Identification of a Polyketide Synthase Gene in the Synthesis of Phleichrome of the Phytopathogenic Fungus Cladosporium phlei

  • Phleichrome, a pigment produced by the phytopathogenic fungus Cladosporium phlei, is a fungal perylenequinone whose photodynamic activity has been studied intensively. To determine the biological function of phleichrome and to engineer a strain with enhanced production of phleichrome, we identified the gene responsible for the synthesis of phleichrome. Structural comparison of phleichrome with other fungal perylenequinones suggested that phleichrome is synthesized via polyketide pathway. We recently identified four different polyketide synthase (PKS) genes encompassing three major clades of fungal PKSs that differ with respect to reducing conditions for the polyketide product. Based on in silico analysis of cloned genes, we hypothesized that the non-reducing PKS gene, Cppks1, is involved in phleichrome biosynthesis. Increased accumulation of Cppks1 transcript was observed in response to supplementation with the application of synthetic inducer cyclo-(${_L}-Pro-{_L}-Phe$). In addition, heterologous expression of the Cppks1 gene in Cryphonectria parasitica resulted in the production of phleichrome. These results provide convincing evidence that the Cppks1 gene is responsible for the biosynthesis of phleichrome.

High-Level Production of Astaxanthin by Fed-Batch Culture of Mutant Strain Phaffia rhodozyma AJ-6-1

  • The production of a carotenoid astaxanthin, a growth-associated principal pigment, is limited in a batch cultivation, because a high glucose concentration severely inhibits the cell growth and also influences the carotenoid production. Therefore, a fermentation strategy including effective chemicals for the high-level production of cells and astaxanthin by a mutant strain Phaffia rhodozyma AJ-6-1 was developed in a fed-batch culture. First, a production medium for maximizing the cell and astaxanthin yields was formulated and optimized. Using this optimized medium, the highest cell and astaxanthin concentrations obtained were about 38.25 g/1 and 34.77 mg/1, respectively. In addition, an attempt was made to increase the amount of astaxanthin using effective chemicals such as ethanol and acetic acid, which are known at an inducer and/or precursor of carotenoid synthesis. When either 10g/1 ethanol or 5 g/1 acetic acid was added to investigate the resulting astaxanthin content, a relatively high astaxanthin concentration or 45.62 mg/l and 43.87 mg/1, respectively, was obtained, and the cell concentrations also increased slightly under these conditions. Therefore, these results imply that a fed-batch culture of the mutant strain P. rhodozyma AJ-6-1 could be effectively employed in the commercial production of astaxanthin, although the factors affecting the productivity remain to be elucidated.

Heme Derived from Corynebacterium glutamicum: A Potential Iron Additive for Swine and an Electron Carrier Additive for Lactic Acid Bacterial Culture

  • To investigate the potential applications of bacterial heme, aminolevulinic acid synthase (HemA) was expressed in a Corynebacterium glutamicum HA strain that had been adaptively evolved against oxidative stress. The red pigment from the constructed strain was extracted and it exhibited the typical heme absorbance at 408 nm from the spectrum. To investigate the potential of this strain as an iron additive for swine, a prototype feed additive was manufactured in pilot scale by culturing the strain in a 5 ton fermenter followed by spray-drying the biomass with flour as an excipient (biomass: flour = 1:10 (w/w)). The 10% prototype additive along with regular feed was supplied to a pig, resulting in a 1.1 kg greater increase in weight gain with no diarrhea in 3 weeks as compared with that in a control pig that was fed an additive containing only flour. To verify if C. glutamicum-synthesized heme is a potential electron carrier, lactic acid bacteria were cultured under aerobic conditions with the extracted heme. The biomasses of the aerobically grown Lactococcus lactis, Lactobacillus rhamosus, and Lactobacillus casei were 97%, 15%, and 4% greater, respectively, than those under fermentative growth conditions. As a potential preservative, cultures of the four strains of lactic acid bacteria were stored at $4^{\circ}C$ with the extracted heme and living lactic acid bacterial cells were counted. There were more L. lactis and L. plantarum live cells when stored with heme, whereas L. rhamosus and L. casei showed no significant differences in live-cell numbers. The potential uses of the heme from C. glutamicum are further discussed.

