Genetic Diversity of Penicillium isolates Isolated from Pears with Postharvest Decay in Storage (수확 후 저장 배에서 분리한 Penicillium속 균의 유전적 다양성)

  • This study was carried out to identify the genetic diversity of Penicillium isolates that were isolated from pears with postharvest decay in storage. URP-PCR was used to detect DNA diversity of 84 Penicillium isolates. Based on URP-PCR profiles, 18 Penicillium isolates were selected and their PCR polymorphic bands were produced by additional primers URP1F, URP2R, URP2F, and URP4R. UPGMA cluster analysis using the polymorphic bands showed four clustered groups and futhermore cultural and morphological features characterized the 18 Penicillium isolates. Group 1 was dominant, which occupies 70% in the four clustered groups and identified as P. expansum based on ITS sequence and morphological features.

  • This paper was concerned with the investigation on the species of fungi detected in indoor environments. Mycological survey on the wall of buildings and houses in Seoul was performed from December 26, 1983 to February 11, 1984. The collected fungi with the sterilized cotton rod were caltivated and isolated in the Potato-Detrose Agar medium with chloramphenicol. The fungi detected in hospital were Cladosporium sp., Penicillium sp., Alternaria sp., Chaetomium sp. and others. The fungi detected in household were Penicillium sp., Alternaria sp., Aspergillus sp., Cladosporium sp., Rhizopus sp., Trichoderma sp. and others. The kitchen of household was more contaminated by fungi than the living room and others. In hospital, it was much contaminated by the same species of the aboves on the wall of lobby and refectory. As the above results were recognized, the higher humidity and the more poor ventilation, the more species and amount of fungi were contaminated. Further studies on the identification of pathogenic fungi should be continued. The proper regulation for the hygienic maintenance of indoor environment in household, hospital and other public buildings should be recommended as well.

  • Nine hundred and eighty fungal strains were isolated from lesions of Trifolium repens L. and various weeds. Among them, F40362, F40496, F40497 strains were selected by the screening of herbicidal activity on Trifolium repens L. and Zoysia japonica. Selected three strains showed selective activity between Trifolium repens L. and Zoysia japonica, and were identified as Penicillium sp.

  • A strain of Penicillium sp. which produces considerable amount of $\beta$-galactosidase was selected from extracellular $\beta$-galactosidase-producing fungi isolated from soil. The enzyme was found to be very stable in neutral pH range. Maximum enzymatic activity was reached after 72 hr of incubation in a wheat bran medium at 3$0^{\circ}C$. Productivity of the enzyme appeared not to be affected by the addition of carbon sources to the medium but the activity of the enzyme was increased from 14% to 85% by the addition of various nitrogen sources. The enzyme extracted from the wheat-bran culture of the Penicillium sp. was purified to 5050-fold by ammonium sulfate fractionation, SP-Sephadex C-50 chromatography, Ultrogel AcA 44 filtration and hydroxyapatite chromatography. The purified $\beta$-galactosidase was homogeneous on ultracentrifugation and disc electrophoresis.

  • 1. Of the 35 various grains, 446 colonies of fungi were isolated. Among the 446 colonies, 423 were possible to identify into 17 genera. 2. Of the 10 bean-pastes, fungi were isolated 193 colonies. It can be identified 11 genera. 3. It will be noted that Aspergillus sp., Penicillium sp., Mucor sp., Cladosporium sp. were predominated genera. 4. A total of 155 colonies of Aspergillus sp. isolated from various grains were obtained from the samples. Asp. versicolor was the frequent species and 47 colonies isolated. 5. Penicillium where was isolated from various grains, obtained 115 colonies. Pen. camemberti was the most frequent species. 6. Penicillium palitans was most excellent amylase producing fungi and which was 1ml per 6.300 unit.

