Stock Characterization of the Fleshy Prawn (Penaeus chinensis) in the Yellow Sea by Intraspecific Sequence Variation of the Cytochrome c Oxidase Subunit I Gene

  • To determine the amount of genetic variation among populations of Penaeus chinensis (Osbeck) in the Yellow Sea, 342 bp region of the mitochondrial cytochrome c oxidase subunit I gene was amplified and sequenced. Six haplotypes, which differ by from one to four nucleotide sustitutions, were detected from 34 individuals of 4 populations examined. Mean sequence divergence between pairs of haplotypes was $0.68\%$. Most individuals from 4 populations were shared by the most common genotype. This genotype was distributed evenly in the Korean and Chinese populations. This result is in accordance with findings observed using RFLPs analysis of mtDNA (Hwang et al., 1997). Therefore, it is suggested that P. chinensis should be treated as one unit stock in the Yellow Sea.

Production of Healthy Shrimp(Penaeus chinensis, Penaeus japonicus) Seedling Free from White Spot Baculovirus(WSBV) (White Spot Baculovirus(WSBV) 미감염 새우(Penaeus chinensis, Penaeus japonicus) 종묘 생산)

  • For the production of healthy shrimp(Penaeus chinensis, Penaeus japonicus) seedlings free from the white spot baculovirus(WSBV), we tried to disinfect shrimp eggs with iodine. A relative hatching rate of over 50% has resulted from the treatments of fertilized eggs of fresh shrimps(P. chinensis) with an effective povidon-iodine concentration of 20-200 ppm for 30 seconds and 60 seconds, respectively. The 60 sec treatment group with a 20 ppm effective povidon-iodine concentration showed nearly a 50% relative hatching rate. The over 50 ppm treatment group, however, did not result in hatching show a relative hatching rate. These hatching rates were similar to those in the fertilized eggs of kuruma prawns treated with povidon-iodine solutions. In case of washing fertilized eggs with UV-irradiated seawater 33.3% relative hatching rate in fresh shrimps was much lower than 55.2% in kuruma shrimps. Treatment fertilized eggs of 2 species of shrimp with 200 ppm iodine for 30 sec did not affect the survival rate of the larvae until the early stage of the post larvae even though the hatching rates were low. In order to produce healthy shrimp seedlings non-infected by WSBV, the shrimp eggs should be washed with UV-irradated seawater and sterilized with a low povidon-iodine concentration for a short time.

  • The virions of causative virus for white spot syndrome in cultured Fresh shrimp, Penaeus chinensis were rod-shaped, double envelope. An average size of the virion was 70 nm in diameter and $250\~300$ nm in length. Histopathological test of affected stomach, heart, and lymphoid organ revealed nuclear hypertrophy. Infectivity trials carried out by injection and feeding with purified virus revealed high cumulative mortality to healthy shrimp. The twenty one different protein species were detected in the analysis of virion. The length of total DNA from the purified virus particles were detected as a single band, double-stranded DNA molecule of approximately 114 kb.

  • Infection rates with white spot baculovirus(WSBV) in wild-caught shrimp spawners appeared to be 52% in Penaeus chinensis and 20% in Penaeus japonicus when diagnosed by polymerase chain reaction (PCR). Infection rates of WSBV from inhabitant organisms in shrimp farm showed 37% in Upogebia major and 29% in Calliamassa japonica. The viruses maintained their survivorship in seawater at $4^{\circ}C$, but lost it at $25^{\circ}C$.

  • The gill morphology and ultrastructure of the fleshy shrimp, Penaeus chinensis were investigated by light and electron microscopy. Fleshy shrimp has dendrobranchiate gills. Gill has a longitudinal septum dividing them into afferent and efferent channel. Each gill lamella is covered by multi-layered thin cuticle of different electron density. The lamella basal cell is squamous and contains cytoplasm of electron dense. Simple epithelial layer consists of squamous epithelium contained large nucleus. The lamella pillar structures are characterized by the axial microtubules and lateral membrane interdigitations Secretory cells of AB-PAS negative are multicellular gland. In active gland each cell boundary is not apparent and the cytoplasm contains smooth endoplasmic reticula, mitochondria, membrane-bounded secretory vesicles of low electron density and granular resettes. In inactive gland each cell boundary is apparent and the cytoplasm is occupied with numerous small granules of electron dense. The well-developed rough endoplasmic reticula and Golgi apparatus are observed in the unicellular gland of alcian blue positive.

