Comparative Analysis of Phenolic Compound of Mutant Lines of Sorghum (Sorghum bicolor)

  • Sorghum (Sorghum bicolor) is increasingly important as a biomass crop worldwide. Its genetic diversity provides a large range of biochemical composition suitable for various uses as bioplastics. Phenolic compounds are the main compounds of lignocellulosic residues, which can be used as a source of active components for their use in active packaging materials. In this research, we investigated the total phenolic content (TPC) and the total flavonoid content (TFC) among 60 mutant lines (early heading, high biomass and dwarfness) and their original cultivars. Sixty sorghum mutant lines were developed by treatment with gamma-ray or proton irradiation in 14 sorghum cultivars. The levels of TPC and TFC of 14 original cultivars were ranging from 3.27 to 11.54 mg/100 g and 2.39 to 6.74 mg/100 g, respectively. The TPCs of the mutant lines were ranging from 1.92 to 13.10 mg/100 g with average content of 6.35 mg/100 g. The TFCs of the mutant lines were ranging from 1.72 to 8.30 mg/100 g with average content of 4.20 mg/100 g. Three mutant lines derived from gamma-ray showed significant lower TPC and TFC than those of the original cultivar. While, five mutant lines showed significant higher TPC and TFC. These findings will be useful for the selection of sorghum genotypes with improved phenolic compounds.

Phenol-Rich Compounds Sweet Gel: A Statistically More Effective Antibiotic than Cloxacillin Against Pseudomonas Aeruginosa

  • Objectives: The purpose of this study was to obtain a natural antibiotic from Phenol-rich compounds; for the dressing and the treatment of chronic wounds. Methods: The Phenol-rich compound sweet gel was prepared by blending four natural herbal extracts, Acacia catechu (L.F.), Momia (Shilajit), Castanea sativa, and Ephedra sinica stapf, with combination of a sweet gel medium, including honey, maple saps, Phoenix dactylifera L. (date), pomegranate extract and Azadirachta indica gum as a stabilizer. The combinations were screened by using a well-diffusion assay with cloxacillin as a control. Pseudomonas spp. was tested with our novel antimicrobial compound. The zones of inhibition in agar culture were measured for each individual component and for the compound, and the results were compared with those of the control group which had been treated with cloxacillin. Data were expressed as means ${\pm}$ standard deviations. Quantitative analyses were performed using the paired t-test. Results: The antibiotic effect of the Phenol-rich compound sweet gel was statistically shown to be more significant than that of cloxacillin against Pseudomonas aeruginosa (P < 0.05). Conclusion: Our novel approach to fighting the antibiotic resistance of Pseudomonas proved to be successful. The Phenol-rich compound sweet gel was found to be suitable for use as an alternative medicine and bioactive dressing material, for the treatment of patients with various types of wounds, including burns, venous leg ulcers, ulcers of various etiologies, leg ulcers on the feet of diabetic, unhealed graft sampling sites, abscesses, boils, surgical wounds, necrotic process, post-operative and neonatal wound infection, and should be considered as an alternative to the usual methods of cure.

Agricultural Characters, Phenolic and Nutritional Contents, and Antioxidant Activities of Pigeon Pea (Cajanus cajan) Germplasms Cultivated in the Republic of Korea

  • Pigeon pea (Cajanus cajan) is an important legume species that produces seeds that are rich in phenolic compounds and dietary nutrients. The aim of the present study was to evaluate the agricultural characters, phenolic and nutritional contents, and antioxidant capacities of seeds from 82 pigeon pea germplasms cultivated in the Republic of Korea. The accessions exhibited compact (24.39%), semi-spreading (74.39%), or spreading (1.22%) growth habits and determinate (89.02%), indeterminate (8.54%), or semi-determinate (2.44%) flowering patterns. Days to 75% maturity ranged from 30 to 72 d, and yield per plant ranged from 6.00 to 148.60 g. Meanwhile, total phenolic, crude protein, crude fiber, and dietary fiber contents ranged from 16.42 ± 0.62 to 29.67 ± 0.43 mg gallic acid equivalent per g of dried extract, from 16.76 ± 6.74% to 22.61 ± 0.05%, from 4.70 ± 0.24 to 8.63 ± 0.02%, and from 12.98 ± 0.71 to 33.19 ± 1.50%, respectively. In addition, DPPH radical-scavenging capacity ranged from 1.61 ± 0.10 to 16.04 ± 2.30 mg ascorbic acid equivalent per g of dried extract, and Trolox equivalent antioxidant capacity ranged from 3.03 ± 0.86 to 42.24 ± 0.72 mg Trolox equivalent per g of dried extract. Phenolic content was correlated with both DPPH radical-scavenging capacity (r = 0.63) and Trolox equivalent antioxidant capacity (r = 0.29). Nine accessions (IT170290, IT170291, IT170270, IT170276, IT170379, IT170386, IT170388, IT170418, and IT170340) exhibited early maturity, compact and erect growth habits, and above average antioxidant activities and phenolic and protein contents. In addition, accessions IT170290 and IT170291 were especially promising pigeon pea germplasms to grow, owing to various favorable characteristics (e.g., high yield and dietary fiber content). Hence, these accessions could be useful cultivars to the Republic of Korea if considered in future agricultural systems.

