Graft cultivation of Paeonia and Moutan was attempted to improve the harvest yields of medicinal parts of the plants. When Paeonia root was grafted with Moutan stem, the harvest yields of underground roots showed 35% increase, with Paeonia radix and Moutan cortex ratio of 3 : 1 paeoniflorin and paeonol contents among grafted Paeonia radix (11.71 and 0.35 mg/g, respectively), Paeonia radix (9.91 and 0.80 mg/g, respectively), and Moutan cortex (10.81 and 2.84 mg/g, respectively) were not significantly different. Paeonia radix grafted with Moutan stem was cultivated to investigate the ecological properties and to compare index components, including minerals, fatty acids, and organic acids. The contents of 10 major minerals among the samples were very similar, whereas those of Fe were different, showing 86.06, 127.36, 75.05, and 140.63 mg/g for (Paeonia radix, Paeonia radix grafted, Moutan cortex, and Moutan cortex grafted) contents and GC profiles of 11 fatty acids including linoleic, palmitic, linolenic, and oleic acids and those of 6 organic acids including oxalic, citric, and malic acids were very similar. Results of sensory evaluations were very similar between Paeonia radix and Paeonia radix grafted or Moutan cortex and Moutan cortex grafted showed astringent taste, and stronger sensory intensity of Paeonia radix than Paeonia radix grafted showed bitter taste, and stronger sensory intensity of Moutan cortex than Moutan cortex grafted.
This experiment was carried in order to compare some propagation methods and the effective components in the root of Paeonia albiflora pallas, Paeonia moutan Sims and Paeonia albiflora pallas root grafted with Paeonia mouton sims The optimum seeding time of August 20 to september 10 for increasing germination ratio. The growing and developing of Paeonia albiflora Pall. for root dividing methods was advantageous to practice at Autum than at spring season. The shoot cutting method during the flowering time was proper to increase the rooting ratio with the aid of Ruton, the plant growth regulator but not emerged of apical bud. The grafting stages between the Paeonia albiflora pall. and Paeonia moutan Sims was available to practice during the September for increasing the thickness growth of root. Among the effective components of the paeony the paeoniflorin was shown high, while paeonol in paeony moutan. The paeoniflorin of herbaceous paeony / paeony moutan was increased about 75.9% than herbaceous paeony alone, and the other compoents are similar to those of herbaceous paeony root.
From the fresh unripe fruits of Paeonia moutan Sim. (Paeoniaceae), paeoniflorin and its acyl congeners, benzoylpaeoniflorin and benzoyloxypaeoniflorin, along with ${\beta}-sitosterol$ and methyl gallate were isolated. All compounds were identified on the basis of spectral data and chemical reactions. However, paeonol was not detected from this plant parts. These results suggested that the chemical components of the unripe fruits were virtually similar to those of root barks.

This paper was intented to find out the effect of graft of Paeonia moutan scion into Paeonia albiflora stock on the yield and the pharmaceutical components of the root-Paeoniflorin, Albiflorin, Oxypaeoniflorin and Paeonol. The roots of Paeonia albiflora(PA), Paeonia moutan(PM) and Paeonia albiflora grafted in Paeonia moutan (M/A), were collected after being cultivated for three years in Yoenki, Chungnam, Korea. The following results were obtained ; 1. The growth characteristics of the underground part of M/A were similar to those of PA. 2. The yield of M/A was 3-5 times as that of PA in appearance. 3. The amount of Paeoniflorin in M/A was 0.2-0.5% higher than that in PA. The content of paeonol in M/A was not increased by grafting of paeonia moutan with high concentration of paeonol.
The genus Paeonia belongs to the family Paeoniaceae having significant medicinal and ornamental importance. The present investigation was undertaken with an aim to understand phylogenetic relationships of three Paeonia species (P. lactiflora, P. obovata, and P. suffruticosa) that are widely distributed in China, Korea, and Japan, using nuclear ribosomal DNA (nrDNA) internal transcribed spacer (ITS) sequence and to compare the phylogeny results with investigations reported earlier using existed sequences of the same species. The size variation obtained among sequenced nrDNA ITS region was narrow and ranged from 722 to 726 bp. The highest interspecific genetic distance (GD) was found between P. lactiflora and P. suffruticosa or P. obovata. The phylogram obtained using our nrDNA ITS sequences showed non-congruence with previous hypothesis of the phylogeny between section Paeonia and section Moutan of genus Paeonia. This result was supported by the phylogenetic relations showed in the phylogram constructed with existed sequences in NCBI. The present study suggested that P. obovata belonging to section Paeonia was phylogenetically closer to P. suffruticosa representing section Moutan of genus Paeonia than P. lactiflora belonging to section Paeonia. The main reason of the paraphyly of section Paeonia is thought to be nucleotide additivity directly caused by origin hybridization. This study provides more sequence sources of genus Paeonia, and will help for further studies in intraspecies population, and their phylogentic analysis and molecular evolution.
