This study aimed to investigate the hepatoprotective effect of Dendropanax morbifera (D. morbifera) leaf hot-water extract on carbon tetrachloride (CCl4)-treated HepG2 cells. Treatment with D. morbifera leaf hot-water extract increased the cell viability of CCl4-treated HepG2 cells without inducing cytotoxicity. The levels of alanine transaminase (ALT) and aspartate transaminase (AST) released by CCl4-treated cells were 27.6 U/L and 52.4 U/L, respectively, and were significantly higher than those in untreated control cells (10.0 U/L and 15.2 U/L, respectively). Moreover, the level of γ-glutamyl transpeptidase (GGT) was 5.4 times higher, while that of glutathione was 44.0% lower in CCl4-treated cells than in control cells. However, treatment with D. morbifera leaf hot-water extract resulted in a dose-dependent decrease in the levels of ALT, AST, and GGT, and an increase in the level of glutathione. Moreover, the malondialdehyde (MDA) content in CCl4-treated HepG2 cells was effectively reduced after treatment with D. morbifera leaf hot-water extract. Additionally, overproduction of intracellular lipids induced by CCl4 treatment was effectively inhibited by D. morbifera leaf hot-water extract treatment. Furthermore, DCFDA staining showed that overproduction of reactive oxygen species (ROS) induced by CCl4 treatment was effectively reduced by treatment with D. morbifera leaf hot-water extract. Our results indicate that owing to its beneficial effects, D. morbifera leaf extract has considerable potential as a functional food material for liver protection.
Hepatoprotective effects of Monascus pilosus mycelial ethanol extract (MPME) were examined in high-fat diet induced-obese rats. The rats were randomly divided into 2 groups; normal control (NC) and a high-fat and high cholesterol diet group (HFC). The HFC diet group was fed a 5L79 diet supplemented with 15% lard and 1% cholesterol for 3 weeks for induction of obesity. And then, the rats were divided into 4 groups (n=5); the NC, a HFC diet obesity control group (HF), 0.5% MPME supplemented HFC diet group (MPM), and 2% conjugated linoleic acid (CLA) supplemented HFC diet group for 7 weeks. Whereas the daily weight gain of NC and HFC groups were 3.48 g and 4.48 g, respectively, those of MPM and CLA were 3.09 g and 4.38 g, respectively. Furthermore, activity of serum alanine and aspartic aminotransferase in HF was markedly higher than those of NC group, but, the activity in MPM and CLA was significantly lower than HF. Hepatic reduced glutathione content in MPM and CLA was higher than HF. On the contrary, hepatic lipid peroxide content in MPM and CLA was significantly lower than HF. In conclusion, although the precise mechanisms of the hepatoprotective effects of the MPME in this study are unknown, our study provides experimental evidence that MPME may prevent obesity and hepatic damage by high-fat and high cholesterol diet via inhibition of lipid absorption and induction of reactive oxygen spices scavenging enzyme such as superoxide dismutase.
