Respones of Photobacterium phosphoreum to toxic substances (독성물질 농도에 따른 Photobacterium phosphoreum의 bioluminescence 변화)

  • Photobacterium phosphoreum was used for the study of bioluminescence response to toxic substances including phenol, As2O3, SoO2, and CrO3 in view of developing monitoring system. measurement of inhibition of bioluminescence in P. phosphoreum has been proposed as a sensitive and raped procedure to monitor toxic substances. The concentration of toxic substance causing 50% light reduction(EC50) in bioluminescence intensity was determined with free and immobilized P. phosphoreum, The minimum inhibitory concentrations (MICs) for bioluminescence emission were found to be 400ppm for As2O3, 800ppm for phenol, 60ppm for SeO2 and 60ppm for CrO3 , respectively. The linear correlation between Gamma value and the concentration of toxic substances was obtained and EC50 wa calculated from the linear correlation. The free cells were shown to be more sensitive to toxic substances than cells immobilized on Sr-alginate and Ca-alginate. However, the linear regression curves were derived from the Sr-alginate immobilized cells indicating the immobilization method in s useful tool for monitoring of toxic substances under the more stable condition of bioluminescence.

Enhancement of Nitrendipine Bioavailability in Rats by its Solid Dispersion with $Hydroxypropyl-{\beta}-Cyclodextrin$ after Oral Administration (흰쥐에 경구 투여시 히드록시프로필-베타-시클로덱스트린과 니트렌디핀 고체분산에 의한 생체이용률 증가)

  • Nitrendipine, a slightly soluble calcium channel blocking agent forms a solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$, which exhibits better dissolution characteristics than the uncomplexed drug. The dissolution rate of nitrendipine was markedly increased in solid dispersion system in pharmacopeial disintegration media at pH 1.2 and pH 6.8. Four different dosage forms of nitrendipine were administered to rats: (a) nitrendipine in the solution of PEG 400; (b) nitrendipine solid dispersion system with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 by solvent evaporation method and administered in capsule form; (c) physical mixture of nitrendipine with $hydroxypropyl-{\beta}-cyclodextrin$ in a molar ratio of 1:2 and administered in capsule form; (d) nitrendipine alone administered in capsule form. Relative bioavailability after the oral administration of various dosage forms to rats with a dose of 10 mg/kg equivalent to nitrendipine was compared with that of nitrendipine in the solution of PEG 400. The AUC of solid dispersion was significantly bigger than that of nitrendipine powder. $T_{max}$ of solid dispersion was significantly shorter and $C_{max}$ was higher than that of nitrendipine powder. These results indicate that the bioavailability of nitrendipine could be improved markedly by inclusion complexation. An interesting correlation also appears to exist between the in vitro dissolution data and the area under the plasma concentration-time curves.

Circadian Changes in the Pharmacokinetics of Acebutolol Orally Administered to Rabbits

  • Circadian variations of acebutolol and its main metabolite, diacetolol pharmacokinetics were studied after a single oral administration of acebutolol (10 mg/kg) to eight rabbits at 10 : 00 AM (in the morning) and 10 : 00 PM (at night). The plasma concentration profiles of acebutolol were significantly different (P<0.05) between 10 : 00 AM and 22 : 00 PM, suggesting circadian variations of pharmacokinetic behaviors. A significant circadian rhythm of pharmacokinetic parameters was noted in rabbits, showing higher total body clearance (CL/F), and lower the area under the plasma concentration-time curves (AUC) of acebutolol than that at night. The half-life ($t_{1/2}$) of acebutolol and diacetolol were also significantly shorter in the morning than at night (P<0.05). Metabolite-parent AUC ratio at night significantly decreased compared to in the morning, implying that night time could inhibit acebutolol metabolism than in the morning. From this study there was an administration-time difference of acebutolol pharmacokinetics in the rabbits. The optimized dosing regimen of acebutolol can be decided by considering circadian rhythm so that the effective therapies are established for patients.

