Impairments of Learning and Memory Following Intracerebroventricular Administration of AF64A in Rats

  • Three types of learning and memory tests (Morris water maze, active and passive avoidance) were performed in rats following intracerebroventricular infusion of ethylcholine aziridium (AF64A). In Morris water maze, AF64A-treated rats showed the delayed latencies to find the platform iron 6th day after the infusion. In pretrained rats, AF64A caused the significant delay of latency at 7th days but not 8th day. In the active avoidance for the pretrained rats, the escape latency was significantly delayed in AF64A-treatment. The percentages of avoidance in AF64A-treated rats were less increased than those in the control. Especially, the percentage of no response in the AF64A-treated rats was markedly increased in the first half trials. In the passive avoidance, AF64A-treated rats shortened the latency 1.5 h after the electronic shock, but not 24 h. AF64A also caused the pretrained rats to shorten the latency 7th day after the infusion, but not 8th day. These results indicate that AF64A might impair the learning and memory. However, these results indicate that the disturbed memory by AF64A might rapidly recover after the first retrain. Furthermore, these results suggest that AF64A may be a useful agent for the animal model of learning for Spatial cognition .

The effects of regular exercise on capsaicin-induced pulpal pain and pain-induced changes in passive avoidance learning and memory in rats

  • Background: Pulpal pain is one of the most common and severe orofacial pain conditions with considerable adverse effects on physiological processes including learning and memory. Regular exercise is known to be effective on cognitive function as well as pain processing in the central nervous system. Here, the possible effects of regular exercise on pulpal pain response as well as pain-induced changes in learning and memory efficiency in rats were investigated. Methods: Twenty-four male Wistar rats were randomly assigned to the control, capsaicin, exercise, and exercise plus capsaicin groups. Rats in exercise groups were forced to run on a treadmill with a moderate exercise protocol for 4 weeks. Capsaicin was used to induce dental pulp pain. Passive avoidance learning and memory performance was assessed by using a shuttle box apparatus. Results: According to the results, regular exercise could decrease the time course of capsaicin-induced pulpal pain (P < 0.001). Moreover, in capsaicin-treated rats, passive avoidance acquisition was impaired as compared to the control (P < 0.05) and exercise (P < 0.001) groups. Additionally, regular exercise before capsaicin injection could attenuate capsaicin-induced memory impairments (P < 0.05). Conclusions: Taken together, the present data showed that regular exercise has inhibitory effects on capsaicin-induced pulpal pain as well as pain-induced cognitive dysfunction in rats.

기억력 감퇴모델에서 산사의 기억력 개선 효과에 관한 연구 (The Fruits of Crataegus pinnatifida Bunge ameliorates Learning and Memory Impairments Induced by Scopolamine)

  • Objectives : In the present study, we assessed the effects of the ethanolic extract of Crataegus pinnatifida Bunge on the learning and memory impairments induced by scopolamine using the passive avoidance and the Morris water maze tasks in mice. Methods : The cognition-enhancing effect of C. pinnatifida was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The ethanolic extract of C. pinnatifida (100, and 200 mg/kg) significantly reversed the scopolamine-induced cognitive impairments in the passive avoidance test (p < 0.05). Moreover, C. pinnatifida (200 mg/kg) also improved escape latencies in training trials and increased swimming times and distances within the target zone of the Morris water maze (p < 0.05). On the Y-maze test, C. pinnatifida (100, and 200 mg/kg) also significantly reversed scopolamine- induced cognitive impairments in mice (p < 0.05). Conclusions : The ethanolic extract of Crataegus pinnatifida dramatically possesses the anti-amnestic and cognitive-enhancing activities related to the memory processes, and these activities were parallel to treatment duration and dependent on the learning models.