Rosa rugosa has traditionally been used as a folk remedy for diabetes. The objective of this study was therefore to demonstrate the inhibition of endothelial dysfunction activities through antioxidants and the anti-glycation of Rosa rugosa roots. Dried roots of Rosa rugosa were boiled in methanol for three hours, evaporated and lyophilized with a freeze-dryer. The methanolic extract of Rosa rugosa roots (RRE) was tested for antioxidant activities by measuring total polyphenol (TP) content, flavonoid content, 1,1-diphenyl-2-picrylhydrazyl free radical-scavenging activity (DPPH) assay, and ferric-reducing antioxidant power (FRAP) assay. The total TP content, flavonoid content, FRAP value, and $DPPHSC_{50}$ are $345.2\;{\mu}g$ gallic acid equivalents/mg dry matter (DM), $128.1\;{\mu}g$ quercetin equivalents/mg DM, 2.2 mM $FeSO_4$/mg DM and $34.2\;{\mu}g$ DM/mL, respectively. Treatment of RRE significantly lowered fluorescent formation due to advanced glycation reaction. In addition, reactive oxygen species (ROS) scavenging assay, monocyte adherent assay and transendothelial electrical resistance (TEER) assay were performed to investigate the possibility that RRE improves endothelial dysfunction-induced diabetic complications. The adhesion of THP-1 to treated HUVEC with RRE ($100\;{\mu}g/mL$; 33% and $500\;{\mu}g/mL$; 75%) was significantly reduced compared to HUVEC stimulated by glyceraldehydes-AGEs (advanced glycation end product). The TEER value ($88\;{\Omega}{\cdot}cm^2$) of stimulated HUVEC by glyceraldehydes-AGEs was reduced compared to non-stimulation ($113\;{\Omega}{\cdot}cm^2$). However, normalization with RRE increased endothelial permeability in a dose-dependent manner ($100\;{\mu}g/mL$; $102\;{\Omega}{\cdot}cm^2$ and $500\;{\mu}g/mL$; $106\;{\Omega}{\cdot}cm^2$). Thus, these results suggest that Rosa rugosa roots could be a novel candidate for the prevention of diabetic complications through antioxidants and inhibition of advanced glycation end product formation.
The composting of layer manure is economical and efficiently process. In this study, the variation of composting characteristics in layer manure was investigated according to air supply capacity. The fermented compost was added in layer manure and mixed with sawdust inside composting reactors. The level of air supply capacity was varied in the range of $50{\sim}200\;{\ell}/m^3/min$. During composting the temperature variations of composting piles was different the temperatures of composting piles for T-1 ($50\;{\ell}/m^3/min$) and T-2 ($100\;{\ell}/m^3/min$) were reached at $40^{\circ}C$ and $50^{\circ}C$ within 2 days, respectively. For T-3 ($150\;{\ell}/m^3/min$) and T-4 ($200\;{\ell}/m^3/min$), their temperatures was $60^{\circ}C$ within same days and maintained during 8 days. Water contents decreased according to the air supply capacity; 8.9%, 15.4%, 18.0% and 18.6% for T-1, T-2, T-3 and T-4. The weight ratios of T-1, T-2, T-3 and T-4 were reduced to 12.8%, 15.6%, 18.1% and 17.9%, respectively. The decreasing volumetric ratios of T-1, T-2, T-3 and T-4 were 18.0%, 21.0%, 22.3% and 22.0%. The oxygen discharge concentrations during composting were 12 ppm for T-1, T-2 and 9 ppm for T-3 and T-4. After composting, fertilizer components such as total nitrogen (TN) and phosphorous pentoxide ($P_2O_5$) were examined at each air supply capacity. Nitrogen contents of the T-1, T-2, T-3 and T-4 were 0.75%, 0.74%, 0.72% and 0.64%. Also, The contents of $P_2O_5$ were 0.35%, 0.40%, 0.38% and 0.42% for T-1, T-2, T-3 and T-4.
In this research, hematite nanoparticles were synthesized by DC thermal plasma process to increase the overall surface area. The effect of binders on hematite electrodes was investigated by changing the type and composition of binders when preparing electrodes. Nitrogen gas was also added to the DC thermal plasma process in order to dope the hematite with N for enhancing photoelectrochemical properties of hematite nanoparticles. The efficiency of water splitting reaction was measured by linear sweep voltammetry (LSV) under solar simulator. In LSV measurements, the onset potential and maximum current density at a fixed voltage were measured. The durability of electrodes was checked by repeating LSV measurements. CMC (carboxymethyl cellulose) binder with 50 : 1 composition exhibits the highest current density of $12mA/cm^2$ and CMC binder with 20 : 1 composition, showing the initial current density of $3mA/cm^2$, endures 20 times of repetitive LSV measurements. Effects of nitrogen doping on hematite nanoparticles were proven to be insignificant.
