Major Foods for folate and Their Folate Contents of Korean Child-bearing Women (가임기 여성의 주요 엽산급원식품 및 동 식품의 엽산함량 분석)

  • In this study, we determined major foods and food groups which contributed to the intake of folate in korean child-bearing women. Folate content of the major foods was also analyzed. A total of 91 healthy women with childbearing potential participated. They were divided into one of three groups by age; A (15∼24 yrs), B(25∼34 yrs) and C (35∼49 yrs). Food consumption data of the subjects were obtained by the 24-hr recall method. The rate of contribution of each food to the total intake of dietary folate was determined. Seventy major foods for folate were selected from the 178 foods they consumed. Two sets of the 70 foods were collected from different markets. After extracting folates from the foods by heating with a HEPES-CHES buffer, the samples were treated with α-amylase, protease and folate intakes was Korean cabbage kimchi followed by laver, soybean sprout, rice, yolmu kimchi, eggs, lettuce, perilla leaves, sea mustard and radish root. Food items that contributed to folate intake were slightly different among the age groups. Although the folate content of Korean cabbage kimchi was 51.6 ㎍/100g, but due to the high amount of intake, it was ranked the first major food prviding folate. High folate foods containing over 100㎍/100g were laver, corn flakes, spinach, mungbean, sesame, quail's eggs, small radish kimchi, kidney beans, leeks and peanuts. However, based on the serving size, spinach, corn flakes, leeks, sweet potato, pepper leaves, quail's eggs, crown daisy, small radish kimchi and perilla leaves contributed above 50㎍ of folate per serving size.

Comparison of Dietary Fiber and Amino Acid Composition in Frequently Consumed Vegetables and Fruits (다소비 채소 및 과일의 식이섬유와 아미노산 조성 비교)

  • This study was conducted to analyze the contents in the dietary fiber and amino acid compositions of 23 vegetables and 6 fruits and to identify the current contents. We generated data on the contents of total, insoluble and soluble dietary fiber (TDF, IDF and SDF), respectively. The TDF and IDF contents were lowest (0.56 g/100 g and 0.44 g/100 g) in wild garlic and highest (5.87 g/100 g and 5.66 g/100 g) in perilla leaves. A total of 18 kinds of amino acids were found in most samples. Essential and non-essential amino acid contents ranged from 53.16-2107.54 mg/100 g and 191.66-2892.28 mg/100 g, respectively. The highest essential and non-essential amino acid content was hot pepper leaves followed by perilla leaves and aralia. They had higher contents of both TDF and amino acids. The results of the study can serve as a fundamental source of information in DF and amino acids for diet planning.

  • The full-length cDNA encoding Perilla frutescens limonene synthase (PFLS) (603 amino acids, GenBank accession no. D49368) was cloned. To elucidate the role of PFLS in gene regulation, we transiently transformed full-length PFLS into tobacco plants. PFLS mRNA was first detected in the intact leaves of the plants at 6 h, and the LS transcript level increased after 12 h in leaves treated with oxidative stress-related chemicals. The transient overexpression of PFLS resulted in increased transcription of NbPR1 and NbSIP in Nicotiana benthamiana leaves. Thus, our result confirmed that the infiltration of PFLS gene act as a transcriptional regulator of NbPR1 or NbSIP genes in the tobacco.

Optimization of Roasted Perilla Leaf Tea Using Response Surface Methodology (반응표면분석을 이용한 들깨잎차 볶음처리의 최적화)

  • Response surface methodology (RSM) was applied in roasting processes of perilla leaves to develop a high quality perilla leaf tea. The Hunter color parameters and electron donating ability were monitored to optimize organoleptic properties of perilla leaf tea. The roasting processes were based on the central composite design with primary variables-roasting temperature $(140{\sim}220^{\circ}C)$, time $(5{\sim}25)$, and reaction variables-sensory test, electron donating ability. From the variables, the roasting condition was optimized using statistical analysis system (SAS) program as developing the functional tea using perilla leaf. Hunter color L and b values of the powdered samples increased with the roasting processes, but Hunter color a value decreased. Electron donating ability was influenced by roasting temperature (p<0.01) and time (p<0.01), and optimum condition selected was at $220^{\circ}C$ for 15 min with coefficient of determinations $(R^2)$ above 0.98. After preference test of perilla leaf tea using parameter of taste, color, and flavor, we can estimate that the optimal roasting condition of preilla leaf for function tea manufacturing are $210{\sim}220^{\circ}C$ for $10{\sim}20$ min by response surface methodology (RSM). Tyrosinase, xanthine oxidase and electron donating ability were 10.14, 14.37 and 59.19% of perilla leaf tea.

