Methyl linoleate was oxidized at 60, 90, 120 and $150^{\circ}C$, respectively, with sparging oxygen for different periods of time. On the basis of the peroxide values determined at four temperatures, four heating times were chosen for the analysis of physicochemical parameters, such as peroxide value, total oxidation products, polymer content, viscosity, refractive index and characteristics of thermal degradation by DSC (Differential Scanning Calorimeter). The content of peroxide linkage (C-O-O-C) polymer and ether or carbon to carbon linkage (C-O-C/C-C) polymer were analyzed by High Performance Size Exclusion Chromatography (HPSEC). The polymer formed at four temperatures was qualitatively identified as dimer. The polymer with peroxide linkage (C-O-O-C) were detected from methyl linoleate oxidized at $60^{\circ}C\;and\;90^{\circ}C$, but they were not detected from methyl linoleate oxidized at $120^{\circ}C\;and\;150^{\circ}C$. The enthalpy changes increased as peroxide value increased whereas maximum degradation temperature decreased. The highest correlation coefficients were obtained between maximum degradation temperature $(T_m)$, exothermic enthalpy changes and peroxide value, peroxide linkage (C-O-O-C) polymer content.
A bacterium which grows on cyclohexanol as sole carbon and energy source was isolated from sludge of industrial areas in Taegu and identified as Acinetobacter calcoaceticus C-15. The growth medium for the optimal culture condition was composed of 0.2% cyclohexanol, 0.11% $NH_4Cl$, 0.05% $KH_2PO_4$, 0.2% $K_2HPO_4$, 0.02% $MgSO_4{\cdot}7H_2O$, and 0.05% yeast extracts. The optimal pH value and temperature for the growth were 7.2 and $33^{\circ}C$, respectively. Specific growth rate of A. calcoaceticus C-15 at $33^{\circ}C$ on the cyclohexanol and cyclohexanone was $0.27hr^{-1}$ and $0.15hr^{-1}$, respectively. Growth yield for cyclohexanol was 1.0. The bacteria utilized ethanol, 1-butanol, 1-pentanol, and cyclohexanol as a carbon source but not methanol, 1-hexanol, m-cresol, glycerol, and cyclohexane. The bacteria grew on benzoate, adipate, acetate, and citrate, but did not on salicylate, phthalate, p-hydroxybenzoate, and gluconate. A calcoaceticus C-15 did not utilize all kind of sugars other than xylose. Cell-free extracts contained $NAD^+$-linked cyclohexanol dehydrogenase which catalized the oxidation of cyclohexanol to cyclohexanone.
Recombinant plasmid shuttle vectors were constructed for genetic studies on the oxidation of carbon monoxide by carboxydobacteria. Two vectors. pYK322 (7.2 kb, Ap'. Tc') and pYK 324 (7.2 kb, Ap', Tc'), were constructed using pBR322 and pYK100. a small plasmid in Pseudomonas carbo,xydovorans. Four plasmids. pYK2IO (5.2 kb. Cm'), pYK220 (5.2 kb, Cmr), pYK230 (5.2 kb, Cm'), and pYK232 (5.2 kb. Cm'), were constructed using pACYC184 and pYK100. Transformation of several carboxydobacteria with pYK322 and pYK220 was round to be efficient when the cells were transformed by the methoti of Bagdasarian and Timmis (Curr. Top. Microbiol. Immunol. 96:47-67. 1982) with several modifications; cells growing on 0.2% succinate were harvested at the mid-exponential phase. 10 mM RbCl in transformation solution was substituted with 100 mM KCI. cclls in transformation solution were incubated for 12 h at 4'C before addition of DNA and heat shock was carried out for 3 min at 45$^{\circ}$C. Plasmid vectors used for transformation, however. were not detected from antibiotics-resistant transformants, suggesting that the vectors may be integrated into the chromosomal DNA.
This study aimed to obtain high Fischer's ratio oligopeptides from goat whey (HFO) and investigate antioxidant property of it. Hydrolysis of goat whey was done with the approach of sequential digestion of pepsin and flavourzyme. With the adsorption of aromatic amino acids by activated carbon, HFO with a Fischer's ratio of 27.070 and a molecular weight of 200-1,000 Da were obtained, and the branched-chain amino acids accounted for 22.87%. Then the antioxidant activity of HFO was evaluated. At the concentrations of 2.0 mg/mL and 0.50 mg/mL, HFO scavenged 77.27% and 99.63% of 1,1-diphenyl-2-picrylhydrazyl and 3-ethylbenzthiazoline-6-sulphonate free radicals respectively. The scavenging rate of HFO against hydroxyl radicals reached 92.31% at the concentration of 0.25 mg/mL. Animal experiments demonstrated that HFO could moderate the changes of malondialdehyde, superoxide dismutase and glutathione peroxidase caused by CCl4-induced oxidative stress in vivo. This study indicated that HFO from goat whey was capable of oxidation resistance both in vivo and in vitro, which provided a scientific basis for the high-value processing and application of goat milk whey.
