Phylogenetic Characteristics of Bacterial Populations Found in Serpentinite Soil (초염기성 사문암 토양 중 세균군집의 계통학적 특성)

  • A phylogenetic analysis of bacterial populations inhabiting soil derived from serpentine was conducted. The samples were collected from adjacent metamorphic rocks and serpentinite soil at Kwangcheon. The pH of the serpentine areas ranged from 8.5 to 9.2. The number of bacteria on the DAL medium which was diluted with $10^{-2}$ of AL medium was 10~100 fold higher than that from the full strength of AL medium, and which indicates that oligotrophs are distributed in the serpentinite soil. Of a total of 76 isolates, 42 isolates were oligotrophic bacteria, which grew only on the DAL medium. Based on a phylogenetic analysis using 16S rDNA sequences, these isolates are found to fall within five major phylogenetic groups: proteobacteria $\alpha$-subdivision (3 strains), $\alpha$-subdivision (7 strains), $\gamma$-subdivision (2 trains); high G+C gram-positive bacteria (19 strains); low G+C grampositive bacteria (14 strains). Bacteria of the genus Streptomyces (high G+C division) and Bacillus (low G+C division) have been considered to form a numerically important fraction of serpentinite soil. Oligotrophic strains categorized as Afipia ($\alpha$-subdivision), Ralstonia, Variovorax ($\beta$-subdivision), Pseudomonas ($\gamma$ -subdivision), Arthrobacter (high G+C division), and Streptomyces (low G+C division).

The Characteristics of Biopellet Produced Upon Reactor Configuration in UASB System (UASB 공법에 있어서 반응조의 형상변화에 따른 입상슬러지의 특성에 관한 연구)

  • Physicochemical and morphological characteristics of biopellets produced in "control" and modified UASB reactor were investigated to compare the reactor performance with regard to the hydrogen partial pressure. The characteristics of biopellet produced in modified UASB reactor operated with high hydrogen partial pressure were better than those of "control" reactor operated with relatively lower hydrogen partial pressure, therefore the hydrogen partial pressure effected greatly on the formation and stability of the biopellet. Furthermore, pellets from the UASB system with modified settler showed a better settleability and biomass holding capacity. The chemical composition of biopellet was distinctively different from that of common bacterial formula, $C_5H_7O_2N$. Biopellets was composed the large fraction of nitrogen in comparison with common anaerobic microbes. These results implicated the existence possibility of polypeptide-type extracellular polymer. The morphological characterization with SEM showed that microorganisms observed at surface of biopellet produced in modified UASB reactor operated with high $P_{H_2}$ condition were very similar in shape and size to the Methanobrevibactor arboriphilus-$H_2$ utilizing methanogen. The microorganisms was distinguished from those of "control" reactor operated with low $P_{H_2}$ condition. From these results, it could be explained the hydrogen partial pressure effects on pelletization mechanism.

Atmospheric concentration and mutagenicity of organic pollutants of suspended particulate in Seoul (서울시 대기중 유기오염물질의 농도와 돌연변이원성에 대한 연구)

  • To evaluate the difference of concentration and mutagenicity of organic pollutants between residential and traffic area of Seoul, air samples were collected in Bulkwang (residential) and Shinchon (traffic) area. Samples were analyzed to measure the concentration of extractable organic matters (EOM) and their subfractions and mutagenicities were tested using Salmonella typhimurium TA 98. The concentrations of polycyclic aromatic hydrocarbons (PAHs) were also measured by gas-chromatography and compared between two areas. The results were as follows ; 1. While the concentration of total suspended particulate (TSP) in residential area was below the environmental standard in annual average, the concentration in traffic area was above the standard and was up to its maximum $256{\mu}g/m^3$ in November. The difference of TSP concentrations in both areas of each month was statistically significant (P<0.05). 2. The concentration of fine particle in traffic area was significantly higher compare to that in residential area and showed statistically significant monthly difference in both areas (P<0.05). The proportion of concentration of fine particle to TSP was 55-68%. 3. Mean concentrations of EOM in residential and traffic areas were $4.3{\mu}g/m^3\;and\;5.3{\mu}g/m^3$ respectively. The proportion of amount of EOM from fine particle to EOM from TSP was 70-88%. 4. While the percentage of polar neutral organic compounds (POCN) of fine particle in Bulkwang's sample was higher compare to Shinchon's sample, the percentage of aliphatic compounds of fine particle in Shinchon's sample was higher compare to Bulkwang's sample. The percentages of PAH fraction were as low as 6-10% in both areas. 5. The mutagenic activity of nit concentration of organic matters extracted from fine particle was higher compare to that of coarse particle and was increased when metabolically activated with S9. Mutagenicities with metabolic activation calculated by unit air volume were significantly different between residential and traffic area, $17\;revertants/m^3$\;and\;22\;revertants/m^3$ respectively. 6. The concentrations of benzo(a)pyrene in fine particle of traffic and residential areas were $3.10ng/m^3\;and\;2.02ng/m^3$ respectively. Sixteen PAHs were higher in samples of traffic area compare to residential area and also concentrations of PAHs in fine particle were higher compare to coarse particle.