  • Let $M_g$ be the moduli space of isomorphism classes of genus g smooth curves. It is a quasi-projective variety of dimension 3g - 3, when $g > 2$. It is known that a complete subvariety of $M_g$ has dimension $< g-1 [D]$. In general it is not known whether this bound is rigid. For example, it is not known whether $M_4$ has a complete surface in it. But one knows that there is a complete curve through any given finite points [H]. Recently, an explicit example of a complete curve in moduli space is given in [G-H]. In [G-H] they constructed a complete curve of $M_3$ as an intersection of five hypersurfaces of the Satake compactification of $M_3$. One way to get a complete curve of $M_3$ is to find a complete one dimensional family $p : X \to B$ of plane quartics which gives a nontrivial morphism from the base space B to the moduli space $M_3$. This is because every non-hyperelliptic smooth curve of genus three can be realized as a nonsingular plane quartic and vice versa. This paper has come out from the effort to find such a complete family of plane quartics. Since nonsingular quartics form an affine space some fibers of p must be singular ones. In this paper, due to the semistable reduction theorem [M], we search singular plane quartics which can occur as singular fibers of the family above. We first list all distinct plane quartics in terms of singularities.

Effects of (100) Orientation of LaNiO3 on the Growth and Ferroelectric Properties of Pb(Zr,Ti)O3 Thin Films (LaNiO3의 (100)배향성이 Pb(Zr,Ti)O3 박막의 결정성장과 강유전성에 미치는 영향)

  • Pb(Zr,Ti)O₃[PZT] thin films were prepared on a highly (100) oriented LaNiO₃[LNO] and a randomly oriented LNO by sol-gel process. The PZT thin films on a highly (100) oriented LNO show a high (100) crystal orientation (F=100 %), those on a randomly oriented LNO show a random crystal orientation (F=60 %). All the PZT layer have a flat and dense microstructure with large columnar grains and their grain size are 25 nm. In the ferroelectric curves at electric field of 40 kV/cm, a highly (100) oriented PZT/LNO samples show coercive field, E/sub c/=10 kV/cm and remanent polarization, P/sub r/=14.5 μC/㎠, while a randomly oriented PZT/LNO sample show E/sub c/=10 kV/cm and P/sub r/=5.4 μC/㎠.

A Modified Back-Titration Method to Measure Soil Titration Curves Minimizing Exchange Acidity and Dilution Effects (토양 적정곡선 측정을 위하여 교환산도와 희석효과를 최소화한 수정역적정법 제안)

  • A back titration method was suggested to determine the exchangeable acidity of soil originated from exchangeable aluminum and adsorbed hydrogen ion in relation to minimizing its titration error. In this method. soils are treated by neutral salt solution whose concentration is the same as soil solution suspended by distilled water. And then. its pH is adjusted at 3, using 1 N HCl solution and back-titrated by 0.1 N NaOH solution containing 1 N NaCl. The results of the suggested back-titration showed the possibility of practical application with linear relationship to that of standard method.

Growth Charateristics of Five Microalgal Species Isolated from Jeju Island and Four Microalgal stock Strans in Hatchery

  • Five microalgal species isolated from the Jeju coast and four microalgal stock strains in hatchery were cultured in order to investigate their adapation to extreme changes in environmental factors such as salinity, water temperatue, adn nutrients. In case of salinity variation, Nitzschia sp. of Bacillariophyceae, Isochrysis galbana of Haptophyceae and Tetraselmis gracilis of Prasinophyceae showed optimum growth at the low salinity of 20 and 25 psu. Amphora coffeaeformis and Chetoceros simplex of Bacillariophyceae, and Pavlova lutheri of Haptophyceae adapted well at the relatively high salinities of 30 and 35 psu. However Phaeodactylum tricornutum of Bacillariophyceae and Chlorella sp. of Chlorophyceae showed euryhaline property In case of water temperature variation, most of all the species studied wer inhibited at 10℃. C. simplex, Nitzschia sp., p. tricornutum, Chlorella sp. and T. gracilis grew well at above 20℃. A. coffeaeformis, I. galbana and P. lutheri adapted also at the high temperature of 30℃. Each microalgal strain showed different growth rates and its maximum biomass. Generally microalgal populations from the Jeju coast grow well in relatively high salinity and high water temperature. Their growth were inhibited at low water temperature, but not likely affected at low salinity. This study indicates that the microalgal populations could not be affected by abnormally low salinity phenomena, which have happened occasionally around the west Jeju coast in summer and have led macrobenthic animals to mass mortality.