밭 잡초(雜草)중에 존재(存在)하는 Allelopathy 관련(關聯) Phenol 화합물(化合物)의 검색(檢索) (Determination of Phenolic Compounds Responsible for Allelopathy in Upland Weeds)

  • 밭 주요(主要) 우점잡초(優占雜草) 중 allelopathy 작용성(作用性)을 보였던 쑥, 냉이, 쇠비름 및 토끼풀에 대하여 이들 잡초(雜草) 중에 존재(存在)하는 phenol화합물(化合物)을 Paper chromatography(PC) 및 High performance liquid chromatography(HPLC)에 의하여 분리(分離) 확인(確認)하였고, 검출(檢出)된 phenol화합물(化合物)이 검정식물(檢定植物)의 발아(發芽) 및 생장(生長)에 미치는 영향(影響)을 검토(檢討)하였다. PC에 의하여 검정(檢定)된 phenol화합물(化合物)은 4초종(草種) 모두에서 methanol추출물(抽出物) 보다는 물추출물(抽出物)에서 많은 종류(種類)가 확인(確認)되었다. 4초종(草種)의 두가지 추출물(抽出物) 모두에서 PC로 분리(分離)된 phenol화합물(化合物)은 ferulic acid이었으며, benzoic acid는 물추출물(抽出物)에서만 확인(確認)되었다. HPLC에 의해서는 hydroquinone, p-hydroxybenzoic, ferulic 및 cinnamic acid가 전초종(全草種)에서 분리(分離)되었다. PC 및 HPLC로 분리(分離) 확인(確認)된 phenol화합물(化合物) 중 p-hydroxyhenzoic, ferulic 및 cinnamic acid는 검정식물(檢定植物)의 발아(發芽) 및 발아후(發芽後) 생육(生育)에 높은 억제효과(抑制效果)를 보였다. 이들 phenol화합물(化合物)은 일반적(一般的)으로 발아(發芽)보다는 유묘(幼苗)의 근생장(根生長)에 더 큰 저해(沮害)를 나타내었다.

유기농 살구, 개암, 오디, 감 및 석류나무 잎의 생육단계별 총 페놀화합물과 항산화 활성변화 (Seasonal Changes of Total Phenolic Compounds and Antioxidant Activity in Leaves of Organic Apricot, Filbert, Mulberry, Persimmon and Pomegranate Trees)

