Response Characteristics of pH-ISFET Urea Sensor (pH-ISFET 요소센서의 감응특성)

  • A preparation method and response characteristics of a urea sensor which consisted of pH-ISFET and urease-immobilized membrane were investigated. The pH-ISFET urea sensor was fabricated by immobilizing BSA and urease with glutaraldehyde on gate of the pH-ISFET. Effects of pH and concentration of working buffer and enzyme load on the pontentiometric response of the pH-ISFET urea sensor were examined. Response characteristics for the determination of urea in synthetic physiological saline solution (pH 7.4) were as follows. That is, the concentration range of linear response, slope of linear response (sensitivity), and response time were 5.0 ${\times}$ $10^{-4}$ ${\sim}$ 5.0 ${\times}$ $10^{-2}$M, 31.6 mV/decade and 4${\sim}$10 min, respectively.

  • As interest in wellness grows, There is a lot of research about monitoring individual health using wearable devices. Accordingly, a variety of methods have been studied to distinguish exercise from daily activities using wearable devices. Most of these existing studies are machine learning methods. However, there are problems with over-fitting on individual person's learning, data discontinuously recognition by independent segmenting and fake activity. This paper suggests a detection method for exercise activity based on the physiological response principle of heart rate up and down during exercise. This proposed method calculates activity intensity and heart rate from triaxial and photoplethysmography sensor to determine a heart rate recovery, then detects exercise by estimating activity intensity or detecting a heart rate rising state. Experimental results show that our proposed algorithm has 98.64% of averaged accuracy, 98.05% of averaged precision and 98.62% of averaged recall.

  • Cytosolic Ca2+ levels ([Ca2+]c) change dynamically in response to inducers, repressors, and physiological conditions, and aberrant [Ca2+]c concentration regulation is associated with cancer, heart failure, and diabetes. Therefore, [Ca2+]c is considered as a good indicator of physiological and pathological cellular responses, and is a crucial biomarker for drug discovery. A genetically encoded calcium indicator (GECI) was recently developed to measure [Ca2+]c in single cells and animal models. GECI have some advantages over chemically synthesized indicators, although they also have some drawbacks such as poor signal-to-noise ratio (SNR), low positive signal, delayed response, artifactual responses due to protein overexpression, and expensive detection equipment. Here, we developed an indicator based on interactions between Ca2+-loaded calmodulin and target proteins, and generated an innovative GECI sensor using split nano-luciferase (Nluc) fragments to detect changes in [Ca2+]c. Stimulation-dependent luciferase activities were optimized by combining large and small subunits of Nluc binary technology (NanoBiT, LgBiT:SmBiT) fusion proteins and regulating the receptor expression levels. We constructed the binary [Ca2+]c sensors using a multicistronic expression system in a single vector linked via the internal ribosome entry site (IRES), and examined the detection efficiencies. Promoter optimization studies indicated that promoter-dependent protein expression levels were crucial to optimize SNR and sensitivity. This novel [Ca2+]c assay has high SNR and sensitivity, is easy to use, suitable for high-throughput assays, and may be useful to detect [Ca2+]c in single cells and animal models.

Recognition Performance of Vestibular-Ocular Reflex Based Vision Tracking System for Mobile Robot (이동 로봇을 위한 전정안반사 기반 비젼 추적 시스템의 인식 성능 평가)

  • This paper presents a recognition performance of VOR (Vestibular-Ocular Reflex) based vision tracking system for mobile robot. The VOR is a reflex eye movement which, during head movements, produces an eye movement in the direction opposite to the head movement, thus maintaining the image of interested objects placed on the center of retina. We applied this physiological concept to the vision tracking system for high recognition performance in mobile environments. The proposed method was implemented in a vision tracking system consisting of a motion sensor module and an actuation module with vision sensor. We tested the developed system on an x/y stage and a rate table for linear motion and angular motion, respectively. The experimental results show that the recognition rates of the VOR-based method are three times more than non-VOR conventional vision system, which is mainly due to the fact that VOR-based vision tracking system has the line of sight of vision system to be fixed to the object, eventually reducing the blurring effect of images under the dynamic environment. It suggests that the VOR concept proposed in this paper can be applied efficiently to the vision tracking system for mobile robot.

  • The period and strength of the pulse on the radial artery are important physiological factors, and they have been used to diagnosis in both Western and Eastern countries for a long time and has been developed as a unique method of diagnosis at each countries. Recently, there are a lot of systems which can give diagnosis information by recording the pulse wave and analyzing the characteristics of the pulse shape. This study describes the Pulse-Wave Measurement System which is able to measure the pulse wave signal using piezoresistive sensor and the pulse wave signal measured by the developed system is transmitted to a computer on the basis of the USB Driver. It has finally shown the the pulse wave signal measured by the sender is appeared to the host PC in real time. The Pulse-Wave Measurement System used the piezoresistive sensor to measure the pulse wave signal and the differential amplifier(AD620) to amplify the pulse wave signal which is small signal. And it used the ADC to convert analog to digital for the measured analog signal and the interface with a computer. It transmitted the measured pulse signal through USB transmission module to the host computer and Labview tool shows it. This Pulse-Wave measurement system will afford comvenience of detecting pulse wave to user related to oriental medicine.

