감마선 조사시 우지 산패억제를 위한 항산화제의 첨가효과 (Effects of Antioxidants on Oxidation of Tallow by Gamma Irradiation)

  • 우지에 항산화제로 ascorbyl palmitate (AP), ${\alpha}-tocopherol\;({\alpha}-Toc)$, BHA 및 ascorbyl $palmitate+{\alpha}-tocopherol\;(AP+{\alpha}-Toc)$을 첨가하여 $0{\sim}10\;kGy$의 선량으로 감마선을 조사하였고, 감마선 조사된 시료는 조사직후와 $50^{\circ}C$에서 저장하면서 저장기간에 따른 과산화물 형성 억제능을 측정하였다. 감마선 조사직후의 과산화물가는 감마선 조사선량의 높아짐에 따라 증가하였다. 그러나 우지에 항산화제를 첨가한 경우에는 감마선 조사에 의한 과산화물 형성을 억제하였고, 억제 활성은 $AP>BHA>AP+{\alpha}-Toc>{\alpha}-Toc$로 AP가 초기산화 억제능을 가진 천연항산화제로 나타났다. 저장기간에 따른 과산화물가는 감마선 조사에 의해 자동산화를 상당히 촉진시키지만 항산화제를 첨가한 경우에는 과산화물 형성을 유의적으로 억제하였으며, 이때의 항산화능은 $BHA>AP+{\alpha}-Toc>{\alpha}-Toc>AP$의 순으로 나타났다. 특히 저장말기까지도 효과가 높았던 천연 항산화제 혼합물인 $AP+{\alpha}-Toc$은 BHA와 유의차가 인정되지 않았다(p>0.05).

항산화제 또는 팜유로 보강된 미강유를 이용한 라면의 산화안정성 (Oxidative Stability of Deep-Fried Instant Noodle Prepared with Ricebran Oil Fortified by Adding Antioxidants or by Blending with Palm Oil)

  • 본 실험에서는 라면제조시 미강유를 튀김유로 사용했을 때, 그 산화 안정성에 대한 ${\alpha}-tocopherol$, BHA, TBHQ, ascorbyl palmitate+구연산의 첨가효과와 미강유에 팜유를 가각 30, 50, 70%(w/w) 혼합한 혼합유의 경우 그 혼합효과 등에 대해서 조사하고자 했다. 이상의 항산화제들은 미강유에 0.02%의 수준으로 첨가했으며, 혼합유의 경우에는 전술한대로 혼합하여 실험에 사용했다. 이상의 기름으로 튀긴 시제라면들을 각각 30분식을 무색, 투명의 polypropylene 봉지에 넣어 $35.0{\pm}0.5^{\circ}C$의 항온기내에서 90일간 저장했다. 일정기간마다 각 시료구와 실험대조구에서 라면시료를 채취한 후 그 속의 유지를 추출하여, 추출유지의 과산화물가, 산가, 요오드가, 유전항수, 지방산조성 등을 측정했다. 그리고 이상의 측정치들의 변과를 토대로 추출유지의 산화 안정성과 시제라면의 저장 안정성을 추정했다. 이상의 결과들을 종합해 보면 다음과 같다. 1. 실험대조구와 각 항산화제 첨가구에서 추출한 유지의 과산화물가, 산가, 유전항수, 지방산 조성의 변화 등에서 판단할 때 ${\alpha}-tocopherol$은 항산화효과가 거의 없었으며, 한편 BHA는 약간의 효과가 있었다. 이에 반해서, ascorbyl palmitate와 구연산을 병용한 경우와 TBHQ의 경우에는 상당한 항산화효과를 보였다. 특히, TBHQ를 0.02% 첨가한 미강유를 사용했을 경우의 그 산화 안정성은 팜유를 사용했을 경우와 거의 같았다. 2. 혼합유 시료구들의 경우, 튀김에 사용한 혼합유 중의 팜유의 비율이 커질수록 그 산화 안정성은 증가했다. 즉, 팜유가 30% 혼합된 혼합유로 만든 시제라면의 추출유지의 산화 안정성은 미강유를 단독 사용했을 경우에 비해서 현저히 증가했다. 그리고 팜유가 70% 함유된 혼합유를 사용했을 경우 그 산화 안정성은 함유를 단독 사용했을 경우의 안정성과 거의 같았다.