The detection of antibiotics in treated wastewater is a global concern as it enters water bodies and causes the development of antibiotic resistance genes in humans and marine life. The study specifically aims to explore the potential of ferrate (VI) in eliminating tetracycline (TCL). The degradation of TCL is optimized with parametric studies, viz., the effect of pH and concentration, which provide insights into TCL elimination. The increase in pH (from 7.0 to 10.0) favors the percentage removal of TCL; however, the increase in TCL concentrations from 0.02 to 0.3 mmol/L caused a decrease in percentage TCL removal from 97.4 to 29.1%, respectively, at pH 10.0. The time-dependent elimination of TCL using ferrate (VI) followed pseudosecond-order rate kinetics, and an apparent rate constant (kapp) was found at 1978.8 L2 /mol2 /min. Coexisting ions, i.e., NaNO3, Na2HPO4, NaCl, and oxalic acid, negligibly affect the oxidation of TCL by ferrate (VI). However, EDTA and glycine significantly inhibited the elimination of TCL using ferrate (VI). The mineralization of TCL using ferrate (VI) was favored at higher pH, and it increased from 18.57 to 32.52% when the solution pH increased from pH 7.0 to 10.0. Additionally, the real water samples containing a relatively high level of inorganic carbon spiked with TCL revealed that ferrate (VI) performance in the removal of TCL was unaffected, which further inferred the potential of ferrate (VI) in real implications.

Genomic analysis indicated that the genome of Drosophila melanogaster contains more than 80 cytochrome P450 genes. To date, the enzymatic activity of these P450s has not been extensively studied. Here, the biochemical properties of CYP6A8 were characterized. CYP6A8 was cloned into the pCW vector, and its recombinant enzyme was expressed in Escherichia coli and purified using Ni2+-nitrilotriacetate affinity chromatography. Its expression level was approximately 130 nmol per liter of culture. Purified CYP6A8 exhibited a low-spin state in the absolute spectra of the ferric forms. Binding titration analysis indicated that lauric acid and capric acid produced type I spectral changes, with Kd values 28 ± 4 and 144 ± 20 µM, respectively. Ultra-performance liquid chromatography-mass spectrometry analysis showed that the oxidation reaction of lauric acid produced (ω-1)-hydroxylated lauric acid as a major product and ω-hydroxy-lauric acid as a minor product. Steady-state kinetic analysis of lauric acid hydroxylation yielded a kcat value of 0.038 ± 0.002 min-1 and a Km value of 10 ± 2 µM. In addition, capric acid hydroxylation of CYP6A8 yielded kinetic parameters with a kcat value of 0.135 ± 0.007 min-1 and a Km value of 21 ± 4 µM. Because of the importance of various lipids as carbon sources, the metabolic analysis of fatty acids using CYP6A8 in this study can provide an understanding of the biochemical roles of P450 enzymes in many insects, including Drosophila melanogaster.
Remediation of wastewater contaminated with metal(II)-complexed species (Cu(II)-NTA (NTA: nitrilotriacetic acid), Cu(II)-EDTA (EDTA: ethylenediamine tetraacetic acid) and Cd(II)-EDTA is attempted using the potential applicability of ferrate(VI). Kinetics of pollutant degradation is obtained with the removal of ferrate(VI) studied at wide range of pH (8.0-10.0) and the concentration of metal(II)-complexed species (0.3 to 15.0 mmol/L) employing a constant dose of ferrate(VI) i.e., 1.0 mmol/L. Pseudo-first-order and pseudo-second-order rate constants were obtained in the reduction of ferrate(VI) which was then employed to obtain the overall rate constants of the pollutant degradation. The mineralization of NTA and EDTA was obtained with the change in TOC (total organic carbon) values collected by the ferrate(VI) treated pollutant samples. Decrease in pH and molar pollutant concentrations was greatly favored the percent mineralization of NTA or EDTA by the ferrate(VI) treatment. The treated pollutant samples were filtered and subjected for AAS (atomic absorption spectrophotometric) analysis to assess the simultaneous removal of copper and cadmium from aqueous solutions at the studied pH as well at the elevated pH 12.0. Results show that an enhanced removal of cadmium or copper was achieved at pH 12.0. Overall, ferrate(VI) possesses multifunctional application in wastewater treatment as it oxidizes the degradable impurities and removes metallic impurities by coagulation process.