Some Properties of Partially Purified Alkaline Inorganic Pyrophosphatase from Mung-bean (Phaseolus radiatus) Seedling (발아초기(發芽初期) 녹두의 Alkaline Inorganic Pyrophosphatase의 효소적(酵素的) 성질(性質)에 대하여)

  • Time course of alkaline inorganic PPase activity with different parts of mung-bean sprout and some properties of partially purified enzyme from mung-bean leaves were investigated. The enzyme activity in leaf, root and cotyledon showed a tendency to increase at an early stage and then decrease gradually as germination continued. However, the crude homogenate of epicotyl showed the continuous decline of the enzyme activity but that of hypocotyl showed the continuous increase. In particular, the enzyme activity of leaf fraction was about 2-4 times as high as those of other fractions. The specific activity of the leaf enzyme was increased 86-fold, with a 23.9% yield, upon purification procedures. The purified enzyme from leaves had the Rm value of 0.35 and was not homogenous when judged by disc gel electrophoresis. Using tetrasodium pyrophosphate as a substrate, the apparent Km value for the partially purified enzyme was 0.89 mM. The enzyme was highly specific for $Mg^{2+}$ $CU^{2+}$ and $Fe^{2+}$ was also showed the activating effect of 56% and 55% with $Mg^{2+}$, respectively. However, $Ca^{2+}$), $Zn^{2+}$, $Mn^{2+}$, $Co^{2+}$ and $Ni^{2+}$ acted as inhibitors for the enzyme. The pH optimum for the enzyme shifted from 9.0 to 8.0 as the concentration of $Mg^{2+}$ was increased. The enzyme from mun-bean leaf was the most active at $50^{\circ}C$ and considerably stable on heat.

Antitumor effect of Ganoderma lucidum : Cytotoxicity and Tumor Growth Delayt(1) (영지버섯의 항암효과 :세포독성과 종양의 성장억제에 미치는 영향(1))

  • Purpose :. To investigate the effect of aqueous extract of Ganoderma lucidum(G.I.) on the surival of tumor cells in vitro and on the growth of tumors in vivo. Materials and Methods : Dried G.I. was made into powder, extracted with distilled water, filtered and diluted from a maximum concentration of 100 mg/ml in sequence. The cytotoxicity of G.I, in vitro was evaluated from its ability to reduce the clonogenicity of SCK tumor cells. For the tumor growth delay study, about $2{\times}10^5$$ of SCK tumor cells were subcutaneously inoculated in the legs of A/J mice. The first experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. from the first day after tomor inoculation for 10 days. The second experimental group of mice were injected i.p. with 0.2ml of 250 mg/kg of G.I. either once a day for 10 days or twice a day for 5 days beginining from the 7th day after tumor inoculation Results : 1. Cytotoxicity in vitro;survival fraction, as judged from the curve, at G.I. concentration of 0.5, 1,5, 10, 25, 50 and 100 mg/ml were 1.0, $0.74{\pm}0.03$, $0.18{\pm}0.03$, $0.15{\pm}0.02$, $0.006{\pm}0.002$, 0.015 and 0.0015, respectively. 2. Tumor growth delay in vivo; a) the time required for the mean tumor volume to grow to $1,000mm^3$ was 11 days in the control group and 14 days in the experimental group. b) the time required for tumor volume to increase 4 times was 11 days in the control group while it was 10.5 and 12 days in the groups injected with G.I. once a day and twice a day from the 7th day after tumor inoculation respectively. Conclusion : Aqueous extracts of G.I. showed a marked cytotoxicity on the SCK mammary cells in vitro. Tumor growth delay was statistically signiricant when G.I. in-jection was started soon after tumor inoculation, but it was not significant when injection was started after the tumors were firmly established.