  • 들깨(Perilla frutescens var. japonica)는 꿀풀과(Lamiaceae)에 속하는 1년생초로써 키는 60-90cm 정도이며, 줄기는 네모지고 곧게 서며 긴 털이 있다. 잎은 마주나고 달걀 모양원형으로 뾰족하며 밑부분은 둥글다. 잎은 길이 7-12cm, 나비 5-8cm로 톱니가 있고 앞면은 녹색이지만 뒷면에는 자줏빛이 돈다. 들깨는 식용계란, 돼지고기, 양식어류 등의 생산을 위한 기능성 보조식품의 사료와 한방에서는 만성위염, 기침, 위산과다 등의 약용으로도 쓰이고 있다. 또한 들깻잎은 특유의 향취로 인해 육류나 생선회, 영양탕 등의 음식점에서 상추와 함께 대량소비가 되고 있다.   • Antimutagenic activity of the enzymatic browning reaction products (EBRPs) was investigated by using the spore rec-assay with Bacillus subtilis strains H17 $(rec^+)\;and\;M45 (rec^-)$. The EBRPs tested were prepared from the reactions of five different kinds of polyphenols with polyphenol oxidase isolated from the leaves Perilla frutescens. In the spore rec-assay, most of the polyphenolic compounds tested showed positive, whereas only their tested compound showed negative respectively. In addition of polyphenol oxidase inhibitors such as cysteine, glutathione and ascorbic acid to the reaction mixtures consisted with the polyphenol oxidase and polyphenols, the mutagenic effects were increased in the spore recassay. These results show that the activity of polyphenol oxidase may play an important role in the reduction of mutagenicity of polyphenols.

  • The purpose of this study was to investigate the contents of apigenin and luteolin in vegetables mainly distributed and consumed in Korea. In this study, the contents of apigenin, apigenin-7-O-glucoside, luteolin, and luteolin-7-O-glucoside in vegetables were surveyed by using liquid chromatography coupled to mass spectrometry (LC-MS/MS). According to the analysis of 27 items (91 samples) in vegetables, the content of total apigenin (the sum of apigenin and apigenin-7-O-glucoside) was quantified in 8 out of the 27 items in vegetables, followed by pepper leaves, parsley, celery, chamnamul, foremost mugwort, and perilla leaves. The content of total luteolin (the sum of luteolin and luteolin-7-O-glucoside) was found in 11 of the 27 items in vegetables, followed by pepper leaves, dandelion, celery, red lettuce, foremost mugwort, and perilla leaves. Celery was divided into stalks and leaves for comparing the contents of apigenin and luteolin. Celery showed higher contents of apigenin and luteolin in leaves than in stalks.

Effect of Temperature and Food Source on the Egg and Larval Development of Tobacco Cutworm, Spodoptera litus Fabricius (온도 및 기주조건이 담배거세미나방(Spodoptera litura)의 난 및 유충발육에 미치는 영향)

