.3% at 1000 ppm. As a result, the suitable $CO_2$ concentration of growth and yield of Pleurotus ferulae was showed as 1000 ppm.

Development of Fruitbody in the Artificial Oak Sawdust Cultures of Phellinus gilvus Mushroom (마른진흙버섯(Phellinus gilvus)의 톱밥재배 기술에 관한 연구)

  • Present experiments were conducted to determine the possibility of artificial culture with oak sawdust block of Phellinus gilvus Mushrooms, Mycelial growth in sawdust block (oak 90 + rice bran 10, V/N) after 25 days. It took 12 days to make fruitbody from burying of sawdust block to pinhead formation. The fruitbodies produced the total fresh weight 577 g (dried weight 97 g) in a block.

  • A Sciarid fly, Lycoriella mali was a serious pest in commercial production of the cultivated mushroom, Pleurotus ostreatus. It was found in light trap and compost samples taken from mushroom houses during all growing seasons. The fly population 'was increased with the development of mushroom growth, the larvae occurred about 30 days after spawn inoculation in compost. The highest population was developed spring cultivation season (March to June) and decreased during summer season (August to September). The larvae feed on mycellium and compost; injury to the growing mushroom mycellium and the subsequent consumption of the primordia; they entered and tunneld sporospore stem and infect pinhead formation which would be resulted in decrease of mushroom yield.

  • To investigate the effect of inorganic nutrient supplement in oyster mushroom (Pleurotus ostreatus), we have conducted some study on cultural and growth characteristics of fruitbody formation and chemical composition of media and fruitbody. When supplemented with slow releasing fertilizer, contamination rate was not different from non-supplemented medium, days for incubation time and first pinhead were faster than non supplemented medium. And fruitbody yield and biological efficiency were increased $10{\sim}28%,\;7{\sim}20%$ respectively, but biological efficiency was decreased when increased supplement ratio. The chemical compositions (total carbon, total nitrogen, ammonia nitrogen, nitrate nitrogen, potassium and phosphate) of slow releasing fertilizer supple mented medium and fruitbody were compared with non-supplemented.