This study was performed to test the production of combustion toxic gases by Pinus rigida specimens treated with various types of methylpiperazinomethyl-bis-phosphonic acid $M^{n+}$ ($PIPEABPM^{n+}$) and methylpiperazinomethyl-bis-phosphonic acid (PIPEABP). Three coats of 15 wt% $PIPEABPM^{n+}$ and PIPEABP solutions were applied to plates of Pinus rigida at room temperature. After drying the treated specimens, the production of combustion toxic gases was examined using a cone calorimeter (ISO 5660-1). The specimens treated with $PIPEABPM^{n+}$ showed lower carbon monoxide production ($CO_{2\;peak}$; 0.0136~0.0178% at 532~678 s) than the PIPEABP plates, except for the specimen treated with $PIPEABPFe^{3+}$. In addition, the peak carbon dioxide production ($CO_{2\;peak}$) was lower (0.03648~0.3648% at 373~433 s) than that of the PIPEABP-treated plate. Notably, oxygen production was much higher than 15%, which can be fatal to humans. Therefore, the resulting risk could be eliminated. The results indicate that the combustion toxicities were partially decreased due to treatment of the virgin plate with $PIPEABPM^{n+}$.
There is tendency of increasing number and decreasing age of patients who are indicated for Rastelli operation for their cyanotic congenita heart disease. So there is the need to find the creiterion which saves the patients from early postoperative hemodynamic disturbances. We reviewed the 26 patients who had been performed Rastelli operation at Seoul national University hOipital from January 1981 to June 1988. mean age of the patients was 7.8 $\pm$ 3.4 years (range 2.5-1.5 years), mean body surface area(BSA) 0.79 $\pm$ 0.25 $m^{2}$(range 0.49-1.51 $m^{2}$) and mean hematocrit 57.95 $\pm$ 12%(range 48-80%). We diveded these patients into survived group and died group before postoperative 72 hours, and analysed preoperative arterial oxygen saturation($SaO_{2}$), the ratio of diameter of fight pulmonary artery to ascending aorta(RA:/AA), the ratio of both right and left pulmonary artery diameter to descending thoracic aorta(RPA+LPA/DTA), pulmonary artery index(PA index), cardiopulmonary bypass time, aorta cross-clamping time, postoperative perfusion state and total amount of dopamine infused postoperatively. The results showed that RPA+LPA/DTA and PA index were statistically significatn factors to influence early postoperative cardiac death rate (P<0.05). Especially there were good linear correlations between PA index(X) and perpheral perfusion index(Y)(Y = -1.15 + 0.02 X, r = 0.86, P<0.01) and between PA index(X) and total amount of dopamine infused before postoperative 72 hours(mg/kg, Y)(Y = 61.94 - 0.15 X, r = - 0.80, P<0.01). Also there were tendencies that the higher RPA + LPA/DTA(Y), the betterperipheral perfusion (X) and the lower need of dopamine(X), but no statistical significance. (Y = 0.78 + 1.60 X, r = 0.49, P>0.05) And the discrimant analysis showed that patients with PA index over 221 $mm^{2}$/BSA could undergo correction with 25 per cent of error rate. In conclusion, early postoperative hemodynamic states could be predicted by preoperatively measured PA index, and which can be used as a criterion for Rastelli operation performed on cyanotic congenital heart disease.
