Trait Variation and Molecular Characterization of Ds insertional rice lines (벼 Ds 삽입변이 계통의 특성변이 및 분자생물학적 특성)

  • The 1,874 rice lines were selected from 3,000 Ds insertional mutant pool by Basta herbicide treatment and were surveyed for trait variation and molecular characteristics of genes knocked out by Ds insertion. Compared with "Donjin", an original japonica cultivar used for transformation, Ds insertion mutant pool showed large variation in major agronomic traits including tiller, panicle, and heading etc. Southern blot analysis demonstrated that these lines on the average had two Ds copies in Donjin genome, resulting in 38.4% of one copy, 32.5% of two copies, 16.7% of three copies, and 11.3% of over four copies. GUS analysis showed that 3.9% of lines (73/1,860) had tissue-specific expression in leaves, nodal parts, floral organs such as stigma and pollen, and roots. Data set obtained from agricultural trait variation and molecular characteristics for individual Ds insertional lines would provide researchers with more information for understanding the function of unknown rice genes controlling economically important traits.

HSP90 inhibitor, AUY922, debilitates intrinsic and acquired lapatinib-resistant HER2-positive gastric cancer cells

  • Human epidermal growth factor receptor 2 (HER2) inhibitors, such as trastuzumab and lapatinib are used to treat HER2-positive breast and gastric cancers. However, as with other targeted therapies, intrinsic or acquired resistance to HER2 inhibitors presents unresolved therapeutic problems for HER2-positive gastric cancer. The present study describes investigations with AUY922, a heat shock protein 90 (HSP90) inhibitor, in primary lapatinib-resistant (ESO26 and OE33) and lapatinib-sensitive gastric cancer cells (OE19, N87, and SNU-216) harboring HER2 amplification/over-expression. In order to investigate whether AUY922 could overcome intrinsic and acquired resistance to HER2 inhibitors in HER2-positive gastric cancer, we generated lapatinib-resistant gastric cancer cell lines (OE19/LR and N87/LR) by continuous exposure to lapatinib in vitro. We found that activation of HER2 and protein kinase B (AKT) were key factors in inducing intrinsic and acquired lapatinib-resistant gastric cancer cell lines, and that AUY922 effectively suppressed activation of both HER2 and AKT in acquired lapatinib-resistant gastric cancer cell lines. In conclusion, AUY922 showed a synergistic anti-cancer effect with lapatinib and sensitized gastric cancer cells with intrinsic resistance to lapatinib. Dual inhibition of the HSP90 and HER2 signaling pathways could represent a potent therapeutic strategy to treat HER2-positive gastric cancer with intrinsic and acquired resistance to lapatinib.

  • For the removal of carbon dioxide from the chemical process, a chemical absorption method is typically used industrially. Development of new processes for the removal of carbon dioxide by the chemical absorption method has been developing new absorbents by using various absorbents. Thermodynamic data of the sorbent mixture in the new process design using hybrid absorbent is essential to reduce the equipment cost and operating costs of the process. In this study, densities of water+diisopropanolamine (DIPA), DIPA+MDEA(Methyldiethanolamine) binary systems and Water+DIPA+MDEA ternary system were measured over the full range of composition at temperatures from 303.15 K to 333.15 K by using an Anton Paar digital vibrating tube densimeter (DMA4500). The experimental excess volumes have been obtained from the experimental density results and have been fitted using the Redlich-Kister-Muggianu expression. The parameters obtained from the binary excess volume data were used for the correlation of ternary system with one additional ternary parameter for each isotherm. All investigated binary and ternary systems are completely miscible, because the values of excess volume are negative under the examined conditions.

The effects of antioxidant and anti-aging treatment of UVB-irradiated human HaCaT keratinocytes with ethanol extracts of colored rice varieties (유색미 에탄올 추출물의 산화방지 효과와 UVB에 유도된 인간유래 피부각질형성세포에서의 항노화 효과)

  • In this study, we investigated the antioxidant activities and anti-aging efficacy in terms of suppression of matrix metalloproteinases (MMPs) in UVB-irradiated HaCaT cells by adding the ethanol extracts of Josaengheogchal (JE) and Shintoheug rice (SRE). In the electron-donating ability and ABTS radical-scavenging assays, we observed that both JE and SRE had scavenging activities and in a collagenase inhibition assay, both extracts showed inhibition effects of over 73% at $1,000{\mu}g/mL$ concentration. The expression of MMP-1 and -3, when the extracts were treated with UVB $50mJ/cm^2$, irradiated human HaCaT keratinocytes, was analyzed by western blotting and reverse-transcription polymerase chain reaction (RT-PCR). The results showed that MMP-1 and -3 proteins and mRNAs were downregulated in a concentration-dependent manner in response to both extracts. Therefore, we expect that these compounds have a potential for the use as functional ingredients with anti-aging effects in the cosmetic and food industries.