입국으로 제조한 증류주의 숙성에 따른 품질 특성 (Quality Characteristics of Distilled Liquor Produced Using Ipguk (Koji) During Aging)

  • 연구에서는 입국 증류주를 항아리와 스테인리스 용기에 숙성시키며 숙성 시간별 증류주의 품질 특성을 검토하였다. 알코올의 경우 초기 39.8-40.9% 알코올이 숙성 180일에 항아리는 38.0-38.9%를 스테인리스 용기는 39.1-39.7%를 나타내었다. 항아리와 스테인레스 용기 모두 숙성 기간이 증가할수록 TBA값이 감압과 상압 증류주 모두에서 증가하였다. 증류주의 n-propanol, isobutanol, isoamyl alcohol는 항아리와 스테인리스 용기 모두 숙성 기간이 증가할수록 감압과 상압 증류주 모두 감소하였으며 A/B의 비율은 감압과 상압 모두 비슷한 1.32-1.35, A/P는 2.17-2.22, B/P는 1.62-1.68의 비율을 나타내었다. 입국을 이용한 증류주의 향기 성분 분석 결과 증류 초기에 감압 증류에서 높은 검출 결과를 나타낸 것은 isoamyl alcohol>isobutyl alcohol>ethyl palmitate>decanoic acid>ethyl octanoate 순서 였으며 상압 증류에서 높은 검출 순서는 isoamyl alcohol>decanoic acid>ethyl palmitate>isobutyl alcohol 순 이었다. 항아리에 숙성을 한 증류주의 경우 숙성 기간이 경과하면서 관능 점수가 감압, 상압 증류주 모두 상승하였으며 가장 높은 관능을 보인 것은 감압 증류 600 mmHg로 종합 기호도에서 $6.98{\pm}0.28$으로 가장 높았다.

Allomyrina dichotoma larva extract attenuates free fatty acid-induced lipotoxicity in pancreatic beta cells

  • BACKGROUD/OBJECTIVES: Allomyrina dichotoma larva (ADL), one of the many edible insects recognized as future food resources, has a range of pharmacological activities. In a previous study, an ADL extract (ADLE) reduced the hepatic insulin resistance of high-fat diet (HFD)-induced diabetic mice. On the other hand, the associated molecular mechanisms underlying pancreatic beta-cell dysfunction remain unclear. This study examined the effects of ADLE on palmitate-induced lipotoxicity in a beta cell line of a rat origin, INS-1 cells. MATERIALS/METHODS: ADLE was administered to high-fat diet treated mice. The expression of apoptosis-related molecules was measured by Western blotting, and reactive oxidative stress generation and nitric oxide production were measured by DCH-DA fluorescence and a Griess assay, respectively. RESULTS: The administration of ADLE to HFD-induced diabetic mice reduced the hyperplasia, 4-hydroxynonenal levels, and the number of apoptotic cells while improving the insulin levels compared to the HFD group. Treatment of INS-1 cells with palmitate reduced insulin secretion, which was attenuated by the ADLE treatment. Furthermore, the ADLE treatment prevented palmitate-induced cell death in INS-1 cells and isolated islets by reducing the apoptotic signaling molecules, including cleaved caspase-3 and PARP, and the Bax/Bcl2 ratio. ADLE also reduced the levels of reactive oxygen species generation, lipid accumulation, and nitrite production in palmitate-treated INS-1 cells while increasing the ATP levels. This effect corresponded to the decreased expression of inducible nitric oxide synthase (iNOS) mRNA and protein. CONCLUSIONS: ADLE helps prevent lipotoxic beta-cell death in INS-1 cells and HFD-diabetic mice, suggesting that ADLE can be used to prevent or treat beta-cell damage in glucose intolerance during the development of diabetes.

Purification and Characterization of the Lipase from Acinetobacter sp. B2

  • Industrial development has increase consumption of crude oil and environmental pollution. A large number of microbial lipolytic enzymes have been identified and characterized to date. To development for a new lipase with catalytic activity in degradation of crude oil as a microbial enzyme, Acinetobactor sp. B2 was isolated from soil samples that were contaminated with oil in Daejon area. Acinetobactor sp. B2 showed high resistance up to 10 mg/mL unit to heavy metals such as Ba, Li, Al, Cr, Pb and Mn. Optimal growth condition of Acinetobactor sp. B2 was confirmed $30^{\circ}C$. Lipase was purified from the supernatant by Acinetobactor sp. B2. Its molecular mass was determined to the 60 kDa and the optimal activity was shown at $40^{\circ}C$ and pH 10. The activation energies for the hydrolysis of p-nitrophenyl palmitate were determined to be 2.7 kcal/mol in the temperature range 4 to $37^{\circ}C$. The enzyme was unstable at temperatures higher than $60^{\circ}C$. The Michaelis constant $(K_{m})\;and\;V_{max}$ for p-nitrophenyl palmitate were $21.8{\mu}M\;and\;270.3{\mu}M\;min^{-1}mg\;of\;protein^{-1}$, respectively. The enzyme was strongly inhibited by $Cd{2+},\;Co^{2+},\;Fe^{2+},\;Hg^{2+},\;EDTA$, 2-Mercaptoethalol. From these results, we suggested that lipase purified from Acinetobactor sp. B2 should be able to be used as a new enzyme for degradation of crude oil, one of the environmental contaminants.

What is the Key Step in Muscle Fatty Acid Oxidation after Change of Plasma Free Fatty Acids Level in Rats?