Paeonol (2-hydroxy-5-methoxyacetophenone) obtained by silica gel column chromatography of the essential oil extracted from Paeonia moutan (Paeoniaceae) was methylated by dimethylsulfate to yield methylpaeonol (2,5-di-O-methylacetophenone). Both compounds inhibited nitric oxide (NO) foundation in lipopolysaccharide-induced macrophage RAW 264.7 cells in nitrite assay. In the western blotting assay, it was shown that both compounds also decreased inducible nitric oxide synthase (iNOS)-and cyclooxygenase-2(COX-2) formation. Methylpaeonol produced more potently inhibited NO-, iNOS and COX-2 formation in the assays than paeonol. These results suggest that paeonol is in part responsible for anti-inflammatory activity of Paeonia moutan, and that synthesis of paeonol derivatives may produce a promising candidate for andtiifnalmmatory agent.
The application of PCR analysis on the herbal medicine Moutan Cortex (Paeonia suffruticosa Andrews) was evaluated by the comparison of the genetic relationship based on the DNA sequence with Paeoniae Radix (Paeonia lactiflora Pallas) following development of specific primers. Moutan Cortex and Paeoniae Radix were distinguished through the PCR analysis based on the internal transcribed spacer (ITS-PCR) from nuclear ribosomal DNA region. The 294 bp PCR products both of Moutan Cortex and Paeoniae Radix was amplified by MIF1 and MIR1. And a Moutan Cortex specific 225 bp PCR amplification product was amplified by MIF2 and MIR1 primers. The 225 bp sequence could be successfully amplified from Mortan Cortex of dried herbal preparations. PCR analysis based on ITS (ITS-PCR) may be an efficient tool for the discrimination of Moutan Cortex.
Previously, the methanolic extract of Paeonia moutan seeds was found to potently inhibit soybean lipoxy-genase (SLO). Hence to isolate SLO inhibitor, the defattd methaniolic extract of the seeds was consecutively partitioned wiht ether, ethyl acetate,n-butanol ,adn water. The ether souble fraction showing strong inhibitory activity against SLO was further fractionated into a strongly acidic, a weakly acidic, and a neutral fractions. The strongly acidic components of the ether extract were successively subjected to chromatography on a silica gel, Sephadex LH-20, and preparative HPLC. Four phenolic compounds were isolated , and twio of them showing a strong SLO inhibition activity were identified as luteolin (IC50=2.32$\mu\textrm{g}$/ml) and 5,6,4'-trihydroxy-7,3'- dimethoxylflavone (IC50=0.31$\mu\textrm{g}$/ml) by UV, IR, 1H-& 13C-NMR, and MS spectroscopy. In addition, two flavonoids showed significantly antioxidative activity as strong as that of of $\alpha$-tocopherol (p<0.05) in the autoxidation system of linoleic acid. These results suggest that luteolin and 5,6,4'-trihydroxy-7,3'-dimethoxy-flavone may be used as a potential source of anti-inflammatory agents with antioxidative activity.
The MeOH extract of the bark of Paeonia moutan showed potent inhibitory effect on the mushroom tyrosinase activity in vitro. The activity-guided fractionation of t he MeOH extract resulted in the isolation of three active compounds. The chemical structures of these compounds were elucidated by chemical and spectroscopic evidence as catechin, 1,2,3,6-tetra-O-galloyl-${\beta}$-D-glucose and 1,2,3,4,6-penta-0-galloyl-${\beta}$-D-glucose, respectively. Among them, the inhibitory activity by 1,2,3,6-tetra-galloyl-${\beta}$-D-glucose on mushroom tyrosinase was more potent $(IC_{50}=3.5\;{\mu}M)$ than that of kojic acid $(IC_{50}=8.7\;{\mu}M)$ ,but catechin enhanced the mushroom tyrosinase activity 50% in the concentration of 34.5M.

Squalene synthase catalyzes the reductive dimerization of two molecules of farnesyl diphosphate to form squalene at the final branch point of the cholesterol biosynthetic pathway. Due to the unique position of this enzyme in the pathway, its inhibitors may have advantages as antihypercholesterolemic agents. Therefore, selective inhibitors of squalene synthase do not prevent the formation of the essential branch products of the isoprene pathway, such as dolichol, coenzyme-Q, and prenylated proteins, as might be expected for inhibitors of enzymes earlier in the pathway; for example, lovastatin and mevalotin. The current study reports that CJ90002, a pentagalloylglucose isolated from Paeonia moutan SIM (Paeoniaceae), which is an important Chinese crude drug used in many traditional prescriptions, was a potent inhibitor of rat microsomal squalene synthase, and also a potent inhibitor of cholesterol biosynthesis in vitro. In addition, the intraperitoneal and oral administration of CJ90002 had a significant lowering effect on plasma cholesterol levels in hamsters.