  • Because of the great concern over the possibility of contamination from the rod-shaped nuclear virus (PRDV) from Japan and white spot virus (WSSV) from Taiwan, most eggs used in Korean shrimp farms are currently obtained from local broodstock. In addition, the screening of imported broodstock for any viral presence at the National Fisheries Research and Development Institute is also mandatory. Nonetheless, massive mortality from white spot syndrome continues in Korea. In the present study, we present an improved PCR method to use tissue-extracted DNA instead of viral DNA extracted from a purified virus based on a sucrose density gradient, and produced results within 8 h. In 1998, this modified PCR method was able to detect that diseased Penaeus japonicus were infected within 8 h. In 1998, this modified PCR method was able to detect that diseased Penaeus japonicus were infected only with PRDV, while Fenneropenaeus chinensis were infected with both PRDV and WSSV. In 1999, PRDV and WSSV were detected in F. chinensis with signs of infection, but not with WSSV alone.

Genetic Distances between Two Cultured Penaeid Shrimp (Penaeus chinensis) Populations Determined by PCR Analysis

  • Genomic DNA samples were obtained from cultured penaeid shrimp (Penaeus chinensis) individuals such as fresh shrimp population (FSP) and deceased shrimp population (DSP) from Shinan regions in the Korean peninsula. In this study, 233 loci were identified in the FSP shrimp population and 162 in the DSP shrimp population: 33 specific loci (14.2%) in the FSP shrimp population and 42 (25.9%) in the DSP population. A total of 66 (an average of 9.4 per primer) were observed in DSP shrimp population, whereas 55 unique loci to each population (an average of 7.9 per primer) in the FSP shrimp population. The Hierarchical dendrogram extended by the seven oligonucleotides primers indicates three genetic clusters: cluster 1 (FRESH 01, 02, and DECEASED 12, 13, 15, 16, 17, 19, 20, 22) and cluster 2 (FRESH 03, 04, 05, 06, 07, 08, 09, 10, 11, and DECEASED 14, 18, 21). Among the twenty-two shrimp, the shortest genetic distance that exposed significant molecular differences was between individuals 20 and 16 from the DSP shrimp population (genetic distance=0.071), while the longest genetic distance among the twenty-two individuals that established significant molecular differences was between individuals FRESH no. 02 and FRESH no. 04 (genetic distance=0.477). In due course, PCR analysis has revealed the significant genetic distance among two penaeid shrimp populations.

  • White spot syndrome virus (WSSV) causes the most serious epizootic in cultured penaeid shrimp. The epizootic started in 1992, and spread through east and south east Asia and into other shrimp growing countries of the region. WSSV has also been reported from Penaeus chinensis cultured in Korean peninsula (Park et al., 1998; Kim et al., 1999). However, this paper deals with the WSSV infection in cultured and wild P. chinensis, purification and characterization of the pathogen, and the occurrence of other concomitant infection. (omitted)

  • The mitochondrial DNA (mtDNA) restriction fragment length polymorphi는 (RFLPs) of five populations were analyzed to delineate the stocks of Penaeus chinensis (Osbeck) in the Yellow Sea. Comparison of P. chinensis with P. japonicus to clarify the nucleotide divergence between two species was also carried out. Based on the fragment patterns, three composite haplotypes were analyzed in P. chinensis mtDNA as four naplotypes were in P. japonicus. Most individuals of each P. chinensis population are shared by one dominant haplotype. Another two haplotypes haying variations at the C/a I and hull sites were also distributed evenly in the Korean and Chinese populations. It is suggested that the gene exchange occurring between populations in the Yellow Sea is frequent. Average length of the mtDNA molecule was estimated to be about 16.44 kb in P. chinensis and 16.31 kb in P. japonicus, Sequence divergence (p) of mtDNA between two species estimated by using Upholt's (1977) fomula was $13.7\%$.

  • Penaeid shrimp (Penaeus chinensis), economically important aquacultural species, belonging to family Penaeidae including genus Fenneropenaeus, widely inhabit the West Sea and South Sea in the Korean Peninsula and the Gulf of Pohai in China under the natural ecosystem. Consequent of the rapid increase in seed production, there is a need to understand the genetic traits and composition of this fish species in order to evaluate exactly the patent genetic effects induced by seed production operations.