  • This study was conducted to determine the effect of temperatures and food sources on the egg and larval developmentof the tobacco cutworm, Spodoptera litura Fabricius. The hatchability of egg masses of S. liturawas 100% on the leaf of soybean, perilla and sweet potato in any given temperature regimes, while the hatchabilitywas only 65-8796 when reared on the pulp paper and decreased as temperature increased. Egg durationwas not significantly different among different food sources within each temperature. However, egg duration at32$^{\circ}$C was shorter than that at 24$^{\circ}$C and 28$^{\circ}$C. During the early larval development, at 28$^{\circ}$C and 32$^{\circ}$C the larvafed on sweet potato leaf was heavier than those fed on soybean and perilla leaves and the opposite case wastrue during mid-larval development stage. However, larval weight at 24$^{\circ}$C was heavier on sweet potato leafthan that on soybean and perilla leaves until 12 days after hatching. This result was probably due to relativelyslower developmental rate at 24$^{\circ}$C compared to 28$^{\circ}$C and 32$^{\circ}$C. The mean larval mortality was 68.896, 44.5%and 33.8% at 24$^{\circ}$C. 28$^{\circ}$C and 32"C, respectively. The lowest mortality was observed on soybena leaf and followedby perilla and sweet potato leaves, and artificial diet regardless of temperature conditions. The durationwas the shortest when they fed on soybean leaf, and followed by perilla and sweet potato leaves and artificialdiet. Larval durations were 23.6-30.4 days at 24$^{\circ}$C. 18.6-22.3 days at 28$^{\circ}$C and 14.5-18.0 days at 32$^{\circ}$C. Thethreshold temperatures of egg and larva of S. litura were about 6.l"C and 10.9"C, respectively.t 6.l"C and 10.9"C, respectively.pectively.

Growth Modeling of Perilla frutescens (L.) Britt. Using Expolinear Function in a Closed-type Plant Factory System (완전제어형 식물공장에서 선형지수함수를 이용한 들깨의 생육 모델링)

  • Growth modeling in plant factories can not only control stable production and yield, but also control environmental conditions by considering the relationship between environmental factors and plant growth rate. In this study, using the expolinear function, we modeled perilla [Perilla frutescens (L.) Britt.] cultivated in a plant factory. Perilla growth was investigated 12 times until flower bud differentiation occurred after planting under light intensity, photoperiod, and the ratio of mixed light conditions of 130 μmol·m-2·s-1, 12/12 h, red:green:blue (7:1:2), respectively. Additionally, modeling was performed to predict dry and fresh weights using the expolinear function. Fresh and dry weights were strongly positively correlated (r = 0.996). Except for dry weight, fresh weight showed a high positive correlation with leaf area, followed by plant height, number of leaves, number of nodes, leaf length, and leaf width. When the number of days after transplanting, leaf area, and plant height were used as independent variables for growth prediction, leaf area was found to be an appropriate independent variable for growth prediction. However, additional destructive or non-destructive methods for predicting growth should be considered. In this study, we created a growth model formula to predict perilla growth in plant factories.

Inhibitory Activity of Nitric Oxide Synthase and Peroxynitrite Scavenging Activity of Extracts of Perilla frutescens (들깨 잎 추출물의 Nitric Oxide Synthase 저해활성 및 Peroxynitrite 소거활성)

  • Activated microglia by neuronal injury or inflammatory stimulation overproduce nitric oxide (NO) by inducible nitric oxide synthase (iNOS) and reactive oxygen species (ROS) such as superoxide anion, resulting in neurodegenerative diseases. The toxic peroxynitrite (ONOO$^-$), the reaction product of NO and superoxide anion further contributes to oxidative neurotoxicity. We tried to evaluate the effects of two kinds of varieties of Perilla frutescens var japnica Hara on the NO production in lipopolysaccharide (LPS)-activated microglia. The perilla cultivars of Namcheondeulkkae (NC) and Boradeulkkae (BR) were developed by pure line from the local variety and by a cross between 'deulkkae' and 'chajogi', respectively. Spirit, hexane, chloroform and butanol fractions of the leaves of NC and BR inhibited the production of NO in LPS-activated microglia. The fractions of BR showed stronger activity than NC and the spirit extracts was the most potent in both cultivars. The solvent fractions of BR suppressed the expression of protein and mRNA of iNOS in LPS-activated microglial cells. Moreover, the extracts of NC and BR showed the activity of peroxynitrite scavenging in cell free bioassay system. These results imply that Namcheondeulkkae and Boradeulkkae might have neuroprotective activity through the inhibition of NO production by activated microglial cells and peroxynitrite scavenging activity.
