MicroRNA-186 targets SKP2 to induce p27Kip1-mediated pituitary tumor cell cycle deregulation and modulate cell proliferation

  • Pituitary tumors are usually benign but can occasionally exhibit hormonal and proliferative behaviors. Dysregulation of the G1/S restriction point largely contributes to the over-proliferation of pituitary tumor cells. F-box protein S-phase kinase-interacting protein-2 (SKP2) reportedly targets and inhibits the expression of $p27^{Kip1}$, a well-known negative regulator of G1 cell cycle progression. In this study, SKP2 expression was found to be upregulated while $p27^{Kip1}$ expression was determined to be downregulated in rat and human pituitary tumor cells. Furthermore, SKP2 knockdown induced upregulation of $p27^{Kip1}$ and cell growth inhibition in rat and human pituitary tumor cells, while SKP2overexpression elicited opposite effects on $p27^{Kip1}$ expression and cell growth. The expression of microRNA-186 (miR-186) was reported to be reduced in pituitary tumors. Online tools predicted SKP2 to be a direct downstream target of miR-186, which was further confirmed by luciferase reporter gene assays. Moreover, miR-186 could modulate the cell proliferation and $p27^{Kip1}$-mediated cell cycle alternation of rat and human pituitary tumor cells through SKP2. As further confirmation of these findings, miR-186 and $p27^{Kip1}$ expression were downregulated, while SKP2 expression was upregulated in human pituitary tumor tissue samples; thus, SKP2 expression negatively correlated with miR-186 and $p27^{Kip1}$ expression. In contrast, miR-186 expression positively associated with $p27^{Kip1}$ expression. Taken together, we discovered a novel mechanism by which miR-186/SKP2 axis modulates pituitary tumor cell proliferation through $p27^{Kip1}$-mediated cell cycle alternation.

Prophylactic Effect of Pegfilgrastim on Febrile Neutropenia in Patients with Non-Hodgikin's Lymphoma (비호지킨림프종 환자의 발열성 호중구감소증에 대한 페그필그라스팀의 예방효과)

  • Objective: Pegfilgrastim is recently introduced that is long acting G-CSF for prophylaxis of febrile neutropenia. Treatment of non-Hodgikin's lymphoma (NHL) with R-CHOP is classified with relative high risk of febrile neutropenia. The study evaluated the prophylactic effect of pegfilgrastim to reduce the incidence of febrile neutropenia associated with R-CHOP of patient in NHL. And the risk factors associated with the incidence of FN and related events were evaluated. Methods: This retrospective study reviews the Electronic Medical Record of 68 NHL patients who received R-CHOP chemotherapy in single center between September 2013 and August 2014. These patients were classified who receive prophylaxis pegfilgrastim or no prophylaxis. Results: Sixty eight patients received R-CHOP with NHL. In 144 cycles of patients receiving pegfilgrastim, compared with no prophylaxis 178 cycles, had a lower incidence of febrile neutropenia (5.5% vs. 23.6%, p = 0.001), grade 3 or grade 4 neutropenia (14.4% vs. 89.8%, p < 0.001) and neutropenia related events (p < 0.05). The risk of febrile neutropenia after prophylaxis was significantly associated with age ${\geq}65$ (OR: 5.87, 95% CI 1.07-32.27, p = 0.042), $IPI{\geq}3$ (OR: 7.2, 95% CI 1.31-39.6, p = 0.023), S.alb < 3.5 g/dL (OR: 31.01, 95% CI 6.32-152.17, p < 0.0001). In multiple logistic regression analysis, lower baseline serum albumin (OR: 21.1, 95% CI 3.8-116.98, p = 0.001) was significantly associated with occurrence of febrile neutropenia. Conclusion: The study recommends prophylactic pegfilgrastim through risk assessment of febrile neutropenia in patients with non-Hodgikin's lymphoma receiving R-CHOP.

Depressive Symptoms in Patients with Parkinson's Disease (파킨슨병 환자에서의 우울증상)

  • Objectives: The aim of this study was to investigate 1) the prevalence of depressive symptoms, 2) the severity of depressive symptoms, 3) the correlation of depressive symptoms with clinical variables, and 4) factors that contribute to depressive symptoms in patients with Parkinson's disease. Methods: One hundred eighteen patients with Parkinson's disease referred from the Parkinson's Disease Clinic of Dong-A University Hospital, Busan, Korea, completed a self-administered questionnaire package, which included basic demographic data, the Beck Depression Inventory, the Parkinson's disease quality of life questionnaire, the Symptom Checklist-90-Revision(SCL-90-R), and the Spielberger's State-Trait Anxiety Inventory. In addition, a structured interview and a complete neurological examination, including the Hoehn and Yahr stage, the motor part of the Unified Parkinson's Disease Rating Scale(some selected scales of UPDRS part III), the Schwab and England Activities of Daily Living scale(ADL), and the Korean version of Mini-Mental State Examination were performed. Results: 1) Based on BDI score, subjects were divided into four groups:severely(40.7%), moderately(13.6%) and mildly(12.7%) depressive and non-depressive(33.1%). 2) The severity of depressive symptom in Parkinson's disease was positively correlated with Hoehn and Yahr(H & Y) stage(r=0.34, p<0.0001), the severity of motor symptom(r=0.35, p<0.0001), and trait anxiety inventory(r=0.33, p<0.001). On the other hand, the severity of depressive symptom was negatively correlated with educational level(r=-0.34, p<0.001), ADL(r=-0.37, p<0.0001) and Parkinson's disease quality of life (PDQL)(r=-0.69, p<0.0001). Among several clinical variables, the PDQL was the most influential factor predicting whether the depressive symptom was present or not. Conclusion: This study suggests that depressive symptom is very prevalent among patients with Parkinson's disease. Data from this study indicate that medical staffs who take care of patients with Parkinson's disease should pay attention to finding and treating depressive symptom among their patients. With appropriate psychiatric intervention, patient's depressive symptom can be minimized or alleviated and thus, the quality of life in these patients is likely enhanced.

