  • Objectives of this study were to select inbred lines which have a good eating quality and desirable segregates during inbreeding of waxy corn. The 64 inbred lines showed a large variance in their kernel shape and weight. 100-kernel weight, pericarp thickness, kernel length, kernel width, and kernel thickness ranged $11.7\~37.3g,\;11\~77{\mu}m,\;5.8\~9.6mm,\;6.5\~10.0mm$, and $4.1\~6.8mm$, respectively. The physicochemical analysis of 64 waxy corn inbred lines showed crude protein, crude fat, free sugar, and amylopectin content ranging $8.7\~15.8\%,\;2.3\~5.8\%,\;1.1\~11.0\%,\;and\;78.5\~93.8\%$, respectively. The texture property analysis of 64 inbred lines by texture analyzer showed a big difference. Gumminess, hardness, and chewiness of 64 inbred lines ranged $91\~383,\;181\~394,\;and\;73\~370$, respectively. The principal component analysis for 14 characteristics related to kernel quality showed that $73.1\%$ of the total variation could be attributed to the first five principal components. Biological meaning of the principal component was explained clearly by the correlation coefficient between principal components and characters. The first principal component appeared to correspond to small kernel and bad eating quality, The second principal component appeared to correspond to large kernel and good eating quality. The 64 inbred lines were classified into 8 groups by the cluster analysis using the first and second principal component. Among the groups, group VII and VIII included inbred lines with good eating quality that had thin pericarp thickness, low protein content, large kernel, and soft tenderness.

  • Amino acid substitution matrices are essential tools for protein sequence analysis, homology sequence search in protein databases and multiple sequence alignment. The PAM matrix was the first widely used amino acid substitution matrix. The BLOSUM series then succeeded the PAM matrix. Most substitution matrixes were developed by using the statistical frequency of substitution between each amino acid at blocks representing groups of protein families or related proteins. However, substitution of amino acids is based on the similarity of physiochemical properties of each amino acid. In this study, a new approach was used to obtain major physiochemical properties in multiple sequence alignment. Frequency of amino acid substitution in multiple sequence alignment database and selected attributes of amino acids in physiochemical properties database were merged. This merged data showed the major physiochemical properties through principle components analysis. Using factor analysis, these four principle components were interpreted as flexibility of electronic movement, polarity, negative charge and structural flexibility. Applying these four components, BAPS was constructed and validated for accuracy. When comparing receiver operated characteristic ($ROC_{50}$) values, BAPS scored slightly lower than BLOSUM and PAM. However, when evaluating for accuracy by comparing results from multiple sequence alignment with the structural alignment results of two test data sets with known three-dimensional structure in the homologous structure alignment database, the result of the test for BAPS was comparatively equivalent or better than results for prior matrices including PAM, Gonnet, Identity and Genetic code matrix.

  • Human chorionic gonadotropin (hCG) is a heterodimeric glycoprotein hormone consisting of non-covalently linked two subunits, the ${\alpha}$ and ${\beta}$ subunit. It has been used as a infertility drug for ovulation to mimic luteinizing hormone $(LH).^{1)}$ A stable cell line was established by transfection of Rc/CMV-i-dhfr-hCG, expression vector containing hCG ${\alpha}-$ and ${\beta}-genes$, into dihydrofolate reductase-deficient CHO cells and subesquent methotrexate-mediated gene amplification. Anchorage-dependent CHO cells were adapted into a serum-free and/or animal component-free suspension medium through gradual serum weaning for the hCG production. The established cell line showed typical morphological characteristics and growth profile of CHO cells, and could produce FSH with passage-to-passage consistency. The high density perfusion culture of the CHO cells was carried out in Celligen Plus bioreactor equipped with a spin-filter as a internal cell retention device. The cell density reached up to $>1x10^{7}$ cells/ml in less than 7 days and a perfusion-control strategy based on cellular consumption rates of glucose was $established.^{2)}$ Biologically active recombinant hCG was purified by a series of chromatographic steps including anion exchange chromatography and hydrophobic interaction chromatography to homogeneity. The highly purified recombinant hCG was characterized for physicochemical, immunological and biological properties.

  • Sesame seed is known as a good nutritional source containing high oil (51%) and protein (20%). Sesame oil contains a very high oxidative stability compared to other vegetable oils. To obtain basic information for quality evaluation, imported and domestic sesame seeds were investigated to measure general components (ash, protein, moisture and oil), fatty acid composition and lignan content. Although the protein contents were the highest in domestic sesame seeds, yet the lipid contents were the highest in imported sesame seeds. Unsaturated fatty acids such as oleic acid and linoleic acids were the highest in the domestic sesame seeds. Lignan contents, the most important component known as antioxidant, were significantly higher in domestic sesame seeds than other imported sesame seeds. These results suggest that domestic sesame seed may have the best quality in terms of the functional components.