  • 유기농으로 관리된 살구, 개암, 오디, 감 및 석류나무로부터 5월부터 10월까지 약 1개월 간격으로 잎을 채취하여 총 페놀화합물과 항산화 활성도를 측정한 결과는 다음과 같다. 살구 잎의 총 페놀화합물 함량에 있어서 5월에 18.9 mg으로부터 점차 증가되다가 7월에 최대치에 도달된 후 감소되었고 10월에 9.7 mg으로 최소치를 나나내었다. 항산화력은 5월에 13.9%로 나타났고 6월 및 7월까지 계속 증가추세를 보였으며 이후 급격히 저하되어 10월에 7.1%로 최저치를 보였다. 개암 잎의 총 페놀화합물 함량에 있어서 5월에 59.1 mg이던 것이 6월에 약간 감소되었다가 7월에 80.6 mg으로 피크를 보인 후 8월에 감소세로 돌아섰고 뒤이어 10월에 33.5 mg으로서 급격히 낮아졌다. 항산화 활성에 있어서도 5월 이후 꾸준하게 증가되어 7월에 64.3%로 최고점에 도달한 후 8~10월까지 지속적으로 감소되었다. 오디나무잎의 총 페놀화합물 함량에 있어서 전반적으로 다른 과종에 비하여 현저히 낮은 수준을 나타내었다. 5월부터 6월까지 계속 증가되다가 7월 이후 2.1 mg까지 급격히 떨어졌고, 8~10월에는 1 mg 수준까지 낮아졌다. 항산화력에 있어서도 전반적으로 10% 이하의 낮은 수준을 나타내었고 특히 7월 이후에는 3% 수준의 낮은 활성도를 보였다. 감나무 잎의 총 페놀화합물 함량에 있어서 5월에 66.2 mg으로 가장 높은 함량을 보였고 이후 6월부터 8월에 이르도록 점차 감소되다가 10월에 현저히 저하되었다. 그러나 항산화력은 5월보다 6월이 더 높았으며 이후 7월 및 8월에 이르도록 뚜렷이 감소되다가 10월에는 완만히 낮은 상태를 유지하였다. 석류나무 잎의 총 페놀화합물 함량에 있어서 조사된 살구, 개암, 오디 및 감나무 잎에 비하여 월등하게 높은 함량을 생육기간 동안 유지하고 있었고, 특히 5월에 238.3 mg으로 최대치를 나타낸 후 6~8월까지 근소하게 낮아진 상태를 유지하다가 10월에 128.9 mg으로 급격히 감소되었다. 한편 항산화력은 생장이 왕성한 봄~여름까지 95% 이상의 높은 상태를 유지하다가 10월에 91.1%로 약간 낮아지는 경향을 보였다. 총 페놀화합물과 항산화력 간의 상관관계에 있어서, 오디나무 잎의 상관계수가 가장 높았고, 다음으로 감, 개암, 및 살구나무 잎의 순이었다. 석류 잎은 상관계수가 0.506으로서 낮아서 총 페놀화합물과 항산화력 간에 상관성이 없는 것으로 나타났다.

  • For the investigation of medicinal resources from Taraxacum species, the studies were carried out to evaluate the pharmaco-constituents in the aerial part of Taroxacum hallaisanensis, an endemic plant of Korea. From BuOH fraction of the MeOH extract, compound 1 (protocatechuic acid, $C_7H_6O_4,\;3,4-dihydroxy\;benzoic\;acid)$, compound 2 $[C_{22}H_{31}O_6,\;luteolin-7-O-{\alpha}-_L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-_D-glucopyranoside]$, and compound 3 $[C_{15}H_{20}O_6,\;luteolin-7-O-{\beta}-_D-glucopyranoside]$ were isolated by column chromatographic separation using polyamide and ODS-gel. The structures were elucidated by means of physico-chemical evidences($^1H-NMR,\;{12}^C-NMR$, IR, EI-Mass, FAB-Mass and GC).

Control Efficacy of Phloretin Isolated from Apple Fruits Against Several Plant Diseases

  • In the course of a searching natural antifungal compounds from plant sources, we found that the methanol extract ($3,000\;{\mu}g/ml$) of Malus domestica fruits had potential of control against rice blast (Magnaporthe grisea) and tomato late blight (Phytophthora infestans). Under bioassay-guided purification, we isolated phloretin, a phenolic compound, with in vivo antifungal activity against M. grisea. By 1-day protective application of phloretin ($500\;{\mu}g/ml$), the compound strongly inhibited the disease development of M. grisea and P. infestans on rice and tomato seedlings, respectively. And red pepper anthracnose caused by Colletotrichum coccodes also was moderately suppressed. However, rice sheath blight (Rhizoctonia solani AG1), and barley powdery mildew (Blumeria graminis f. sp. hordei) were hardly controlled. In addition, the compound showed in vitro antifungal activity against some plant pathogenic fungi including Phytophthora capsici, Alternaria panax, Sclerotinia sclerotiorum, R. solani AG4, and M. grisea. This is the first report on the antifungal activity of phloretin against plant pathogenic fungi.

