Effects of Vesicular-Arbuscular Mycorrhizae on The Growth of American Ginseng (V. A. Mycorrhizae가 미국삼의 생육에 미치는 영향)

  • Newly sprouted American ginseng (Panax quinquefolium L.) seedlings were transplanted to forest pots with mycorrhizae-infested soil and grown in screenhouse for 2 years. Growth patterns, mortality rate and fresh root weight were investigated. Plants in VAM soil had lower mortality rales than control. In soils infested with two species of mycorrhizal fungi (Glomu deseyicola, frappe, Bloss and Merge and G. intraradices, Schenck and Smith), 28-35% of plants produced 3-prongs in the second season and significantly increased fresh root weight by 41 to 43%.

Diversity of Fungal Endophytes in Various Tissues of Panax ginseng Meyer Cultivated in Korea

  • Endophytic fungi were isolated from various tissues (root, stem, petiole, leaf, and flower stalk) of 3- and 4-year-old ginseng plants (Panax ginseng Meyer) cultivated in Korea. The isolated endophytic fungi were identified based on the sequence analysis of the internal transcribed spacer (ITS), 1-5.8-ITS 2. A morphological characterization was also conducted using microscopic observations. According to the identification, 127 fungal isolates were assigned to 27 taxa. The genera of Phoma, Alternaria and Colletotrichum were the most frequent isolates, followed by Fusarium, Entrophospora and Xylaria. Although 19 of the 27 taxa were identified at the species level, the remainder were classified at the genus level (6 isolates), phylum level (Ascomycota, 1 isolate), and unknown fungal species (1 isolate). Endophytic fungi of 13 and 19 species were isolated from 3- and 4-year-old ginseng plants, respectively, and Phoma radicina and Fusarium solani were the most frequently isolated species colonizing the tissues of the 3- and 4-year-old ginseng plants, respectively. The colonization frequency (CF%) was dependant on the age and tissue examined: the CFs of the roots and stems in the 3-year-old ginseng were higher than the CF of tissues in the 4-year-old plants. In contrast, higher CFs were observed in the leaves and petioles of 4-year-old plants, and endophytic fungi in the flower stalks were only detected in the 4-year-old plants. In conclusion, we detected diverse endophytic fungi in ginseng plants, which were distributed differently depending on the age and tissue examined.

  • Previously, we found the operon random primer (OP-5A) that is characteristic the genus Panax by randomly amplified polymorphic DNA (RAPD) analysis. However, OP-5A primer is limited to apply on the differentiation of only crude herbal plants. To construct more sensitive and unique primers on the genus Panax, ginseng-specific DNA profile (350 bp) that was amplified by OP-5A primer were inserted in a plasmid vector in the TA cloning method and sequenced. We designed the PCR primers (Forward: 5"-AGGGGTCTTGCTAT AGCGGAAC-3", Reverse: 5"-AGTCTTAATTTCATATTTTCGTATG-3") and identified the unique ginseng band (350 bp) in commercial granule products including ginseng extracts as well as crude ginseng plants by nascent PCR.(omitted)

  • This study was conducted to investigate the metallothionein (MT) induction by Panax ginseng C. A. Meyer in cadmium chloride intoxication. The results were as follows: Generally, detoxicatlon effects by Panax ginseng C. A. Meyer extract increased proportionally to the increase of cadmium concentrations. When a 8 mg/g dosage of cadmium was administered, formation of the cadmium and EDTA complex showed the highest antitoxic effect. Also, when a 4 mg/g dosage of cadmium was administered, Panax ginseng C. A. Meyer extract showed the highest antitoxic effect in metallothionein induction. According to the above results, Panax ginseng C. A. Meyer extract administered with cadmium increased metallothionein concentration and decreased the toxicity of cadmium in liver.

  • The composition and concentration of ginsenosides and the free sugars in panax ginseng(Korea ginseng), panax quinquefolium (American ginseng) and panax pseudoginseng var. notoginseng (Sanchi ginseng), were investigated. The major ginsenosides and the order of their amount in panax ginseng are Rbl, Rc Rgl, Re, Rb2 Rd and these are about 90% of total ginsenosides, but major ginsenosides of American and Snachi ginseng art Rbl, Re, Rg1 (about 91% of total) ansi Rgl, Rbl, Re (about 93% of total) respectively. Sanchi ginseng was observed in higher concentration of panaxatriol than panaxadiol unlike panax and American ginseng. Free sugars in white ginseng are fructose, glucose, maltose and sucrose. Whereas, in red ginseng rhamnose and xylose were also detected as free sugar.

  • This study was conducted to know the elects of light intensity of polyethylene net shading on the growth status, photosynthesis and root yield of ginseng plants. Polyethylene net shading of loft transmittance was the best one among light intensities of polyethylene net used in view of photosynthesis and decreasing of early leaf defoliation. According to increase of light intensity under the shading chlorophyll contents of ginseng leaves were decreased. As it was increased over 2 mg/g Photosynthesis and total saponin of leaves showed on the decrease remarkably. The rate of alternaria blight of ginseng plants showed the positive correlation between light intensity and leaking rate. The shading of 10% transmittance in root yield was increased by 40% in 6-year-old ginseng plant as compared with common straw shading, due to decreased missing plant and increased root weight.

  • Carlaviruses were isolated from naturally infected medicinal plants, and identified by means of test plants and electron microscopy. Mottle symptoms were shown on leaves of Panax ginseng, Aralia cordata, Xanthium strumarium, Taraxacum officinale, Aconitum carmichaeli, and Bupleurum longiradiatum var. breviradiatum. Ring spot on leaves of Abutilon avicennae and ring mosaic or vein clearing on leaves of Sambucus sieboldiana were also shown. These viruses had rather narrow host ranges by mechanical inoculation. The virus particles were scattered or aggregated in cytoplasm of infected host plant leaves. The carlaviruses for which the name panax virus S (PaVS), aralia virus S (ArVS), xanthium mottle virus (XaVS), taraxacum virus S (TaVS), aconite mottle virus (AcMV), bupleurum virus S (BuVS) and abutilon ring spot virus (AbRSV) were proposed, had flexuous particles with width 13 nm and length $620{\sim}720nm$. A reported elder ring mosaic virus was isolated from leaves of Sambucus sieboldiana with ring mosaic or vein-clear symptoms.

  • The inheritance of violet and green stems was examined in F1 F2 of violetstem variant x yellow-berry variant in Panax ginseng C. A. Meyer, and the characters of F1 and F2 plants were investigated. From these results, it was shown that most of the characters of F1 and F2 plants were simillar to the female plants. However, reciprocal crosses between violet and green stems yielded progeny of violet-stem variant. Thus the cross V♀x♂Y gave all violet-stem seedlings, and the cross Y♂ x Y♀ gave all violet-stem seedlings. And all of the crosses segregated in F2 in a ratio of 3 violet to 1 green.