  • The purpose of this study was to discern the critical point in skeletal muscle fatty acid oxidation by changing plasma free fatty acids (FFA) level in rat. In the study, 3 key steps in lipid oxidation were examined after changing plasma FFA level by acipimox. The rates of both palmitate and palmitoylcarnitine oxidation were decreased by decrease of plasma FFA level, however, carnitine palmitoyl transferase (CPT) 1 activity was not changed, suggesting CPT1 activity may not be involved in the fatty acid oxidation at the early phase of plasma FFA change. In the fasted rats, ${\beta}-hydroxy$ acyl-CoA dehydrogenase (${\beta}$-HAD) activity was depressed to a similar extent as palmitate oxidation by a decrease of plasma FFA level. This suggested that ${\beta}-oxidation$ might be an important process to regulate fatty acid oxidation at the early period of plasma FFA change. Citrate synthase activity was not altered by the change of plasma FFA level. In conclusion, the critical step in fatty acids oxidation of skeletal muscles by the change of plasma FFA level by acipimox in fasting rats might be the ${\beta}-oxidation$ step rather than CPT1 and TCA cycle pathways.

Neuraminidase Inhibitors from Mushroom Microphorus affinis

  • In the course of screening anti-influenza agents from natural products, four neuraminidase inhibitors were isolated from the methanol extract of mushroom Microphorus affinis by purification using solvent partition, silica gel column chromatography, Sephadex LH-20, and semi-preparative HPLC. The chemical structures of these compounds were identified as ${\alpha}-lupeol$, methyl linoleate, methyl palmitate, and methyl oleate by means of spectral data including GC-MS, $^1H-,\;and\;^{13}C-NMR\;with\;IC_{50}$ values of 5.65, 7.07, 7.12, and $7.52\;\mu\textrm{M}$, respectively. They did not inhibit other glycosidases such as glucosidase, mannosidase, and galactosidase, indicating that they were relatively specific inhibitors of neuraminidase. The relationship between the fatty acid structure and inhibitory activity was investigated. The result showed that, in the case of an aliphatic linear hydrocarbon skeleton, at least one carboxyl (presumably any carbonyl) moiety and sixteen carbons were the necessary requirements for potent inhibition, whereas saturated, unsaturated, free, and ester forms did not have any significant effect on the activity.

산씀바귀의 Triterpenoid 성분조성 (Triterpenoid constituents of the Herbs of Lactuca raddeana)

  • 국화과 식물인 산씀바귀(Lactuca raddeana)를 식물화학적 방법에 의하여 연구한 바 primaylong chain alcohol인 1-hexacosanol과 1-tetracosa not을, triterpene acetate로 ${\beta}-amyrin\;acetate,\;{\alpha}-amyrin acetate$, lupeol acetate, pseudotaraxasterol acetate, taraxasterol acetate 및 germanicol acetate등을, fatty acyl triterpene은 이들의 구성 triter pone alcohol이 triterpone acetate의 경우와 같았으며 acyl moiety는 myristate, palmitate, stearate및 arachidate로 각각 나타났다.

  • Two diterpenes and one new sucrose isovaleryl ester having inhibitory effects on melanogenesis in B16 mouse melanoma were isolated from Euphorbiae Lathyridis Semen which has been used in traditional medicine for cancer, tumors and warts. New sucrose isovaleryl ester was identified as ${\alpha}-D-glucopyranoside$, $3,4,6-tris-O-(3-methyl-l-oxobutyl)-{\beta}-D-fructofuranosyl$, 2,6-bis(3-methylbutanoate) and two diterpenes were identified as ingenol-20-palmitate and 5,10-diacetyl-3-benzoyllathyrol$(Euphorbia\;factor\;L_3)$ from their spectral data.

국내산 참당귀 추출물의 휘발성 향기성분

  • Volatile flavor components of extracts produced from the domestic angelica root, which are oleoresin and absolutes type, for tobacco flavoring materials were isolated by solvent extraction method and were analyzed by GC and GC/MSD. And then volatile flavor components of oleoresin were compared with volatiles isolated from absolutes. A total of 65 components were identified in the angelica root extracts, from which 41 components were identified in the oleoresin volatiles, contained 15 hydrocarbons, 12 alcohols, 6 acids, 10 esters and 2 miscellaneous components. The major components were hexadecanoic acid (7.79%), methyl palmitate (6.49%), ethyl palmitate (2.02 %) and sesquiterpenes and sesquiterpene alcohols, such as elemol (2.92 %), ${\gamma}$-selinene (2.19%), $\beta$-selinene (2.02%), $\alpha$-eudesmol (3.49%) and $\beta$-eudesmol (6.12%). On the other hand, volatiles of absolutes, from which 60 components were identified, contained 28 hydrocarbons, 14 alcohols, 5 acids, 10 esters and 3 miscellaneous components. The major components were hyrocarbons, such as undecane (5.11 %), dodecane (3.10%) and pentadecane (1.14 %), and $\alpha$-muurolene (1.64 %), ${\gamma}$-selinene (1.49%), $\beta$-selinene (2.12 %), $\alpha$-eudesmol (2.25%), $\beta$-eudesmol (4.87%), hexadecanoic acid (12.67%) and hexanoic acid (1.87 %).