Solid Culture of Phosphate-solubilizing Fungus, Penicillium sp. PS-113 (인산가용화 사상균 Penicillium sp. PS-113의 고체배양)

  • A fungus, Penicillium sp. PS-113, isolated from soil showed the high phosphate-solubilizing activity in patato dextrose broth-rock phosphate to produce free phosphates to the culture broth with the concentrations of 585 ppm against rock phosphate. In this medium, the optimum temperature and initial pH to solubilize rock phosphate were 30$^{\circ}C$ and pH 7.5, respectively. In order to make the mass production of the conidia from this fungus, we cultured in on various solid-based media like barley, corn, wheat, rice, rice bran, and compost. As a result, the fungus highly produced conidia ranging from 2.1 to $5.1{\times}10_9$ conidia/g${\cdot}$media on these solid media except compost-based medium, which was 0 times less than others. Effects of inoculation of the phosphate solubilizing fungus as a biofertilizer were studied in perlite-based pot cropped with Zea mays Suwon 19. Inoculation of Penicillium sp. PS-113 increased in plant height (1.4 times), plant weight (5.2~8.1 times) and root length (1.1~1.2 times) at 60-day cultivation, compared to Hogland solution either without $NH_4H_2PO_4$ or displace $NH_4H_2PO_4$ to powdered rock phosphate, a phosphorus source for plant growth.

  • Microorganisms growing on Dried-Persimmon have heed isolated ana identified. Fluorescent substance were extracted from a putrefactive Dried-Persimmon after invaded Toxigenic Fungi, and compared with Aflatoxin by Thin layer chromatography and u. v. absorption spectra. The results obtained were summarized as follows: 1) Fungal invasion was frequently appeared at the beginning of storage, and after then Bacteria invasion was followed. 2) Several Genera of microorganisms (Aspergillus sp., Escherichia sp., Mucor sp., Alternaria sp., Penicillium sp. ) were observed in Dried-persimmon during storage. Aspergillus sp., one of all Genus was predominant. 3) Two strains (Aspergillus flavus Group, Penicillium citrimum Series) of 6 Fungi had Fluorescent substance, which was presumed Aflatoxin-like substance. 4) The Rf value of T. L. C. ana λ max of u. v. absorption spectra showed the same value as the standard of Aflatoxin. It is suppose that the Fluorescent substance in Dried-Persimmon is a Aflatoxin-like substance.

Isolation and Characterization of Xylanase from a Novel Strain, Penicillium menonorum SP10

  • Xylanase has been applied in various sectors, such as biomass conversion, paper, pulp, textiles, and pharmaceutical industries. This study aimed to isolate and screen potential xylanase-producing fungi from the soil of Suphan Buri Province, Thailand. Fifteen fungi were isolated, and their xylanase activities were tested by the qualitative method. The result showed that isolate SP3, SP10 and SP15 gave high xylanase activity with potency index (PI) of 2.32, 2.01 and 1.82, respectively. These fungi were selected for the xylanase quantitative test, isolate SP10 performed the highest xylanase activity with 0.535 U/mL. Through molecular methods using the 𝛽-tubulin gene, isolate SP10 was identified as Penicillium menonorum. The xylanase characteristics from P. menonorum SP10 were determined, including the xylanase isoforms and the optimum pH and temperature. The xylanase isoforms on SDS-PAGE indicated that P. menonorum SP10 produced two xylanases (45 and 54 kDa). Moreover, its xylanase worked optimally at pH 6 and 55 ℃ while reaching 61% activity at 65 ℃. These results proposed P. menonorum SP10 as a good candidate for industrial uses, especially in poultry feed and pulp industries, to improve yield and economic efficiency under slightly acidic and high-temperature conditions.

  • Farnesylhydroquinone (1) has been isolated from the mycelium of a marine-derived fungus of the genus Penicillium. The structure of the compound (1) has been elucidated by spectral method. The compound 1 exhibits potent radical scavenging activity ($IC_{50}{\;}12.5{\;}{\mu}M$) against the DPPH.

Isolation of Erythritol Producing Microorganisms from Nature (자연계로부터 Erythritol 생산 균주의 분리)

  • For the purpose of obtaining microorganisms producing high amount of erythritol, the screening test was carried out. Productivity of erythritol was analyzed by paper chromatography and HPLC' methods. Among more than two hundred isolates, one strain(KJX1) was selected as an erythritol prtducer from thc soil of corn shock. The isolated strain was identified as Pmicilliurn sp. KJ81 from the morphological and physiological characteristics. Penicillium sp. KJ81 showed white to green colony color, two- to three-stage branching conidiophcvc and flask-shaped phialides.