The Antioxidant and Anti-aging Effects of Treatment with Schisandra chinensis Seeds Fractions in UVB-irradiated Human HaCaT Cells (자외선 B에 유도된 사람유래 HaCaT cells에서의 오미자 종자 분획물의 항산화 및 항노화 효과)

  • In this study, we investigated the antioxidant and anti-aging activities of Schisandra chinensis seed fractions by adding them to UVB-irradiated HaCaT cells and analyzing the suppression of matrix metalloproteinases (MMPs). An electron-donating assay, ABTS radical scavenging assay, and hydrogen peroxide scavenging assay showed that the ethyl acetate fraction of S. chinensis seed (SCEAf) has scavenging activities. A collagenase inhibition activity assay showed that SCEAf had inhibitory effects of over 92.3% at $500{\mu}g/ml$ concentration. An MTT assay was performed to determine the cytotoxicity of SCEAf in HaCaT cells and the results showed that SCEAf increased cellular viability in a concentration-dependent manner. In addition, SCEAf was found to increase the viability of cells irradiatged by UVB $50mJ/cm^2$. To examine the anti-aging effects of SCEAf on HaCaT cells irradiated with UVB $50mJ/cm^2$, the expression of MMP-1 and -3 was analyzed by Western blotting and reverse-transcription polymerase chain reaction. The results showed that MMP-1 and -3 proteins and mRNA levels were downregulated in a concentration-dependent manner in response to SCEAf. These results suggest that SCEAf can prevent aging and alleviate aging symptoms by inhibiting collagenase activity in skin keratinocytes damaged by UVB. Therefore, S. chinensis seeds may have the potential for use as functional ingredients with anti-aging effects in the cosmetic and food industries.

  • Korea follows the slope design criteria during construction. It was enacted by the Ministry of Land, Transport and Maritime Affairs. There are cases where the Soil-nail is designed as a measure to secure slope stability. The arrangement of the soil-nail may be arranged at equal intervals or may be arranged differently depending on the soil failure model. The optimum design of the countermeasure method is determined by securing stability of the slope through optimization of dimensions and shape. However, when uniform nails are placed at low elevations in slopes, the standard safety factor is exceeded, which may hinder economic design. It is preferable to arrange the reinforcement of the nails over the entire slope. When the horizontal spacing of the nails was topology optimized according to the slope height, it was possible to minimize the amount of reinforcement while satisfying the standard safety factor. Since the active load is reduced in the section where the slope height is lowered, the safety factor after reinforcement may be excessively increased. Therefore, the phase optimization method is proposed as an economical optimal design method using the reinforcing shape density. In addition, a relational expression was designed to optimize the horizontal spacing by slope height.

Skin Anti-aging and Anti-wrinkle Effects of Pinus koreaiensis Seed Oil (해송자 오일의 피부 항노화 및 주름 개선 효과)

  • Objectives : The aim of this study was to investigated the anti-aging and anti-wrinkles effects of the pinus koreaiensis seed(PKS) oil. Methods : The anti-oxidant effect was performed by beta-carotene bleaching assay and the intracellular proteome was analyzed expression of each 15 proteins by 2-D electrophoresis. And fatty acid was analysed by gas chromatography. Anti-wrinkle effect was analyzing human skin by the PRIMOS system. Results : Fatty acid analysis of PKS oil has shown oleic acid was 49.7% and linoleic acid was 34.1%. And the antioxidant effect was about 125% compared with alpha-tocoperol(0.1%) by beta carotene bleaching assay. In 2D PAGE analysis, fifteen protein changes in five mechanisms which was collagen synthesis pathway, MMPs, ECM-cell interaction, cytokine, antioxidant enzymes were analyzed. In case of anti-wrinkle effect was proved in vivo by analyzing human skin by the PRIMOS system. The analysis results of eye wrinkles for 4 weeks showed an improvement effect of over 6%. Conclusions : In this study, the amount of protein change in the five mechanism through the cell experiment and the skin anti wrinkle efficacy by the human in vivo test were investigated. As a result pinus koreaiensis seed oil by supercritical extraction could be used as a anti-aging and anti-wrinkle substance for the skin.