A Study on the Galactooligosaccharides Produced from Transgalactosylation with ${\beta}-Galactosidase$ of Aspergillus and Kluyveromyces (A. oryzae와 K. fragilis 유래의 ${\beta}-Galactosidase$ 혼합처리에 의한 전이 Galactooligo당의 생성에 관한 연구)

  • These studies examined the production of oligosaccharides by ${\beta}-galactosidase$originated from Aspergillus oryzae and Kluyveromyces fragilis mixed. In addition, heat resistance and acid stability of transgalactosylated oligosaccharides were measured. When ${\beta}-galactosidase$ from A. oryzae, K. fragilis and mixed ${\beta}-galactosidase$ were added to 30%(w/v) lactose solution, maximum production of transgalactosylated oligosaccharides were 26.9%, 37.05 and 27.2%, respectively. The ratios of disacchride, trisacchride and tetrasaccharide in transgalactosylated oligosaccharides were 20.5 : 5.4 : 0.6, 20.4 : 10.5 : 4.2 and 21.0 : 4.1 : 1.9, respectively. Nine different oligosaccharides were recovered with 30% and 40% ethanol fractions. When the 30% ethanol fraction was treated at $150^{\circ}C$ for 10 min more than 90% of oligosaccharides remained stable. More than 90% of the oligosaccharides were stable at $130^{\circ}C$ for 3 min with pH 3.0, whereas there of Kluyveromyces was more than 90% with pH 3.5.

  • Hydrogenated amorphous carbon nitride[a-C:H(N)] films were deposited on p-type Si(100) at room temperature with bias voltage of 200 V by DC saddle-field plasma-enhanced chemical vapor deposition. Effects of the ratio of $N_2$ to $CH_4$($N_2/CH_4$), in the range of 0 and 4 on such properties as optical properties, microstucture, relative fraction of nitrogen and carbon, etc. of the films have been investigated. The thickness of the a-C:H(N) film was abruptly decreased with the addition of nitrogen, but at $N_2/CH_4$>0.5, the thickness of the film gradually decreased with the increase of the $N_2/CH_4$. The ratio of N to C(N/C) of the films was saturated at 0.25 with the increase of $N_2CH_4$. N-H, C≡N bonds of the films increased but C-H bond decreased with the increase of $N_2CH_4$.Optical band gap energy of the film decreased from 2.53 eV at the ratio of $N_2CH_4$=4.

Sugar Content and Protein Fractionation in Human Pleural Fluid (늑막액의 당 및 단백분획상)

  • Previous studies concerning the usefulness of pleural fluid glucose levels in differentiating causes of pleural effusions have been conflicting. Gelenger and Wiggers (1949), Calnan et al(1951) and Barber et al(1957) concluded that the lower the level of pleural fluid glucose, the more likely was tuberculosis, and that tuberculosis was unlikely if the pleural fluid glucose level was more than 80 mg/100 ml. Light and Ball(1973), however, reported that in the great majority of tuberculous pleural fluids the glucose concentration was high rather than low, concluded that the pleural fluid glucose levels were not useful in the differential diagnosis of pleural effusion. In this study, pleural fluid glucose was determined in 46 pleural effusions from various causes to evaluate the usefulness in the differential diagnosis of pleural effusion. In addition, the protein concentration and the electrophoretic patterns of protein and amylases in pleural fluid was compared with that of serum. And the results were as follows. 1. The mean glucose concentration of pleural fluid was 80.8 mg/100 ml in 22 tuberculous origin, 92.5 mg/100 ml in 12 cancer patient and 70.4 mg/100 ml in 10 undiagnosed cases. In 2 cases of paragonimiasis the pleural fliud glucose levels were low (mean, 32.0 mg/100 ml). The percentage of pleural fluid protein to serum is about 75% in all disease groups and the protein level of tuberculous pleural fluid was significantly correlated with that of serum. 2. The disc eletrophoretic patterns of pleural fluid were almost similar with that of serum in all disease groups but the prealbumin fraction was not observed in pleural fluid. 3. With the isoelectric focusing, 4 to 7 isoamylase was observed in serum and the isoelectric point was ranged from pH 5.8 to 7.8 and isoelectic point of main fracticn is pH 7.2. The isoelectic focusing patterns of amylase of pleural fluid were identical to that of serum in all disease group. With the above results it is concluded that the pleural fluid is exudate of serum and that the glucose levels of pleural fluid are not useful in the differential diagnosis of pieural effusions.

