저장미곡중의 균류에 관한 연구 (Studies on the Fungi in Stored Rice)

  • 미곡저장중 균류에 의한 미곡의 손실을 방지하기 위하여 1978년 12월 부터 1979년 6월까지 7개월간 싸일로, 평창고 및 통가리에 저장한 미곡시료(밀양23호)에서 균류를 분리, 동정하였다. 한국산 벼 및 현미에서 분리된 30종의 균류중, Aspergillus속 11종(A. caespitosus, A. condidus, A. chevalieri, A fischeri, A. fumigatus, A. fiavus, A. nidulans, A. oryzae, A. ruber, A. sydowii, A. versicolor), Penicillium 속 5 종 (P. atramentosum P. chrysogenum, P. cyaneofulvum, P. nototum, P. steckii), Alternaria 속 2종 (Al. faesiculata, Al. grisea) Curvalaria 속 2종 (C, interseminata, C. tetromea), Trichothecium roseum, Nigrospora sphaerica, Rhizopus nigricans, Fusarium spp., Mucor spp., Helminthosporium spp., 및 Gliocladiopsis spp. 각 1종, 총 27종이 동정, 확인되었다. 한국산 벼에서 분리된 표면균류는 A. flavus와 A. candidus가 대부분이었으며, 현미에서는 A. sydowii가 많이 출현하였다. 또한 저장중 변질미에는 A. candidus, A. versicolor 및 A. glaucus group들이 주로 많았다.

제주도 야생버섯 hyphosphere 토양에서 분리된 국내 미기록 진균 11종 보고 (Eleven previously unrecorded fungal species isolated from hyphosphere soil supporting wild mushrooms in Jeju Island)

  • 야생버섯의 결실에 미치는 미생물의 영향을 파악하기 위하여 제주도 곶자왈 지역에서 발견된 야생버섯, 끈적버섯 (Cortinarius violaceus), 달걀버섯 (Amanita hemibapha), 색시졸각버섯 (Laccaria vinacelavellanea), 흰알광대버섯 (Amanita verna)의 근권토양 균류를 조사하였다. 형태학적 특성 조사와 ITS, LSU rDNA, β-tubulin 유전자 서열에 대한 분자생물학적 분석을 바탕으로 동정하는 과정에서 국내 미기록 진균 11종을 분리하였다. 이들 진균은 Arthrinium kogelbergensis, Kalmusia longisporum, Keithomyces carneum, Neopyrenochaeta cercidis, Penicillium ranomafanaense, Phomatodes nebulosa, Pyrenochaeta nobilis, Tollypocladium album, Talaromyces kendrickii, Talaromyces qii, Umbelopsis gibberispora이다. 이들 11종에 진균에 대한 형태적 특성과 계통발생학적 위치를 기술하였다.

Isolation and Cytotoxic Potency of Endophytic Fungi Associated with Dysosma difformis, a Study for the Novel Resources of Podophyllotoxin

  • Endophytic fungi are promising sources for the production of podophyllotoxin-an important anticancer compound, replacing depleted medical plants. In this study, the endophytes associated with Dysosma difformis-an ethnomedicinal plant species were isolated to explore novel sources of podophyllotoxin. Fifty-three endophytic fungi were isolated and identified by morphological observation and ITS-based rDNA sequencing, assigning them to 27 genera in 3 divisions. Fusarium was found the most prevalent genus with a colonization frequency of 11.11%, followed by Trametes (9.26%) and Penicillium (7.41%). Phylogenetic trees were constructed for the endophytic fungi community in two collection sites, Ha Giang and Lai Chau, revealing the adaptation of the species to the specific tissues and habitats. Cytotoxic activity of endophytic fungal extracts was investigated on cancer cell lines such as SK-LU-1, HL-60, and HepG2, demonstrating strong anti-cancer activity of six isolates belonging to Penicillium, Trametes, Purpureocillium, Aspergillus, and Ganoderma with IC50 value of lower than 10 ㎍/mL. The presence of podophyllotoxin was indicated in Penicillium, Trametes, Aspergillus and for the first time in Purpureocillium and Ganoderma via high-performance liquid chromatography, which implied them as a potential source of this anticancer compound.

한국 토괴중의 사상균에 관한 연구. I. Penicillium속과 자양균 Eupenicillium javanicum에 관하여 (Hyphomycetes from Korean Soil. I. The Genus Penicillium with a Teleomorphic State Eupenicillium javanicum.)

  • 한국사양으로부터 1978년부터 1979년에 걸쳐서 123균주를 분리하였다. 분리된 균주중 Penicillium속 16종이 동정되었고, 이 중에서 한 균주만이 Eupenicillium javanicum인 자양균이었다. 이 균의 특징은 엷은 노란색의 폐자기를 가지며 그 크기는 $120\mu에서\;150\mu$ 정도이며, 그 내부에는 자양을 만든다. 자양포자의 구형이고, 반달모양이며 적도판에 골을 형성하고 있으며 그벽은 거친 모양이다. 그 직경은 $2~5\mu$ 정도이었다. 여기서 보고되는 Penicillium속의 모든 종은 한국에서는 미기록종들이다.