  • The surface film, which is formed on graphite negative electrodes during the initial charging, is a key component in lithium secondary batteries. The battery reactions are strongly affected by the nature of the surface film. It is thus very important to understand the physicochemical properties of the surface film. On the other hand, the surface film formation is a very complicated interfacial phenomenon occurring at the graphite/electrolyte interface. In studies on electrode surfaces in lithium secondary batteries, in-situ experimental techniques are very important because the surface film is highly reactive and unstable in the air. In this respect electrochemical atomic force microscopy (ECAFM) is a useful tool for direct visualizing electrode/solution interfaces at which various electrochemical reactions occur under potential control. In the present review, mechanism of surface film formation and its correlation with electrolyte are summarized on the basis of in-situ ECAFM studies for understanding of the nature of the surface film on graphite negative electrodes.

  • Industrial gas drying, dilute gas mixtures purification, air fractionation, hydrogen production from steam reformers and petroleum refinery off-gases, etc are conducted by using adsorptive separation technology. The pressure swing adsorption (PSA) has certain advantages over the other methods, such as absorption and membrane, that are a low energy requirement and cost-effectiveness. A key component of PSA systems is adsorbents that should be highly selective to a gas being separated from its mixture streams and have isotherms suitable for the operation principle. The six standard types of isotherms have been examined in this review, and among them the best behavior in the adsorption of $CO_2$ as a function of pressure was proposed in aspects of maximizing a working capacity upon excursion between adsorption and desorption cycles. Zeolites and molecular sieves are historically typical adsorbents for such PSA applications in gas and related industries, and their physicochemical features, e.g., framework, channel structure, pore size, Si-to-Al ratio (SAR), and specific surface area, are strongly associated with the extent of $CO_2$ adsorption at given conditions and those points have been extensively described with literature data. A great body of data of $CO_2$ adsorption on the nanoporous zeolitic materials have been collected according to pressure ranges adsorbed, and these isotherms have been discussed to get an insight into a better $CO_2$ adsorbent for PSA processes.

  • In order to screen new tyrosinase inhibiting principle which is expected to be a new biofunctional skin whitening cosmetics, we have isolated 600 strains of the marine- derived fungi and investigated tyrosinase inhibiting activity for their acetone extracts. The significant activities (>70% Inhibition) were observed in the extract of 10 strains of fungi (MFA7, MFA27, MFA58, MFA317, MFA318, MFA345, MFA412, MFA552, MFA562, MFA581). These active strains were cultured in SWS medium with 1 L scale and the resulting broth and mycelium were extracted to afford mycelium extract (000M) and broth extract (000B), respectively. Tyrosinase inhibiting activity for all extracts has been tested. As the results, the broth extracts of 4 strains (27B, 58B, 552B and 581B) exhibited relatively high levels of activity of $IC_{50}$ values of $3.0-19.0\;{\mu}g/mL$. The active component of 581B was purified by assay-guided isolation to yield the known kojic acid (1), and its structure was determined by physicochemical evidence. Kojic acid showed the significant tyrosinase inhibitory activity with $IC_{50}$ values of $12.0\;{\mu}M$.

  • Objectives: Chrysotile is mineralogically distinct from amphiboles, displaying a notably different chemical structure. The thin sheets that form chrysotile fiber lead to the ability of the lung/macrophage system to decompose the chrysotile fibers. This study was performed in order to compare the physicochemical characteristics of Korean asbestos with those of Canadian amphiboles. Materials: An acid solubility test for each test substance was done to compare pH 4.5 and pH 1.2 distilled water. Asbestos fibers which had been placed in acid solutions for five days, five weeks and weeks were analyzed with a transmission electron microscope equipped with an energy dispersive X-ray spectrometer (TEM-EDS). Results: The composition element (Mg) of Korean chrysotile, Korean anthophyllite and Canadian amosite significantly decreased from 5 days and also decreased significantly after 5 weeks and 10 weeks. Only the composition (Mg) of Canadian crocidolite did not change under any conditions. From 5 days, the Mg of Korean chrysotile, Korean anthophyllite and Canadian amosite were significantly lower than before the acid treatment, but there were no changes over time or by the pH of the acid solutions. Particularly after 10 weeks, the composition (Mg) of Korean chrysotile in the pH 1.2 acid solution showed a rapid reduction of 15.86%. Conclusions: Korean chrysotile was very weak in an acid environment, beginning to show significant changes after 5 days. The Mg component rapidly decreased after 10 weeks in the pH 1.2 acid solution.