Effect of Panax ginseng Extract on Passive Avoidance Retention in Old Rats

  • Female rats of two groups (6 and 27 months) were tested in the passive avoidance test to investigate the age-dependency of the learning ability. The results showed a significantly better avoidance behavior in the young adult animals compared to the older ones. The influence of a 13-day treatment with Panax ginseng (30 mg/kg/d, oral) on 27 month old rats caused a considerably prolonging of the latency time in comparison to the untreated control group of the same age. In the open field the treated rats exhibited neither an altered locomotion nor exploration nor a changed emotional reactivity which could explain the improved avoidance reaction.

Effects of Chongmyung-tang on Learning and Memory Performances in Mice

  • Chongmyung-tang(CMT, 聰明湯), oriental herbal medicine which consists of Polygaglae Radix(遠志), Acori Graminei Rhizoma(石菖蒲) and Hoelen(白茯神) has effect on amnesia, dementia. In order to evaluate effect of CMT on memory and learning in mice, CMT extract was used for studies. This paper describes the effects of CMT extract on memory and learning processes by using the passive and active avoidance performance tests, novel object recognition task and water maze task. The CMT extract ameliorated the memory retrieval deficit induced by ethanol in the passive avoidance responses but did not affect ambulatory activity of normal mice. These results suggest that CMT has an ameliorating effect on memory retrieval impairment. CMT extract decreased spontaneous motor activity(SMA) in the latter sessions of memory registration in active avoidance responses. These results suggest that CMT has partly transquilizing or antianxiety effects. In novel object recognition task to measure visual recognition memory, CMT-administered mice enhanced in long term memory for 1-3 days. In water maze task to measure spatial learning, which requires the activation of NMDA receptors in the hippocampus, spatial learning in CMT-administered mice was faster than in wild-type mice. These results suggest that CMT enhances memory and activates NMDA receptors.

  • The present study examined the effects of carbon monoxide (CO) intoxication and aging on learning and memory deficit in young($5{\sim}8$ weeks) and aged($52{\sim}66$ weeks) mice, using the step down and step through passive avoidance failure techniques. We also investigated the effects of ginseng saponins on memory deficit. Significant decrease in memory registration, retention and retrieval function in young mice and decrease in memory registration and retention function in aged mice were observed. Normal young mice were apt to perform to a great degree of passive avoidance response than normal aged mice, but there was no difference between both groups by CO exposure. Administration of ginseng saponins showed an improvement on passive avoidance failure induced by CO exposure.

  • This study was performed to examine the effect of red ginseng total saponin and extract on memory in mice using one trial step-down type passive avoidance method. Red ginseng total saponins (No. 1: PD/PT ratio=1.24, No. 2: PD/PT ratio=1.47) were prepared with the different mixing ratio by using the parts of red ginseng. In single administration of total saponin No. 1 (100 mg/ kg, bw) or No. 2 (50 mg/kg, bw) increased the latency time as compared with control group but was not statistically significant. Treatment of total saponin No. 1 (50 mg/kg, bw) for 10 days produced an increase of latency time but was not statistically significant. In scopolamine-induced memory deficient mice total saponin No. 1 (50 mg/kg, bw) and No. 2 (100 mg/kg, bw) significantly improved the latency time. These results show that red ginseng total saponin may improve the memory of sco-polamine-induced memory deficient mice and have nootropic activity.

Activation of Adenosine A2A Receptor Impairs Memory Acquisition but not Consolidation or Retrieval Phases

  • Several lines of evidence indicate that adenosine $A_{2A}$ agonist disrupts spatial working memory. However, it is unclear which stages of learning and memory are affected by the stimulation of adenosine $A_{2A}$ receptor. To clarify these points, we employed CV-1808 as adenosine $A_{2A}$ agonist and investigated its effects on acquisition, consolidation, and retrieval phases of learning and memory using passive avoidance and the Morris water maze tasks. During the acquisition phase, CV-1808 (2-phenylaminoadenosine, 1 and 2 mg/kg, i.p.) decreased the latency time in passive avoidance task and the mean savings in the Morris water maze task, respectively. During the consolidation and retrieval phase tests, CV-1808 did not exhibited any effects on latency time in passive avoidance task and the mean savings in the Morris water maze task. These results suggest that CV-1808 as an adenosine $A_{2A}$ agonist impairs memory acquisition but not consolidation or retrieval.