The compounds associated with corrosion, in metallic archaeological samples of carbon steel of insular origin were evaluated to establish their degree of deterioration and structural damage against air pollution. The iron phases present in samples of archaeological artifacts were detected by Raman spectroscopy and confocal Raman microcopy. These samples mainly exhibited ing mainly ${\beta}$-FeO(OH) type goethite oxyhydroxides and small amounts of akaganeite ${\alpha}$-FeO(OH) lepidocrocite ${\gamma}$-FeO(OH) due to dominant chloride in a marine environment and non-stoichiometric oxyhydroxides Fe (II + / III +) as indicators of early corrosion. Some parts showed the presence of magnetic maghemite indicating high corrosion. ${\gamma}$-FeO(OH) is a precursor of phases associated with advanced marine corrosion. By studying its decomposition by Raman spectroscopy, it was synthesized with the following sequence: ${\gamma}-FeO(OH){\rightarrow}{\alpha}-FeO(OH)+{\gamma}-FeO(OH)$, ${\rightarrow}{\gamma}-Fe_2O_3+Fe_3O_4$. Ferric compounds provided evidence for the effect of intensity of laser on them, constituting a very useful input for the characterization of oxidation of iron in this type of artifacts. Thus, destructive analysis techniques should be avoided in addition to the use of small amounts of specimen.
Soil column experiments were evaluated effects of silver nanoparticles (i.e., 0, 2.5, 5, and 10 mg/L) on the microbial viability which is strongly associated with the degradation of organic matter, pharmaceutically active compounds(PhACs) and biological oxidation of nitrogenous compounds during river bank filtration. The addition of silver nanoparticles resulted in almost no change in the aqueous matrix. However, the intact cell concentration decreased with addition of silver nanoparticles from 2.5 to 10 mg/L, which accounted for 76% to 82% reduction compared to that of control (silver nanoparticles free surface water). The decrease in adenosine triphosphate was more pronounced; thus, the number and active cells in aqueous phase were concurrently decreased with added silver nanoparticles. Based on the florescence excitation-emission matrix and liquid chromatograph - organic carbon detection analyses, it shows that the removal of protein-like substances was relatively higher than that of humic-like substances, and polysaccharide was substantially reduced. But the extent of those substances removed during soil passage was decreased with the increasing concentration of silver nanoparticles. The attenuation of ionic PhACs ranged from 55% to 80%, depending on the concentration of silver nanoparticles. The attenuation of neutral PhACs ranged between 72% and 77%, which was relatively lower than that observed for the ionic PhACs. The microbial viability was affected by silver nanoparticles, which also resulted in inhibition of nitrifiers.
A sample of soil humic acid (HA) was divided by ultrafiltration (UF) into five fractions of different molecular size (UF1: > 300, UF2: 100~300, UF3: 30~100, UF4: 10~30, UF5: 1~10 kilodaltons). Apparent average molecular weight (Mw) of the HA fractions were measured using high performance size exclusion chromatography (HPSEC), and the chemical and structural properties of the five HA fractions were characterized by elemental compositions (H/C, O/C and w ((2O + 3NH)/ C)) and ultraviolet-visible absorption ratios (SUVA, A4/6). The organic carbon normalized-sorption coefficients (Koc) for the binding of phenanthrene to the HA fractions were determined by fluorescence quenching and relationship between the sorption coefficients and structural characteristics of the HA fractions were investigated. The elemental analysis and UV-vis spectral data indicated that the HA fractions with higher molecular weights have grater aliphatic character and lower contents of oxygen, while the HA fractions with lower molecular size have greater aromatic character and molecular polarity that correspond to greater SUVA and internal oxidation values (w). The log Koc values (L/kg C) were gradual increased from 4.45 for UF5 to 4.87 for UF1. The correlation study between the structural descriptors of the HA fractions and log Koc values of phenanthrene show that the magnitude of Koc values positively correlated with $M_w$ and H/C, while negatively correlated with the independent descriptors of the O/C, w, SUVA and A4/6.