Anaerobic digestion(AD) has successfully been used for many applications that have conclusively demonstrated its ability to recycle biogenic wastes. AD has been successfully applied in industrial waste water treatment, stabilsation of sewage sludge, landfill management and recycling of biowaste and agricultural wastes as manure, energy crops. During AD, i.e. organic materials are decomposed by anaerobic forming bacteria and fina1ly converted to excellent fertilizer and biogas which is primarily composed of methane(CH4) and carbon dioxide(CO2) with smaller amounts of hydrogen sulfide(H2S) and ammonia(NH3), trace gases such as hydrogen(H2), nitrogen(N2), carbon monoxide(CO), oxygen(O2) and contain dust particles and siloxanes. The production and utilisation of biogas has several environmental advantages such as i)a renewable energy source, ii)reduction the release of methane to the atomsphere, iii)use as a substitute for fossil fuels. In utilisation of biogas, most of biogas produced from small scale plant e.g. farm-scale AD plant are used to provide as energy source for cooking and lighting, in most of the industrialised countries for energy recovery, environmental and safety reasons are used in combined heat and power(CHP) engines or as a supplement to natural. In particular, biogas to use as vehicle fuel or for grid injection there different biogas treatment steps are necessary, it is important to have a high energy content in biogas with biogas purification and upgrading. The energy content of biogas is in direct proportion to the methane content and by removing trace gases and carbon dioxide in the purification and upgrading process the energy content of biogas in increased. The process of purification and upgrading biogas generates new possibilities for its use since it can then replace natural gas, which is used extensively in many countries, However, those technologies add to the costs of biogas production. It is important to have an optimized purification and upgrading process in terms of low energy consumption and high efficiency giving high methane content in the upgraded gas. A number of technologies for purification and upgrading of biogas have been developed to use as a vehicle fuel or grid injection during the passed twenty years, and several technologies exist today and they are continually being improved. The biomethane which is produced from the purification and the upgrading process of biogas has gained increased attention due to rising oil and natural gas prices and increasing targets for renewable fuel quotes in many countries. New plants are continually being built and the number of biomethane plants was around 100 in 2009.

In this study, a technique of speciation and determination of the trace inorganic arsenic(As(III) and As(V)) in water sample using HPLC-DRC-ICP-MS has been developed. Isocratic mobile phase of 10 mM ammonium nitrate and 10 mM ammonium phosphate monobasic was used and methanol(5 v/v%) was used as flushing solvent. Selection of the best flow rate of reaction gas, O$_2$, and optimization of the parameters such as pH and flow rate of mobile phase, and injection volume of sample for the separation and detection of arsenic species were carried out. The oxygen flow rate of 0.5 mL/min, pH of 9.4 and flow rate of 1.5 mL/min of mobile phase, and injection volume of sample of 100 $\mu$L were found to be the best parameters for the speciation and determination of arsenic species. The analytical features of the method were detection limit 0.10 and 0.08 $\mu$g/L, precision(RSD) 4.3% and 3.6%, and recovery 95.2% and 96.4% for As(III) and As(V), respectively. Analysis time was 4 minutes per sample. Linear calibration graphs with r$^2$ = 0.998 were obtained for both As(III) and As(V). Speciation analysis of arsenic species in the raw water samples collected from the tributary streams to Han River and main stream of Paldnag were performed by the proposed method. The concentrations of As(III) ranged from 0.10 to 0.22 $\mu$g/L and As(V) concentrations ranged from 0.44 to 1.19 $\mu$g/L, and 93.5% of total arsenic was found to be As(V).
Cells can resist and even recover from stress induced by acute hypoxia, whereas chronic hypoxia often leads to irreversible damage and eventually death. Although little is known about the response(s) to acute hypoxia in neuronal cells, alterations in ion channel activity could be preferential. This study aimed to elucidate which channel type is involved in the response to acute hypoxia in rat pheochromocytomal (PC12) cells as a neuronal cell model. Using perfusing solution saturated with 95% $N_2$ and 5% $CO_2$, induction of cell hypoxia was confirmed based on increased intracellular $Ca^{2+}$ with diminished oxygen content in the perfusate. During acute hypoxia, one channel type with a conductance of about 30 pS (2.5 pA at -80 mV) was activated within the first 2~3 min following onset of hypoxia and was long-lived for more than 300 ms with high open probability ($P_o$, up to 0.8). This channel was permeable to $Na^+$ ions, but not to $K^+$, $Ca^+$, and $Cl^-$ ions, and was sensitively blocked by amiloride (200 nM). These characteristics and behaviors were quite similar to those of epithelial sodium channel (ENaC). RT-PCR and Western blot analyses confirmed that ENaC channel was endogenously expressed in PC12 cells. Taken together, a 30-pS ENaC-like channel was activated in response to acute hypoxia in PC12 cells. This is the first evidence of an acute hypoxia-activated $Na^+$ channel that can contribute to depolarization of the cell.