First Detection of Penicillium fellutanum from Stored Rice in Korea

  • A representative isolate KU53 of the predominant Penicillium species was obtained from rice samples from rice processing complexes of National Agricultural Cooperative Federation in Korea. In this study, isolate KU53 was identified by its morphological and molecular characteristics. The macro- and microscopic characteristics of isolate KU53 were compared with the P. fellutanum reference isolate KCTC16913 on different media; isolate KU53 was generally identical to those of the reference isolate KCTC16913. In a molecular-based identification, the ${\beta}$-tubulin and translation elongation factor 1-alpha sequences of isolate KU53 was most closely related to those of P. fellutanum. Thus, isolate KU53 from stored rice could be identified as P. fellutanum, some isolates of which are known to produce mycotoxin-related metabolites. To our knowledge, this is the first detection of P. fellutanum from stored rice in Korea.

Isolation and Characterization of Penicillium crustosum, a Patulin Producing Fungus, from Apples

  • Patulin is a food mycotoxin which induces genotoxicity and acute intestinal disease in infants. Patulin mainly originates from fruit putrefactive moulds, especially in apples, which necessitates the maintenance of strong safety standards against patulin for fresh and processed apples. To investigate the patulin producing moulds in Korean apples, 16 morphological types of fungi were isolated from Korean apples and a patulin producing fungus was identified based on a sequence analysis of the region of internal transcribed spacers (ITS5-5.8S-ITS4 region, 505 base pair) and the 26 rRNA D1/D2 region (527 base pair). Morphological analyses were also performed. The isolated patulin producing fungus was found to a representative species of Penicillium crustosum. The maximal patulin production ability of the isolated fungus (P. crustosum) and the patulin producing standard strain (P. griseofulvum, ATCC 46037) in an SY broth medium were 0.32 and 2.46 mg/L, respectively.

Detection of Mold by Enzyme-Linked Immunosorbent Assay

  • To develop an enzyme-linked immunosorbent assay (ELISA) for detecting mold, we produced anti-mold polyclonal antibodies by immunizing extracellular polysaccharide (EPS) of Aspergillus flavus or Penicillium citrinum in rabbits subcutaneously. Using the purified antibody (Ab) and Ab-HRP conjugate, a sandwich ELISA for EPS was established. The standard curve of the ELISA showed the detection limit for P citrinum EPS to be $0.003{\;}\mu\textrm{g}/ml$. The cross-reactivities of the anti-P citrinum EPS Ab toward components of P citrinum such as EPS, liquid, and solid culture mycelium were 100, 10.5, and 0.58%, respectively, and those toward components of A. flavus such as EPS, liquid and solid culture mycelium, and spore were 300, 0.67, 0.29, and 0%, respectively. When the reactivities toward culture broths of 59 mold strains were tested by the sandwich ELISA, most of the Aspergillus (16 of 18) and Penicillium (14 of 16) strains along with one of the two Cladosporium strains gave positive signals in the culture broths even when diluted 1,000 fold, while the rest of species such as Fusarium, Absidia, Alternaria, and Candida gave negative signals. When the water extracts of 30 corn samples were analyzed by the sandwich ELISA, the EPS in the com could be detected in the concentration range of $0.1-1.6{\;}\mu\textrm{g}/g$.

Five New Records of the Family Aspergillaceae in Korea, Aspergillus europaeus, A. pragensis, A. tennesseensis, Penicillium fluviserpens, and P. scabrosum

  • During an investigation of the fungi from the Aspergillaceae family obtained from different environmental sources in Korea, we isolated six strains, including CNUFC WJC9-1, CNUFC BPM36-33, CNUFC MSW6, CNUFC ESW1, CNUFC TM6-2, and CNUFC WD17-1. The morphology and phylogeny of these isolates were analyzed based on their partial b-tubulin (BenA) and calmodulin (CaM) gene sequences. Based on the morphological characteristics and sequence analyses, the isolates CNUFC WJC9-1, CNUFC BPM36-33, CNUFC TM6-2, and CNUFC WD17-1 were identified as A. europaeus, A. pragensis, Penicillium fluviserpens, and P. scabrosum, respectively, and isolates CNUFC MSW6 and CNUFC ESW1 were identified as A. tennesseensis. To the best of our knowledge, the species A. europaeus, A. pragensis, A. tennesseensis, P. fluviserpens, and P. scabrosum have not been previously reported in Korea.

Promotion of Tricholoma matsutake mycelium growth by Penicillium citreonigrum

  • Tricholoma matsutake has been the most valuable ectomycorrhizal fungi in Asia because of its unique flavor and taste. However, due to the difficulty of artificial cultivation, the cultivation of T. matsutake has relied on natural growth in forests. To cultivate the T. matsutake artificially, microorganisms in fairy rings were introduced. In this study, we isolated 30 fungal species of microfungi from the soil of fairy rings. Among them, one single fungal strain showed a promoting effect on the growth of T. matsutake. The growth effect was confirmed by measuring the growth area of T. matsutake and enzyme activities including a-amylase, cellulase, and b-glucosidase. In comparison with control, microfungal metabolite increased the growth area of T. matsutake by 213% and the enzyme activity of T. matsutake by 110-200%. The isolated fungal strain was identified as Penicillium citreonigrum by BLAST on the NCBI database. The Discovery of this microfungal strain is expected to contribute to artificial cultivation of T. matsutake.