Characterization and Anti-Gastric Ulcer Activity of Bamboo Salt (죽염의 특성 분석과 항위궤양효과)

  • Bamboo salt has been used as a traditional remedy for gastric ulcer and gastro-intestinal disorders. It is produced by baking the salt packed in bamboo cylinder nine times under the fire of pine tree. Three of commercially available bamboo salt products (bamboo salt A, B, and C) were characterized by qualitative and quantitative analyses using inductively coupled plasma (ICP) spectrometer, ion chromatograph (IC), X-ray diffractometer (XRD), and electron microscope (EM). Compared with crude salt, the contents of iron, silicon, potassium, and phosphate in the bamboo salt products were higher whereas the sulfate content was lower. Water-insoluble fraction of bamboo salts contained the following compounds; MgO, $SiO_2,\;Mg_2Si0_4,\;and\;CaMgSi0_4$. The study on the microscopic structures of the bamboo salts were shown to have smooth surface and fused shape compared with crude salt. Among the three bamboo salt products, product A was used to test a possible inhibitory effect on gastric acid secretion. Each test material (bamboo salt A, crude salt, and reagent-grade NaCl) was given orally to Sprague-Dawley rats at doses of 0.2, 1.0, and 2.0 g/kg for 28 days before pyrolus ligation. Twenty four hours after the last administration of the test materials, volume, pH, total acidity, and pepsin activity of gastric juice were measured by the Shay-ligation method. No significant differences were observed in the secretion of gastric acid between treated groups (bamboo salt-, crude salt- and reagent-grade NaCI-treated groups) and control group (distilled water-treated group). This result demonstrated that bamboo salt did not exert anti-ulcer activities in experimental animals used in the present study.

Hydrolysis of Agricultural Residues and Kraft Pulps by Xylanolytic Enzymes from Alkaliphilic Bacillus sp. Strain BK

  • An alkaliphilic bacterium, Bacillus sp. strain BK, was found to produce extracellular cellulase-free xylanolytic enzymes with xylan-binding activity. Since the pellet-bound xylanase is eluted with 2% TEA from the pellet of the culture, they contain a xylan-binding region that is stronger than the xylan-binding xylanase of the extracellular enzyme. The xylanases had a different molecular weight and xylan-binding ability. The enzyme activity of xylanase in the extracellular fraction was 6 times higher than in the pellet-bound enzyme. Among the enzymes, xylanase had the highest enzyme activity. When Bacillus sp. strain BK was grown in pH 10.5 alkaline medium containing xylan as the sole carbon source, the bacterium produced xylanase, arabinofuranosidase, acetyl esterase, and $\beta$-xylosidase with specific activities of 1.23, 0.11, 0.06, and 0.04 unit per mg of protein, respectively. However, there was no cellulase activity detected in the crude enzyme preparation. The hydrolysis of agricultural residues and kraft pulps by the xylanolytic enzymes was examined at 50$^{\circ}C$ and pH 7.0. The rate of xylan hydrolysis in com hull was higher than those of sugarcane bagasse, rice straw, com cop, rice husk, and rice bran. In contrast, the rate of xylan hydrolysis in sugarcane pulp was 2.01 and 3.52 times higher than those of eucalyptus and pine pulp, respectively. In conclusion, this enzyme can be used to hydrolyze xylan in agricultural residues and kraft pulps to breach and regenerate paper from recycled environmental resources.