육미지황탕(六味地黃湯)이 국소뇌허혈유발 기억장애(記憶障碍) 모델 흰쥐에 미치는 영향 (Effect of Yukmijihwangtang on Learning and Memory Impairment in Transient Focal Cerebral Ischemia Rat Model)

  • Objectives: This study investigated the effect of Yukmijihwangtang on cerebral ischemia-induced learning and memory impairment by middle cerebral artery (MCA) occlusion in rats. Methods: The ability of learning and memory of rats was measured using the eight-arm radial maze and the passive avoidance test, and profile of cholinergic neuron was assessed in the medial septum and hippocampus region by immuno-histochemistry. Results: 1. No differences were found between groups in the number of correct choices in acquisition performance during the eight-arm radial maze task. 2. No differences were found between groups on day 1 in the error rate in acquisition performance, which is defined as the number of enters into the same arm more than once within five minutes. After 5 to 6 days of test, the number of errors was significantly reduced in the Yukmijihwangtang group (forebrain ischemia group with Yukmijihwangtang treatment), compared with the ischemia group. 3. The memory processes significantly improved in the Yukmijihwangtang group according to results of the passive avoidance test. 4. The appearance of AchE (acetylcholinesterase) in the CA1 region of hippocampus significantly decreased in the ischemia group, compared with the sham group (untreated group). The appearance of AchE in the same region significantly increased in the Yukmijihwangtang group, compared with the ischemia group. 5. The appearance of ChAT (choline acetyltransferase) in the CA1 region of the hippocampus and medial septum decreased in the ischemia group, compared with the sham group. The appearance of ChAT in the same region significantly increased in the Yukmijihwangtang group, compared with the ischemia group Conclusions: This study provides evidence that Yukmijihwangtang is effective for reviving the ability of learning and memory and damaged neurons in rats with experimental cerebral ischemia.

선택적 세로토닌 재흡수차단제들이 만성 경도 스트레스 후의 백서에서 수동적 회피학습에 미치는 영향 (Effects of Selective Serotonin Reuptake Inhibitors on the Retention of Passive Avoidance Learning after Chronic Mild Stress in Rats)

  • The study was designed to evaluate the significant roles of SSRI in rat of depression model. Chronic exposure to mild unpredictable stress has been found to depress the consumption of sweet 1% sucrose solutions in the Sprague-Dawley rats. We applied the variety of 11 types of stress regimens and identified depressive behaviours(developed by Willner) in 70 Sprague-Dawley rats. Rats in experiments were stratified into 6 groups, ie ; 3 kinds of SSRI(paroxetine, fluoxetine, sertraline), clomipramine, choline and saline control. Memory function was evaluated by passive avoidance learning and retention test. The authors determined how long memory retention would remain improved with 24 hour, 1 week, 2 weeks, 3 weeks, and 4 weeks at training-testing interval in depressive states of the Sprague-Dawley rats. The results were as follows ; 1) There were no significant differences between the 6 groups at the 24 hour training-testing interval. 2) The paroxetine treated group showed significant differences from the control group at the 1 week and 2 weeks training-testing interval. 3) The paroxetine and the fluoxetine treated groups showed singificant differences from the control group at 3 week training-testing interval. 4) The paroxetine and the choline treated groups showed significant differences from the control group at 4 week training-testing interval. In summary, paroxetine had an effect on long term memory processing from 1st week to 4th week. Also, fluoxetine(at 3rd week) and choline(at 4th week) had effect on long term memory processing. Sertraline, clomipramine were ineffective on memory processing during 4 weeks observation. Possible explanations why paroxetine had early effect on memory processing than the other selective serotonin reuptake inhibitors are rapid bioavailability, which is the characteristics of pharmacokinetics of paroxetine. In clinical situation, author carefully suggest that SSRI would be beneficial to improve the memory function caused by depressive neurochemical changes.