Inhibitory Effects of Licochalcone A and Isoliquiritigenin on Monocyte Adhesion to TNF-$\alpha$-activated Endothelium

  • Numerous natural herbal compounds have been reported to inhibit adhesion and migration of leukocytes to the site of inflammation Licorice extracts, which have been widely used in traditional Chinese medicinal preparation, possess various pharmacological effects. Isoliquiritigenin, a biogenetic precursor of flavonoids with various pharmacological effects, is a natural pigment present in licorice. We attempted to explore whether licorice extracts and isoliquiritigenin mitigate monocyte adhesion to tumor necrosis factor-$\alpha$ (TNF-$\alpha$)-activated human umbilical vein endothelial cells (HUVEC). In addition, it was tested whether the inhibition of monocyte adhesion to the activated HUVEC accompanied a reduction in vascular cell adhesion molecule-l expression(VCAM-l). Dry-roasted licorice extracts in methylene chloride but not in ethanol markedly interfered with THP-l monocyte adhesion to INF-$\alpha$-activated endothelial cells. licochalcone A compound isolated from licorice extract in methylene chloride appeared to modestly inhibit the interaction of THP-l monocytes and activated endothelium. In addition, isoliquiritigenin abolished the monocyte adhesion with attenuating VCAM-l protein expression on HUVEC induced by INF-$\alpha$. These results demonstrated that non-polar components from dry-roasted licorice extracts containing licochalcone A as well as isoliquiritigenin were active in blocking monocyte adhesion to cytokine-activated endothelimn, which appeared to be mediated most likely through the inhibition of VCAM-l expression on HUVEC. Therefore, licorice may hamper initial inflammatory events on the vascular endothelium involving induction of endothelial cell adhesion molecules.

  • Gametogenesis and the reproductive cycle of the marsh clam, Corbicula leana(Prime} were investigated monthly Hyongsan estuary, Korea from January to December 1998 by histological observation. C. leana had separate sexes, and oviparous. The gonads were located between the sub-region of mid-intestinal gland in visceral cavity and reticular cennective tissue. The ovary and testis were composed of a number of ovarian and testicular tubules, respectively, Mature oocytes were characterized by germinal vesicles with nucleoli and their sizes ranged $70\;to\;80{\mu}m$ in diameter. A number of mesenchymal tissues and pigment granular cells were distributed in the growing oocytes and spermatocytes during early developmental stages. With the further development of gonad, these tissuse and cells gradually disappeared. The monthly changes in the fatness index were closely related to reproductive cycle. Minimun size for sexual maturity was reached over 10.0 mm in shell length. The reproductive cycle could be classified into 6 stages : multiplicative (March to April): early growing and growing (May to June), mature (July to August), spent (September), degenerative (October) and recovery (November to February).

Pyrrolo[3,2-b]pyrrole-Based Copolymers as Donor Materials for Organic Photovoltaics

  • A new accepter unit, pyrrolo[3,2-b]pyrrole-2,5-dione, was prepared and utilized for the synthesis of the conjugated polymers containing electron donor-acceptor pair for OPVs. Pyrrolo[3,2-b]pyrrole-2,5-dione unit, regioisomer of the known pyrrolo[3,4-c]pyrrole-1,4-dione, is originated from the structure of stable synthetic pigment. The new conjugated polymers with 1,4-diphenylpyrrolo[3,2-b]pyrrole-2,5-dione, thiophene and carbazole were synthesized using Suzuki polymerization to generate P1 and P2. The solid films of P1 and P2 show absorption bands with maximum peaks at about 377, 554 and 374, 542 nm and the absorption onsets at 670 and 674 nm, corresponding to band gaps of 1.85 and 1.84 eV, respectively. To improve the hole mobility of the polymer with 1,4-bis(4-butylphenyl)-pyrrolo[3,2-b]-pyrrole-2,5-dione unit, which was previously reported by us, the butyl group at the 4-positions of the N-substituted phenyl group was substituted with hydrogen and methyl group. The field-effect hole mobility of P2 is $9.6{\times}10^{-5}cm^2/Vs$. The device with $P2:PC_{71}BM$ (1:2) showed $V_{OC}$ value of 0.84 V, $J_{SC}$ value of 5.10 $mA/cm^2$, and FF of 0.33, giving PCE of 1.42%.

  • When a piece of the dorsal skin(cat fish) was observed under a light microscope, melanophores were horizontally expanded and had a few processes filled with melanosomes. They were isolated from one another. Some melanophores looked black, while others were d&k brown. On observation with a transmission electron microscope, the epidermal melanophore of cat fish was highly branched and small segments of processes were found frequendy m the vidnity of the interecellular spaces. The cross section of the processes of melanophore was almost circular, and often invested by a thin layer of epidermal cells. Some processes, however, occurred free m the wide interecellular spaces or at the cytoplasm of the superilcial layers. In the mature melanophore, the cell organelles including melanosomes, mitochondria and free ribosomes were prominent in the perinuclear portions. Many melanosomes were spherical or elipsoidal in shape. Each melanosomes was surrounded by a limiting membrane. The processes of the mature melanophore were well developed, but the processes of the immature melanophores were incompletely developed.