Extending the Storage Periods of Zanthoxylum schinifolium Seed Oil using Sodium Bicarbonate and Ascorbic Acid (중탄산나트륨과 아스코르브산을 이용한 산초유의 저장기간 연장)

  • Morus alba, Anti-obesity, C57BL/6 Mice, Expression, Flavonoid, Gene, Mulberry Background: The seed oil of Zanthoxylum schinifolium S. et Z. (sancho) is a traditional cooking oil that has long been sold at a very high price however, depending on the method of extraction and storage, this oil becomes rancid occurs very quickly. Therefore, this study aimed to find a material that prevents rancidity and improves the storage properties of sancho oil. Methods and Results: Sancho oil was extracted using an extraction press, and acid values were compared with commercially available vegetable oils, sancho oil had a higher acid value than other vegetable oils. A very high acid value was observed in sancho oil stored for 6 months, regardless of temperature, requiring an effective storage method. The high acid value and the decrease in turbidity of sancho oil are dependent on the days of sedimentation. Treatment with sodium bicarbonate by concentration resulted in minimal changes in acid value over time. However, minor differences were detected among the treatment concentrations. Ascorbic acid was added to maximize the effect of sodium bicarbonate, and it was observed that ascorbic acid did not improve the antioxidant effect. The sodium bicarbonate and ascorbic acid mixture resulted in minimal change in acid value at temperature up to 25℃. Conclusions: Sancho oil becomes rancid very quicky and requires efficient storage techniques. Sodium bicarbonate and ascorbic acid have been proven to be useful as safe anti-racidity agents without causing harm to humans.

Anti-inflammatory Effect of Berchemia berchemiaefolia Leaves through Inhibition of NF-κB and MAPK Signaling Activation in LPS-stimulated RAW264.7 Cells (망개나무 추출물의 NF-κB 및 MAPK 신호전달 억제를 통한 항염증 효과)

  • Berchemia berchemiaefolia (Makino) Koidz. is found not only in korea, but also in japan and is known as a rare plant all over the world. In this study, we evaluated anti-inflammatory effect of B. berchemiaefolia (Makino) Koidz. leaves (BBK-L) in LPS-stimulated RAW264.7 cells. BBK-L inhibited the generation of NO through the suppression of iNOS experession. BBK-L attenuated the expression of iNOS, COX-2, TNF-α, IL-1β, IL-6 induced by LPS. BBK-L decreased LPS-mediated IκB-α degradation inhibite which resulted in the inhibition of NF-κB activation in RAW264.7 cells. In addition, BBK-L suppressed ERK1/2, JNK phosphorylation induced by LPS. BBK-L showed anti-inflammatory effect through blocking the generation of the inflammatory mediators such as NO, iNOS, COX-2, TNF-α, IL-1β, IL-6 via the inhibiting of NF-κB and MAPK signaling activation. These results suggests that BBK-L may have great potential for the development of anti-inflammatory drug to treat acute and chronic inflammatory disorders.

Molecular Mechanism of ABC Transporter Mdr49A Associated with a Positive Cross-Resistance in Transgenic Drosophila (형질전환 초파리를 이용한 Mdr49A 유전자의 살충제 교차저항성 기능 구명)

  • The ATP-binding cassette (ABC) transporter superfamily represents the largest transmembrane protein that transports a variety of substrates across extra- and intra-cellular membranes. In insects, the ABC transporter proteins play crucial roles in insecticide resistance. To date, no studies have investigated the involvement of ABC transporter gene for cross-resistance to insecticide chemistries. Here, we studied such possible mechanisms against six conventional insecticides using transgenic Drosophila melanogaster strains carrying Mdr49 transcript variant A. For the 91-R and 91-C strains of Drosophila melanogaster, although they have a common origin, 91-R has been intensely selected with DDT for over 60 years, while 91-C has received no insecticide selection. Our transgenic analyses showed that overexpression of 91-R-MDR49 transcript variant A along with three amino acid variations can yield a relatively low degree of cross-resistance to carbofuran (2.0~6.7-fold) and permethrin (2.5~10.5-fold) but did not show cross-resistance to abamectin, imidacloprid, methoxychlor, and prothiofos as compared to the Gal4-driver control strain without transgene expression. These results indicate that the overexpression of Mdr49A in itself leads to a cross-resistance and three amino acid changes have additional effects on positive cross-resistance to carbofuran and permethrin.