Torque Curves and Cardiovascular Response to Isometric Exercise at the Elbow Joint in Normal and Hemiplegic Subjects (정상인과 편마비 환자의 주관절 등척성 운동시 우력양상과 심혈관계에 미치는 영향)

  • The isometric torque of the elbow flexor and extensor muscles were measured for 6 seconds at a joint angle of 90$^{\circ}$ , in 10 normal subjects (control group) and 10 hemiplegic subjects(patient group), using the Cybex NORMTM System. The peak torque, the time to peak torque were measured for each exercise. In addition, heart rate and blood pressure were recorded simultaneously at rest and immediately following exercise completion at 1 and 3m mutes. Statistical analysis was performed using SPSS 8.0 for Windows software and mean and standard deviations were calculated. The results are as follows. 1) In the patient involved group. the isometric values for flexors and extensors were significantly lower than in the normal nondominant group(p<.05). 2) The extensor to flexor strength ratio in the isometric mode was 121.0% in the patient involved group compared with 78.7%in the normal nondominant group, a significant difference(p<.05). 3) The mean increment ratio was increased 19.0% for systolic blood pressure and 25.2% for disatolic blood pressure in the patient group. 4) The mean increment ratio was increased 36.0% heart rate in the patient group.

An NMR Study on Complexation of Cesium Ion by p-tert-Butylcalix[6]arene Ethyl Ester

  • Complexation of cesium ion by p-tert-butylcalix[6]arene ethyl ester was studied by NMR spectroscopy in nonpolar $CDCl_3$ and polar acetone-$d_6$ and the results were compared with each other. Analysis of temperature dependent $^1H$ spectra and titration curves reveals that both solvents result in a 1 : 1 cone-form complex with nonpolar $CDCl_3$yielding a more tightly bound one than acetone-$d_6$. Unexpectedly, at very low temperature, we have found that two phenyl ring proton peaks of equal intensity appear both in $CDCl_3$and in acetone-$d_6$ solution which gradually collapse and eventually coalesce into a single line as temperature is raised. This observation could be interpreted in terms of the chemical exchange through direct and/or indirect interconversion between two equivalent conformations possible the complex in both solvents over the temperature range observed. And broadening of $^{133}Cs$ (I = 7/2) nmr line with increasing temperature has also been observed, indicating the exchange of $^{133}Cs$ ion between the complex and the solvent. From numerical fitting of lineshape changes for one-dimensional $^1H$ and $^{133}Cs$ spectra, the exchange rate constants and other relevant parameters for this conformational interconversion and the complex-solvent exchange were deduced.

Use of water retention curves predicted from particle-size distribution data for simulation of transport of Benzo[a]pyrene in soil

  • Water retention curve (WRC), one of soil hydraulic properties, is often approximated by property-transfer models (PTMs). Using the PTMs, we can estimate the WRCs from other physical properties such as particle-size distribution (PSD). The objective of this work was to investigate the performance of two PTMs with different origins for numerical simulations on transport of Benzo[a]pyrene in a soil. To do this, we chose both PTMs with different origins, i.e., (1) the lognormal distribution model (L anti NL models), and (2) the modified $Kov\'{a}cs$ model (MK model). The MK model showed tile worse performance in estimation of the WRCs. When transport of B[a]P was simulated, the MK model predicted to move farther than the L and NL models did, indicating that transport of B[a]P in a soil can be greatly influenced by the choice of PTMs.