Trametes versicolor 의한 triphenyl methane계 염료의 분해 (Biodegradation of triphenyl methane dyes by white rot fungus, Trametes versicolor)

  • 구름버섯(Trametes versicolor)은 phenolic compound인 CV와 MG를 효과적으로 탈색할 수 있었으며 고체와 액체배양 상태 모두에서 CV보다 MG를 더 효과적으로 탈색시켰다. 구름버섯에 의한 두 색소의 탈색 과정에서 phenolic compounds를 분해하는 것으로 알려진 세 가지 효소 중 laccase의 활성이 가장 높았다. MnP 역시 적은 수치지만 활성을 나타냈으며 LiP의 활성은 나타나지 않았다. 따라서 구름버섯에 의한 합성염료의 분해과정에서 laccase가 주로 사용되고 MnP는 탈색과정에서 보조적인 작용을 하는 것으로 추정된다. 그러나 CV의 경우 MnP가 활발하게 염료분해에 관여하는 것으로 판단된다. 또한 MG가 대부분 탈색되었을 때의 laccase 활성(0.16 U/mg)이 CV가 대부분 탈색되었을 때의 활성(0.23 U/mg)보다 현저하게 낮은 것으로 보아 구름버섯이 CV를 탈색시키는데 더 높은 활성의 laccase가 필요로 하는 것이 밝혀졌다. 본 실험에서 한국산 구름버섯 종의 CV와 MG 탈색능력이 확인되었으며 앞으로 한국산 구름버섯을 이용한 triphenyl methane계에 속하는 합성염료의 분해에 관한 친환경적 처리기술 개발에 도움이 될 것으로 기대된다.

유황시용(硫黃施用)이 배추 품질(品質)과 수량(收量)에 미치는 영향(影響) (Effect of Sulphur on the Yield and Some Quality of Chinese Cabbage)

  • 배추에 유황(硫黃)을 시용(施用)한 후(後) 수량(收量), 수확기(收穫期) 토양(土壤)과 엽중(葉中) 유기(有機), 무기성분(無機成分)의 변화(變化)와 품질(品質) 개선(改善) 효과(效果)를 연구(硏究)한 결과(結果)는 다음과 같다. 1. 유황(硫黃)을 배추에 10~30kg/10a 시용(施用)하였을 때 약 10% 증수(增收)되었다. 2. 토양(土壤) 분석(分析) 결과(結果) 유황(硫黃) 시용(施用)으로 유효(有效) 유황(硫黃). 함량(含量)이 증가(增加)하고 그 반면 pH가 낮아졌다. 3. 유황(硫黃) 시용(施用)에 따라 배추 엽중(葉中) nitrate의 함량(含量)이 감소(減少)되었다. 4.유황시용(硫黃施用)으로 배추의 맛과 향미(香味)에 관련(關聯)되는 water-solubl pectin, phenolic compound, sugar, vitamin C의 함량(含量)이 증가(增加)되었으며 식미(食味) 검사(檢査)에서는 유황시용(硫黃施用)으로 배추의 독특(獨特)한 향기(香氣), 맛과 조직감(組織感)에서 훨씬 향상(向上)되었다.

Enhancement of Polyphenol Content and Antioxidant Activity of Brown Alga Eisenia bicyclis Extract by Microbial Fermentation

  • The objective of this study was to select an effective microbial strain to improve the functional qualities of Eisenia bicyclis water extract by fermentation. For this purpose, several microorganisms isolated from traditional Korean fermented foods were inoculated and cultivated in E. bicyclis water extract. Ultimately, yeast strain YM-1 was selected for further study based on its total phenolic compound (TP) content and antioxidant activity, which were enhanced by microbial fermentation. The extract fermented by YM-1 exhibited a superior TP content and 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity compared to extracts fermented by other microbes. The highest TP content and DPPH radical scavenging activity were observed after one day of YM-1 fermentation. Yeast strain YM-1 was identified as Candida utilis based on an analysis of its physiological characteristics. During fermentation of the extract by C. utilis YM-1, no significant difference was observed in the proximate composition, including moisture, crude lipid, crude protein, and crude ash. Fermentation by C. utilis YM-1 resulted in enhanced biological activity, including increases in the TP content and antioxidant activity. Thus, fermentation by C. utilis YM-1 is an attractive strategy for developing value-added food ingredients.