Drosophila CrebB is a Substrate of the Nonsense-Mediated mRNA Decay Pathway that Sustains Circadian Behaviors

  • Post-transcriptional regulation underlies the circadian control of gene expression and animal behaviors. However, the role of mRNA surveillance via the nonsense-mediated mRNA decay (NMD) pathway in circadian rhythms remains elusive. Here, we report that Drosophila NMD pathway acts in a subset of circadian pacemaker neurons to maintain robust 24 h rhythms of free-running locomotor activity. RNA interference-mediated depletion of key NMD factors in timeless-expressing clock cells decreased the amplitude of circadian locomotor behaviors. Transgenic manipulation of the NMD pathway in clock neurons expressing a neuropeptide PIGMENT-DISPERSING FACTOR (PDF) was sufficient to dampen or lengthen free-running locomotor rhythms. Confocal imaging of a transgenic NMD reporter revealed that arrhythmic Clock mutants exhibited stronger NMD activity in PDF-expressing neurons than wild-type. We further found that hypomorphic mutations in Suppressor with morphogenetic effect on genitalia 5 (Smg5) or Smg6 impaired circadian behaviors. These NMD mutants normally developed PDF-expressing clock neurons and displayed daily oscillations in the transcript levels of core clock genes. By contrast, the loss of Smg5 or Smg6 function affected the relative transcript levels of cAMP response element-binding protein B (CrebB) in an isoform-specific manner. Moreover, the overexpression of a transcriptional repressor form of CrebB rescued free-running locomotor rhythms in Smg5-depleted flies. These data demonstrate that CrebB is a rate-limiting substrate of the genetic NMD pathway important for the behavioral output of circadian clocks in Drosophila.

  • Daily doses of phenylmercuric acetate arranged in $30{\gamma}\;(group\;I)$, 3{\gamma}\;(group\;II)$ and $0.3{\gamma}\;(group\;III)$ were administered respectively to rabbits for 90 days. The chief histopathological changes in the organs and the analytical data on mercury residues in the excretion and liver were as follows. 1. Kidney: In group I, severe degrees of vacuolization and cloudy swelling were occurred in the epithelial cells of proximal convoluted tubules and severe cloudy swelling and coagulative necrosis were observed in the proximal straight tubules. There were many hyaline casts in the collecting tubules. In group II, moderate degrees of vacuolization and cloudy swelling were observed in the epithelial cells of proximal convoluted tubules and moderate cloudy swelling and coagulative necrosis were encountered in the proximal straight tubules. A little numbers of hyaline casts were located in the lumen of collecting tubules. In group III, slight degree of cloudy swelling were observed in the epithelial cells of proximal convoluted and straight tubules. 2. Liver: In group I, cloudy swelling, fatty changes and coagulative necrosis were observed in the central zone of hepatic lobules. Dissociation of hepatic cell cords was encountered. Hyperplsia of hepatic cells were remarkable in group II. No Pathological changes were observed in group III. 3. Spleen: Deposition of hemosiderin pigment was prominant in group I and small amount of the pigment was observed in group II. There were no pathological changes in group III. 4. Adrenal, colon and heart: No pathological changes were detected in all 3 groups. 5. In an average about 76.5% of mercury was excreted from group I, 85.4% from group II and 79.8% from group III. 6. Mercury content in the liver was 0.0348 g in group I, 0.00378 g and 0.00066 g in group II and group III respectively. 7. In general, as to increased mercury doses the concentration of mercury accumulation in the liver became higher, how·ever, the accumulation quantity against a total amount of mercury doses showed an adverse trend. In other word, the quantity of mercury accumulation was not increased proportionately by higher dose of mercury.

Evidence Suggesting that the Deposition of Pigments into Yolks is Independent of Egg Production: Enhanced Pigmentation of Yolks by Feeding Hens with Canthaxanthin Biosynthesized by Microbials (난황 내 색소의 축적은 산란율과 무관함을 제시하는 증거: 균체가 생성하는 Canthaxanthin의 급여에 의해 강화된 난황의 착색)

  • Pigments in the diet affect yolk colors. Due to variations in both the bioavailability of pigments in chickens and their amounts occurring in the feed ingredients, concern about egg quality arises in terms of yolk color. In this study, the effects of pigments, produced through cell culture in the laboratory, on yolk colors were determined for 4 weeks in laying hens receiving one of the 6 dietary treatments: control diets containing 1) no synthetic pigments (CON); 2) canthaxanthin (4 ppm) purchased from BASF (BASF); 3) cultured cells so that the diet had canthaxanthin at 4 ppm (CX); 4) cultured cells so that the diet had lycopene at 30 ppm (LP); 5) canthaxanthin (4 ppm) that was purified from cultured cells (SPCX); or 6) lycopene (30 ppm) that was purified from cultured cells. Relation between deposition of pigments into yolks and egg production was also tested. Yolk color of eggs from chickens fed dietary CX was significantly enhanced, which was slightly but significantly below that of BASF. Results from other treatments were lower than those of CX. Deposit rates of pigments into yolks were: BASF > CX > SPCX > LP > SPLP. The amounts of pigments, with the exception of SPLP, in feed were not changed during the storage for 4 weeks at $25^{\circ}C$. Egg production rates varied among treatments during the initial phase of the study but became relatively uniform at the later stage, except for CON and LP groups. The results of the present study indicate that the deposition of pigments into yolks is independent of egg production.