MicroRNA-3200-5p Promotes Osteosarcoma Cell Invasion via Suppression of BRMS1

  • Tumour metastasis is one of the most serious challenges of cancer as it is the major cause of mortality in patients with solid tumours, including osteosarcoma (OS). In this regard, anti-metastatic genes have potential for metastasis inhibition strategies. Recent evidence showed the importance of breast cancer metastasis suppressor 1 (BRMS1) in control of OS invasiveness, but the regulation of BRMS1 in OS remains largely unknown. Here, we used bioinformatics analyses to predict BRMS1-targeting microRNAs (miRNAs), and the functional binding of miRNAs to BRMS1 mRNA was evaluated using a dual luciferase reporter assay. Among all BRMS1-targeting miRNAs, only miR-151b, miR-7-5p and miR-3200-5p showed significant expression in OS specimens. Specifically, we found that only miR-3200-5p significantly inhibited protein translation of BRMS1 via pairing to the 3'-UTR of the BRMS1 mRNA. Moreover, we detected significantly lower BRMS1 and significantly higher miR-3200-5p in the OS specimens compared to the paired adjacent non-tumour bone tissues. Furthermore, BRMS1 and miR-3200-5p levels were inversely correlated to each other. Low BRMS1 was correlated with metastasis and poor patient survival. In vitro, overexpression of miR-3200-5p significantly decreased BRMS1 levels and promoted OS cell invasion and migration, while depletion of miR-3200-5p significantly increased BRMS1 levels and inhibited OS cell invasion and migration. Thus, our study revealed that miR-3200-5p may be a critical regulator of OS cell invasiveness.

Factors Influencing on the Attitude toward withdrawal of life-sustaining treatment of Nursing College Students (간호대학생의 연명치료 중단에 대한 태도에 영향을 미치는 요인)

  • This study was a descriptive study to investigate the factors influencing on attitude toward withdrawal of life-sustaining treatment of nursing students' consciousness of biomedical ethics, good death recognition and meaning of life. The subjects of this study were 293 nursing students attending college at P city. Data were collected for two weeks from May 1, 2018 to May 11, 2018. The statistical method was performed with SPSS WIN 22.0 using a complex sampling analysis, pearson's correlation coefficients, and a multiple regression analysis. The result of this study was significant positive correlation between attitude toward withdrawal of life-sustaining treatment and consciousness of biomedical ethics(r=.266, p<.001), good death recognition(r=.373, p<.001), meaning of life(r=.122, p=.037). The meaning of life was significant positive correlation consciousness of biomedical ethics(r=.294, p<.001), good death recognition(r=.230, p<.001). The good death recognition was significant positive correlation consciousness of biomedical ethics(r=.306, p<.001). Factors Influencing on the attitude toward withdrawal of life-sustaining treatment was good death recognition(${\beta}=.32$, p<.001) and consciousness of biomedical ethics(${\beta}=.16$, p=.004). These factors explained about 16% of the variance. Therefore it is necessary to develop an educational program to positively raise the attitude of consciousness of biomedical ethics and good death recognition.

Analysis of correlation between the inspiratory capacity of the National softball players and the bone density, bon mass, muscle power, muscle endurance (국가대표 소프트볼선수들의 흡기능력과 골밀도 및 골 질량 그리고 체간의 근력 및 근지구력의 상관관계 분석)

  • PURPOSE: A prospective study was conducted to investigate the correlation between the inspiration ability, bone mineral density, lumbar muscle strength, and muscular endurance for the national softball athletes in the national training center. METHODS: The general characteristics of study subjects, inspiration ability, bone mineral density, muscle strength, and muscular endurance data were analyzed using descriptive statistics. In addition, the Pearson product moment correlation was performed to investigate the correlation between the inspiratory capacity, bone mineral density, muscle strength, and muscular endurance. RESULTS: The inspiration, flow rates, and volume were not correlated with the bone mass and bone mineral density. Inspiration and the flow rates and volume were not correlated with the bone mass and bone mineral density. On the other hand, inspiration was correlated with the Extensor muscles (r=.464, p=.006) at an angular velocity of 60°/s and the flexors (r=.463, p=.006) and extensor muscles (r=.615, p<.001) at an angular velocity of 180°/s. The flow rate was also correlated with the extensor muscles (r=.444, p=.009) at an angular velocity of 60°/s and with flexor muscles (r=.432, p=.011) and extensor muscles (r=.589, p<.001) at an angular velocity of 180°/s. Finally, the volume was correlated at the extensor muscles at an angular velocity of 180°/s (r=.534, p=.001). CONCLUSION: The correlation between the inspiratory capacity, bone mineral density, muscle strength, and muscular endurance of softball athletes did not correlate with the bone mass and bone density. On the other hand, the lumbar muscle strength increased with increasing inspiratory capacity.

  • In this paper, a new class of diagram algebras which are subalgebras of signed brauer algebras, called the Walled Signed Brauer algebras denoted by ${\overrightarrow{D}}_{r,s}(x)$, where $r,s{\in}{\mathbb{N}}$ and x is an indeterminate are introduced. A presentation of walled signed Brauer algebras in terms of generators and relations is given. The cellularity of a walled signed Brauer algebra is established. Finally, ${\overrightarrow{D}}_{r,s}(x)$, is quasi- hereditary if either the characteristic of a field, say p, p = 0 or p > max(r, s) and either $x {\neq}0$ or x = 0 and $r{\neq}s$.