Development and physiological assessments of multimedia avian esophageal catheter system

  • We developed multimedia esophageal catheters for use with birds to measure and record ECG and angular velocity while anesthesized, at rest, and in flight. These catheters enable estimates of blood pressure based on readings given by an angular velocity sensor and by RR intervals of ECG affected by EMG. In our experiments, the catheters had the following characteristics: 1. Esophageal catheters offer a topological advantage with 8-dB SNR improvement due to elimination of electromyography (EMG). 2. We observed a very strong correlation between blood pressure and the angular velocity of esophageal catheter axial rotation. 3. The impulse conduction pathway (Purkinje fibers) of the cardiac ventricle has a direction opposite to that of the mammalian pathway. 4. Sympathetic nerves predominate in flight, and RR interval variations are strongly suppressed. The electrophysiological data obtained by this study provided especially the state of the avian autonomic nervous system activity, so we can suspect individual's health condition. If the change of the RR interval was small, we can perform an isolation or screening from the group that prevent the pandemics of avian influenza. This catheter shall be useful to analysis an avian autonomic system, to perform a screening, and to make a positive policy against the massive infected avian influenza.

Quality Estimation of Net Packaged Onions during Storage Periods using Machine Learning Techniques

  • Onions are a significant crop in Korea, and cultivation is increasing every year along with high demand. Onions are planted in the fall and mainly harvested in June, the rainy season, therefore, physiological changes in onion bulbs during long-term storage might have happened. Onions are stored in cold room and at adequate relative humidity to avoid quality loss. In this study, bio-yield stress and weight loss were measured as the quality parameters of net packaged onions during 10 weeks of storage, and the storage environmental conditions are monitored using sensor networks systems. Quality estimation of net packaged onion during storage was performed using the storage environmental condition data through machine learning approaches. Among the suggested estimation models, support vector regression method showed the best accuracy for the quality estimation of net packaged onions.

  • Blood pressure is one of the key physiological parameters for determining human health, and can prove whether human cardiovascular function is healthy or not. In general, what we call blood pressure refers to arterial blood pressure. Blood pressure fluctuates greatly and, due to the influence of various factors, even varies with each heartbeat. Therefore, achievement of continuous blood pressure measurement is particularly important for more accurate diagnosis. It is difficult to achieve long-term continuous blood pressure monitoring with traditional measurement methods due to the continuous wear of measuring instruments. On the other hand, radar technology is not easily affected by environmental factors and is capable of strong penetration. In this study, by using machine learning, tried to develop a linear blood pressure prediction model using data from a public database. The radar sensor evaluates the measured object, obtains the pulse waveform data, calculates the pulse transmission time, and obtains the blood pressure data through linear model regression analysis. Confirm its availability to facilitate follow-up research, such as integrating other sensors, collecting temperature, heartbeat, respiratory pulse and other data, and seeking medical treatment in time in case of abnormalities.

A Pumilio Activity Sensor Reveals Bag-of-Marbles Inhibition of Pum Activity in the Drosophila Ovary

  • Pumilio (Pum) is an RNA-binding protein and translational repressor important to diverse biological processes. In the Drosophila ovary, Pum is expressed in female germline stem cells (GSCs), wherein it acts as an intrinsic stem cell maintenance factor via repressing target mRNAs that are as yet mostly unknown. Pum recognizes the Pum binding sequence (PBS) in the mRNA 3'UTR through its C-terminus Puf domain. Translational repression is mediated through its N-terminal domain, but the molecular mechanism remains largely unknown. We previously showed that Bag-of-marbles, a critical differentiation-promoting factor of female GSCs, physically interacts with the N-terminus of Pum. We further showed that this interaction is critical to Bam inhibition of Pum repressive action in cultured cells, but the physiological relevance was not addressed. Here we design an in vivo GFP translational reporter bearing the PBS in its 3'UTR and show that GFP expression is reduced in cells wherein Pum is known to be active. Furthermore, we demonstrate in pum mutant ovary that this GFP repression requires Pum, and also that the sensor faithfully monitors Pum activity. Finally, we show that forced expression of Bam inhibits Pum-mediated repression, validating that Bam inhibits Pum activity in vivo.

Development of Personalized Respiratory Training Device with Real-time Feedback for Respiratory Muscle Strengthening

  • Objective: The practice of breathing exercises involves altering the depth and frequency of respiration. Strengthening respiratory muscles plays a crucial role in maintaining overall health and well-being. The efficiency of the respiratory system affects not only physical activity but also various physiological processes including cardiovascular health, lung function, and cognitive abilities. The study evaluated the reliability of the developed device for inspiratory/expiratory training using pressure sensors and Bluetooth connectivity with a smartphone application. Design: Design & development research Methods: The research methodology involved connecting a custom-made respiratory sensor to an IMT-PEP BIC Breath device. Various pressure conditions were measured, and statistical analyses were performed to assess reliability and consistency. Results showed high Intraclass Coefficient Correlation (ICC) values for both inspiratory and expiratory pressures, indicating strong test-retest reliability. The device was designed for ease of use and wireless monitoring through a smartphone app. Results: This study conducted at expiratory pressure confirmed the proper operation of the IMT/PEP breathing trainer at the specified pressure setting in the product. The pressure sensor demonstrated high test-retest reliability with an ICC value of 0.999 for both expiratory and inspiratory pressure measurements. Conclusions: The developed respiratory training device measured and monitored inspiratory and expiratory pressures, demonstrating its reliability for respiratory training. The system could be utilized to record training frequency and intensity, providing potential benefits for patients requiring respiratory interventions. Further research is needed to assess the full potential of the device in diverse populations and applications.
